Abstract:OBJECTIVE To explore the effects of rosiglitazone on serum levels of lipids and inflammatory factors in atherosclerotic rats.METHODS 55 male Wistar rats were randomly divided into 3 groups,the normal group(N,n=20) with ordinary diet for 12 weeks,the model group(M,n=20) and the treatment group(RSG,n=15) with intra-abdominal injection of a single dose of vitamin D3(400,000 IU·kg-1) and atherogenic diet for 6 weeks,then be administered with normal saline(10 mg·kg-1·d-1) and rosiglitazone(3 mg·kg-1·d-1),respectively for 6 weeks.Before and after treatment the serum levels of total cholesterol(TC),triglyceride(TG),low density lipoprotein-cholesterol(LDL-C),high density lipoprotein-cholesterol(HDL-C) and pathomorphology in heart tissue were detected.The levels of serum C-reactive protein(CRP),monocyte chemoattractant protein-1(MCP-1) and intercellular adhesion molecule-1(ICAM-1) were determined by ELISA. RESULTS 6 weeks later the model group rats showed high serum levels of TC,TG,LDL-C,CRP,MCP-1,ICAM-1 and low serum level of HDL-C than that of the normal group(P0.05).Coronary atherosclerotic plaques had been observed.After treatment for 6 weeks,compared with the model group rats,rosiglitazone could significantly reduce the levels of serum TC,TG,LDL-C,CRP and MCP-1(P0.05 or P0.01),which were still higher than that of the normal group(P0.05).However,the changes of the serum HDL-C and ICAM-1 levels has no difference(P0.05).The pathomorphology degree of coronary atherosclerosis has decreased significantly.CONCLUSION Rosiglitazone can reduce the serum levels of TC,TG,LDL-C and inhibit the production of CRP and MCP-1,which may account for the anti-arterioisclerosis effects.

Abstract:Objective The study was to observed the effects of rosiglitazone plus metformin on serum high C reaction protein and IL-6 and the effect of insulin resistance in type 2 diabetes mellitus.Methods It takes a comparative study involving 48 cases who are suitable for this study.Patients accepted the rosiglitazone added metformin over a 12-week treatment.Fasting plasma glucose(FPG),serum insulin(FINS),glycosylated hemoglobin(HbA1c),postprandial 2 h(2hPPG) blood glucose,and insulin(2hpINS),C reaction protein and IL-6 were determined at the first and last day of this study.HOMA2model formula evaluation was used in testing insulin sensitivity.Results After a 12-weeks' treatment,FPG,HbA1c and C reaction protein of patients were lower than those before at baseline respectively(P < 0.05).IL-6 were without significant statistical differences.Insulin resistance index(HOMA2IR) was lower after treatment(P < 0.05).Other the result of metabolic parameter has no statistical differences.Conclusion Rosiglitazone added biguanides combined treatment of type 2 diabetes are effective not only in reducing in blood glucose levels and improving insulin sensitivity,But also decreasing serum hCRP,IL-6 no has significant statistical differences.
Efficacy and safety of rosiglitazone hydrochloride in the treatment of patients with type 2 diabetes mellitus

Abstract:Objective: To evaluate the clinical efficacy and safety of rosiglitazone hydrochloride in the treatment of patients with type 2 diabetes mellitus. Methods: One hundred and thirty two patients with type 2 diabetes mellitus who met 1999 WHO diagnostic criteria of diabetes mellitus were assigned to receive rosiglitazone hydrochloride (4 mg/d) or rosiglitazone maleate (4 mg/d) in a randomized, double blind, double dummy, multicentre clinical trial. The trial consisted of a 2-week washing period and a 16-week treatment period. The levels of fasting and postprandial plasma glucose,glycosylated hemoglobin,fasting insulin and C peptide of the patients in two groups were determined before and after the treatment. Results: One hundred and twenty-six patients completed the trial (64 patients in control group and 62 patients in treatment group). After the treatment, the levels of fasting and postprandial plasma glucose,glycosylated hemoglobin,fasting insulin were decreased significantly compared with those of the baseline (P<0.01). However, the differences were not statistically significant between the two groups (P>0.05). The levels of C peptide in two groups were also decreased after 16-week treatment,but the differences were not significant compared with the baseline. The results of blood routine and biochemical examinations (including liver function) and electrocardiogram did not change significantly. Conclusion: Once-daily regimen with rosiglitazone hydrochloride provides effective control of plasma glucose levels for the patients with type 2 diabetes mellitus. Compared with rosiglitazone maleate, rosiglitazone hydrochloride is equally safe and effective.
The Effect and Their Significance of Rosiglitazone on Serum CRP and Fg in Subjects with Impaired Glucose Tolerance

