Abstract:Molecular imprinting technique is a borderline subject which combines the advantages of the subjects of high polymer chemistry, analytical chemistry and science of material. Molecular imprinting technique is a novel technique based on mimicking specification of antibody-antigen. The study of fluorescent molecular imprinting polymer sensors is a significant direction in the development of current sensors with a bright prospect. Fluorescent molecular imprinting polymer sensors combine the recognition ability and predetermined selectivity of molecular imprinting polymers with the high sensitivity of fluorescence analysis. This review summarizes the progress of fluorescent molecular imprinting polymer sensors in recent years. The mechanism of preparation, methods for monitoring binding sites and applications on organic small molecules and ions of fluorescent molecular imprinting polymer sensors are described in detail. Additionally, the development trend of fluorescent molecular imprinting polymer sensors is also discussed.
Polymerization-assisted signal amplification for electrochemical detection of biomarkers.

Abstract:We present a novel immunosensor by using polymerization-assisted signal amplification strategy coupled with electrochemical detection. A sandwich immunoassay process was used to immobilize a polymerization reaction center, the initiator-conjugated polyclonal prostate specific antigen (PSA) or polyclonal carcinoembryonic antigen (CEA) antibodies on the surface of the electrode. Activator generated electron transfer for atom transfer radical polymerization (AGET ATRP) subsequently triggered the local accumulation of glycidyl methacrylate (GMA) monomers. Growth of long chain polymers provided excess epoxy groups for electrochemical tags aminoferrocene (FcNH(2)) coupling, which in turn significantly increased the loading of the signal molecules and enhanced the electrochemical readouts. The detection limit was similar to 0.14 pg mL(-1) for PSA and similar to 0.10 pg mL(-1) for CEA in PBS buffers. The proposed immunosensor was highly sensitive, selective and has a good match to the clinical electrochemiluminescent method. This suggested that the polymerization-assisted immunosensing strategy could be used as an effective method to significantly enhance signal output of the sandwich immunoassays and acted as a promising platform for the clinical screening of cancer biomarkers.
Electrochemical biosensing using amplification-by-polymerization.

Abstract:A novel signal amplification strategy for electrochemical detection of DNA and proteins based on the amplification-by-polymerization concept is described. Specifically, a controlled radical polymerization reaction is triggered after the capture of target molecules on the electrode surface. Growth of long chain polymeric materials provides numerous sites for subsequent aminoferrocene coupling, which in turn significantly enhances electrochemical signal output. Activators generated electron transfer for atom transfer radical polymerization (AGET ATRP) is used in this study for its high efficiency in polymer grafting and better tolerance toward oxygen in air. 2-Hydroxyethyl methacrylate (HEMA) and glycidyl methacrylate (GMA) are examined to provide excess hydroxyl or epoxy groups for aminoferrocene coupling. A limit of detection of 15 pM and 0.07 ng/mL is demonstrated for DNA and ovalbumin, respectively. More than 7-fold signal enhancement in ovalbumin detection has been achieved comparing to the unamplified method. In addition, a more than 5 orders of magnitude of dynamic range is achieved with a linear correlation coefficient (R-2) of 0.997 for DNA, and a more than 3 orders of magnitude with R-2 of 0.999 for ovalbumin. Together, the results, show that the coupling of amplification-by-polymerization concept with electrochemical detection offers great promises in providing a sensitive and cost-effective solution for biosensing applications.
A Novel Electrochemical Immunosensor Incorporating a Pyrrole/4-(3-Pyrrolyl) Butyric Acid Conducting Polymer

Abstract:A novel amperometric immunosensor was fabricated using an electropolymerized conducting polymer composite of pyrrole and 4-(3-pyrrolyl) butyric acid as the probe immobilization matrix. The biomolecule was immobilized via covalent bonding on the polymer surface with a high density of carboxyl groups. Instead of directly attaching the probe to the polymer surface, the streptavidin-biotin affinity complex was introduced to improve the bioactivity, orientation, and the amount of immobilized material. Polyvinyl alcohol was employed to improve the hydrophilicity of the polymer surface and further decrease the nonspecific protein adsorption. As a result, high sensitivity and specificity were achieved with a wide linear dynamic range. Using mouse immunoglobulin G as a model protein, the immunosensor exhibited a linear relationship between 7ng/mL and 20 mu g/mL with a limit of detection of 7ng/mL.

