Abstract:Anion channels play a very important role in regulating plant growth and development,nutrient uptake and(adverse) environment adaptation such as drought,salt stress,Al toxicity and P deficiency.They located on different (organelles) and their characteristics were different.Thus,their regulatory mechanisms,which were complex,were different.The extensive localization,cell function,primary characters and possible signal transduction process of intervention of(anion) channel were comparative reviewed in this article in order to facilitate the further study.In contrast to the large amount of information on physiological role of anion channels,the molecular biochemistry knowledge is quite few.In addition,this paper introduces the possible protein components of S-type anion channel in guard cell protoplast membrane and the genes encoding the transporter facilitating Al induced malate efflux in wheat root apex for the purpose of exploring the research technique on anion channel based on molecular biology

Abstract:Plasma membrane H+-ATPases (PM H+-ATPases) are critical proton pumps that export protons from the cytoplasm to the apoplast. The resulting proton gradient and difference in electrical potential energize various secondary active transport events. PM H+-ATPases play essential roles in plant growth, development, and stress responses. In this review, we focus on recent studies of the mechanism of PM H+-ATPases in response to abiotic stresses in plants, such as salt and high pH, temperature, drought, light, macronutrient deficiency, acidic soil and aluminum stress, as well as heavy metal toxicity. Moreover, we discuss remaining outstanding questions about how PM H+-ATPases contribute to abiotic stress responses.

Abstract:The plasma membrane H(+)-ATPase is the pump that provides the driving force for transport of numerous solutes in plant cells, and plays an essential role for the growth and maintenance of cell homeostasis. Recent investigations using guard cells with respect to blue-light-induced stomatal opening uncovered the regulatory mechanisms of the H(+)-ATPase, and revealed that the phosphorylation status of penultimate threonine in the C-terminus of H(+)-ATPase is key step for the activity regulation. The same regulatory mechanisms for the H(+)-ATPase were evidenced in hypocotyl elongation in response to ABA and auxin, suggesting that the phosphorylation of the penultimate threonine is a common regulatory mechanism for the H(+)-ATPase. We also present the data that the activity of the H(+)-ATPase limits the plant growth. Typical structure of the H(+)-ATPase in the C-terminus was acquired in the transition of plants from water to the terrestrial land.

Abstract:Plasma membrane H+-ATPases (PMAs) pump H+ out of the cytoplasm by consuming ATP to generate a membrane potential and proton motive force for the transmembrane transport of nutrients into and out of plant cells. PMAs are involved in nutrient acquisition by regulating root growth, nutrient uptake, and translocation, as well as the establishment of symbiosis with arbuscular mycorrhizas. Under nutrient stresses, PMAs are activated to pump more H+ and promote organic anion excretion, thus improving nutrient availability in the rhizosphere. Herein we review recent progress in the physiological functions and the underlying molecular mechanisms of PMAs in the efficient acquisition and utilization of various nutrients in plants. We also discuss perspectives for the application of PMAs in improving crop production and quality.

Abstract:Vacuolar-type adenosine triphosphatase (V-ATPase, VHA) is a highly conserved, ATP-driven multisubunit proton pump that is widely distributed in all eukaryotic cells. V-ATPase consists of two domains formed by at least 13 different subunits, the membrane peripheral V1 domain responsible for ATP hydrolysis, and the membrane-integral V0 domain responsible for proton translocation. V-ATPase plays an essential role in energizing secondary active transport and is indispensable to plants. In addition to multiple stress responses, plant V-ATPase is also implicated in physiological processes such as growth, development, and morphogenesis. Based on the identification of distinct V-ATPase mutants and advances in luminal pH measurements in vivo, it has been revealed that this holoenzyme complex plays a pivotal role in pH homeostasis of the plant endomembrane system and endocytic and secretory trafficking. Here, we review recent progress in comprehending the biochemical properties and physiological functions of plant V-ATPase and explore the topics that require further elucidation.

Abstract:Increase of crop yield is always the desired goal, manipulation of genes in relation to plant growth is a shortcut to promote crop yield. The plasma membrane (PM) H+-ATPase is the plant master enzyme; the energy yielded by ATP hydrolysis pumps H+ out of cells, establishes the membrane potential, maintains pH homeostasis and provides the proton-motive force required for transmembrane transport of many materials. PM H+-ATPase is involved in root nutrient uptake, epidermal stomatal opening, phloem sucrose loading and unloading, and hypocotyl cell elongation. In this review, we summarize the recent progresses in roles of PM H+-ATPase in nutrient uptake and light-induced stomatal opening and discuss the pivotal role of PM H+-ATPase in crop yield improvement and its potential application in agricultural production by modulating the expression of PM H+-ATPase in crops.