Abstract:Objective To investigate the changes of plasma C-reaction protein(CRP),fibrinogen(Fg) and their significance in subjects with impaired glucose tolerance after the treatment with rosiglitazone.Methods 60 subjects with IGT were divided into two groups:the control group(diet and excise) and treatment group(diet and excise with rosiglitazone 4mg daily).before and after treating for 12 weeks,plasma hsCRP,Fg,blood lipid and fasting insulin were measured,height,weight,waist and hip circumference and blood pressure also were recorded,then Homa-IR,BMI and WHR were calculated.Results(1)Rosiglitazone resulted in a significant reduction in CRP,Fg and HOMA-IR SBP,DBP,TG and LDL-C after the 12 weeks(P0.05);In the rosiglitazone-treated group,reductions in hsCRP and Fg were significantly positive relationship with the changes of Homa-IR and serum fasting insulin.Conclusion Rosiglitazone significantly reduces the level of plasma CRP and Fg and improve insulin resistance.
Effects of rosiglitazone on serum retinol-binding protein 4 and hepatic fat contents in patients with type 2 diabetes mellitus

Abstract:Objective To investigate the changes of serum retinol-binding protein 4,hepatic fat content,carbohydrate and lipid metabolism in patients with type 2 diabetes mellitus before and after the treatment with rosiglitazone.Methods Seventeen subjects newly-diagnosed with type 2 diabetes mellitus,aged between 35 to 69 years,were treated with rosiglitazone(4-8 mg/d)for 24 weeks.The wight,waist and hip circumference were measured before and after the rosiglitazone treatment.Fasting plasma glucose(FPG),2 hours plasma glucose(2hPG),HbA1c,lipid,retinol-binding protein 4(RBP4)and adiponectin(APN)were determined at the same time.Hepatic fat content was measured by ultrasound histogram.B-cell function was evaluated by arginine stimulating test,insulin sensitivity was estimated by homeostasis model assessment(HOMA).The trial adopted a self-controlling methods.Results Compared with baseline levels,the levels of FPG,2hPG,HbA1c and RBP4 were significantly decreased,and APN level was increased during rosiglitazone treatment(all P<0.01).After treatment,the attenuation coefficients of liver adipose content in the longitudinal section under the xiphoid of left hepatic lobe,the section between the ribs of right hepatic lobe,oblique section under the ribs of right hepatic lobe,and its mean value decreased by 19.5%,26.7%,22.0% and 22.9%,respectively.Conclusion Rosiglitazone can decrease serum RBP4 level,reduce the hepatic fat content in patients with type 2 diabetes,and improve insulin resistance.(Shanghai Med J,2008,3:767-770)
Effect of rosiglitazone on the expression of platelet CD40 ligand in insulin-resistant rats

Abstract:Objective To investigate the effect of rosiglitazone on the expression of platelet CD40 ligand (CD40L) in insulin-resistant rats, and to further determine the relationship between CD40L and insulin resistance. Methods 60 healthy male SD rats [(200±20)g] were randomly divided into 4 groups: control group (C), high fat group (HF), low dose rosiglitazone group (LR) and high dose rosiglitazone group (HR). Rats in group C were fed normal chow diet, and the others were given high fat chow diet. After 12 weeks, high dose of rosiglitazone (10mg/kg) was given to rats in group HR and low dose of rosiglitazone (5 mg/kg) was given to rats in group LR for 4 weeks. Rats in group HF and group C were given 0.9% sodium chloride solution. The level of sCD40L was measured by ELISA and the expression of platelet membrane CD40L was detected by immunoprecipitation and Western blot. The insulin resistance (IR) index was calculated by homeostasis model assessment (HOMA). Results HOMA-IR, sCD40L level and platelet membrane CD40L expression were higer in group HF than in group C (9.8±3.2 vs. 5.9±1.7, 367.3 ±35.3 vs. 232.3±120.6, 2.1±0.4 vs. 1.4±0.2, respectively, all P<0.05). Compared with the group HF, HOMA-IR, sCD40L level and platelet membrane CD40L expression were obviously decreased in group HR(5.4±1.1, 276.9±54.0, 1.4±0.3, respectively, all P<0.05), and there were no significant differences in HOMA-IR, sCD40L level and platelet membrane CD40L expression between group HF and group LR (P>0.05). Conclusions In insulin-resistant rats, the level of sCD40L and the expression of platelet membrane CD40L were higher. After treatment with high dose of rosiglitazone, sCD40L level and platelet membrane CD40L expression were decreased with the improvement of insulin resistance.

Abstract:OBJECTIVE:To observe the effect of high-fat diet and rosiglitazone intervention on the function of pancreatic alpha cell of SD rats.METHODS:36 normal male SD rats, 8-week old, were randomly divided into 3 groups i.e., a normal chow group (CC, n = 12), an isocaloric high-fat diet group (CF, n = 12), and a rosiglitazone-treated group (Ro, n = 12, rosiglitazone 3 mg.kg(-1).d(-1) and isocaloric high fat diet). Triglyceride (TG) was measured every 4 weeks after feeding for 6 weeks. After 28 weeks, the secretion of insulin and glucagon (Gg) was assessed with intravenous glucose tolerance test (IVGTT) at 0, 3, 5, and 10 minutes. (3)H-2-deoxyglucose ((3)H-2-DG) uptake by tissues was measured to evaluate the insulin sensitivity.RESULTS:The ratio of intra-abdominal fat mass and body weight was higher in the rats of CF and Ro group than that in the rats of CC group. At the first 10 min of IVGTT, the Gg level was higher in the CF group than that in CC group [(119.3 +/- 12.4, 82.3 +/- 6.4, 72.2 +/- 5.8, 68.2 +/- 9.1) ng/L vs (96.8 +/- 9.1, 67.6 +/- 5.9, 57.9 +/- 5.3, 55.3 +/- 6.9) ng/L, P < 0.05] and Ro group [(78.4 +/- 6.0, 59.4 +/- 4.0, 49.9 +/- 6.2, 40.9 +/- 6.0) ng/L, P < 0.01], the level was even lower in the latter group than in CC group (P < 0.01). There was no difference of insulin level among the 3 groups. By using quantitative image analysis, the integrated A (area x A) of alpha cells was significantly higher in the CF group and Ro group as compared with that in the CC group (1661 +/- 130 and 1532 +/- 132 vs 1188 +/- 104, P < 0.05). In contrast, there was no difference among the 3 groups in the integrated A of beta cells.CONCLUSIONS:High-fat feeding induces insulin resistance in rats, which is associated with pancreatic alpha cell proliferation and abnormal Gg secretion.

Abstract:Objective To observe the relationship between the expressions of genes that involve in insulin signal system and tissue glucose metabolism in high fat diet rats and the effect of rosiglitazone intervention. Methods SD rats were randomly divided into three groups: normal chow feeding (NC), high fat diet (HF) and rosiglitazone treatment (RSG).Glucose utilization and uptake were measured by glucose area under curve(GluAUC) of IVGTT and the rate of tissue uptake of 3H-2-deoxyglucose.Gene expression was investigated by using RT-PCR method. Results Compared with NC group, HF group showed the 24.1% increase in GluAUC, the decreases in the uptake of 3H-2-deoxyglucose in skeletal muscle by 36.7% and in adipose tissue by 48.1% and the decreases in IRS1 mRNA of skeletal muscle by 64.8% and in adipose tissue by 45.7%. Compared with HF group, rosiglitazone treatment had the 12.9% decrease in GluAUC and the increases in skeletal muscle and adipose tissue uptake of 3H-2-deoxyglucose by 66.3% and 66.7%,respectively. Conclusions The decreases in tissue glucose uptake and utilization are related to the lowered level of IRS1 mRNA of high fat diet rats.Rosiglitazone treatment enhances the ability of glucose uptake and utilization.

Abstract:After treatment for 3 months in the patients with impaired glucose tolerance (IGT), plasma glucose, insulin and homeostasis model assessment for insulin resistance (HOMA-IR) were decreased both in rosiglitazone therapy group and in the group treated with rosiglitazone combined with body weight control.Those parameters in the latter group were decreased more significantly than those in the former.Rosiglitazone ameliorates the insulin resistance in patients with IGT, which is further improved when combined with body weight control.

