Abstract:Objective. To detect the effect and cytotoxicity of luteinizing hormone-releasing hormone-Pseudomonas aeruginosa exotoxin 40 (LHRH-PE40) on target cells using LHRH receptors (LHRHR). Methods. The affinity of LHRH-PE40 and LHRH binding to LHRHR on the membrane surface of target cells were measured by enzyme linked immunosorbent assay. Morphological observations with light microscope were used to analyze its receptor pathway, with Spiegelmer, and cytotoxicity. IC50 values of LHRH-PE40, which caused 50% inhibition of tumor cell growth were evaluated by MTT assay. The target cells were exposed to LHRH-PE40 and its cytotoxicity was analyzed by scanning and transmission electron microscopies, agarose gel electrophoresis, and flow cytometry. Results. LHRH-PE40 killed target cells by LHRHR pathway. The morphological changes in these cells showed decreased cell size, cytoplasmic membrane blebbing, and chromatin condensation and margination. At a certain concentration and time point, HeLa cells were also induced to undergo programmed cell death. Conclusion. LHRH-PE40 induced target cells apoptosis via LHRHR.
Effect of Luteolin and its combination with chemotherapeutic drugs on cytotoxicity of cancer cells]

Abstract:To investigate the effect of Luteolin alone or combination with chemotherapentic drugs on the cytoxicity of cancer cells.Cultured A549, Hela, MCF-7, AGS, MGC-803, Caco2 and HepG2 cells were treated with Luteolin or the combination of Luteolin with other chemotherapeutic agents (Bexarotene, Cisplatin and Bleomycin). Cell viability was measured by MTS assay and IC(50) was calculated.The IC(50) of Bexarotene to Hela cells was 2 micromol/L, but with the combination of 5 micromol/L of Luteolin that reduced to 0.2 micromol/L. However, the combination of Bexarotene and Luteolin did not show significant benefit in MGC-803, HepG2 cells, Caco2 and MCF-7 cells. The IC(50) of Cisplatin to Hela cells was over 30 micromol/L,but it decreased to 3 micromol/L in the presence of 5 micromol/L Luteolin; Luteolin also sensitized Cisplatin in MGC-803, HepG2 and A549 cells studied. The IC(50) of Bleomycin to Hela cells was over 100 micromol/L, but it was about 1 micromol/L in the presence of 5 micromol/L Luteolin. A549 cells were resistant to Bleomycin with an IC(50) of 100 micromol/L, 10 micromol/L Luteolin greatly enhanced the cytotoxicity of Bleomycin to the cells with the IC(50) of about 10 micromol/L. The inhibitions of MGC-803, HepG2, A549 and AGS cells didn't change by combination of Luteolin.Low concentration of Luteolin has little toxic effect on the cancer cell lines tested in the study, but it can sensitize chemotherapeutic drugs in various cancer cell lines.
Experiment Study on the Competition for the Cell Receptor of Gastric Cancer and Normal Cells with Luteinizing Hormone-Releasing Hormone and Aeruginosa Exotoxin 40

Abstract:Objective To study whether luteinizing hormone-releasing hormone aeruginosa exotoxin 40 (LHRH-PE40) can be expressed in the gastric carcinoma cells or the normal cells and detect the competitive inhibition. Metheods The cellular membrane of the gastric carcinoma cell line BGC-823 and human normal kidney cell line 293 was made. Each of them was analyzed by radioactive ligand of LHRH-PE40 marked by 125I and their competitive binding with LHRH was detected. Results The binding and competition of aglucone-receptor were not found in human kindy cell line. There was binding and competition of aglucone-receptor in gastric carcinoma BGC-823 cells. The cellular affinity of BGC-823 was as follows:Kd= (37.82± 1.42) nmol/L, capacity of Bmax = (475.00± 17.86) pmol/mg. Conclusion LHRH might be a useful target of immunotherapy in gastric carcinoma. LHRH-PE40 has an evident anti-tumor efficacy in vitro.

Abstract:We developed a new assay of BCl-x(L) inhibitors based on fluorescence resonance energy transfer that occurs between an AEDANS-labeled Bak-BH3 peptide and three tryptophans in the BH1 and BH2 domains of Bcl-x(L). The method can tolerate up to 5% DMSO, and it was validated with several BCl-x(L) inhibitors. It can be adapted to screen for compounds targeting other Bcl-2 family proteins.

