Abstract:A 2-month fumigation experiment was conducted with opened top chambers to study the effects of different concentration (0.1, 0.5, and 4.0 microl x L(-1)) NO2 on the growth and photosynthesis of 1-year Cinnamomum camphora seedlings. Fumigation with 0.1 and 0.5 microl NO2 x L(-1) promoted the growth of the seedlings, while with 4.0 microl NO2 x L(-1) was in adverse. The diurnal variation of net photosynthetic rate (P(n)) presented two-peaks, with an obvious depression in midday. 0.5 microl NO2 x L(-1) increased the P(n), the maximum of P(n) reached 8.542 micromol CO2 x m(-2) s(-1); 4.0 microl NO2 x L(-1) decreased the P(n) in most period of time; while the effect of 0.1 microl NO2 x L(-1) varied with time period. Fumigation with 0.5 and 4.0 microl NO2 x L(-1) increased the maximal and minimal values of stomatal conductance (G(s)) and intercellular CO2 concentration (C( i)), while 0.1 microl NO2 x L(-1) increased the maximal and minimal values of C(i) but decreased the maximal and minimal values of G(s). At the middle and late stages of fumigation, the mean P(n) of the seedlings treated with 0.5 microl NO2 x L(-1) was significantly higher than that treated with 0.1 and 4 microl NO2 x L(-1). At the early stage of fumigation, 0.5 and 4.0 micro NO2 x L(-1) significantly decreased the maximal PS II efficiency (F(v)/F(m)); and at the late stage, 4.0 microl NO2 x L(-1) still decreased the F(v)/F(m) significantly.
Effects of Exogenous Nitric Oxide Donor SNP on Membrane Lipid Peroxidation and Activity of Antioxidative Enzyme in Rice Seedlings under Cd Stress

Abstract:The growth of rice seedling,membrane lipid peroxidation and the specific activities of antioxidant enzymes(superoxide dismutase,catalase,ascorbate peroxidase and guaiacol peroxidase) in rice seedlings treated with a varying concentrations of SNP under 30 mg/L Cd2+ stress were evaluated.The results showed that pretreatment with SNP alleviated significantly the oxidative damage caused by Cd stress in the leaves and roots of rice seedlings,reduced the contents of H2O2 and malondialdehyde(MDA),and decreased the activities of superoxide dismutase(SOD),catalase(CAT),ascorbate peroxidase(APX),and guaiacol peroxidase(GPX) in the leaves and roots of rice seedlings under Cd stress.Nitric Oxide(NO) acted in plants with a concentration-dependent manner,lower concentrations of NO acting as an efficient scavenger in breaking the oxidative chain.Higher NO concentrations could provoke a major oxidative insult,since this molecule is a nitrogen reactive species.
Influence of Nitrogen Supply on Growth, Antioxidant Capacity and Cadmium Absorption of Kenaf (Hibiscus cannabinus L.) Seedlings

Abstract:Kenaf (Hibiscus cannabinus L.) is considered suitable for the remediation of cadmium (Cd)-contaminated farmlands, because of its large biomass and resistance to Cd stress. The addition of nitrogen (N) fertilizer is an important measure used to increase crop yields, and it may also affect Cd accumulation in plants. To clarify the effects of different forms and concentrations of N on plant growth and Cd absorption in kenaf, a hydroponic experiment was conducted using three N forms (NH4+–N, NO3−–N and urea–N) at four concentrations (0, 2, 4 and 8 mM, 0 mM as control) under Cd stress (30 μM). The plant growth, the antioxidant enzyme activity and the Cd contents of various parts of the kenaf seedlings were measured. The results showed that the N form had the greatest impact on the growth of the kenaf and the absorption and transport of the Cd, followed by the interaction effect between the N type and the concentration. Compared to the control, the addition of N fertilizer promoted the growth of kenaf to varying degrees. Among all the treatments, the use of 2 mM of NO3−–N enhanced the biomass and Cd accumulation to the greatest extent compared to CK from 2.02 g to 4.35 g and 341.30 μg to 809.22 μg per plant, respectively. The NH4+–N significantly reduced the Cd contents of different parts but enhanced the translocation factors of Cd stem to root (TF S/R) and leaf to stem (TF L/S) by 34.29~78.57% and 45.10~72.55%, respectively. The peroxidase (POD), superoxide dismutase (SOD) and catalase (CAT) enzyme activities of the kenaf increased with the N treatments, especially with NH4+–N. Overall, applying low concentrations of NO3−–N can better promote the extraction of Cd by kenaf.