Abstract:Objective To investigate the effects of Rosiglitazone(ROS)on the levels of serum adiponectin,high sensitive C-reactive protein(hsC-RP),tumor necrosis factor-α(TNF-α),interleukin-6(IL-6)in patients with impaired glucose tolerance(IGT)and metabolic syndrome(MS).Methods 53 outpatients with IGT(7.8 mmol/L≤2-hour postpradial plasma glucose(PPG)<11.1 mmol/L)and MS,which came from department of endocrinology,Nanjing Drum Tower Hospital from Oct.2004 to Jan.2005,were treated with ROS or remedial life-style intervention randomly,and the levels of serum adiponectin,hsC-RP,TNF-α,IL-6 were measured both before and after 12 weeks'treatment.Results ROS 4mg daily improved HbA1c and lipid,significantly increased the levels of adiponectin,and decreased the levels of hsC-RP,TNF-αand IL-6(P<0.01).Whereas no significant improvement was found in the control group(P>0.05).Conclusion (1)ROS can improve insulin sensitivity of the patients with MS.(2)ROS can increase the levels of adiponectin,decrease the serum levels of hsC-RP,TNF-α,IL-6,and has anti-inflammatory effect.

Abstract:To observe the effects of rosiglitazone and metformin in rats on insulin resistance induced by high-fat diet.Normal 8-week old male SD rats were divided into four groups. They were normal chow group (NC, n = 11), high-fat diet (HF, n = 11), metformin-treated (HF + Met, n = 11) and rosiglitazone-treated group (HF + Ros, n = 11). Rosiglitazone 3 mg x kg(-1) x d(-1) and metformin 300 mg x kg(-1) x d(-1) were given orally to HF + Ros and HF + Met group, respectively. After feeding for 8 weeks, serum insulin, adiponectin, glucose (BG), triglyceride (TG) and free fatty acid (FFA) were measured in all the rats. Insulin sensitivity was measured with glucose infusion rate (GIR) and determined by using euglycemic-hyperinsulinemic clamp method.High-fat diet induced obesity in SD rats after feeding for 8 weeks. High-fat diet decreased adiponectin level by 43.7% (P < 0.01) and GIR by 51.3% (P < 0.01) as compared with the NC group. Metformin decreased body weight by 8.4% (P < 0.01) and TG level by 40.5% (P < 0.01). Metformin significantly increased GIR by 58.9% (P < 0.01) when compared with the HF group. Rosiglitazone caused an apparent reduction of FFA (-25.3%, P < 0.05) and TG level (-54.0%, P < 0.01). At the same time, rosiglitazone increased adiponectin by 60% (P < 0.01), and improved insulin sensitivity by 149.6% (P < 0.01) as compared with the HF group.(1) High-fat diet induces insulin resistance in SD rats; this was associated with an increase in visceral fat and a decrease in the level of adiponectin; (2) Metformin treatment improved insulin sensitivity accompanied by a decrease in body weight and TG level; (3) Rosiglitazone treatment ameliorates IR in a greater extent and is accompanied by a reduction of FFA, TG and an increase of adiponectin levels.
Effects of Rosiglitazone on Glycemia and Blood Pressure in Type 2 Diabetic Patients

Abstract:Objective To investigate the effects of tartrate and maleate rosiglitazone on lowering the plasma glucose level and changes of blood pressure in type 2 diabetics with or without hypertension.Methods 222 patients with type 2 diabetes were randomly assigned to groups of receiving maleate and tartrate rosiglitazone(4~8 mg/d-1)for 12 weeks.Blood pressure was measured at the 4th,8th and 12th week.Fasting and 2 hour postprandial plasma glucose levels,and insulin levels at baseline and 12th week were also determined.The effects of these two agents on lowering the plasma glucose level and improving insulin sensitivity were compared,at the same time blood pressure reduction was analyzed in the hypertension group(n=60) and normotension group(n=162).Results(1) The fasting and 2 hour postprandial plasma glucose,HbA_1c,and insulin levels at baseline were comparable between two groups(P0.05).All these parameters reduced after treatment with rosiglitazone for 12 weeks,no significantly difference was found between groups(P0.05).(2)Blood pressure reduced gradually after treatment with rosiglitazone in the hypertension group.The mean systolic blood pressure reduced by(7.26±1.73)mmHg after 4 weeks,(9.37±1.95)mmHg after 8 weeks,and(10.6±1.86)mmHg after 12 weeks.The mean diastoLic blood pressure reduced by(4.35±1.01) mmHg after 4 weeks,(5.42±1.07) mmHg after 8 weeks,and(6.4±1.04)mmHg after 12 weeks.Blood pressure did not change in the normotension group,with the mean of systolic and diastolic blood pressure reduced only by(1.79±1.08)mmHg and(1.34±0.64)mmHg,respectively.Reduction of blood pressure was significantly difference between groups of normotension and hypertension(P0.01).(3) The reduction of blood pressure was related to the level of blood pressure at baseline,and the reduction of 2 hour postprandial insulin levels.Conclusions Rosiglitazone decreases blood glucose and improves insulin resistance in type 2 diabetes.It also reduces blood pressure at some extent in diabetics with hypertension.
Comparison of Clinical Effects Between Rosiglitazone and Pioglitazone in the Treatment of Type 2 Diabetes of Eldly Patients

