Abstract:Samples suffer from X-ray radiation damage during the protein crystal diffraction data collection process.Radiation damage disrupts covalent bonds in molecules,causing breakage and delocalization of disulphide bonds,even disrupting the lattice structure,and is one of the major reasons for errors occurring during the collection of accurate diffraction data in protein crystallography.Through analyzing the single-wavelength anomalous diffraction(SAD)data of lysozyme and Acutolysin-C protein,the impact of radiation damage accumulated on samples in the exposure process on the diffraction data and on the signal of anomalous diffraction was discussed.