Abstract:Employing nutrient solution hydroponic method,the effects of exogenous nitric oxide(NO)on the growth and active oxygen metabolism in cucumber(Cucumis sativus L.)seedlings under NaCl stress were investigated.The results indicated that NaCl treatment significantly inhibited the growth of cucumber seedlings,while exogenous NO could significantly alleviate the inhibitory effects of NaCl stress on seedling growth.Especially,0.1 mmol/L SNP treatment exhibited better effects than 0.5 mmol/L SNP treatment on alleviating NaCl stress.Under 0.1 mmol/L NaCl stress,adding 0.1 mmol/L exogenous NO could significantly decrease the generation rate of O·-2and MDA content,significantly improve soluble protein content and enhance the activities of SOD,POD and CAT,thus reducing the damage of salt stress to cucumber seedlings.

Abstract:Effects of exogenous nitric oxide(NO)on some physiological characteristics of leaves and ray florets in chrysanthemum Zaohuang at flowering stage under heat stress were investigated.The results indicated that 0.2 mmol·L-1 exogenous NO donor,named sodium nitroprusside(SNP)had obviously protective effects on leaves and ray florets oxidative damage in heat stressed chrysanthemum.Compared to the control,NO content and activities of antioxidant enzymes including superoxide dismutase,peroxidase,catalase and ascorbic acid peroxidase increased significantly in chrysanthemum plants applied with SNP,while superoxide anion produciton activity,hydrogen peroxide and malondialdehyde content decreased significantly.NO acted as an important signal molecule,might alleviate lipid peroxidation through activating antioxidant enzyme system to improve the heat resistance of chrysanthemum.
A intermediate concentration of atmospheric nitrogen dioxide enhances PSII activity and inhibits PSI activity in expanded leaves of tobacco seedlings

Abstract:Nitrogen dioxide (NO2) is a major air pollutant that affects plant growth, development and yields. Previous studies have found that atmospheric NO2 changes plant photosynthesis in a concentration-dependent manner. Low concentrations of NO2 (4.0 μL L-1) can increase photosynthetic rates, while high concentrations of NO2 (16.0 μL L-1) can have an inhibitory effect. However, the specific effects of a critical intermediate concentration of NO2 on the photosynthetic apparatus of plants has remained unknown. Therefore, in this study, tobacco seedlings at three-leaf ages were fumigated with a intermediate concentration of 8.0 μL L-1 NO2 for 15 days to determine the effects on leaf weight, leaf number per plant, chlorophyll content, net photosynthetic rate, the reaction center activity of photosystems I and II (PSI and PSII, respectively) and core protein gene expression (PsbA and PsaA). Fumigation with 8.0 μL L-1 NO2 increased the number of leaves per plant and the weight of leaves, and the leaves became dark green and curly after 10 days of fumigation. During NO2 fumigation for 15 days, the chlorophyll content, PSII maximum photochemical efficiency (Fv/Fm), electron transfer rate (ETR) and non-photochemical quenching (NPQ) increased most in the oldest leaves (Lmax leaves), but decreased PSI activity (∆I/Io). The Fv/Fm, ETR and NPQ in the youngest leaves (Lmin leaves) were lower than those of Lmax leaves, but the actual photochemical efficiency (ΦPSII) of PSII increased most and ∆I/Io was the highest in these samples. The Fv/Fm, ETR, NPQ and ΦPSII in the leaves at the middle leaf age (Lmid leaves) were lower than those of Lmin and Lmax leaves, but the relative fluorescence intensity of point L (VL) and the relative fluorescence intensity of point K (VK) decreased the most in these samples. Thus, this critical concentration of atmospheric NO2 increased the activity of PSII and inhibited PSI activity in expanded leaves of tobacco seedlings.