Abstract:AIM To compare the clinical effects of rosiglitatine with pioglitazone in the treatment of type 2 diabetic patients.METHODS Sixty-four patients who had been treated with food control,exercise and fixed oral anti-dia-betic drugs for one month,but their fasting plasma glucose level was still over 7 rranol·L-1.They were randomized divided into two groups.One was added with rosiglitazone,the other was added with pioglitazone.All of them were treated for 12 weeks.RESULTS After treatment for 12 weeks,①patient's fasting plasma glucose,postprandial plasma glucose,HbA1c and insulin resistance index were significant decreased in both groups;②triglyceride and total cholesterol were decreased in pioglitazone group(P＜0.05),but those in rosiglitagone group were no significant reduction(P＞0.05);③high density lipoprotein-cholesterol was elevated in both groups(P＜0.05).CONCLUSION Rosiglitazone or pioglita zone can be used to control blood glucose,but pioglitazone shows more efficiency on triglyceride control.

Abstract:Objective To investigate the effect of rosiglitazone on non-alcoholic fatty liver (NA and to explore the relationship between insulin resistance and NAFL. Methods Animal model of NAFL was established by feeding Sprague-Dawley rats a high-fat diet for 8 weeks. The model rats were then randomized into rosiglitazone-treated and untreated groups. The animals were sacrificed after being treated with rosiglitazone or vehicle for 4 weeks. The histological changes of liver were examined, and liver function, fasting plasma glucose, fasting serum insulin, serum lipid profile, leptin, and adiponectin were measured. Results As compared with untreated group, hepatic steatosis and liver function were significantly improved in rosiglitazone-treated group. Alanine aminotransferase (ALT) was 54±19 U/L vs 101±24 U/L, aspartic acid aminotransferase (AST) 151±37 U/L vs 198±48 U/L, and alkaline phosphatase (ALP) 87±16 U/L vs 115±39 U/L, respectively (P<0.01). Serum adiponectin level was higher, and serum leptin level and insulin resistance index (HOMA-IR) were lower in rosiglitazone-treated group than in untreated group. HOMA-IR was 6.9±1.8 vs 12.0±1.2 (P<0.01). Serum triglyceride, total cholesterol and low density lipoprotein-cholesterol level were significantly decreased in rosiglitazone-treated group as compared with untreated group (P<0.01). Conclusions Insulin resistance might play important role in the pathogenesis of NAFL. Rosiglitazone effectively reverses NAFL in animal model.
Combined Treatment of Rosiglitazone and Metformin for Blood Glucose, Blood Lipid, Blood Pressure and Body Mass of Type 2 Diabetic Patients Complicated by Blood Lipid Disturbance