Abstract:Electrochemiluminescence (ECL) has been widely utilized in immunoassay. The amplification of ECL signal can be effectively realized through the modification of functional nanomaterials, thereby obtaining accurate detection results. In this work, a hybrid material AuNPs@Fe-Zr-MOL was prepared based on Au nanoparticles (AuNPs) and metal-organic layers (MOLs), a novel kind of ultra-thin metal-organic frameworks, for the amplification of the ECL signal of luminol. In this hybrid material, Fe-Zr-MOL was synthesized by introducing the catalytic active site of Fe2+ into Zr-MOL through the coordination interaction. Fe-Zr-MOL could enhance the ECL signal of luminol effectively owing to its peroxidase-like activity. In particular, in comparison with three-dimensional MOFs, Fe-Zr-MOL could expose more active sites for luminol catalysis benefiting from its planar structure, thus enhancing the conductivity availably. While, another key member AuNPs could not only improve the efficiency of electron transport, but also be used for antibody conjugation. Further, a sandwich immunosensor was constructed based on antibody-conjugated AuNPs@Fe-Zr-MOL for the detection of alpha fetoprotein (AFP). The results showed that the as-prepared immunosensor could enhance the amplification of ECL signal effectively and realize effective immunodetection of AFP. This work extends the application of MOLs in nanozyme catalyzed amplification of ECL signal and the construction of immunosensor.

Abstract:An intramolecular self-enhanced electrochemiluminescent derivative is prepared by grafting polystyrene (PS)-based polymer chains with pendant Ru(II) luminophore from poly(ethylenimine) (PEI) on the surface of palladium nanocages (PdNCs). In this way, the Ru(II) luminophore and its co-reactive group (amine groups in PEI) exist in the same polymer molecule, which shortens the electronic transmission distance between them and enhances the luminous stability. Meanwhile, through atom transfer radical polymerization (ATRP), the loading amount of Ru(II) luminophore is greatly increased. Therefore, the obtained electrochemiluminescent derivative (PdNC-PEI-PSRu) has high luminous efficiency and stability. Furthermore, due to their special nanostructures of porous walls and hollow interiors, PdNCs have great advantages in high specific surface areas and good electrocatalytic ability, which make them act as an excellent immobilized platform for PEI and detection antibody. Based on the sandwiched immunoreactions, a sensitive "signal on" electrochemiluminescence immunosensor is constructed for the detection of carbohydrate antigen 15-3 (CA 15-3). As a result, a wide linear range from 0.01 U mL(-1) to 120 U mL(-1) is acquired with a relatively low detection limit of 0.003 U mL(-1).

Abstract:Co-reactant electrochemiluminescence (ECL) is a simple and effective method for sensitive detection with amplified ECL signals. However, the intermolecular interaction between the luminescent reagents and their corresponding co-reactants, which is widely applied, has disadvantages in poor stability, low efficiency of electron transfer and relatively high loss of energy. In this work, an intramolecular self-enhanced ECL is proposed to settle this problem. Firstly, palladium nanowires (PdNWs) are synthesized with a green procedure in which Lentinan (LNT), one of β-glucans with a triple helical conformation (t-LNT) in aqueous solution and single chains (s-LNT) at a temperature higher than 130°C, is used as stabilizer and reducing agent. The abtined PdNWs are applied to immobilize polyamidoamine (PAMAM) dendrimer which further reacts with tris (4, 4′-dicarboxylicacid-2, 2′-bipyridyl) ruthenium (II) dichloride to form a new electrochemiluminescent derivative (PdNWs–PAMAM–Ru). In this way, the Ru (II) luminophore and its co-reactive groups (amine groups in PAMAM) exist in the same complex, by which the electronic transmission distance is shortened and the luminous properties including stability and efficiency are enhanced. Moreover, due to the high specific surface areas and good electro-catalytic ability of PdNWs, the obtained PdNWs–PAMAM–Ru can be also applied to immobilize detection antibody (Ab2). Then, a sandwiched and sensitive ECL immunosensor is fabricated for the detection of carcinoembryonic antigen (CEA) with a wide linear ranged from 0.001ngmL−1 to 80ngmL−1 and a low detection limit of 0.3pgmL−1.