Abstract:A pot experiment was conducted to study the effects of sodium nitroprusside (SNP, a nitric oxide [NO] donor) on cadmium (Cd) toxicity in peanut plants. SNP solution was poured into Cd-contaminated soil at sowing, seedling, flowering, and pod setting stages, respectively. The results showed that exogenous NO increased the biomass and yield of peanuts and improved chlorophyll content, photosynthesis (Pn), and transpiration (Tr). Cd-induced oxidative damages were eliminated by exogenous NO, reflected by decreased accumulation of superoxide anion (), hydrogen peroxide (H2O2), and malondialdehyde (MDA). Moreover, exogenous NO increased Cd-decreased activities of superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), and soluble protein and proline contents also reached a normal level. Furthermore, exogenous NO improved the uptake of nutrient elements and reduced Cd accumulation in the roots, shoots, and kernels under Cd stress. These data suggest that SNP application at sowing and seedling stages had better alleviation effects on Cd toxicity in peanut plants.

Abstract:Atmospheric nitrous oxide (N 2 O) increase contributes substantially to global climate change due to its large global warming potential. Soil N 2 O emissions have been widely studied, but plants have so far been ignored, even though they are known as an important source of N 2 O. The specific objectives of this study are to (1) reveal the effects of nitrogen and biochar addition on plant functional traits and N 2 O emission of Cinnamomum camphora seedlings; (2) find out the possible leaf traits affecting plant N 2 O emissions. The effects of nitrogen and biochar on plant functional traits and N 2 O emissions from plants using C. camphora seedlings were investigated. Plant N 2 O emissions, growth, each organ biomass, each organ nutrient allocation, gas exchange parameters, and chlorophyll fluorescence parameters of C. camphora seedlings were measured. Further investigation of the relationships between plant N 2 O emission and leaf traits was performed by simple linear regression analysis, principal component analysis (PCA), and structural equation model (SEM). It was found that nitrogen addition profoundly increased cumulative plant N 2 O emissions (+109.25%), which contributed substantially to the atmosphere's N 2 O budget in forest ecosystems. Plant N 2 O emissions had a strong correlation to leaf traits (leaf TN, P n , G s , C i , T r, WUE L , α, ETR max , I k , Fv/Fm , Y (II), and SPAD ). Structural equation modelling revealed that leaf TN, leaf TP, P n , C i , T r, WUE L , α, ETR max , and I k were key traits regulating the effects of plants on N 2 O emissions. These results provide a direction for understanding the mechanism of N 2 O emission from plants and provide a theoretical basis for formulating corresponding emission reduction schemes.

Abstract:A greenhouse hydroponic experiment was performed using Cd-sensitive (cv. Dong 17) and Cd-tolerant (Weisuobuzhi) barley seedlings to evaluate how different genotypes responded to cadmium (Cd) toxicity in the presence of sodium nitroprusside (SNP), a nitric oxide (NO) donor. Results showed that 5 μM Cd increased the accumulation of O 2 •− , H2O2, and malondialdehyde (MDA) but reduced plant height, chlorophyll content, net photosynthetic rate (P n), and biomass, with a much more severe response in the Cd-sensitive genotype. Antioxidant enzyme activities increased significantly under Cd stress in the roots of the tolerant genotype, whereas in leaves of the sensitive genotype, superoxide dismutase (SOD) and ascorbate peroxide (APX), especially cytosol ascorbate peroxidase (cAPX), decreased after 5–15 days Cd exposure. Moreover, Cd induces NO synthesis by stimulating nitrate reductase and nitric oxide synthetase-like enzymes in roots/leaves. A Cd-induced NO transient increase in roots of the Cd-tolerant genotype might partly contribute to its Cd tolerance. Exogenous NO dramatically alleviated Cd toxicity, markedly diminished Cd-induced reactive oxygen species (ROS) and MDA accumulation, ameliorated Cd-induced damage to leaf/root ultrastructure, and increased chlorophyll content and P n. External NO counteracted the pattern of alterations in certain antioxidant enzymes induced by Cd; for example, it significantly elevated the depressed SOD, APX, and catalase (CAT) activities in the Cd-sensitive genotype after 10- and 15-day treatments. Furthermore, NO significantly increased stromal APX and Mn-SOD activities in both genotypes and upregulated Cd-induced decrease in cAPX activity and gene expression of root/leaf cAPX and leaf CAT1 in the Cd-sensitive genotype. These data suggest that under Cd stress, NO, as a potent antioxidant, protects barley seedlings against oxidative damage by directly and indirectly scavenging ROS and helps to maintain stability and integrity of the subcellular structure.

