Abstract:Background: Our previous study shows that satellite glial cells (SGCs) of dorsal root ganglia (DRG) are in close proximity to adrenergic fibers that are marked by expression of tyrosine hydroxylase, an enzyme involved in norepinephrine (NE) synthesis.Several adrenergic receptors (AR) are present in DRG neurons but their expression is less known in SGCs.Using PCR and a variety of inhibitors, we demonstrate that beta (β)2AR mediates NEinduced phosphorylation of transcription factor cAMP response element-binding on serine 133 (p-CREB S 133 ) in cultured DRG glial cells that are predominantly SGCs.The mechanisms of β2AR-mediated DRG glial activity and the function of β2AR in colitis have not been defined.AIMS: 1) Identify the location of β2AR in DRG; 2) examine the activity of DRG glial cells regulated by β2AR; and 3) explore the role of β2AR in colitis and visceral pain.Methods: 1) Fluorescent In Situ Hybridization (FISH); 2) calcium (Ca 2+ ) imaging; 3) western blot, real-time PCR, luciferase assay, and immunohistochemistry; 4) plasmid transfection; 5) induction of colitis by 2,4,6-Trinitrobenzenesulfonic acid (TNBS) in rats; 6) H&E stain; and 7) colonic pain analysis.Results: Using luciferase assay and CREB S142A mutation, we found that NE-induced CREB transcriptional activity was positively correlated with CREB phosphorylation at S 133 and negatively correlated with CREB phosphorylation at S 142 , and was mediated by β2AR but not αAR or β1AR.β2AR-mediated NE signaling in DRG glial cells was Ca 2+ independent.The level of p-CREB S 142 was also reduced in cultured DRG glial cells following NE treatment or in thoracolumbar DRGs examined on day 7 following TNBS treatment.Similar to NE treatment, the level of p-CREB S 133 was increased in SGCs of DRG on day 7 following TNBS treatment.Using FISH, we revealed that β2AR was exclusively expressed by SGCs in DRG.Simultaneous TNBS treatment and intravenous (i.v.) injection of β2AR agonist Formoterol enhanced TNBS-induced tumor necrosis factor alpha (TNFα) up-regulation in DRG on day 3, and exacerbated TNBS-induced colonic damage and pain on day 3 and 7 following drug treatment.Simultaneous TNBS treatment and β2AR antagonist ICI-118551 injection (i.v.) inhibited TNFα up-regulation in DRG, and reduced TNBS-induced colonic damage and pain on day 3 and 7 following drug treatment.CONCLU-SION: Activation of β2AR enhances SGC activity to promote neuroinflammatory process in DRG, and exaggerates colonic damage and pain.Inhibition of β2AR suppresses neuroinflammatory process in DRG, and reduces colonic inflammation and pain.A recent study also reported that β2AR agonist impaired type 2 immune responses in the intestine, and β2AR deficiency reduced worm burden after infection (Moriyama et al., Science.359:
