Abstract:Objective To investigate the diagnostic value of texture analysis derived from conventional MR imaging in differentiating benign and malignant breast lesions. Methods Thirty-six patients with malignant breast lesion and 33 patients with benign breast lesion were retrospectively analyzed in our study. All patients underwent conventional MR imaging including axial T1WI, T2WI, and contrast-enhanced T1WI before surgery. Texture features were calculated from manually drawn ROIs by using MaZda software. The feature selection methods included mutual information (MI), Fishers coefficient, classification error probability combined with average correlation coefficients (POE + ACC) and the combination of the above three methods(FPM). These methods were used to identify the most significant texture features in discriminating benign breast lesion from malignant breast lesion. The statistical methods including raw data analysis (RDA), principal component analysis (PCA), linear discriminant analysis (LDA) and nonlinear discriminant analysis (NDA) were used to distinguish malignant breast lesion from benign breast lesion. The results were shown by misclassification rate. Results In the three kinds of sequences, the texture features for differentiating malignant breast lesion and benign breast lesion were mainly from T2WI which had the lowest misclassification rate 4.35%(3/69). The misclassification rates of the feature selection methods were similar in MI, Fisher coefficient and POE+ACC (15.94%to 56.52%for MI;17.39%to 56.52%for Fisher coefficient and 17.39%to 56.52%for POE+ACC). However, the misclassification rate of the combination of the three methods (4.35%to 53.62%for FPM) was lower than that of any other kind of method. In the statistical methods, NDA (4.35% to 27.54%) had lower misclassification rate than RDA (33.33% to 56.52%), PCA (33.33% to 53.62%) and LDA (15.94% to 44.93%). Conclusion Texture analysis of conventional MR imaging can provide reliably objective basis for differentiating benign from malignant breast lesions.

Abstract:Rationale and Objectives. To investigate the feasibility using quantitative morphology/texture features of breast lesions for diagnostic prediction, and to explore the association of computerized features with lesion phenotype appearance on magnetic resonance imaging.Materials and Methods. Forty-three malignant/28 benign lesions were used in this study. A systematic approach from automated lesion segmentation, quantitative feature extraction, diagnostic feature selection using an artificial neural network (ANN), and lesion classification was carried out. Eight morphologic parameters and 10 gray level co-occurrence matrix texture features were obtained from each lesion. The diagnostic performance of selected features to differentiate between malignant and benign lesions was analyzed using receiver-operating characteristic analysis.Results. Six features were selected by an ANN using leave-one-out cross validation, including compactness, normalized radial length entropy, volume, gray level entropy, gray level sum average, and homogeneity. The area under the receiver-operating characteristic curve was 0.86. When dividing the database into half training and half validation set. The selected morphology feature ''compactness'' was associated with shape and margin in the Breast Imaging Reporting and Data System lexicon, round shape and smooth margin for the benign lesions, and more irregular shape for the malignant lesions. The selected texture features were associated with homogeneous/heterogeneous patterns and the enhancement intensity. The malignant lesions had higher intensity and broader distribution on the enhancement histogram (more heterogeneous) compared to the benign lesions.Conclusion. Quantitative analysis of morphology/texture features of breast lesions was feasible, and these features could be selected by an ANN to form a classifier for differential diagnosis. Establishing the link between computer-based features and visual descriptors defined in the BI-RADS lexicon will provide the foundation for the acceptance of quantitative diagnostic features in the development of computer-aided diagnosis.
[Diffusion-Weighted Imaging Texture Features in Differentiation of Malignant from Benign Nonpalpable Breast Lesions for Patients with Microcalcifications-Only in Mammography].