Abstract:The effect of exogenously applied nitric oxide on the heat tolerance of Chrysanthemum morifolium was investigated by applying the NO donor sodium nitroprusside (SNP). We found that the SNP partially alleviated the heat stress by slowing down the reduction of photosynthetic pigment content and net photosynthetic rate. SNP treatment also lowered the increase in the non-photochemical quenching of fluorescence and malondialdehyde content and maintained higher activities of superoxide dismutase, peroxidase, catalase and ascorbate peroxidase.
Nickel oxide nanoparticles cause substantial physiological, phytochemical, and molecular-level changes in Chinese cabbage seedlings

Abstract:Nickel oxide nanoparticles (NiO NPs) are utilized in various industries and their release into the environment may lead to the pollution of agricultural areas. However, assessing the toxicity of NiO NPs in major food crops is difficult due to the limited information available on their toxicity. The present investigation was carried out to evaluate how NiO NPs affect plant growth, photosynthetic efficiency, and phytochemical content, as well as changes at the transcriptional level of these phytochemicals in Chinese cabbage seedlings. Chlorophyll, carotenoid, and sugar contents were reduced, while proline and the anthocyanins were significantly upregulated in NiO NPs-treated seedlings. The levels of malondialdehyde, hydrogen peroxide, and reactive oxygen species, as well as peroxidase (POD) enzyme activity, were all enhanced in seedlings exposed to NiO NPs. The levels of glucosinolates and phenolic compounds were also significantly increased in NiO NPs-treated seedlings compared to control seedlings. The expression of genes related to oxidative stress (CAT, POD, and GST), MYB transcription factors (BrMYB28, BrMYB29, BrMYB34, and BrMYB51), and phenolic compounds (ANS, PAP1, and PAL) were significantly upregulated. We suggest that NiO NPs application stimulates toxic effects and enhances the levels of phytochemicals (glucosinolates and phenolic compounds) in Chinese cabbage seedlings.
Effects nitroprusside,a nitric oxide donor,on carbon and nitrogen metabolism and the activity of the antioxidation system in wheat seedings under salt stress

Abstract:Seeds of two wheat cultivars (Huaimai and Yangmai) were pre-soaked with 0. 1 mmol/L SNP (sodium nitroprusside, a nitric oxide donor) to study the effects of nitric oxide (NO) on the activity of the antioxidation system, carbon and nitrogen metabolism, and activities of proteinase in wheat seedlings growing with 120 mmol L-1 NaCl. Exogenous NO significantly reduced the 02 (superoxide anion) release rate as well as H202 content. The activities of SOD (superoxide dismutase) and CAT (catalase) increased, which resulted in a decrease in malondialdehyde (MDA) content in leaves of the seedlings growing under salt stress. In addition, exogenous NO increased the contents of chlorophyll, carotenoids, and the total soluble sugars. Compared with the salt stress treatment alone, the NO pre - treatment significantly increased the content of soluble protein and enhanced the activities of both endopeptidase and carboxypeptidase in leaves. Thus, NO effectively contributed to a better balance between carbon and nitrogen metabolism in seedlings growing under salt stress, and this pre-treatment increased growth rates, as increases in plant height, fresh and dry weight were observed. In addition, Huaimai 17 was more responsive to exogenous NO than Yangmai 12 in this study.
Nitric oxide maintains postharvest quality of navel orange fruit by reducing postharvest rotting during cold storage and enhancing antioxidant activity

