Abstract:A one-pot two-step reaction of 4-aminocoumarin with arylglyoxal monohydrates and p-toluenesulfonates has been developed, which offers 3-alkoxy-substituted chromeno[4,3-b]pyrrol-4(1H)-ones in moderate to good yields. At first, arylglyoxal monohydrates and 4-aminocoumarin were converted into 3-hydroxychromeno[4,3-b]pyrrol-4(1H)-one intermediates in AcOH under reflux. Then the title compounds in an in situ reaction of the intermediates with p-toluenesulfonates catalyzed by DBU in refluxing toluene were obtained.

Abstract:A concise and efficient synthetic approach was developed for the synthesis of functionalized chromeno[4,3-b]pyrrol-4(1H)-ones via a one-pot, three-component reaction of 4-aminocoumarin, arylglyoxal monohydrates, and aromatic amines promoted by KHSO4 in toluene under reflux condition. The attractive features of this protocol are operational simplicity, short reaction time, easy purification of products and good to excellent yields.

Abstract:A facile and efficient domino cyclization between 3-aminocoumarins and arylglyoxal monohydrates in the presence of para-toluenesulfonic acid (p-TSA) as a promoter was employed for the synthesis of two libraries of chromeno[3,4-b]pyrrol-4(3H)-one derivatives. Simple workup procedures were used to obtain the products in moderate to excellent yields. The solvents toluene and ethanol served important roles in the selective formation of the pyrrolocoumarin derivatives characterized by their different substitution patterns.

Abstract:The one-pot U-4CR and intramolecular O-alkylation sequence starting from 2-aminophenols in combination with α-bromoalkanoic acids, aldehydes, and isocyanides under controlled microwave heating has been established for a rapid access to highly functionalized 3,4-dihydro-3-oxo-2H-1,4-benzoxazines. With appropriate substitutions on the 1,4-benzoxazines, a microwave-assisted Cu-catalyzed intramolecular amidation was performed to furnish a novel class of heterocyclic conjugates of 1,4-benzoxazines with a 2-oxindole linked through a C–N single bond.

Abstract:A novel three-component reaction of 2-oxo-2H-chromene-3-carbaldehydes with isocyanides and anilines was developed for one-pot assembly of biologically intriguing chromeno[4,3-b]pyrrol-4(1H)-ones, which can also be transferred into diverse polycyclic fused scaffolds through further synthetic manipulations. The described method tolerates a broad substrate scope and proceeds in moderate to good yield via a sequential multicomponent reaction and intramolecular Michael cyclization.

Abstract:A highly diastereoselective [3 + 2] cycloaddition of aziridines with difluorinated silyl enol ethers has been developed. This approach provides a facile methodology for highly functionalized gem-difluorinated pyrrolidines in good to excellent yields with good functional group tolerance. A one-pot, two-step approach for synthesis of structurally interesting fluorinated pyrroles has also been developed through a cycloaddition/aromatization/desulfonation sequence. Moreover, readily available substrates, mild reaction conditions, and easy scale-up synthesis show practical advantages.

Abstract:A one-pot synthesis of pyrrolo[3,2-d]pyridazine derivatives via zirconocene-mediated four-component coupling of one Si-tethered diyne, two nitriles, and one azide is reported. When TMSN3 was used, pyrrole-2,3-diones were formed in good yields. Further condensation of these highly functionalized pyrrole-2,3-diones with hydrazine and hydroxylamine afforded useful pyrrole-fused heterocycles.

Abstract:4,6-Diarylpyrimidin-2(1 H)-ones were effectively synthesised by utilising chlorotrimethylsilane (TMSCI) as an efficient promoter in the cyclisation condensation of arylketones, substituted benzaldehydes and urea by a one-pot, three-component reaction under air in DMF/CH3CN. The clean, mild reaction conditions, operational simplicity and high yields were attractive features of the reaction which enables a facile preparative procedure for building the pyrimidine ring.

Abstract:We developed a novel protocol for the preparation of functionalized chromeno[2,3-c]pyrrol-9(1H)-one derivatives (CMPOs) 3 & 3' from 3-formylchromones with proline derivatives. This strategy involved an unprecedented cascade reaction entailing simple heating of a mixture of substrates 1 with 2 and toluene, followed by oxidation with oxygen and catalysis by CuBr. As a result, a series of CMPOs 3 & 3' were produced via cleavage of two bonds (2 C−C) and formation of three bonds (1 C−N and 2 C−C bonds) in a single step. This protocol can be used to synthesize functionalized CMPOs for combinatorial and parallel syntheses of CMPOs in a one-pot reaction.

Abstract:A novel one-pot pseudo-three-component reaction of 2-amino-4H-chromen-4-ones with 4-benzylidene-5-alkyl-2-aryl-2,4-dihydro-3H-pyrazol-3-ones was investigated for the synthesis of the spiro[chromeno[2,3-b]pyridine-3,4'-pyrazole] derivatives. This procedure involved Michael addition, elimination, and hetero-Diels-Alder sequences, affording a series of spiropyrazolone-chromeno[2,3-b]pyridines in good yields that possessed cis relationships between two aryl groups and the carbonyl of the pyrazolone unit.
