Abstract:An effective N-ligand for copper-catalyzed N-arylation of 1H-1,2,4-triazole has been developed. In the presence of Cu2O, Cs2CO3, N-ligand-B. 1H-1,2,4-triazole were coupled with toluence to afford the corresponding products in moderate condition and with excellent yields.

Abstract:A general and efficient copper-catalyzed benzylation reaction of triazolopyridine with N-tosylhydrazones was developed. This reaction forms a C(sp(2))-C(sp(3)) bond through cross-coupling, and represents an exceedingly practical method to afford 3-benzylated triazolopyridines in moderate to good yields. A proposed mechanistic pathway underlying this reaction was outlined. This catalytic transformation should enable broad synthetic applications in functionalization chemistry, allowing the synthesis of new pharmaceutically relevant triazolopyridine derivatives.

Abstract:Cu-II-catalysed diazo transfer and Cu-I-catalysed azide-alkyne 1,3-dipolar cycloaddition (CuAAC) "click chemistry" were used to synthesis C7N aminocyclitol-derivatised 1,2,3-triazoles. In the course of this work, the -N=N- moiety was transferred onto C7N aminocyclitols such as validamine, valienamine and valiolamine by employing imidazole-1-sulfonyl azide as the diazo transfer reagent with catalysis by Cu-II, Zn-II and Ni-II, in moderate to good yields. The obtained azidocyclitols were coupled with various terminal alkynes under modified Meldal's conditions with good to excellent yields. The stereo- and regiochemistry of the products were confirmed by 2D-NMR (NOESY and HMBC). One-pot syntheses of the corresponding 1,2,3-triazoles, as safer and more efficient procedures, were also investigated and gave moderate to good yields.

Abstract:A regioselective electrochemical C–H amination method to synthesize N2-substituted 1,2,3-triazole using easily accessible ethers has been developed. Various substituents, including heterocycles, have a good tolerance, and 24 examples were obtained in moderate to good yields. Control experiments and DFT calculation investigations demonstrate that the electrochemical synthesis undergoes a N-tosyl 1,2,3-triazole radical cation process promoted by the single-electron transfer of the lone pair electrons of the aromatic N-heterocycle, and the desulfonation is responsible for the high N2-regioselectivity.

Abstract:A copper-catalyzed formation of C-N/N-N bonds using N-phenylbenzamidines with aryl nitriles has been developed and affords a route to 1,3,5-trisubstituted 1H-1,2,4-triazoles in moderate to excellent yields. The method is operationally simple and environmentally benign with a large substrate scope available and represents an economical path for numerous C-N/N-N bond formations.

Abstract:Copper-catalyzed intermolecular annulation of 2-amine-[1,3,5]triazines and aryl nitriles for the synthesis of [1,2,4]triazolo[1,5-a][1,3,5]triazines via N-C bond formation and oxidative N-N coupling [oxidative 3 + 2 cyclization] is presented. A wide range of aryl nitriles, including electron-rich benzonitriles, electron-poor benzonitriles, 2-cyanothiophene, and 4-cyanopyridine, were all functionalized with 2-amine-[1,3,5]triazines. Furthermore, amidation of 2-amine-[1,3,5]triazines via Cu-catalyzed C-CN bond cleavage of phenylacetonitriles is also demonstrated. The reaction occurred in moderate to satisfactory yields and tolerated alkyl- or aryl-substituted 2-amine-[1,3,5]triazines. Aniline, aminopyridine, and aminopyrimidine also afforded the desired products.
Copper-Promoted One-Pot Sandmeyer-Type Reaction for the Synthesis of N-Aryltriazoles.

Abstract:Herein, we report the copper-catalyzed one-pot Sandmeyer-type reaction of aromatic amines with triazoles to afford N-aryl-1,2,3-triazoles. Diazonium salts, formed from aromatic amines and tert-butyl nitrite in the presence of fluoroboric acid, reacted with triazoles in a copper-catalyzed Sandmeyer-type reaction. The reaction proceeded under mild conditions to afford N-aryltriazoles in moderate to good yields. This method is amenable to a wide range of aromatic amines and triazoles and shows diverse functional group tolerance. Inhibition of the reaction upon the addition of free radical scavengers suggested a radical pathway, in which the aryl radical, copper, and triazole formed a complex that underwent reductive elimination to give aryltriazole compounds; this is consistent with the mechanism underlying the Sandmeyer reaction. Thus, we demonstrate a new effective strategy for the construction of C-N bonds via Sandmeyer-type reactions and a valuable alternative approach for the synthesis of aryltriazole derivatives.