Abstract:Current study aimed to evaluate the effects of fumigation with 15 μl L-1 nitric oxide (NO) on the quality of 'Newhall' navel orange fruit during storage at 5 oC. Fruits quality parameters, decay rate, antioxidant capacity, antioxidant enzymes activities and disease resistance related enzymes activities were evaluated. NO treatment significantly inhibited the increasing weight loss, maintained soluble solid content (SSC), titratable acid (TA) and vitamin C (VC) contents (P<0.05). Furthermore, NO treatment significantly decreased the respiration rate, suppressed superoxide anion (O2-·) and hydrogen peroxide (H2O2) generation (P<0.05). Antioxidant capacity, superoxide dismutase (SOD), catalase (CAT) and peroxidase (POD) activities in NO-treated navel oranges were significantly higher than controls (P<0.05). Moreover, exogenous NO significantly enhanced the activities of phenylalanine ammonia-lyase (PAL), polyphenoloxidase (PPO), β-1,3-glucanase (GLU) and chitinase (CHT) (P<0.05). Application of NO would be a promising method in maintaining the quality of postharvest navel orange in the future.

Abstract:The present study was aimed to assess the effect of nitric oxide (NO) and the interaction of NO with hydrogen peroxide (H2O2) on plant tolerance to low temperatures in cucumber seedlings. Exogenous NO significantly increased the endogenous NO content, initial and total activities of ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBisCO), RuBisCO carboxylation rate (Vc,max), RuBP regeneration rate (Jmax) and the transcript levels of related genes in cucumber seedlings under low temperatures (11 °C/7 °C); however, the effect of NO was blocked by PTIO (NO scavenger). In addition, the SNP treatment significantly improved the contents of glucose, fructose, sucrose, starch, and the activities of sucrose phosphate synthase (SPS), acid invertase (AI), sucrose synthase (SS), as well as the expression levels of SUCROSE PHOSPHATE SYNTHASE 1&2 (SPS1, SPS2), SUCROSE TRANSPORTER 1&2 (SUT2, SUT4), β-starch hydrolase (BAM), and invertase gene (INVERTASE) in cucumber leaves under low temperatures, and the positive effect of NO was impaired by PTIO. Furthermore, we found that the H2O2, induced by NO, participated in NO-induced elevation of ascorbic acid (AsA), glutathione (GSH), and increased activities of related enzymes in the AsA-GSH cycle at low temperatures. However, the positive effect of NO was blocked by l-NAME (NOS inhibitor), PTIO, DPI (inhibitor of NADPH oxidase), and DMTU (reactive oxygen species scavenger). Taken together, our findings indicate that NO increased the low temperature tolerance of cucumber seedlings via H2O2 by improving the efficiency of the Calvin cycle, which in turn increased the carbohydrates content and accelerated the AsA-GSH cycle to enhance ROS scavenging.

Abstract:The present study designed to illustrate correlation between cadmium induced stress and plant growth, photosynthetic pigments, morphological and physiological attributes. To study these parameters 2 weeks old seedling of B. juncea were subjected to 50 µM Cd, 100 µM Cd and 100 µM SNP separately and in combination with SNP. After 96 h, the treated plant were harvested to analyze the cellular homeostasis and metal tolerance mechanism via examining growth, stress parameters, enzymatic and non enzymatic antioxidants and expression level of NR. Higher level of Cd (100 µM) significantly increased accumulation of reactive oxygen species and malonaldehyde content in comparison to 50 µM Cd. Exogenous supplementation of SNP (100 µM) to 50 µM Cd treated plant had an additive effect on plant growth by improving the level of proline, photosynthetic pigments and activities of enzymatic antioxidants which was confirmed by histochemical staining for NADPH-d and NO fluorescence from DAF-DA staining in roots of B. juncea. Applying SNP to 50 µM Cd exposed B. juncea roots enhanced NR activity by 1.36 folds and increased NO production by 1.12 folds than individual Cd treated roots. In addition, semi quantitative RT-PCR study revealed the induction of BjNR was more pronounced in 50 µM Cd treated roots in comparison to 100 µM Cd treated roots. The present finding revealed NO confers increased B. juncea tolerance to Cd stress by stimulation of antioxidants and reestablishment of cellular redox status. Different biochemical analysis showed that plant growth, photosynthetic pigment and antioxidants were positively correlated with NO and it's negatively correlated with oxidative stress biomarkers. Therefore, NO is gaseous signalling molecule with potential role in Cd detoxification mechanism in B. juncea.