Abstract:BACKGROUND: Insulin resistance (IR) is associated with type 2 diabetes mellitus and its macrovascular complications. Euglycemic agent (rosiglitazone and metformin) can ameliorate IR, but its influences on other cardiovascular risk factors, including blood lipid, blood pressure and body mass need to be further studied.OBJECTIVE: To observe the effects of rosiglitazone and metformin on the blood glucose, blood lipid, blood pressure and body mass of patients with type 2 diabetes mellitus complicated by blood lipid disturbance.DESIGN: A clinical observation.SETTING: Department of Endocrinology, Nanjing First Hospital, Nanjing Medical University.PARTICIPANTS: Forty-two patients with type 2 diabetes mellitus complicated by dyslipideamia, who received treatment in the Department of Endocrinology, Nanjing First Hospital, Nanjing Medical University between June and September 2006 were recruited in this study. The involved patients, 27 male and 15 female, averaged (58±2)years ranging from 30 to 70 years old. Inclusive criteria:①Type 2 diabetes mellitus all met the diagnostic criteria of American Diabetes Association (2006).② Diagnostic criteria of blood lipid disturbance : total cholesterol(TC)＞ 4.68 mmol/L, triglyceride (TG) ＞ 1.7 mmol/L, low density lipoprotein cholesterol (LDL-C) ＞ 2.68 mmol/L, high density lipoprotein cholesterol (HDL-C) ＜ 0.9 mmol/L (female), ＜ 1.0 mmol/L (male). Informed consents were obtained from all the patients.METHODS：①Drug treatment：Dudng the 12-week treatment,all the patients received 0．75 g metformin(Lot No．051110 and 051214,0.25 g，lablel,Jiangsu Hengrui Medicine Co.，Ltd)and 4 mg rosiglitazone (Lot No．0511101,2 mg／tablet，Jiangsu Hengrui Medicine Co.，Ltd) daily．②Physical and laboratory measurements：Body mass index，blood pressure were taken，and blood lipid profile(TC,TG,C-LDL and C-HDL)，fasting and postprandial blood glucose were measured by colodmetdc method before,8 and 12 weeks after treatment．MAIN OUTCOME MEASURES：①Fasting and postprandial blood glucose level change．②Blood lipid，blood pressure and body mass index change．RESULTS：Forty-two patients with type 2 diabetes mellitus complicated by dyslipideamia were recruited，and three dropped out the experiment，one patient suffered from severe watery stool, one patient from severe abdominal distention,one patient from urinary tract infection, and the other thirty-nine patients completed the observation in the final analysis．Compared with pretraatment data,fasting and postprandial blood glucose levels 12 weeks after treatment were decreased(P＜0．01),TC increased(P＜0．05)，LDL-C decreased(P＜0．01)，HDL-C increased(P＜0．01),blood pressure did not change(P＞0．05) and body mass index increased(P＜0．01)．CONCLUSION： Combinatin of rosiglitazone and metformin significantly decreases blood glucose，decreases LDL-C,increases TC，HDL-C,body mass index,and has no obvious effects on TG and blood pressure．It is safe and bearable for type 2 diabetes mellitus patients with dyslipideamia．
Effects of Rosiglitazone on Metabolic Parameters and Large Artery Sclerosis in Nondiabetic Patients with Metabolic Sydrome

Abstract:Objective To observe the effects of rosiglitazone on metabolic parameters,carotid intimamedia thickness( IMT) ,brachial-ankle pulse wave velocity( baPWV) and ankle-brachial index( ABI) in nondiabetic patients with metabolic syndrome. Methods Seventy-nine nondiabetic patients with metabolic syndrome were randomly divided into treatment group ( n = 41) and control group ( n = 38) . The patients in treatment group were treated with rosiglitazone on the basis of life-style intervention,those in control group were treated with life-style intervention for 9 months. All patients were followed up every 3 months. Body mass index ( BMI) ,waist circumference( WC) ,systolic blood pressure( SBP) ,diastolic blood pressure( DBP) ,fasting blood glucose ( FBG) ,triglyceride( TG) ,total cholesterol( TC) ,highdendity lipoprotein ( HDL-C) ,Low-density lipoprotein ( LDL) ,high sensitivity C-reactive protein ( hsCRP) ,HbA1C,fasting insulin( FIns) ,HOMA-IR,IMT,baPWV and ABI were measured in both groups before treatment and at the 6th,9th month after treatment. Results ( 1) After treatment with rosiglitazone for 6 months,FPG,TG,HDL-C,hsC RP,HbA1C,Fins and HOMA-IR in treatment group were improved ( P <0. 01) . FPG,HDL-C,hsC RP,HbA1C,Fins and HOMA-IR got further improvement at the 9th month after treatment( P < 0. 01) . After 9 months,FPG,HDL-C,hsC-RP,HbA1C,Fins and HOMA-IR were changed significantly in treatment group compared to those in control group( P < 0. 01) . ( 2) After treatment with rosiglitazone for 6 months,baPWV and ABI in treatment group were improved ( P < 0. 01) . IMT was not changed until the 9th month after treatment. IMT,baPWV,ABI were changed significantly in treatment group compared to those in control group after 9 months ( P < 0. 01) . ( 3) The reduction of HbA1C and HOMA-IR was independently related to the improvement of ABI by multivarient analysis. Conclusion In nondiabetic patients with metabolic syndrome,rosiglitazone can significantly improve insulin resistance,correct metabolic disorders,has anti-inflammatory effect and retard atherosclerosis at some extent.