Abstract:Electrochemiluminescence (ECL) has been engineered to perform various tasks in the area of immunoassays and molecular diagnosis. However, there is still substantial potential for developments of ECL assay with high efficiency to achieve trace analysis. Herein, we demonstrate a polymer-amplified ECL assay via construction of linear Ru(bpy)32+–polymer. This new polymer material compensates for the relatively low ECL intensity from single ECL luminophore and realizes a stable and controllable labeling process. The polymer-amplified ECL assay achieved a remarkable sensitivity of 100 amol. The wide-ranging applications of the polymer-amplified ECL assay for Hepatitis B virus, carcinoembryonic antigen, 16sRNA, and thrombin also demonstrate its superiority. Hence, the polymer-amplified ECL assay possesses the potential to create a new paradigm in amplified ECL assays that could provide outstanding performance for biomedical analysis.

Abstract:Novel luminescence-functionalized metal-organic frameworks (MOFs) with superior electrogenerated chemiluminescence (ECL) properties were synthesized based on zinc ions as the central ions and tris(4,4'-dicarboxylicacid-2,2'-bipyridyl)ruthenium(II) dichloride ([Ru(dcbpy)3](2+)) as the ligands. For potential applications, the synthesized MOFs were used to fabricate a "signal-on" ECL immunosensor for the detection of N-terminal pro-B-type natriuretic peptide (NT-proBNP). As expected, enhanced ECL signals were obtained through a simple fabrication strategy because luminescence-functionalized MOFs not only effectively increased the loading of [Ru(dcbpy)3](2+), but also served as a loading platform in the ECL immunosensor. Furthermore, the proposed ECL immunosensor had a wide linear range from 5 pg mL(-1) to 25 ng mL(-1) and a relatively low detection limit of 1.67 pg mL(-1) (signal/noise=3). The results indicated that luminescence-functionalized MOFs provided a novel amplification strategy in the construction of ECL immunosensors and might have great prospects for application in bioanalysis.
A Novel Electrochemiluminescence Immunosensor Based on Functional Β-Cyclodextrin-ferrocene Host-Guest Complex with Multiple Signal Amplification

Abstract:The shortened distance between luminophor and coreactant can effectively enhance luminous efficiency in electrochemiluminescence (ECL) system owing to the improvement of electron transfer rate. Here, a novel sandwich-type ECL immunosensor based on N-(aminobutyl)-N-(ethylisoluminol) (ABEI) as luminophor was constructed by integrating hollow gold nanoparticles (HAuNPs), L-cysteine (L-Cys) and ferrocene (Fc) in a complex for multiple signal amplification. The HAuNPs with large specific surface areas and superior conductivity were first used as scaffolds to load abundant coreactant L-Cys, aiming to further assemble catalyzer Fc stably. Through host-guest recognition, the beta-cyclodextrin (beta-CD) functionalized ABEI thus subsequently decorated on Fc, bringing in shortened distance between luminophor ABEI and coreactant L-Cys with high ECL signal. Moreover, the catalyzer Fc and HAuNPs could dramatically promote decomposition of another coreactant H2O2 to produce abundant superoxide radical (O-2(center dot-)) and hydroxyl radical (OH center dot) for further signal amplification. Taking beta 2-microglobulin (beta 2 M) as model analyte, the developed immunosensor exhibited a superior performance for detection beta 2 M in the range of 1.0 x 10(-3) 1.0 x 10(3) ngmL(-1) with a detection limit of 2.6 x 10(-1) pg mL(-1). The developed strategy realized solid-state ECL detection and multiple ECL signal amplification, providing a novel sensitive detection method for beta 2 M, which may show a great promise for the clinical application. Abstract:Developing novel luminophores for electrochemiluminescence (ECL) that could operate smoothly under low-potential excitation while incorporating creative enhancement strategies is a challenging but very attractive initiative to achieve sensitive detection of disease markers. Herein, carbonized polymer dots (L-CPDs) with low cathode excitation potential were successfully synthesized and used for dual amplified sensitive electrochemiluminescence immunoassay of cytokeratin 19 fragment antigen 21–1 (CYFRA21–1). l -CPDs were first used as ECL luminophores and have an unprecedentedly low excitation potential of -0.95 V that has not been equipped for other cathodic carbon-base dots, avoiding side reactions, bioactive damage and electrode polarization due to high potential, thus obtaining excellent sensor performance. In addition, zeolitic imidazolate framework-8 (ZIF-8) synthesized at room temperature as a nanoconfinement carrier encapsulated l -CPDs and reduced the non-radiative energy loss of the luminophore, which led to substantially improved the ECL signal. The ECL efficiency of the encapsulated luminophores is 2.7 times higher than that of monomeric l -CPDs at the same mass concentration. Platinum composite iron oxide (Fe 3 O 4 /Pt) was used as a co-reaction acceleration platform to strengthen the ECL response. The sandwich ECL immunosensor fabricated achieved sensitive detection of CYFRA21–1 in the detection range of 150 fg/mL–110 ng/mL with a detection limit of 126 fg/mL. In conclusion, this work provides a consideration direction of developing ideal luminophores complemented by amplification strategies in ECL systems construction to achieve accurate and sensitive detection of targets.