Anti-Cancer Agents in Medicinal Chemistry

Abstract:Background: Peniciketal A (Pe-A) is a spiroketal compound isolated from saline soil-derived fungus Penicillium raistrickii. However, its role for biological processes has not been clarified. In this study, we for the first time investigated the anticancer effects and the underlying mechanisms of Pe-A in A549 lung cancer cells. Metheds: Cell proliferation was tested by MTT assay and colony formation assay. Flow cytometry was performed to examine the cell cycle, apoptosis and mitochondrial membrane potential. Invasion and migration were analyzed using transwell assay and wound healing analysis. Immunofluorescence staining and western blotting were used to evaluate the protein expression. Results: Pe-A effectively inhibited proliferation, with IC50 values was 22.33 mu M for 72 h. Mechanistic studies revealed that Pe-A caused cell cycle arrest at the G0-G1 phase by decreasing cyclinD1 expression and induced apoptosis through accelerating the mitochondrial apoptotic pathway. Moreover, Pe-A significantly inhibited A549 cell migration and invasion by reducing the protein levels of MMP-2 and MMP-9, while the Epithelial-Mesenchymal Transition (EMT) property was also changed. Importantly, Pc-A exhibited much lower toxicity towards L02, normal liver cells, and MRCS, normal fibroblast cells, compared to A549 cells. Conclusion: Collectively, the current results indicate that Pc-A may offer effective potentials and insights for lung cancer treatment and drug design.

Abstract:Background and objective: At present clinical researches has proven that the chemotherapeutic effect on advanced non-small cell lung cancer (NSCLC) has gotten to plateau. Biotherapy combined with chemotherapy could improve curative effect on advanced NSCLC obviously. The project is aiming at exploring the killing effect and the mechanism of rmhTNF-α in combination with gemcitabine on human lung adenocarcinoma cell line A549. Methods: CCK-8 was used to detect the inhibition ratio of rmhTNF-α and gemcitabine cell line A549; cell curve of growth was used to illustrate the influence of rmhTNF-α and gemcitabine on the proliferation of A549; FCM was used to detect the cell cycle and the apoptosis ratio of cell line A549 treated with either rmhTNF-α or gemcitabine or those combined. The change in cellular morphology and cell ultramicromorphology was observed by light microscope and transmission electron microscope respectively. Results: The result of CCK-8 and cell curve of growth demonstrated that rmhTNF-α not only inhibited the proliferation of A549, but also enhanced the killing effect of gemcitabine; FCM showed that rmhTNF-α promoted S cell cycle arrest and reduced proportion of G2/M cell arrest; also showed different apoptosis ratio was most significant in the group of rmhTNF-α combination with gemcitabine; the changes of morphology was observed obviously by light microscope and transmission electron microscope. Conclusion: rmhTNF-α enhances the the killing effect of gemcitabine on human lung adenocarcinoma cells A549 by inducing apoptosis and promoting cell cycle arrest.
Functional Identification of the Stable Transfection C5ar Cell Line Molt-4

Abstract:The complement C5 anaphylatoxin receptor is a member of the seven transmembrane-spanning G protein-coupled receptor superfamily that signals through G alpha i and G alpha 16. C5aR is mostly expressed on neutrophils, macrophages and endothelial cells. C5a and C5aR interaction plays an important role in numerous biological effects such as in vivo cytokine storm which results in inflammatory damage. Considering the limitation of collection of human peripheral blood neutrophils and their short half life, the stably transfected cell line for studying the biological effects of C5aR is needed. In this study, we transfected C5aR gene into Molt-4 cell line and examined the function of ectopic C5aR. Our results showed stable expression of the C5aR in Molt-4 cell line and their interaction with human C5a induced ERKI/2 phosphorylation, Ca++ influx. This stable transfected cell line may provide a useful tool for studying signal pathways related to C5a and C5aR interplay and antibody development specific for C5aR.

