Effect of support hydrophobicity on the lipase immobilization and the synthesis of feruloylated monoacyl- and diacylglycerols

Abstract:The influences of support hydrophobicity on the adsorption of Candida Antarctica lipase B (CALB) and its catalytic efficiency in the transesterification of ferulic acid ethyl ester with triolein were investigated. Four kinds of methyl-modified silica aerogels, whose contact angle with water were 42.40°, 61.03°, 116.6°, 131.0°, were chosen as the carrier. It was found that the support which was very hydrophilic or hydrophobic may achieve the maximum CALB loading, while the support whose hydrophobicity was in the middle adsorbed less lipase. The loading amount reached 400 mg/g. Raising temperature helped improve lipase loading, but it may also lead to denaturalization, and the effect of pH was not obvious. The immobilized CALB was used for the synthesis of feruloylated monoacyl- and diacylglycerols through the transesterification of ferulic acid ethyl ester with triolein. The results showed that the support hydrophobicity had effect on lipase selectivity. No FDO or FMO was synthesized by the hydrophilic support with lipase. This is probably because the conformation changes of lipase caused by the interactions of protein molecule and the functional group of the support.