Aggregation-induced electrochemiluminescence based on intramolecular charge transfer and twisted molecular conformation for label-free Immunoassay

Abstract:Organic emitters with exceptional properties exhibit significant potential in the field of aggregation-induced electrochemiluminescence (AIECL); however, their practicality is impeded by limited ECL efficiency (ΦECL). This paper investigates a novel type of AIECL emitter (BDPPA NPs), where an efficient intramolecular charge transfer (ICT) effect and highly twisted conformation contribute to a remarkable enhancement of ECL. The ICT effect reduces the electron transfer path, while the twisted conformation effectively restricts π-π stacking and intramolecular motions. Intriguingly, compared to the standard system of [Ru(bpy)32+]/TPrA, bright emissions with up to 54 % ΦECL were achieved, enabling direct visual observation of ECL through the co-reactant route. The label-free immunosensor exhibited distinguished performance in detecting SARS-CoV-2 N protein across an exceptionally wide linear range of 0.001-500 ng mL-1, with a remarkably low detection limit of 0.28 pg mL-1. Furthermore, this developed ECL platform exhibited excellent sensitivity, specificity, and stability characteristics, providing an efficient avenue for constructing platforms for bioanalysis and clinical diagnosis analysis.
Synergistic Amplification Effect for Electrochemiluminescence Immunoassay Based on Dual Coreactants Coupling with Resonance Energy Transfer.

Abstract:Sensitive electrochemiluminescence (ECL) bioanalysis could be achieved by exploring the amplification principle or strategy. This work described the first proof-of-concept study for coupling dual coreactants (K2S2O8 and H2O2) with localized surface plasmon resonance (LSPR) of silver nanoparticles (AgNPs) enhancement as an effective signal amplification strategy for ECL immunosensor. Specifically, AgNPs as an excellent conductor and nanocarriers were employed as the antibody (Ab(2)) immobilization film on the glassy carbon electrode (GCE) for the subsequent sandwich-type immunoreaction between the target protein and antibody (Ab(1))-g-C3N4 resulted in the ECL generation. On the one hand, due to the perfect overlap between the plasmon absorption spectrum of AgNPs and the ECL emission spectrum of g-C3N4, a plasmon-enhanced ECL phenomenon was proved in g-C3N4 AgNPs system result from high efficient energy transfer. On the other hand, it was validated that the ECL signal of the constructed biosensor was greatly strengthened by K2S2O8 and H2O2 as dual coreactants, which was ca. 3.5 and 7 times stronger than that of K2S2O8 or H2O2 as an individual coreactant, respectively. Consequently, this synergistic amplified ECL immunosensor performed well with great stability and repeatability for squamous cell carcinoma antigen (SCCA) detection in the range from 0.001 to 1000 ng mL(-1). Considering the favorable analytical feedback in actual serum samples assay, this proposed method indicates a great potential for bioassay application.

Abstract:An immunosensor based on the electrochemiluminescence (ECL) of peroxydisulfate was firstly proposed by coupling the cooperation of two enzymes to in situ generate coreactant with palladium nanoparticles (PdNPs) as catalyst for the ECL reaction. PdNPs were previously synthesized, which successfully attached to functional carbon nanotubes (FCNTs), to bind the secondary antibody and bienzyme (horseradish peroxidase and glucose oxidase). Then the prepared bioconjugates were introduced to the electrode via sandwich immunoreactions. Accordingly, a dramatically amplified ECL signal was obtained for that GOD catalyzed glucose to produce H2O2 which was subsequently reduced by HRP to in situ generate O-2, then PdNPs as catalyst for the ECL reaction of peroxydisulfate/O-2. The present immunosensor was used to detect alpha-1-fetoprotein (AFP) and showed a wide linear range of 1 x 10(-5)-100 ng mL(-1), with a low detection limit of 3.3 fg mL(-1)(S/N=3). This new signal amplification strategy for preparation of the ECL immunosensor could be easily realized and has a potential application in ultrasensitive bioassays. (C) 2012 Elsevier B.V. Abstract:In this work, N-(aminobutyl)-N-(ethylisoluminol) (ABEI), an analogue of luminol, is served as both the reductant and luminescence reagent to synthesize ABEI capped Pd@Au core-shell nanoparticles (ABEI-Pd@AuNPs). The nanoparticles not only exhibit inherent electrochemiluminescence (ECL) property, but also possess advantages of noble-metal nanomaterials such as outstanding electronic property, high specific surface area and good biocompatibility. In order to enhance the luminescence efficiency, ferrocene monocarboxylic acid (Fc) as catalyzer is grafted on the surface of ABEI-Pd@AuNPs with the aid of l-cysteine (l-Cys). When the Fc is electrochemically oxidized to ferricinium cation species (Fc+), the decomposition of H2O2 which existed in detection solution can be catalyzed by Fc+ to generate oxygen-related free radicals, resulting effective signal amplification for ABEI-H2O2 system. For potential applications, the Pd@Au core-shell nanoparticles bifunctionalized by ABEI and catalyzer are employed as nano-carriers to immobilize detection antibody (Ab2). Based on sandwiched immunoreactions, a “signal-on” ECL immunosensor is developed for detection of human collagen type IV (Col IV), a potential biomarker associated with diabetic nephropathy. Consequently, the proposed immunosensor provides a wide linear detection ranging from 1pgmL−1 to 10ngmL−1 with a relatively low detection limit of 0.3pgmL−1 (S/N=3).
Atomically Fe-anchored MOF-on-MOF nanozyme with differential signal amplification for ultrasensitive cathodic electrochemiluminescence immunoassay

Abstract:The successful application of electrochemiluminescence (ECL) in immunoassays for clinical diagnosis requires stable electrodes and high-efficient ECL signal amplification strategies. Herein, the authors discovered a new class of atomically dispersed peroxidase-like nanozymes with multiple active sites (CoNi-MOF@PCN-224/Fe), which significantly improved the catalytic performance and uncovered the underlying mechanism. Experimental studies and theoretical calculation results revealed that the nanozyme introduced a Fenton-like reaction into the catalytic system and the crucial synergistic effects of definite active moieties endow CoNi-MOF@PCN-224/Fe strong electron-withdrawing effect and low thermodynamic activation energy toward H2O2. Benefiting from the high peroxidase-like activity of the hybrid system, the resultant ECL electrode exhibited superior catalytic activity in the luminol-H2O2 system and resulted in an ≈17-fold increase in the ECL intensity. In addition, plasmonic Ag/Au core-satellite nanocubes (Ag/AuNCs) were designed as high-efficient co-reactant quenchers to improve the performance of the ECL immunoassay. On the basis of the differential signal amplification strategy (DSAS) proposed, the immunoassay displayed superior detection ability, with a low limit of detection (LOD) of 0.13 pg mL-1 for prostate-specific antigen (PSA). The designed atomically anchored MOF-on-MOF nanozyme and DSAS strategy provides more possibilities for the ultrasensitive detection of disease markers in clinical diagnosis.
A Compatible Sensitivity Enhancement Strategy for Electrochemiluminescence Immunosensors Based on the Biomimetic Melanin-Like Deposition.

Abstract:In this work, a compatible strategy was demonstrated for the enhancement of detection sensitivity of sandwich-type electrochemiluminescence (ECL) immunosensors. The enhanced signal response was based on the combination of biomimetic melanin-like deposition with the effective ECL quenching ability of quinone-rich biopolymers. Gold nanoparticle-loaded horseradish peroxidase (HRP) was used as a catalytic label for the secondary antibodies. The intrinsic catalytic property of HRP toward hydrogen peroxide (H2O2) generates reactive oxygen species, which highly promote the autopolymerization of catecholamines. The resulting fast deposition of quinone-rich biopolymers approaching the luminophor-incorporated sensing platform achieves an obvious ECL quenching. A broad-spectrum tumor marker alpha fetoprotein (AFP) was selected as a model analyte to demonstrate the feasibility of the proposed strategy. Under optimal conditions, a very low detection limit of 0.056 pg mL-1 was obtained. Two orders of magnitude enhancement was achieved in contrast to the signal response without the step of catalytic biopolymer deposition. The combination of compatible HRP labeling with unique melanin-like deposition has potential as a universal strategy in other ECL bioassays.