Abstract:OBJECTIVE:To evaluate the application of MR diffusion-weighted imaging(DWI) texture features in differentiation of malignant from benign nonpalpable breast lesion for patients with microcalcifications-only in mammography.METHODS:The clinical and MR-DWI data of 61 patients with microcalcifications, who underwent three-dimensional positioning of breast X-ray wire from October 2012 to December 2015 in Zhejiang Cancer Hospital, were retrospectively analyzed, including 38 patients with malignant lesions and 23 patients with benign lesions. Two radiologists independently drew the regions of interest (ROI) on DWI for image segmentation, and 6 histogram features and 16 grayscale symbiosis matrix (GLCM) texture features were extracted on each ROI. The random forest algorithm was applied to select the features and built the classification model. The leave-one-out cross-validation (LOOCV) was used to validate the classifier, and the performance of the classifier was evaluated by ROC curve.RESULTS:Six features were selected, including histogram features of mean, variance, skewness, entropy, as well as contrast (0°) and correlation (45°) in GLCM. The histogram features of mean, variance, skewness and entropy were significantly different between the benign and malignant breast lesions (all P<0.05). The AUC of the model was 0.76, and the diagnostic accuracy, sensitivity and specificity were 77.05%, 84.21% and 65.21%, respectively.CONCLUSIONS:The texture feature analysis of DWI can improve the diagnostic accuracy of differentiating benign and malignant breast nonpalpable lesions with microcalcifications-only in mammography. Histogram features of mean, variance, skewness, entropy of DWI may be used as important imaging markers.
The Utility of Texture Analysis Based on Breast Magnetic Resonance Imaging in Differentiating Phyllodes Tumors From Fibroadenomas

Abstract:Background: The preoperative diagnosis of phyllodes tumors (PTs) of the breast is critical to appropriate surgical treatment. However, reliable differentiation between PT and fibroadenoma (FA) remains difficult in daily clinical practice. The purpose of this study was to investigate the utility of breast MRI texture analysis for differentiating PTs from FAs. Materials and Methods: Forty-two PTs and 42 FAs were enrolled in this retrospective study. Clinical and conventional MRI features (CCMF) and MRI texture analysis were used to distinguish between PT and FA. Texture features were extracted from the axial short TI inversion recovery T2-weighted (T2W-STIR), T1-weighted pre-contrast, and two contrast-enhanced series (first contrast and third contrast). The Mann-Whitney U test was used to select statistically significant features of texture analysis and CCMF. Using a linear discriminant analysis, the most discriminative features were determined from statistically significant features. The K-nearest neighbor classifier and ROC curve were applied to evaluate the diagnostic performance. Results: With a higher classification accuracy (89.3%) and an AUC of 0.89, the texture features on T2W-STIR outperformed the texture features on other MRI sequences and CCMF. The AUC of the combination of CCMF with texture features on T2W-STIR was significantly higher than that of CCMF or texture features on T2W-STIR alone (p < 0.05). Based on the result of the classification accuracy (95.2%) and AUC (0.95), the diagnostic performance of the combination strategy performed better than texture features on T2W-STIR or CCMF separately. Conclusions: Texture features on T2W-STIR showed better diagnostic performance compared to CCMF for the distinction between PTs and FAs. After further validation of multi-institutional large datasets, MRI-based texture features may become a potential biomarker and be a useful medical decision tool in clinical trials having patients with breast fibroepithelial neoplasms.
Discrimination of Malignant and Benign Breast Masses Using Automatic Segmentation and Features Extracted from Dynamic Contrast-Enhanced and Diffusion-Weighted MRI

Abstract:Magnetic resonance imaging exhibits high sensitivity but low specificity for breast cancer. The present study aimed to investigate whether combining morphology, texture features and kinetic features with diffusion-weighted imaging using quantitative analysis improves the accuracy of discriminating malignant from benign breast masses. In total, 104 and 171 malignant lesions in 205 women were included. Additionally, 13 texture and 11 morphology features were computed from each lesion using a semi-automated segmentation method. To increase prediction accuracy, a newly designed classification model, difference-weighted local hyperplane, was used for statistical analysis of the combined effects of the features for predicting lesion type. The mean apparent diffusion coefficient (ADC) value for each lesion was calculated. Diagnostic performances of morphology and texture features, kinetic features and ADC alone and the combination of them were evaluated using receiver operating characteristics analysis. Malignant lesions had lower mean ADCs than benign lesions. By using 10-fold cross validation scheme, combined morphological and kinetic features achieved a diagnostic average accuracy of 0.87. Adding an ADC threshold of 1.37x10(-3) mm(2)/sec increased the overall averaged accuracy to 0.90. A multivariate model combining ADC values with 6 morphological and kinetic parameters best discriminated malignant from benign lesions. Incorporating morphology and texture features, kinetic features and ADC into a multivariable diagnostic model improves the discriminatory power of breast lesions.
Prediction of malignant breast lesions from MRI features: a comparison of artificial neural network and logistic regression techniques

