Effects of intense pulsed light on fibroblast proliferation and possible roles of TGF-β1 and ERK signaling pathway

Abstract:Objective: To investigate the effects of intense pulsed light (IPL) on the proliferation of fibroblasts and the roles of ERK signaling pathway and TGF-β1. Methods: Fibroblasts isolated from human foreskin were cultured and treated with different doses of IPL irradiation. The cell growth was detected by MTT assay, p-ERK1/2 expressions were assessed by Western blot, and TGF-β1 in the supernatant was measured by ELISA. In the intervention study, the fibroblasts were treated either by PD98059 (a specific inhibitor of ERK) alone or by PD98059 plus IPL irradiation, to observe their effects on the proliferation of fibroblasts. Results: Compared with sham- irradiation, the cell growth was not affected by the IPL irradiation at the doses of 10, 18, 27 and 36 J/cm2 (P>0.05). When the dose of IPL irradiation was 72 J/cm2, the cells were markedly proliferated (P<0.05), the p-ERK1/2 expression level decreased and the TGF-β1 expression significantly enhanced (P<0.05). The cell proliferation and p-ERK1/2 expression were obviously inhibited by PD98059 (P<0.05). Conclusions: IPL irradiation at a dose of 72 J/cm2 can induce fibroblast proliferation, which may be due to the increased expression of TGF-β1 but not through the ERK signaling pathway.