Abstract:This experiment aimed to study the response of Chinese cabbage seedling to microcystins-LR(MC-LR) stress.By calculation from regression equation,after exposure to 4.23 mg/L MC-LR for 5 d,the growth of Chinese cabbage seedlings decreased by 50% compared to that of control.Exposing to MC-LR significantly enhanced the intercellular levels of total reactive oxygen species(ROS) and nitric oxide(NO),which could be visually detected by using specific florescent probes DCFH-DA and DAD-2DA,respectively.Nitrate reductase(NR) mainly contributed to NO production under MC-LR stress.NaN3(NR inhibitor) significantly reduced MC-LR-induced increases in ROS production and the transcripts of Cu/Zn-SOD,CAT and APX genes in Chinese cabbages seedlings.It indicated that NO mediated MC-LR-induced oxidative stress in Chinese cabbage seedlings.
Exogenous nitric oxide treatment delays the senescence of postharvest mung bean sprouts by regulating ascorbic acid metabolism

Abstract:Background: This study evaluated the effects of nitric oxide (NO) treatment on ascorbic acid (AsA) metabolism and mung bean sprout quality. It examined changes in the AsA content, enzyme activity associated with AsA metabolism, antioxidant capacity, cell membrane composition, and cellular structure to clarify the effects of NO on mung bean sprouts. Results: Nitric oxide treatment preserved mung bean sprout quality by enhancing significantly the activity of enzymes involved in the l-galactose pathway (including guanosine diphosphate (GDP)glutathione (-d-mannose pyrophosphorylase, GDP-mannose-3',5'-epimerase, GDP-l-galactose phosphorylase, l-galactose-1-phosphate phosphatase, l-galactose dehydrogenase, and l-galactose-1,4-lactone dehydrogenase) and the AsA-glutathione (GSH)(Beijing Solarbio Science and Technology Co.,Ltd., Beijing, China) cycle (including ascorbate peroxidase, ascorbic acid oxidase, glutathione reductase, dehydroascorbate reductase, and monodehydroascorbate reductase) during the germination and storage stage. Increased enzyme activity led to an increase in AsA content and enhanced antioxidant capacity, and reduced the membrane lipid damage in mung bean sprouts. This was demonstrated by higher levels of DPPH radical scavenging capacity, unsaturated fatty acids and phospholipids, along with lower levels of hydrogen peroxide, superoxide anions, and malondiadehyde, in NO-treated mung bean sprouts. Scanning electron microscopy also revealed that NO treatment maintained the integrity of the cellular structure of the mung bean sprouts. Conclusion: Nitric oxide accelerates AsA metabolism effectively by regulating the biosynthesis and regeneration of AsA in mung bean sprouts. These changes increased AsA levels, alleviated membrane lipid damage, delayed senescence, and maintained the quality of mung bean sprouts during storage. © 2024 Society of Chemical Industry.
Effects of Exogenous Nitric Oxide on Reactive Oxygen Metabolism of Walnut Seedlings under Low Temperature Stress

Abstract:[Objective]Effects of exogenous nitric oxide ( NO ) on the active oxygen metabolism system of walnut seedlings under low temperature stress were studied,in order to investigate the possible mechanism of exogenous NO on the cold resistance of walnut. This study aimed at developing a new method for improving the resistance of fruit trees by applying exogenous NO and providing a theoretical basis for the wide application of NO on walnut in the future.[Method]The effects of sodium nitroprusside ( SNP with 200 μmol·L -1 ) ,a NO donor,on the active oxygen metabolism of walnut varieties'Xiangling'and'Luguo No. 12',seedlings with different cold resistance were studied in a artificial climate chamber. The seedling were subjected to low temperature treatments and the leaves were sprayed with SNP. [Result]1) Under normal growth conditions,the effects of spraying SNP on the membrane permeability,chlorophyll content, production rate of superoxide anion radical(O·-2 ),H2O2,MDA and Pro content,and SOD and CAT activity in walnut leaves were not obvious. POD,DHAR,APX and GR activity,AsA and GSH content,GSH/GSSG and AsA/DHA ratio were significantly increased,while DHA and GSH content were reduced. The variation ranges of the two walnut varieties were different. 2) Under low temperature stress,SNP treatment significantly increased the activity of POD,CAT,SOD, APX and GR,increased the content of chlorophyll,Pro,AsA,DHA,GSH and GSSG,and decreased the accumulation of MDA and H2O2,production rate of superoxide anion radical(O·-2 ) and relative permeability of plasma membrane. The variation ranges of the two walnut varieties were different. [Conclusion]Under low temperature stress,exogenous NO treatment could maintain the stability of AsA-GSH circulation system,decrease the accumulation of MDA and H2 O2 , production rate of superoxide anion radical ( O·-2 ) and protect the stability of cell membrane structure by promoting antioxidant enzyme activity and antioxidant content,and there by reduce the damage of the active oxygen to the walnut leaf and enhance the cold resistance.
Nitric oxide promotes adventitious root formation in cucumber under cadmium stress through improving antioxidant system, regulating glycolysis pathway and polyamine homeostasis