Abstract:Objective To investigate the effects of rosiglitazone on vascular lesions in rats with diabetes mellitus,and explore its possible mechanism.MethodsTwenty-four SD rats were randomly divided into normal control group,diabetes mellitus group and diabetes mellitus rosiglitazone treatment group.Rat models of type 1 diabetes mellitus were established in diabetes mellitus group and diabetes mellitus rosiglitazone treatment group by intraperitoneal injection of 60 mg/kg of streptozocin(STZ),and intragastric administration of rosiglitazone(1mg/kg) was conducted in diabetes mellitus rosiglitazone treatment group for 8 weeks from the second week after glucostasis.At the end of the eighth week,heart blood samples were collected from each group of rats,and ELISA was employed to detect the levels of phospholipase A2(PLA2).Besides,thoracoabdominal aorta were obtained,histomorphology and ultrastructure changes of vessel wall were observed,and expression of PLA2 protein of vessel wall was detected by immunohistochemistry.ResultsCompared with normal control group,the vascular structure of rats in diabetes mellitus group was damaged severely,both local PLA2 protein expression and serum PLA2 level increased.The vascular lesions in diabetes mellitus rosiglitazone treatment group were less severe than those in diabetes mellitus group.The expression of PLA2 protein of vessel wall and serum PLA2 level in diabetes mellitus rosiglitazone treatment group were significantly higher than those in control group,and significantly lower than those in diabetes mellitus group(P<0.05).ConclusionRosiglitazone has protective effects on vascular structure of rats with diabetes mellitus,and the mechanism may relate to the inhibition of PLA2 expression and reduction of inflammation.
Rosiglitazone alleviates vascular endothelial dysfunction in type 2 diabetic rats

Abstract:To observe metabolic abnormalities, histology changes and eNOS expression of aorta in type 2 diabetes rat.And to observe intervention effect of rosiglitazone.Methods 80 male Wistar rats were randomized to control group, high diet group, diabetes group, and rosiglitazone treatment group (diabetes plus rosiglitazone treatment).Type 2 diabetes models were developed and rosiglization group was treated with rosiglitazone .Six weeks and twelve weeks after treatment with rosiglitazone, blood glucose, endothlin and nitric oxide were tested.Histology changes of aorta in different groups were observed under microscopy .Meanwhile the protein and mRNA expression of eNOS in aorta were examined.Results 1)Compared with control group, ET in high fat diet group, diabetes group and rosiglitazone group increased significantly , and the level of NO decreased significantly at 6 week and 12 week.At 12 week, ET in diabetes group increased, and NO decreased significantly than that of high fat diet group and rosiglitazone group.2)Histology changes were observed in high fat diet group , diabetes group and rosiglitazone group at 12 week.3)Compared with control group, protein and mRNA expression in high fat diet group , diabetes group and rosiglitazone group were down regulated at 6 week and 12 week.And protein expression in diabetes group was down regulated than that in rosiglitazone group .Conclusions Rosiglization can ease the endothelial dysfunction in type 2 diabetes rat.

Abstract:Objective:To investigate the effects of Rosiglitazone maleate(Ros M)combined with continuous subcutaneous Insulin of infusion(CSII)on endothelium dependent vasodilation in newly-diagnosed type 2 diabetes mellitus(T2DM).Methods:106 newly diagnosed patients with T2DM were randomly divided into therapeutic group(CSII combined with Ros M,n=66)and control group(n=40,simple CSII).The level of blood glucose,HbAIc,endothelium-dependent dialatation(EDD),HOMR-IR,IAI,therapeutic time,amount of Insulin and weight were compared between them 2 weeks before and after treatment.Ros M was used in the therapeutic group for a 3-month follow-up.Results:Two weeks after the treatment,the BG,TG,HbA1c,HOMR-IR dropped significantly(P<0.05).EDD and IAI escalated in both groups(P<0.05),while IAI、EDD were obviously higher,HOMR-IR,therapeutic time,and amount of Insulin was relatively lower in therapeutic group than those in the control group.The 3-month follow-up study showed that the therapeutic group did not show any serious hypoglycaemia and impaired function of heard,liver and kidney.Conclusion:CSII therapy combined with Ros M can increase the IAI,improve endothelium dependent vasodilation,reduce T2DM patients' Insulin resistance,protect blood vessel endothelium function and is favorable for T2DM prognosis.