Abstract:A catalytic and step economic protocol for the construction of N-2-aryl-naphthotriazoles via copper-catalyzed cyclization of aryl amines and aryldiazonium salts is reported. With dioxygen in the air as termial oxidants under copper catalysis, various N-2-aryl-naphthotriazoles were synthesized in high yields. Importantly, this protocol features the sufficient inhibition of the classic C-N-2 bond cleavage process of aryldiazonium tetrafluoroborates under copper catalysis and the undesired dehydrogenative coupling of aryl amines under oxidative conditions. Preliminary mechanistic studies indicated that a radical procedure might not be involved in this transformation. In addition, simple decoration of the obtained compounds delivers novel and attractive tetraphenylethylene moiety containing N-2-aryl-naphthotriazole derivatives.

Abstract:An efficient, regioselective C-5 arylation of thiazolo[3,2-b]-1,2,4-triazoles catalyzed by a simple copper catalyst was developed. This arylation proceeded smoothly and tolerated a variety of functional groups (44 examples). A wide range of functionalized thiazolo[3,2-b]-1,2,4-triazole derivatives were obtained in high yields (up to 99% yield). Possible catalytic cycles of the arylation were also discussed.

Abstract:Efficient post-triazole regioselective N-2 arylation was developed from C-4, C-5 disubstituted-1,2,3-NH-triazoles. Three different approaches had been investigated, including S(N)Ar, Cu(I) catalyzed aryl amidation and Cu(II) mediated boronic acid coupling. The N-2-aryl triazoles were successfully synthesized with excellent yields. The structures were characterized by X-ray crystallography and some N-2-triazole products gave strong fluorescence with various emission controlled by the C-5 groups.

Abstract:C -Amino-1,2,4-triazoles are challenging polynitrogen substrates for metal-catalyzed arylation due to their multidentate character, enhanced coordinating ability and decreased nucleophilicity of the amino group. In the present study, the Buchwald–Hartwig cross-coupling of diverse 3(5)-amino-1,2,4-triazoles with aryl chlorides and bromides delivering (hetero)arylamino-1,2,4-triazoles in good-to-excellent yields under Pd/NHC catalysis was developed. The use of Pd complexes with bulky NHC ligands such as IPr *OMe and TPEDO (1,1,2,2-tetraphenylethane-1,2-diol) as an in situ Pd( II ) to Pd(0) reductant enabled the selective arylation of the NH 2 group even in acidic NH unprotected substrates and deactivated 1-substituted 5-amino- and 4-substituted 3-amino-1,2,4-triazoles. The reaction mechanism and structure–activity relationships were studied with DFT calculations. A significant effect of the position of the N-substituent in the 1,2,4-triazole ring on the favorable reaction pathways was revealed.

Abstract:N-(1-Oxy-pyridin-2-ylmethyl) oxalamic acids (L3-L5) were identified as novel efficient ligands for copper-catalyzed C-N cross-coupling of azoles and aryl halides in water. The N-arylation of imidazoles, indoles and pyrazoles proceeded with moderate to excellent yields and complete selectivity over aromatic amines and phenols. Moreover, L5, which is also efficient in organic solvent with low catalyst loading, can be used to promote the N-arylation reactions with water-sensitive materials. The catalytic mechanism was proposed based on the results of several verification experiments which indicated that the ligands as new-type chelators may coordinate to Cu(I) with two oxygen atoms of N-oxide and amide in the coupling process.

Abstract:A new methodology for the Cu-catalyzed synthesis of 1H-1,2,3-triazoles from 1,1-dibromoalkenes and sodium azide is presented. Aryl dibromoolefins were efficiently converted into the corresponding 1,2,3-triazoles. A comprehensive number of functional groups were compatible with this reaction. 1,2,3-Triazoles were obtained in moderate to excellent yields.

Abstract:Relatively mild and highly efficient CuI-catalyzed N-arylation procedures for nitrogen-containing heterocycles (e.g., imidazoles, benzimidazoles, pyrroles, pyrazoles, indoles, triazoles, etc.) with aryl and heteroaryl halides have been developed. The protocols can be performed easily and tolerate a number of functional groups, such as ester, nitrile, nitro, ketone, free hydroxyl, and free primary amine on the aryl halide.