Abstract:Rationale and objectives: Dynamic contrast-enhanced magnetic resonance imaging is a clinical imaging modality for the detection and diagnosis of breast lesions. Analytic methods were compared for diagnostic feature selection and the performance of lesion classification to differentiate between malignant and benign lesions in patients. Materials and methods: The study included 43 malignant and 28 benign histologically proved lesions. Eight morphologic parameters, 10 gray-level co-occurrence matrix texture features, and 14 Laws texture features were obtained using automated lesion segmentation and quantitative feature extraction. Artificial neural network (ANN) and logistic regression analysis were compared for the selection of the best predictors of malignant lesions among the normalized features. Results: Using ANN, the final four selected features were compactness, energy, homogeneity, and Law_LS, with an area under the receiver-operating characteristic curve (AUC) of 0.82 and accuracy of 0.76. The diagnostic performance of these four features computed on the basis of logistic regression yielded an AUC of 0.80 (95% confidence interval [CI], 0.688-0.905), similar to that of ANN. The analysis also showed that the odds of a malignant lesion decreased by 48% (95% CI, 25%-92%) for every increase of 1 standard deviation in the Law_LS feature, adjusted for differences in compactness, energy, and homogeneity. Using logistic regression with z-score transformation, a model composed of compactness, normalized radial length entropy, and gray-level sum average was selected, and it had the highest overall accuracy, 0.75, among all models, with an AUC of 0.77 (95% CI, 0.660-0.880). When logistic modeling of transformations using the Box-Cox method was performed, the most parsimonious model with predictors compactness and Law_LS had an AUC of 0.79 (95% CI, 0.672-0.898). Conclusion: The diagnostic performance of models selected by ANN and logistic regression was similar. The analytic methods were found to be roughly equivalent in terms of predictive ability when a small number of variables were chosen. The robust ANN methodology uses a sophisticated nonlinear model, while logistic regression analysis provides insightful information to enhance the interpretation of the model features.
Differentiation of Non-puerperal Mastitis from Breast Carcinoma with Non-mass-like Enhancement by Texture Analysis of Contrast-enhanced Magnetic Resonance Imaging

Abstract:Purpose To investigate the feasibility of texture analysis of breast contrastenhanced magnetic resonance imaging in differentiating non-puerperal mastitis and breast carcinoma with non-mass-like enhancement in order to prevent misdiagnosis of nonpuerperal mastitis.Materials and Methods In this retrospective study,the contrastenhanced MRI images of 42 female patients of invasive ductal carcinoma with non-masslike enhancement and 30 female patients of non-puerperal mastitis were analyzed.3234 texture features were generated from manually selected region of interest (ROI) of normal breast tissue and breast lesions.By means of genetic algorithm and linear discriminative analysis,10 texture features were selected based on their stability and accuracy in breast tissue classification.Results With these 10 features,the linear discriminative analysis classifiers had sensitivity of 92.9％ and specificity of 90.0％ in classifying two lesions,and accuracy of 89.6％ in classifying all three types of tissue.The result showed that texture analysis successfully differentiate non-puerperal mastitis and breast carcinoma with nonmass-like enhancement.Conclusion Texture analysis demonstrates the ability of differentiating invasive ductal carcinoma with non-mass-like enhancement,non-puerperal mastitis and normal breast tissue,and provides reliable results for clinical diagnosis.

Abstract:Purpose . The objective of this study is to investigate the use of texture analysis (TA) of magnetic resonance image (MRI) enhanced scan and machine learning methods for distinguishing different grades in breast invasive ductal carcinoma (IDC). Preoperative prediction of the grade of IDC can provide reference for different clinical treatments, so it has important practice values in clinic. Methods . Firstly, a breast cancer segmentation model based on discrete wavelet transform (DWT) and K -means algorithm is proposed. Secondly, TA was performed and the Gabor wavelet analysis is used to extract the texture feature of an MRI tumor. Then, according to the distance relationship between the features, key features are sorted and feature subsets are selected. Finally, the feature subset is classified by using a support vector machine and adjusted parameters to achieve the best classification effect. Results . By selecting key features for classification prediction, the classification accuracy of the classification model can reach 81.33%. 3-, 4-, and 5-fold cross-validation of the prediction accuracy of the support vector machine model is 77.79%~81.94%. Conclusion . The pathological grading of IDC can be predicted and evaluated by texture analysis and feature extraction of breast tumors. This method can provide much valuable information for doctors’ clinical diagnosis. With further development, the model demonstrates high potential for practical clinical use.

