Abstract:Because of the inherent difficulty in differentiating two olefins, the development of metal-catalyzed asymmetric cyclization of 1,6-dienes remains challenging. Herein, we describe the first rhodium(III)-catalyzed asymmetric borylative cyclization of cyclohexadienone-tethered mono-, 1,1di-, and (E)-1,2-disubstituted alkenes (1,6-dienes), affording optically pure cis-bicyclic skeletons bearing three or four contiguous stereocenters with high yields (25-93%), and excellent diastereoselectivities (>20:1 dr) and enantioselectivities (90-99% ee). This mild catalytic approach is generally compatible with a wide range of functional groups, which allows several facile conversions of the cyclization products. Furthermore, on the basis of our SAESI-MS experiment and computational study, a Rh(I)/(III) catalytic cycle is proposed in this tandem reaction, and the Rh(I) active species catalyzes the overall transformation via sequential oxidative addition of B(2)pin(2), olefin insertion, cyclizing conjugate addition, and reductive elimination. The irreversible conjugate addition determines the overall regioselectivity of borylative cyclization, and the ring strain favors the formation of 5,6-bicyclic structure. This highlights the control of ring strain in diene cyclizations, which provides a useful basis for future reaction designs.

Abstract:Reported herein is the first rhodium-catalyzed [4+2+1] cycloaddition of in situ generated ene/yne-ene-allenes and CO to synthesize challenging seven-membered carbo-cycles fused with five-membered rings. This reaction is designed based on the 1,3-acyloxy migration of ene/yne-enepropargyl esters to ene/yne-ene-allenes, followed by oxidative cyclization, CO insertion, and reductive elimination to form the final [4+2+1] cycloadducts. The possible competing [4+1], [4+2], and [2+2+1] cycloadditions were disfavored, making the present reaction an efficient way to access functionalized 5/7 rings.

Abstract:The transition-metal-catalyzed asymmetric cycloisomerization of 1,7-enynes is regarded as a formidable challenge due to the poor ability of 1,7-enynes to serve as bidentate ligands to metal. In this Letter, a highly enantioselective rhodium(I)-catalyzed Alder-ene-type cycloisomerization of 1,7-enynes is disclosed, offering an efficient method for the synthesis of a wide range of fused six-membered cyclic compounds. Abstract:We report the first Rh2(II)-catalyzed asymmetric cycloisomerization of activated enynes to provide cyclopropane-fused tetra-hydropyridines in good yields and excellent enantioselectivities under mild conditions. The activated group, CHZ (Z is electron-withdrawing group (EWG)), in the enyne substrates exerts two synergetic roles, one is to activate alkyne for the cyclopropanation reaction; the other is to introduce the C-H···O interaction between substrate and catalyst (reducing the energy barrier of the reaction). This double-mode activation was supported by both density functional theory (DFT) calculations and experimental tests. This strategy was also extended to other CH2Z (Z can be OH, OMe, F) as activating groups that made the CH2 more acidic so that the substrates could also form increased C-H···O interaction with the catalyst.

Abstract:A catalyst system, [Rh(COD)Cl]2-BINAP-AgSbF6, has been developed as a second-generation catalyst for the cycloisomerization of 1,6-enynes tethered by carbon chains. Cyclopentanes and cyclopentanones, which can contain functional groups, such as the 1,4-dienes, vinyl ether, aldehyde etc., were obtained from readily available starting materials in high yields and selectivities. Both regioselectivities and enantioselectivities are excellent: only 1,4-dienes were observed and in over 99% ee.

Abstract:A rhodium-catalyzed enantioselective hydroboration/cyclization reaction of 1,6-enynes is achieved by employing a spirosiladiphosphine ligand. The process allows the synthesis of five-membered hetero- and carbocycles bearing a boron handle with high levels of activity and selectivity. Various enynes and organoboranes (HBdan, HBpin, HBmp, and HBamm) have been accommodated, and enynes containing terminal alkynes have been integrated into the process for the first time. The high yields and selectivities of the transformation highlight the synthetic utility of these novel spirosiladiphosphine ligands.