Abstract:OBJECTIVE:To determine the sensitivity of tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) induced apoptosis in Hep-2 cells by means of systematically evaluating the cytotoxicity of TRAIL alone and TRAIL in combination with chemotherapeutic agents (cisplatin, paclitaxel) or radiation in Hep-2 cells in vitro, and whether the synergistic killing effects correlated with the expression level of TRAIL receptors and the activity of caspase-8 or caspase-9.METHODS:The cytotoxicities of TRAIL, cisplatin and paclitaxel were investigated by cell counting kit-8 (CCK-8) assay. The expression levels of four TRAIL receptors in Hep-2 cells after treated by TRAIL, chemotherapeutic agents or radiation alone and by combined treatments were measured by flow cytometry and Western blotting respectively. The growth inhibition effects of caspase-8 or caspase-9 inhibitor on Hep-2 cells were determined by CCK-8 assay, and the activities of caspase-8, caspase-9 and caspase-3 were measured by Western blotting.RESULTS:The half maximal inhibitory concentration (IC50) of TRAIL to Hep-2 cells on 24 h was 596.92 microg/L. Cisplatin, paclitaxel and radiation had synergistic inhibitory effects with TRAIL on the growth of Hep-2 cell line. After the activity of caspase-9 was inhibited by Z-LETD-FMK, the inhibition effects of TRAIL, cisplatin and paclitaxel on Hep-2 cells decreased significantly (all P<0.05). The expressions of caspase-8, caspase-9 and the death receptors (DR4 and DR5) increased significantly (all P<0.05) after combined administration.CONCLUSIONS:Hep-2 cells are resistant to the apoptosis induced by TRAIL and TRAIL can induce the apoptosis of Hep-2 cells through the endogenous apoptotic pathway. The increase of death receptors expression by chemotherapeutic agents or radiation could enhance the sensitivity of Hep-2 cells to TRAIL and the synergistic killing effects.
Functional Significance of CD40 and CD40 Ligand Linking on Human Lung Cells

Abstract:OBJECTIVE:To determine the possible functional significance of CD40 and CD40 ligand (CD154) linking on human lung carcinomas and to assess the potential of CD40 as a therapeutic target.METHODS:We evaluated the effect of CD40L on the surface expression of major histocompatibility complex class I (MHC-I), Fas, bcl2, CD54, epidermal growth factor receptor (EGFR), p-glucoprotein (PGP), lung related protein (LRP), cell cycle, cell apoptosis and growth kinetics of 7 lung cancer cell lines (including 1 CD40-transfected cell line GLC-82/CD40) by gene cloning, Western blotting, and flow cytometry etc.RESULTS:Significant increased expression of MHC-I, CD54 and Fas was observed in 4 tumor lines expressing high levels of CD40 after CD40L (0.1 micro g/ml) linked to CD40. CD40L (0.1 micro g/ml or greater) in the fifth day was found to significantly inhibit the proliferation of 4 cell lines expressing high levels of CD40, decreased the percentage of aneuploid cell, and inhibited S-phase cells entering G2/M phase. The effect of CD40 cross-linking was reversible. CD40-moderate and low and negative tumor did not respond to CD40L. All of 7 cell lines show no significant changes in apoptosis in either the experimental (CD40L pulsed) or control (medium only) cell cultures.CONCLUSION:Expression of CD40 on lung tumors cells expressing high level of CD40 (including CD40-transfected cells) may represent a potential therapeutic target.
Functional Modulating Effect of CD40 Ligand on CD40-transfected Human Lung Carcinomas

Abstract:OBJECTIVE:To determine the modulating effect of CD40 ligand (CD40L, CD154) on CD40-transfected human lung carcinomas and to assess the potential of CD40 as a therapeutic target.METHODS:Tumor cells of a CD40-negative lung cancer cell lines (GLC-82) were transfected with a vector expressing CD40 cDNA. The transfected cell line, GLC-82/CD40, was shown to express high levels of CD40. GLC-82/CD40 cells after being exposed to 0.1 micro g/ml CD40L were examined for their surface expression, cell cycle, apoptosis and cell growth by flow cytometry and MTT assay.RESULTS:The expression of MHC-I, ICAM-1 and Fas in GLC-82/CD40 cells was significantly increased, whereas that of EGFR was decreased. Cell proliferation was significantly inhibited by CD40L with an inhibition rate of 30% on day 5, but no change in cell cycle. All of the changes disappeared after 48 h incubation with CD40L. More significant changes were observed in Calu-3 cell lines which expressed high levels of CD40, but the CD40-negative GLC-82 cells were unresponsive to CD40L. None of the 3 cell lines showed significant changes in apoptosis upon CD40L treatment.CONCLUSION:CD40, if over-expressed in tumor cells, could be considered as a potential therapeutic target.
[Effect of PLGA nanoparticles conjugated with anti-OX40/anti-AFP mAbs on cytotoxicity of CTL cells against hepatocellular carcinoma]