Abstract:In plants,plasmalemma H~+-ATPase,participating in substance trans-membrane transport and elongation growth of the cells,the opening and closedown of the stomata and the responses to environmental stress,is a master enzyme.The regulation of its activity involves many factors such as hormones and environmental factors and can occur on many levels such as transcription,translation and enzyme molecule.Thus,the regulation of plasmalemma H~+-ATPase activity plays an important role in physiological activities in plant growth and development.The paper summarizes the research advances about structural characters,physiological functions activity changes and regulation of plasmalemma H~+-ATPase so as to further clarify the relationship of its physiological functions and regulation mechanism with the life activities of plants.

Abstract:Adenosine 5'-triphosphate (ATP) serves as a universal energy source for various life activities in all organisms.Previous research has shown that ATP,the small molecule synthesized in organelles such as mitochondria and chloroplast of plant cells,can be transported to the outside of the cell.Extracellular ATP (eATP) is an important signaling molecule used by plants and functions in a various of physiological processes including growth,development and stress responses in plants.Recently,the first ATP receptor of plants was identified,which is a milestone for our overall understanding of the signal transduction pathway regulated by extracellular ATP.In this review,we summarize past researches on the production and roles of eATP in plant growth,development and stress responses,discuss the discovery,releases,recognization,regulation and signal transduction,and assay methods of eATP and propose several research directions which deserve future investigation.
Advances in Plant PHT1 Phosphate Transporter Family Research

Abstract:[Objectives] Phosphorus (P) is one of the essential macronutrients for plants.Plant PHT1 family proteins have vital roles in plant P absorption,mobilization and re-utilization.Numerous members of the PHT1 family have been cloned from various higher plant species.In this paper,researches in plant PHT1 phosphate transporter family were summarized,including gene expression profiles,gene function and possible regulation mechanism.[Major advances] Plant PHT1 family belongs to a major facilitator superfamily (MFS).The protein structure of PHT 1 genes is extraordinarily conserved,sharing a common topology with 12 membranespanning domains,which are separated into two groups of six domains by a charged hydrophilic loop,with the protein inserted in the plasma membrane.Meanwhile,PHT1 family genes have the same characterized domains as in Pi:H+ co-transporters,sugar transporters,and MFS transporters as well as a conserved amino acid sequence (GGDYPLSATIMSE).Generally,two to four protons enter the plasma membrane simultaneously while plants uptake one phosphate ion via PHT1 family genes,with changes in membrane potential.There are massive members of PHT1 family in plants which are rather different from each other in transport characteristics.The variation of PHT 1 gene sequences can give rise to significant changes of their kinetic parameters of Km value.Until now,a huge number of PHT1 family members have been identified in higher plants such as arabidopsis,rice,soybean,solanaceae etc.The expression profiles of PHT1 family genes vary largely.It is generally recognized that most of the PHT1 family members can be regulated by the signal of low P stress and express in plant roots,while the expression of part of the family members can be detected in other organs of plants,and these genes function distinctly.The transcriptional levels of PHT 1 family genes can be regulated by many factors such as external inorganic P concentration.Some transcriptional factors from MYB family and WRKY family as well as ZAT6 can interact with PHT1 family genes by binding with some special cis-elements existing in the promoter region of plant PHT1 family genes,such as MYCS element,P 1BS element and W-box element,to regulate the expression levels of PHT1 family genes.Besides,the expression levels of PHT1 family genes can also be regulated by arbuscular mycorrhiza fugi (AMF).In addition to transcriptional regulation,post-transcriptional regulation and signal transduction pathway involved in the expression of PHT1 family genes are reported by different research groups.It shows that numerous genes including PHF1,genes containing SPX domain,MicroRNA,and other genes related to protein phosphorylation and chromatin modification are involved in the process of post-transcriptional regulation of PHT1 family members,as well as plant hormones such as auxin,ethylene and cytokinin etc.[Suggestions and Expectations] There is an increasingly number of reports on the mechanism of how plants response to low P stress until now,and some genes are cloned involving P absorption and utilization.The results show that complex mechanism exists in the process of P absorption and utilization as well as signal transduction pathway.Thus,it is a long way to breeding genetically modified cultivars with enhanced P efficiency.In terms of researches on PHT 1 family genes,it has changed recently from model plants to crops and other higher plants.However,further researches might be conducted on biochemistry and structural biology of proteins of PHT 1 family members.In addition,more researches about PHT1 family genes might also be performed towards polyploid species such as Brassica napus,wheat,barley,cotton etc.