Abstract:Through the combined use of computational (density functional theory) and experimental studies, a new [Rh(CO)(2)Cl](2) catalyzed two-component [5+2+1] cycloaddition of ene-vinylcyclopropanes and CO for the synthesis of fused bicyclic cyclooctenones has been designed and experimentally realized. The key point behind this design is to turn a disfavored reductive elimination of (sp(3))C-Rh-C(sp(3)) to an easier migratory reductive elimination of (sp(3))C-Rh-CO-C(sp(3)) by the introduction of CO. These reactions proceed in good yields for substrates with various tether types and substituents, providing a flexible, convenient, efficient, and stereocontrolled method for the construction of bicyclic cyclooctenones.

Abstract:Vinylcyclopropane (VCP) has been well applied as a five-carbon component, rather than a three-carbon component, in transition-metal catalyzed cycloadditions. Here we demonstrate a Rh(l)-catalyzed [3 + 2] reaction of trans-VCP-enes, where VCP acts as a three-carbon synthon to furnish five-membered carbocycles. This novel cycloaddition is efficient in generating bicyclic cyclopentanes in good yields from simple and easily prepared substrates. When cis-VCP-ene is used as the substrate, VCP acts as a five-carbon unit to give a [5 + 2] cycloadduct. Rationalization of the [3 + 2] and [5 + 2] cycloadditions of VCP-enes has been proposed.

Abstract:Transition metal-catalyzed cycloisomerization of 1,6-enynes is a highly effective approach to generating five-membered rings. Among the various catalytic systems developed, the use of chiral ligands in Rh-catalyzed cycloisomerization induced exceptional reactivity and enantioselectivity. In this study, we describe a novel approach to obtained kinds of five-membered rings via Rh-catalyzed cycloisomerization of N-aryl 1,6-enynes, employing a novel spirosilane diphosphine ligand with a significantly enhanced enantioselectivities. Mechanistic experiments and DFT calculations have provided insights into the reaction mechanism.

Abstract:Described herein is an unprecedented example of generating the endocyclic donor–donor vinyl carbene achieved by the dirhodium(II)-catalyzed enynal cycloisomerization. The endocyclic vinyl carbene intermediate could be trapped by a diversity of alkenes, including 1,1-disubstituted alkenes, dienes, and α-methyl styrenes, which could undergo [2 + 1], [4 + 3] cycloaddition and an unexpected formal allylation, respectively, giving spiro and fused polycyclic heterocycles. In these cascade transformations, up to three rings and three stereocenters were constructed in one step with good selectivities. The enantioselective versions were also achieved with up to 84% ee. Mechanistic studies revealed that a concerted intramolecular [2 + 3] cycloaddition was the key step for the generation of vinyl Rh(II) carbene. Detailed studies were conducted to elucidate the intriguing mechanism of the formal allylation.

Abstract:<p>The first example of Rh‐catalyzed kinetic resolution of 1,3‐disubstituted allene‐1,3‐dienes involving intramolecular [4+2]‐cycloaddition has been developed. Followed by C=C isomerization, the reaction finally afforded aza‐[4.3.0]bicyclic compounds with a very high enantio‐ and E‐selectivity. The mechanism has been unveiled by careful control experiments. Abstract:Conjugated enynones can be used as carbene precursors to couple with arylboronic acids in the presence of Rh(I) catalyst. This reaction shows good functional compatibility, and a range of furyl-containing triarylmethanes can be smoothly synthesized from easily available starting materials under mild reaction conditions. Mechanistically, the formation of Rh(I) (2-furyl)carbene species and the subsequent carbene migratory insertion are proposed as the key steps in this reaction.

Abstract:The Rh(I)-catalyzed reaction of siloxyvinylcyclopropanes and diazoesters leads to the formation of siloxyvinylcyclobutane and 1,4-diene derivatives. With [Rh(cod)Cl]2 as the catalyst, the formation of 1,4-diene was favored over the formation of siloxyvinylcyclobutane. By changing the catalyst to [Rh(cod)2OTf], siloxyvinylcyclobutane derivatives are formed with excellent chemoselectivities and in moderate to good yields. The alkene products are also obtained as single E configured isomers. A detailed mechanism for this transformation is proposed on the basis of mechanistic experiments and DFT calculations. The effect of catalysts on the chemoselectivity of these reactions is also examined computationally.