Abstract:Objective: To evaluate the effect of anti-OX40 and anti-AFP antibodies conjugated onto poly(DL-lactide-co-glycolide)-nanoparticles (PLGA-NPs) on the cytotoxic activity of AFP158-166; -specific cytotoxic T lymphocyte (CTL) against hepatocellular carcinoma cells in vitro. Methods: PLGA-NPs were prepared by oil-in-water single emulsion solvent evaporation method and covalently conjugated with anti-OX40 and anti-AFP monoclonal antibodies. Scanning electron microscopy (SEM) was utilized for the characterization of the surface morphology and estimation of the size of the PLGA-NPs. The mean diameter and zeta potential of the nanoparticles were measured by dynamic light scattering (DLS) performed in a Zetasiser Nano Series ZEN3600. Antibody conjugation efficiency was determined using bicinchoninic acid (BCA) protein assay. Dendritic cells (DCs) were induced from human peripheral blood mononuclear cells (PBMCs) in the presence of GM-CSF and IL-4, and loaded with AFP158-166; peptide to generate AFP-specific CTL (CTL/AFP158-166;). WST-1, ELISA and lactate dehydrogenase (LDH) methods were respectively used to examine the effects of the anti-OX40/anti-AFP-NPs on CTL/AFP158-166; proliferation, IL-2 and IFN-γ production, and cytotoxicity against the tumor cells. Results: The obtained nanoparticles were found to be of regular spherical shape and the smooth surface with an average diameter of (300±42) nm and a negative zeta potential of -(25.12±5.34) mV. Approximately 100 μg antibodies were conjugated to every milligram of the nanoparticles with a conjugation efficiency of about 25% as estimated by BCA protein assay. Proliferation and activation analysis revealed that anti-OX40/anti-AFP mAb-NPs significantly induced CTL proliferation and the secretion of IL-2 and IFN-γ. The cytotoxicity assay showed that anti-OX40/anti-AFP-NPs markedly enhanced CTL/AFP158-166; specific killing on HepG2 cells but had no obvious effect on SMMC-7721 cells. Conclusion: Anti-OX40 mAb and anti-AFP mAb conjugated to PLGA-NPs could stimulate CTL/AFP158-166; cell proliferation and cytokine production as well as enhancing their specific killing on AFP-positive hepatocellular carcinoma cells.
Identification of an aptamer through whole cell-SELEX for targeting high metastatic liver cancers.

Abstract:Hepatocellular carcinoma (HCC) is one of the most deadly human cancers due to its ability of invasion and metastasis. Thus, the approaches to identify potential compounds that inhibit invasion and metastasis of HCC are critical for treatment of this disease. In the present study, we used HCCLM9 cells with high metastatic potential and MHCC97L with low metastatic potential as a model system to study the molecular mechanisms of HCC metastasis. By applying cell-Systematic Evolution of Ligands by Exponential enrichment (SELEX) against living cells, we used HCCLM9 as target cells and MHCC97L cells as control to screen a group of HCC metastasis-and cell-specific DNA aptamers. One of selected aptamers, LY-1, could specifically bind to metastatic HCC with a dissociation constant (Kd) in nanomolar range. In vitro studies demonstrated that LY-1 can recognize and bind to membrane protein of metastatic HCC cells. Furthermore, QD605 labeled LY-1 aptamer could recognize HCC cells in both local liver cancer tissues and pulmonary metastatic sites in a xenograft model of HCC with pulmonary metastasis. Further biochemical and immunostaining studies showed that LY-1 could selectively bind to a subpopulation of more metastatic cells in HCCLM9 cells, which express more CK19 and vimentin. Finally, treatment of highly metastatic cells with LY-1 led to reduced migration and invasiveness of HCCLM9 cells in vitro and suppression of xenograft growth in vivo. Taken together, the present study demonstrated the tumor targeting and tumor suppressive effects of LY-1, which could be a promising molecular probe for metastatic HCC and a potential candidate of chemotherapy for metastatic HCC.
Effect of Recombinant Toxin LHRH-PE40 on Proliferation and Apoptosis of Ovarian Cancer Cell