Abstract:Cadmium (Cd) as a potentially toxic heavy metal that not only pollutes the environment but also interferes with plant growth. Nitric oxide (NO) regulates plant growth and development as well as abiotic stress response. However, the mechanism underpinning NO-induced adventitious root development under Cd stress remains unclear. In this study, cucumber ( Cucumis sativus ‘Xinchun No. 4’) was used as the experimental material to investigate the effect of NO on the development of adventitious roots in cucumber under Cd stress. Our results revealed that, as compared to Cd stress, 10 μM SNP (a NO donor) could considerably increase the number and length of adventitious roots by 127.9% and 289.3%, respectively. Simultaneously, exogenous SNP significantly increased the level of endogenous NO in cucumber explants under Cd stress. Our results revealed that supplementation of Cd with SNP significantly increased endogenous NO content by 65.6% compared with Cd treatment at 48 h. Furthermore, our study indicated that SNP treatment could improve the antioxidant capacity of cucumber explants under Cd stress by up-regulating the gene expression level of antioxidant enzymes, as well as reducing the levels of malondialdehyde (MDA), hydrogen peroxide (H 2 O 2 ) and superoxide anion (O 2 .- ) to alleviate oxidative damage and membrane lipid peroxidation. Application of NO resulted in a decrease of the O 2 .- , MDA, and H 2 O 2 level by 39.6%, 31.4% and 60.8% as compared to Cd-alone treatment, respectively. Besides that, SNP treatment significantly increased the expression level of related genes involved in glycolysis processes and polyamine homeostasis. However, application of NO scavenger 2-(4-carboxy -2-phenyl)-4, 4, 5, 5-tetramethy limidazoline -1-oxyl -3-oxide (cPTIO) and the inhibitor tungstate significantly reversed the positive role of NO in promoting the adventitious root formation under Cd stress. These results suggest that exogenous NO can increase the level of endogenous NO, improve antioxidation ability, promote glycolysis pathway and polyamine homeostasis to enhance the occurrence of adventitious roots in cucumber under Cd stress. In summary, NO can effectively alleviate the damage of Cd stress and significantly promote the development of adventitious root of cucumber under Cd stress.

Abstract:The effects of 60-d cadmium (Cd) exposure on enzymatic and non-enzymatic antioxidative system of Oryza sativa L. seedlings at tillering stage were studied using soil culture experiment. Research findings showed that chlorophyll content of Oryza sativa L. declined with the increase in soil metal concentration. Cd pollution induced the antioxidant stress by inducing O2 −1 and H2O2, which increased in plants; at the same time, MDA as the final product of peroxidation of membrane lipids, accumulated in plant. The antioxidant enzyme system was initiated under the Cd exposure, i.e. almost all the activities of superoxide dismutase (SOD), peroxidase, catalase, glutathione peroxidase, and ascorbate peroxidase were elevated both in leaves and roots. The non-protein thiols including phytochelatins and glutathione to scavenge toxic free radicals caused by Cd stress was also studied. The contents of phytochelatins and glutathione were about 3.12–6.65-fold and 3.27–10.73-fold in leaves, against control; and the corresponding values were about 3.53–9.37-fold and 1.41–5.11-fold in roots, accordingly.