Abstract:Phosphorus (P) is an essential macronutrient for plant growth and development. To adapt to low inorganic-phosphate (Pi) environments, plants have evolved complex mechanisms and pathways that regulate the acquisition and remobilization of Pi and maintain P homeostasis. These mechanisms are regulated by complex gene regulatory networks through the functions of Pi transporters (PTs) and Pi starvation-induced (PSI) genes. This review summarizes recent progress in determining the molecular regulatory mechanisms of phosphate transporters and the Pi signaling network in the dicot Arabidopsis (Arabidopsis thaliana) and the monocot rice (Oryza sativa L.). Recent advances in this field provide a reference for understanding plant Pi signaling and specific mechanisms that mediate plant adaptation to environments with limited Pi availability. We propose potential biotechnological applications of known genes to develop plant cultivars with improved Pi uptake and use efficiency.

Abstract:ATP is the primary form of energy for plants, and a shortage of cellular ATP is generally acknowledged to pose a threat to plant growth and development, stress resistance, and crop quality. The overall metabolic processes that contribute to the ATP pool, from production, dissipation, and transport to elimination, have been studied extensively. Considerable evidence has revealed that in addition to its role in energy supply, ATP also acts as a regulatory signaling molecule to activate global metabolic responses. Identification of the eATP receptor DORN1 contributed to a better understanding of how plants cope with disruption of ATP homeostasis and of the key points at which ATP signaling pathways intersect in cells or whole organisms. The functions of SnRK1a, the master regulator of the energy management network, in restoring the equilibrium of the ATP pool have been demonstrated, and the vast and complex metabolic network mediated by SnRK1a to adapt to fluctuating environments has been characterized. This paper reviews recent advances in understanding the regulatory control of the cellular ATP pool and discusses possible interactions among key regulators of ATP -pool homeostasis and crosstalk between iATP/eATP signaling pathways. Perception of ATP deficit and modulation of cellular ATP homeostasis mediated by SnRK1a in plants are discussed at the physiological and molecular levels. Finally, we suggest future research directions for modulation of plant cellular ATP homeostasis.

Abstract:Rice is an important staple crop in China.The yield of rice is closely related to the uptake of plant nutrients. As the major essential elements,nitrogen,phosphorus and potassium,their transport across the plasma membrane is critical for the nutrient absorption efficiency.The plasma membrane H +-ATPase actively drives H + outside the plant cells.Thus the proton gradient across the plasma membrane not only builds up the membrane potential but also forms the proton motive force for the transport of various nutrients.This review illustrates the mechanism of plasma mem-brane H +-ATPase of rice roots involved in the uptake of nitrogen,phosphate and potassium to provide more strategies for improving the nutrition use efficiency.
Genome-Wide Identification, Characterization, and Expression Analyses of P-Type ATPase Superfamily Genes in Soybean

Abstract:P-type ATPases are transmembrane pumps of cations and phospholipids. They are energized by hydrolysis of ATP and play important roles in a wide range of fundamental cellular and physiological processes during plant growth and development. However, the P-type ATPase superfamily genes have not been characterized in soybean. Here, we performed genome-wide bioinformatic and expression analyses of the P-type ATPase superfamily genes in order to explore the potential functions of P-type ATPases in soybean. A total of 105 putative P-type ATPase genes were identified in the soybean genome. Phylogenetic relationship analysis of the P-type ATPase genes indicated that they can be divided into five subfamilies including P1B, P2A/B, P3A, P4 and P5. Proteins belonging to the same subfamily shared conserved domains. Forty-seven gene pairs were related to segmental duplication, which contributed to the expansion of the P-type ATPase genes during the evolution of soybean. Most of the P-type ATPase genes contained hormonal- and/or stress-related cis-elements in their promoter regions. Expression analysis by retrieving RNA-sequencing datasets suggested that almost all of the P-type ATPase genes could be detected in soybean tissues, and some genes showed tissue-specific expression patterns. Nearly half of the P-type ATPase genes were found to be significantly induced or repressed under stresses like salt, drought, cold, flooding, and/or phosphate starvation. Four genes were significantly affected by rhizobia inoculation in root hairs. The induction of two P2B-ATPase genes, GmACA1 and GmACA2, by phosphate starvation was confirmed by quantitative RT-PCR. This study provides information for understanding the evolution and biological functions of the P-type ATPase superfamily genes in soybean.

Abstract:Phosphate is one of the indispensable elements for plant growth and development. The uptake of phosphate from soils and translocation between organs and subcellular compartments require a series of phos-phate transporters with phosphate transport activity. In recent years, with the rapid development of molecular biology and functional genomics technology, researchers have made great progress in clarifying the function of phosphate transporters and its molecular regulation mechanism. In this paper, focused on Arabidopsis and rice, we reviewed the recent progress on the discovery of phosphate transporters, their functions and molecular regu-lation mechanisms. It would provide reference for improving phosphorus absorption and utilization efficiency in crop breeding.