Abstract:Objective:To study the effects of recombinant protein LHRH-PE40 on ovarian cancer cell proliferation and apoptosis.Methods:Proliferation inhibition assay was taken in vitro cultured ovarian cancer SK cell.Different concentration of LHRH-PE40 was added into cell culture solution.The effects of LHRH-PE40 on ovarian cancer cell proliferation were analyzed by enzyme micro-plate reader 1-4 days after adding LHRH-PE40.The effects of LHRH-PE40 on proliferative factor Ki-67,anti-apoptosis gene Bcl-2 and cell cycle in ovarian cancer SK cells were detected by immunohistochemical staining and Cytometry.Results:LHRH-PE40 could inhibit the growth of in vitro cultured epithelial ovarian cancer SK cell,which presented time and concentration dependency.The best action time was 48-72 hours after administration of drugs.Under the concentration of medicine 1μg/ml,the positive cell rate of Ki-67(the rate before using medication was 73%,after was 25%) and Bcl-2(the rate before using medication was 92 %,after was 43%) was decreased after medication,and the ratio of apoptosis was increased(from 0.8% to 3.0%).Under 0.5 μg/ml concentration,the cell ratio of G0/G1 was also increased,in which the cell ratio of S phase was increased and the ratio of G2/M decreased.Conclusions:LHRH-PE40 can kill ovarian cancer cells and promote apoptosis of ovarian cancer cell.
Transient Transfection of Short Hairpin RNA Targeting Epidermal Growth Factor Receptor into Lung Adenocarcinoma A549 Cell Line and Its Effect on Cell Growth and Sensitivity to Cytotoxic Drugs

Abstract:Background and purpose:Up-regulation of expression and activation of epidermal growth factor receptor(EGFR)in non-small cell lung carcinoma(NSCLC)is associated with advanced tumor stage,and resistance to radiochemotherapy.EGFR has been identified as an important target in anticancer therapy by blocking EGFR function with the detailed understanding of the biology of EGFR protein and its downstream signal pathway.In this study,we investigated the effects of short hairpin RNA(shRNA)targeting EGFR on cell growth and its sensitivity to cytotoxic drugs in lung adenocarcinoma A549 cell line.Methods:Recombined plasmids of short hairpin RNA targeting EGFR(pShEGFR)and of negative control shRNA(pShNEG)were respectively transfected into A549 cells,and named A549/pShEGFR,A549/pShNEG,and control cell group A549.The protein level of EGFR was observed by immunofluorescence,and then further quantified by Western blot.The effect of shRNA targeting EGFR on tumor cell growth was assessed by colony formation assay,cell cycle and apoptosis by flow cytometry,and the sensitivity of A549 to cytotoxic drugs by 3-4,5-dimethylthiozol-2yl-2,5-diphenyltetrazolium bromide MTT assay.Results:A549/pShEGFR significantly down-regulated EGFR expression by 74.1% on day 6 after transfection and the colony number by 62.8% in A549(P0.01).Compared with the other two groups,A549/pShEGFR caused G1 arrest(63.2±1.1,64.5±1.4 vs.74.2±0.8;P0.01),induced apoptosis(5.8±1.4,7.7±1.2 vs.25.6±1.2;P0.01),and subsequently increased the sensitivity to cisplatin,doxorubicin and paclitaxel by 6.7,5.5,6.6 fold when compared with recombined plasmid A549 respectively(P0.01).Conclusions:Our data suggested that shRNA could effectively inhibit the growth and enhance the sensitivity of A549 cells to different cytotoxic drugs by downregulating the expression of EGFR gene.

Abstract:Objective To investigate the effects of 4-HPR on adenocarcinoma of the human lung cell line A549 in vitro and initial mechanisms for apoptosis. Methods The growth inhibitory effect of 4-HPR on A549 was observed by MTT method, the cell cycle and apoptosis was assayed by flow cytometry, protein expression on apoptosis was detected by Western blot. Results 4-HPR inhibited the proliferation of A549 cells in a dose-dependent manner.4-HPR induced apoptosis, with the decreasing of bcl-2, Bcl-x(subscript S/L) expression, without marked change of expression of Caspase-3, Caspase-9. Conclusion 4-HPR could inhibit the proliferation of pulmonary adenocarcinoma cell remarkably, induce A549 apoptosis through decreasing Bcl-2, Bcl-x(subscript S/L) expression, and it provides experimental evidence to futher investigation of 4-HPR mechanisms in pulmonary adenocarcinoma theraphy.

Abstract:To study the affinities of antagonistic peptides with HAb18G/CD147 on the hepatocellular carcinoma cells.Fluorescence-activated cell sorting analysis(FACS) was used to detect the expression of HAb18G antigen on HHCC and SMMC7721 cells. Antagonistic peptides(APs) AP-1, AP-2 and AP-6 were labelled with biotin at their N-terminals. The binding abilities of APs to HHCC or SMMC7721 cells were determined by FACS and confocal microscope.HAb18G antigen was highly expressed on the HHCC and SMMC7721 cells. AP-6 has the highest affinity for HHCC and SMMC7721 cells compared with AP-1 and AP-2.Antagonistic peptides of HAb18G/CD147 have high affinities with HAb18G/CD147 on the hepatocellular carcinoma cells.