Assessment of Pathological Grading of Bladder Cancer Using Texture Features from MRI

Abstract:Objective: To explore the value of texture features from MR in differentiating the pathological grade of urothelial carcinoma of the bladder. Methods: A total of 108 lesions from patients with bladder cancer confirmed by postoperative pathology were analyzed retrospectively, including 60 cases of high-grade urothelial carcinoma (HGUG) and 48 cases of low-grade urothelial carcinoma (LGUC). All patients underwent routine MRI examination with the same scanning parameters. Lesions on T2-weighted image were delineated on ITK-Snap software by two radiologists to extract the corresponding volumes of interest(VOI). A.K. (Artificial Intelligence Kit) software was used for texture extraction, and the least absolute shrinkage and selection operator (LASSO) regression was used to select image features with a method of 10 fold cross-validation, and to reduce the dimensionality. The spearman method was used afterwards to condense the image features by removing redundant. Logistic multiple regression correlation analysis was carried out, and the diagnostic efficiency of the characteristic parameters was analyzed by using the area under receiver operating characteristic curve (AUC). Result: 7 optimal features between HGUC group and LGUC group were screened, includeing two histogram features (Quantile0.025, skewness) and five GLCM features (GLCMEntropy_angle45_offset7, GLCMEntropy_angle0_offset7, ShortRunEmphasis_angle45_offset4, LongRunEmphasis_angle0_offset7, LongRunEmphasis_angle45_offset7), the AUC of the training group was 0.955, and the AUC of the testing group was 0. 894, both of which at good diagnostic decision point. Conclusion: Textural features from T2-weighted image can reflect the difference between low- and high-grade bladder cancer effectively.
Value of Conventional MRI Texture Analysis in the Differential Diagnosis of Phyllodes Tumors and Fibroadenomas of the Breast

Abstract:Background: There is substantial overlap in MRI findings between phyllodes tumors (PTs) and fibroadenomas (FAs). Our study was performed to investigate the value of conventional MRI texture analysis in the differential diagnosis of PTs and FAs. Methods: Preoperative MRI data - including axial T1WI, T2WIFS (T2WI with fat suppression), dynamic contrast-enhanced (DCE)-T1WI2min and DCE-T1WI7min (T1WI post-strengthened for 2 and 7 min, respectively, on DCE-MRI) - of 45 patients with PTs and 67 patients with FAs were retrospectively analyzed. MaZda 4.7 software was used to manually draw the maximum ROIs at the same lesion level of the above MRI images. The optimized feature selection methods included Fisher's coefficient, probability of classification error and average correction coefficient (POE + ACC), and mutual information (MI) as well as a combination of the above 3 methods (F + POE + ACC + MI [FPM]), respectively. The misclassification rates of PTs and FAs were compared between texture analysis and subjective diagnosis by radiologists. Results: The DCE-T1WI7min images had the lowest misclassification rate of 10.71% (12/112). The misclassification rate for the radiologists' analysis (31.25%, 35/112) was higher than that of all the texture analysis, and there was a statistically significant difference between the radiologists' misclassification rates and those from the FPM method in terms of the T2WIFS and DCE-T1WI2min images (all p < 0.05), and for the DCE-T1WI7min images by using the Fisher and FPM methods (all p < 0.05). Conclusion: Texture analysis of conventional MRI can be used as an assistant tool in providing a certain objective basis for differentiating PTs from FAs.
Texture Feature Analysis for Classification of Early-Stage Prostate Cancer in mpMRI

Abstract:Magnetic resonance imaging (MRI) has become a crucial tool in the diagnosis and staging of prostate cancer, owing to its superior tissue contrast. However, it also creates large volumes of data that must be assessed by trained experts, a time-consuming and laborious task. This has prompted the development of machine learning tools for the automation of Prostate cancer (PCa) risk classification based on multiple MRI modalities (T2W, ADC, and high-b-value DWI). Understanding and interpreting the predictions made by the models, however, remains a challenge. We analyze Random Forests (RF) and Support Vector Machines (SVM), for two complementary datasets, the public Prostate-X dataset, and an in-house, mostly early-stage PCa dataset to elucidate the contributions made by first-order statistical features, Haralick texture features, and local binary patterns to the classification. Using correlation analysis and Shapley impact scores, we find that many of the features typically used are strongly correlated, and that the majority of features have negligible impact on the classification. We identify a small set of features that determine the classification outcome, which may aid the development of explainable AI approaches.

Machine Learning-Based MRI Texture Analysis to Predict the Histologic Grade of Oral Squamous Cell Carcinoma.

Abstract:OBJECTIVE. This study aimed to explore the performance of machine learning (ML)-based MRI texture analysis in discriminating between well-differentiated (WD) oral squamous cell carcinoma (OSCC) and moderately or poorly differentiated OSCC. MATERIALS AND METHODS. The study enrolled 80 patients with pathologically confirmed OSCC (18 WD OSCCs and 62 moderately or poorly differentiated OSCCs) who underwent pretreatment MRI. ROIs were manually delineated to cover the entire tumor to the greatest possible extent on T2-weighted imaging and contrast-enhanced T1-weighted imaging, and 1118 texture features were extracted. Dimension reduction was performed using reproducibility analysis by two radiologists, collinearity analysis, and feature selection with a minimum-redundancy maximum-relevance algorithm. Models were created using random forest (RF), artificial neural network, and logistic regression (LR) alone and with a synthetic minority oversampling technique (SMOTE). Classifier performance was assessed using 10-fold cross-validation. RESULTS. Dimension reduction steps yielded eight texture features, including four features from each sequence. None of the clinical variables was selected. Among the eight texture features, five and seven texture features showed significant differences between the two groups in the actual data and balanced data, respectively (p < 0.05). All classifiers with SMOTE achieved better performances than those alone. The RF classifier with SMOTE achieved the best performance with an area under the ROC curve of 0.936 and accuracy of 86.3%. CONCLUSION. ML-based MRI texture analysis provides a promising noninvasive approach for predicting the histologic grade of OSCC.
Diagnosis of breast masses from dynamic contrast-enhanced and diffusion-weighted MR: a machine learning approach.

Abstract:PURPOSE:Dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI) is increasingly used for breast cancer diagnosis as supplementary to conventional imaging techniques. Combining of diffusion-weighted imaging (DWI) of morphology and kinetic features from DCE-MRI to improve the discrimination power of malignant from benign breast masses is rarely reported. MATERIALS AND METHODS:The study comprised of 234 female patients with 85 benign and 149 malignant lesions. Four distinct groups of features, coupling with pathological tests, were estimated to comprehensively characterize the pictorial properties of each lesion, which was obtained by a semi-automated segmentation method. Classical machine learning scheme including feature subset selection and various classification schemes were employed to build prognostic model, which served as a foundation for evaluating the combined effects of the multi-sided features for predicting of the types of lesions. Various measurements including cross validation and receiver operating characteristics were used to quantify the diagnostic performances of each feature as well as their combination. RESULTS:Seven features were all found to be statistically different between the malignant and the benign groups and their combination has achieved the highest classification accuracy. The seven features include one pathological variable of age, one morphological variable of slope, three texture features of entropy, inverse difference and information correlation, one kinetic feature of SER and one DWI feature of apparent diffusion coefficient (ADC). Together with the selected diagnostic features, various classical classification schemes were used to test their discrimination power through cross validation scheme. The averaged measurements of sensitivity, specificity, AUC and accuracy are 0.85, 0.89, 90.9% and 0.93, respectively. CONCLUSION:Multi-sided variables which characterize the morphological, kinetic, pathological properties and DWI measurement of ADC can dramatically improve the discriminatory power of breast lesions.
Texture analysis based on PI-RADS 4/5-scored magnetic resonance images combined with machine learning to distinguish benign lesions from prostate cancer

