Abstract:Nucleotide-Binding Site and Leucine-rich Repeat Region (NBS-LRR) are the largest class of disease resistance genes cloned from plants up to date. In this paper, NBS-type resistance genes were isolated and cloned from a high resistance to downy mildew introgression line from C. hystrir ×C. sativus (IL5211S) using a PCR method with degenerate primers designed from conserved regions of the NBS domain. A total of twenty eight sequences were identified in IL5211S and named as cucumber resistance gene analogues (CSRGAs). Polygenetic analysis classified these sequences into four subclasses (CSRGA I-IV). Isolation of these CSRGAs may prove to be useful for developing RGA markers and map-based cloning of R-genes in IL5211S.