Abstract:Cytosolic pH homeostasis is a precondition for the normal growth and stress responses in plants, and H+ flux across the plasma membrane is essential for cytoplasmic pH control. Hence, this review focuses on seven types of proteins that possess direct H+ transport activity, namely, H+-ATPase, NHX, CHX, AMT, NRT, PHT, and KT/HAK/KUP, to summarize their plasma-membrane-located family members, the effect of corresponding gene knockout and/or overexpression on cytosolic pH, the H+ transport pathway, and their functional regulation by the extracellular/cytosolic pH. In general, H+-ATPases mediate H+ extrusion, whereas most members of other six proteins mediate H+ influx, thus contributing to cytosolic pH homeostasis by directly modulating H+ flux across the plasma membrane. The fact that some AMTs/NRTs mediate H+-coupled substrate influx, whereas other intra-family members facilitate H+-uncoupled substrate transport, demonstrates that not all plasma membrane transporters possess H+-coupled substrate transport mechanisms, and using the transport mechanism of a protein to represent the case of the entire family is not suitable. The transport activity of these proteins is regulated by extracellular and/or cytosolic pH, with different structural bases for H+ transfer among these seven types of proteins. Notably, intra-family members possess distinct pH regulatory characterization and underlying residues for H+ transfer. This review is anticipated to facilitate the understanding of the molecular basis for cytosolic pH homeostasis. Despite this progress, the strategy of their cooperation for cytosolic pH homeostasis needs further investigation.

Abstract:Nitrogen, phosphorous and potassium are essential nutrients for plant growth and development. However, their contents in soils are limited so that crop production needs to invest a lot for fertilizer supply. To explore the genetic potentialities of crops (or plants) for their nutrient utilization efficiency has been an important research task for many years. In fact, a number of evidences have revealed that plants, during their evolution, have developed many morphological, physiological, biochemical and molecular adaptation mechanisms for acquiring nitrate, phosphate and potassium under stress conditions. Recent discoveries of many transporters and channels for nitrate, phosphate and potassium uptake have opened up opportunities to study the molecular regulatory mechanisms for acquisition of these nutrients. This review aims to briefly discuss the genes and gene families for these transporters and channels. In addition, the functions and regulation of some important transporters and channels are particularly emphasized.

Abstract:Abstract Phosphorus (P) is an essential macronutrient for plant growth and development. Low inorganic phosphate (Pi) availability is a limiting factor for plant growth and yield. To cope with a complex and changing environment, plants have evolved elaborate mechanisms for regulating Pi uptake and use. Recently, the molecular mechanisms of plant Pi signaling have become clearer. Plants absorb Pi from the soil through their roots and transfer Pi to various organs or tissues through phosphate transporters, which are precisely controlled at the transcript and protein levels. Here, we summarize recent progress on the molecular regulatory mechanism of phosphate transporters in Arabidopsis and rice, including the characterization of functional transporters, regulation of transcript levels, protein localization and turnover of phosphate transporters. A more in-depth understanding of plant adaptation to a changing Pi environment will facilitate the genetic improvement of plant P efficiency.

Abstract:Plasma membrane proton-ATPase (PM H+-ATPase) is a primary H+ transporter that consumes ATP in vivo and is a limiting factor in the blue light-induced stomatal opening signaling pathway. It was recently reported that manipulation of PM H+-ATPase in stomatal guard cells and other tissues greatly improved leaf photosynthesis and plant growth. In this report, we review and discuss the function of PM H+-ATPase in the context of the promotion and upregulation H+-ATPase strategy, including associated principles pertaining to enhanced stomatal opening, environmental plasticity, and potential applications in crops and nanotechnology. We highlight the great potential of the promotion and upregulation H+-ATPase strategy, and explain why it may be applied in many crops in the future.
Discrete subcellular localization of membrane-bound ATPase activity in marine angiosperms and marine algae

Abstract:The subcellular distribution of membrane-bound ATPases was compared among terrestrial plants, seagrasses and marine algae by cytochemical techniques. High ATPase activity was detected in the copiously invaginated plsma membrane that was characteristic of transfer cells but not in the tonoplast of epidermal cells in mature leaves of seagrasses. Magnesium- or Ca(2+)-dependent ATPase activity was induced together with the characteristics of transfer cells during the development of leaf tissues able to resist seawater. Northern hybridization revealed the effective induction of the synthesis of mRNA for plasma-membrane H(+)-ATPase during the development of leaves. Such high ATPase activity was not detected in the smooth plasma membranes of marine macro-algae but was found in the membranes of some cytoplasmic vesicles or microvacuoles, providing evidence of the excretion of salts by exocytosis. It appears, therefore, that two essentially different methods for excreting excess salts have developed separately in these two classes of marine plants. The evolution of mechanisms of salt tolerance in the plant kingdom is discussed in terms of the differential subcellular distribution of ATPase activity.