Abstract:Background: The global morbidity and mortality of prostate cancer (PCa) increase sharply every year. Early diagnosis is essential; it determines survival and outcome. So, this study extracted the texture features of apparent diffusion coefficient images in multiparametric magnetic resonance imaging (mp-MRI) and built machine learning models based on radiomics texture analysis (TA) to determine its ability to distinguish benign from PCa lesions using the Prostate Imaging Reporting and Data System (PI-RADS) 4/5 score.Methods: We enrolled 103 patients who underwent mp-MRI examinations and transrectal ultrasound and magnetic resonance fusion imaging (TRUS-MRI) targeted prostate biopsy and obtained pathological confirmation at our hospital from August 2017 to January 2020. We used ImageJ software to obtain texture feature parameters based on apparent diffusion coefficient (ADC) images, then standardized texture feature parameters, and used LASSO regression to reduce multiple feature parameters; 70% of the cases were randomly selected from the PCa group and the benign prostate hyperplasia group as the training set. The remaining 30% was used as the test set. The machine learning classification model for identifying benign and malignant prostate lesions was constructed using the feature parameters after dimensionality reduction. The clinical indicators were statistically analyzed, and we constructed a machine learning classification model based on clinical indicators of benign and malignant prostate lesions. Finally, we compared the model's performance based on radiomics texture features and clinical indicators to identify benign and malignant prostate lesions in PI-RADS 4/5 score.Results: The area under the curve (AUC) of the R-logistic model test set was 0.838, higher than the R-SVM and R-AdaBoost classification models. At this time, the corresponding R-logistic classification model formula is as follow: Y_radiomics=9.396-7.464*median ADC-0.584*kurtosis+0.627*skewness+0.576*MRI lesions volume; analysis of clinical indicators shows that the corresponding C-logistic classification model formula is as follows: Y_clinical =-2.608+0.324*PSA-3.045*Fib+4.147*LDL-C, the AUC value of the model training set was 0.860, smaller than the training set R-logistic classification model AUC value of 0.936.Conclusions: Radiomics combined with the machine learning classifier model has strong classification performance in identifying benign and PCa in PI-RADS 4/5 score. Various treatments and outcomes for PCa patients can be applied clinically.

Abstract:Objectives To investigate the diagnostic value of MRI-based texture analysis in discriminating common posterior fossa tumors, including medulloblastoma, brain metastatic tumor, and hemangioblastoma. Methods A total number of 185 patients were enrolled in the current study: 63 of them were diagnosed with medulloblastoma, 56 were diagnosed with brain metastatic tumor, and 66 were diagnosed with hemangioblastoma. Texture features were extracted from contrast-enhanced T1-weighted (T1C) images and fluid-attenuation inversion recovery (FLAIR) images within two matrixes. Mann-Whitney U test was conducted to identify whether texture features were significantly different among subtypes of tumors. Logistic regression analysis was performed to assess if they could be taken as independent predictors and to establish the integrated models. Receiver operating characteristic analysis was conducted to evaluate their performances in discrimination. Results There were texture features from both T1C images and FLAIR images found to be significantly different among the three types of tumors. The integrated model represented that the promising diagnostic performance of texture analysis depended on a series of features rather than a single feature. Moreover, the predictive model that combined texture features and clinical feature implied feasible performance in prediction with an accuracy of 0.80. Conclusion MRI-based texture analysis could potentially be served as a radiological method in discrimination of common tumors located in posterior fossa.
Application of MRI Breast Imaging Reporting and Data System in Breast Lesions

Abstract:Objective To study the diagnostic value of MRI breast imaging reporting and data system(BI-RADS) for breast lesions.Methods One hundred and twenty-four subjects were enrolled in this study,including 72 breast cancer cases(73 lesions) and 52 benign cases(59 lesions).All breast lesions were verified by pathology.MRI was performed for every case.MRI sequences included T1WI,T2WI,STIR and dynamic contrast-enhanced MRI scanning(VIBRANT).The sensitivity,specificity and accuracy of MRI BI-RADS were evaluated by ROC analysis.Results The sensitivity,specificity and accuracy of MRI BI-RADS were 91.0%,96.2% and 93.2%,respectively.Conclusions MRI BI-RADS has high sensitivity,specificity and accuracy,and it is useful in the differential diagnosis of malignant and benign lesions,MRI BI-RADS provides an objective canonical classification,reducing subjectivity and is convenient for communication between physicians.

Abstract:ObjectivesPatient-tailored treatments for breast cancer are based on histological and immunohistochemical (IHC) subtypes. Magnetic Resonance Imaging (MRI) texture analysis (TA) may be useful in non-invasive lesion subtype classification.MethodsWomen with newly diagnosed primary breast cancer underwent pre-treatment dynamic contrast-enhanced breast MRI. TA was performed using co-occurrence matrix (COM) features, by creating a model on retrospective training data, then prospectively applying to a test set. Analyses were blinded to breast pathology. Subtype classifications were performed using a cross-validated k-nearest-neighbour (k = 3) technique, with accuracy relative to pathology assessed and receiver operator curve (AUROC) calculated. Mann-Whitney U and Kruskal-Wallis tests were used to assess raw entropy feature values.ResultsHistological subtype classifications were similar across training (n = 148 cancers) and test sets (n = 73 lesions) using all COM features (training: 75 %, AUROC = 0.816; test: 72.5 %, AUROC = 0.823). Entropy features were significantly different between lobular and ductal cancers (p < 0.001; Mann-Whitney U). IHC classifications using COM features were also similar for training and test data (training: 57.2 %, AUROC = 0.754; test: 57.0 %, AUROC = 0.750). Hormone receptor positive and negative cancers demonstrated significantly different entropy features. Entropy features alone were unable to create a robust classification model.ConclusionTextural differences on contrast-enhanced MR images may reflect underlying lesion subtypes, which merits testing against treatment response.Key Points• MR-derived entropy features, representing heterogeneity, provide important information on tissue composition.• Entropy features can differentiate between histological and immunohistochemical subtypes of breast cancer.• Differing entropy features between breast cancer subtypes implies differences in lesion heterogeneity.• Texture analysis of breast cancer potentially provides added information for decision making.

Abstract:Purpose To extract texture features from magnetic resonance imaging (MRI) scans of patients with brain tumors and use them to train a classification model for supporting an early diagnosis. Methods Two groups of regions (control and tumor) were selected from MRI scans of 40 patients with meningioma or glioma. These regions were analyzed to obtain texture features. Statistical analysis was conducted using SPSS (version 20.0), including the Shapiro–Wilk test and Wilcoxon signed-rank test, which were used to test significant differences in each feature between the tumor and healthy regions. T-distributed stochastic neighbor embedding (t-SNE) was used to visualize the data distribution so as to avoid tumor selection bias. The Gini impurity index in random forests (RFs) was used to select the top five out of all features. Based on the five features, three classification models were built respectively with three machine learning classifiers: RF, support vector machine (SVM), and back propagation (BP) neural network. Results Sixteen of the 25 features were significantly different between the tumor and healthy areas. Through the Gini impurity index in RFs, standard deviation, first-order moment, variance, third-order absolute moment, and third-order central moment were selected to build the classification model. The classification model trained using the SVM classifier achieved the best performance, with sensitivity, specificity, and area under the curve of 94.04%, 92.3%, and 0.932, respectively. Conclusion Texture analysis with an SVM classifier can help differentiate between brain tumor and healthy areas with high speed and accuracy, which would facilitate its clinical application.

Abstract:Objective To discuss the application value of texture analysis of conventional MRI in differential diagnosis of orbital lymphoma from inflammatory pseudotumor.Methods The conventional MRI data of 15 patients with lymphoma and 17 patients with inflammatory pseudotumor proven by pathology or clinical follow-up were retrospectively reviewed.The texture features of lesions based on axial T1WI,fat-saturated T2WI and contrast enhanced fat-saturated T1WI were extracted by manually drawn ROIs with software MaZda.The subsets of optimized texture parameters were chosen by four different methods:Fisher coefficient,probability of classification error and average correlation coefficient (POE+ ACC),mutual information measure (MI) and the combination of the above three methods (FPM),respectively.Linear discriminant analysis (LDA) and nonlinear discriminant analysis (NDA) were performed for texture classification.The texture features from the sequence with the best classification result of orbital lymphoma and inflammatory pseudotumor were compared.Results The optimal texture parameters were mainly derived from co occurrence matrix and run-length matrix on T1WI and T2WI.The optimal texture parameters were mainly derived from co-occurrence matrix and histogram on contrast enhanced T1WI.The best classification of MRI texture was obtained within T2WI with lowest classification error of 1.56％ achieved by FPM in combination with NDA.Comparing the texture parameters of orbital lymphoma and inflammatory pseudotumor on T2WI,the angular second moment and long length emphasis were significantly higher in orbital lymphoma (both P＜0.005),while the entropy and short length emphasis were significantly lower in orbital lymphoma (both P＜0.005).Conclusion It is feasible to use texture analysis on conventional MRI for the differentiation of orbital lymphoma and inflammatory pseudotumor.