Abstract:Objective: Programmed death ligand-1 (PD-L1) is expressed on tumor cells and macrophages. The detection of PD-L1 expression in cancer and the treatment by targeting the PD-L1/programmed death-1 (PD-1) are of great clinical significance. This work aims to screen the aptamers with high affinity and specificity for recombinant human PD-1 (rhPD-1)/recombinant human PD-L1 extracellular domain (rhPD-L1). Materials and methods: In this study, we have expressed, purified, prepared, and identified rhPD-1 and rhPD-L1. The rhPD-L1/rhPD-1 aptamers with high afﬁnity and speciﬁcity were obtained by systematic evolution of ligands by exponential enrichment technique. Ten aptamers sequences to rhPD-L1 and 10 aptamers sequences to rhPD-1 were obtained by cloning and sequencing. The afﬁnity and speciﬁcity of candidate aptamers were analyzed by gold nanoparticles-based colorimetric assay, dot blot assay, and electrophoretic mobility shift assay. Results: The aptamers named A6 were picked out as the optimal aptamers that recognize PD-1, speciﬁcally with the Kd value of 47.84 ± 24.78 nM. The aptamers named B10 were picked out as the optimal aptamers that recognize PD-L1, speciﬁcally with the Kd value of 59.72 ± 15.87 nM. Conclusions: The study lays a foundation for the development of detection methods and therapeutic drugs targeting PD-L1/PD-1.
Tumor Homing Cell Penetrating Peptide Decorated Nanoparticles Used for Enhancing Tumor Targeting Delivery and Therapy.

Abstract:Specific targeting ability and good tissue penetration are two critical requirements for tumor targeted delivery systems. Systematical selected peptides from a library may meet these two requirements. RLW was such a cell penetrating peptide that could specifically target to non-small cell lung cancer cells (A549). In this study, RLW was linked onto nanoparticles (RNPs) and then the RNPs were used for lung cancer targeting delivery. A traditional cell penetrating peptide, R8 (RRRRRRRR), was used as control. In vitro cellular uptake study demonstrated that modification with RLW specifically enhanced the uptake by A549 cells rather than human umbilical vein endothelial cells, while modification with R8 increased the uptake by both cells. Furthermore, the modification with RLW specifically elevated the penetration into A549 tumor spheroids rather than glioma cell (U87, used as in vivo control) spheroids. And the in vivo imaging further demonstrated RNPs could target to A549 xenografts rather than U87 xenografts. Importantly, the distribution of RNPs in normal organs was approximately the same as that of unmodified nanoparticles. However, R8 modified nanoparticles elevated the distribution in almost all the tissues. These results demonstrated that RLW was superior in A549 tumor targeted delivery. After loaded with docetaxel, an anti-microtube agent, different formulations could effectively induce the A549 cell apoptosis, and inhibit the growth of A549 spheroids in vitro. While in vivo, RNPs displayed the best antitumor effect. The tumor volume was significantly lower than other groups, which was only 33.3% as that of saline group. In conclusion, in vitro RLW could specifically target to A549 cells and enhance the cytotoxicity of docetaxel. In vivo, RLW could significantly enhance the A549 xenografts targeting delivery and led to improved antitumor effect.

Abstract:This study aims to determine the sensitivity, specificity and accuracy of epidermal growth factor receptor monoclonal antibody (EGFRmAb) modified poly(lactic acid-co-l-lysine) nanoparticles (PLA-PLL-EGFRmAb) NPs delivery system to EGFR positive cancer cells. In the study, a new PLA-PLL-EGFRmAb NPs was prepared. The cellular cytotoxicity, cellular uptake, and the targeted effect for hepatocellular carcinoma of PLA-PLL-EGFRmAb NPs were investigated. In vitro, the findings of Flow cytometry and Confocal Laser scanning Biological Microscopy showed that PLA-PLL-EGFRmAb NPs can bind to hepatocellular carcinoma cells and were uptaken effectively. In vivo in the SMMC-7721 xenograft mouse model, PLA-PLL-EGFRmAb NPs could target to the tumor effectively, which demonstrated a better targeting. These results showed that the PLA-PLL-EGFRmAb NPs have the potential to be used as a target delivery carrier for tumor therapies.
