Abstract:Matrine and oxymatrine are the major bioactive compounds extracted from the root of Sophora flavescens Ait, which have been widely used in traditional Chinese medicines. The objective of the study was to investigate the effects of matrine or oxymatrine on hepatic cytochrome P450 (CYP450) and the underlying molecular mechanisms. Matrine (15, 75 and 150 mg/kg) or oxymatrine (36, 180 and 360 mg/kg) was administered to rats for 14 days and the activities of CYP450 were measured by the quantification of the metabolites from multiple CYP450 probe substrates, using validated liquid chromatography coupled with liquid chromatography-tandem mass spectrometry detection (LC-MS/MS) and high-performance liquid chromatography methods. The mRNA and protein expression levels of CYPs were determined by quantitative real-time reverse-transcription polymerase chain reaction and Western blotting analysis respectively. Interactions between matrine or oxymatrine and human constitutive androstane (CAR), pregnane X receptor were evaluated by means of the reporter gene assay in CV-1 cells. Our study showed that matrine and oxymatrine significantly induced the activity and gene expression of CYP2B1 in a dose-dependent manner; matrine (150 mg/kg) slightly induced the mRNA and protein expression of CYP2E1 and mildly inhibited the mRNA and protein expression of CYP3A1 in rats. Matrine or oxymatrine could activate human CAR and induce the CYP2B reporter construct in CV-1 cells. These results reveal that matrine and oxymatrine can induce the activity and expression of CYP2B1/2 in rats, and the underlying mechanism may be related to the activation of CAR.

Abstract:PURPOSE:Matrine, one of the main active components of extracts from the dry roots of Sophora flavescens, has potent anti-tumor activity in various cancer cell lines. However, the activity of matrine against osteosarcoma remains unclear. In the present study, we examined the effects of matrine on human osteosarcoma cells and explored the underlying mechanism.METHODS:Four human osteosarcoma cell lines: MG-63, U-2OS, Saos-2, and MNNG/HOS were treated by matrine and subjected to MTT assay, annexin V-FITC/PI double staining, and TUNEL assay. The activation of caspases and the expression of pro-apoptotic and anti-apoptotic factors were examined by qRT-PCR and Western blot. In addition, MNNG/HOS xenograft tumors were established in female nude BALB/c mice, and matrine was intraperitoneally (i.p.) administered to evaluate the anti-cancer capacity of matrine in vivo.RESULTS:We found that matrine inhibited the proliferation and induced apoptosis of the four osteosarcoma cell lines in vitro and induced the activation of caspase-3, -8, and -9 in a dose-dependent manner. Furthermore, the pro-apoptotic factors Bax and Fas/FasL were upregulated, and the anti-apoptotic Bcl-2 was downregulated. More importantly our in vivo, studies showed that administration of matrine decreased tumor growth in a dose-dependent manner. Immunohistochemistry analysis demonstrated the downregulation of Bcl-2 and upregulation of Bax and Fas/FasL in MNNG/HOS tumor tissues following matrine treatment, consistent with the in vitro results.CONCLUSION:Our results demonstrate that matrine inhibits the proliferation and induces apoptosis of human osteosarcoma cells in vitro and in vivo. The induction of apoptosis appears to occur through the upregulation of Fas/FasL and Bax, downregulation of Bcl-2, and activation of caspase-3, -8, and -9, which then trigger major apoptotic cascades.

Abstract:Objective To investigate the inhibitory effect of matrine on the invasiveness of human malignant melanoma cell line A375,and the mechanism of this effect.Methods MTT assay and Annex- in-V-FIFT/PI double staining were performed to examine the effects of matrine on the proliferation and apoptosis of A375 cells.Semi-quantitative RT-PCR was used to evaluate the mRNA expression of hep- aranase in the treated cells.The adhesion and invasiveness of A375 cells were tested by cell-matrigel adhe- sion assay and matrigel invasion assay,respectively.Results Matrine (≥0.5 mg/mL) significantly inhibit- ed the proliferation of A375 cells,and induced apoptosis of these cells;both effects were in a dose-depen- dent manner.The heparanase-mRNA expression in A375 cells were significantly decreased by 0.125～0.5 mg/mL matrine,as were the adhesion and invasiveness of these cells (all P＜0.01);all the effects were in a dose-dependent manner (P＜0.01).Conclusions Matrine inhibited the adhesion and invasiveness of A375 cells in vitro.These effects could be due to the inhibition of cellular proliferation,induction of cellu- lar apoptosis and down-regulation of heparanase-mRNA expression.

Abstract:OBJECTIVE To investigate the effects of matrine on cellular proliferation and apoptosis in human malignant melanoma cell line A375 in vitro. METHODS The A375 cells were cultured with different concentrations of matrine.The inhibition of cellular proliferation was determined by measuring MTT dye absorption of the living cells. The apoptosis was assessed by the Annexin-V-FIFT/PI affinity assay. The morphological changes of A375 cells were observed by light and electron microscopy. RESULTS Matrine significantly inhibited the proliferation of A375 cells in a dose-and time-dependented manner. The outcome of flow cytometry showed that matrine induced more Annexin-V stained cells in a dose-dependented manner,which was proved by light and electron microscopy. CONCLUSION Matrine plays an inhibitory role in the tumor cell proliferation and induces the cell apoptosis in malignant melanoma cells.

Abstract:OBJECTIVETo study the inhibition effect on invasion and metastasis related factors in differentiation of SMMC-7721 cells induced by matrine.METHODHuman hepatoma cell lines SMMC-7721 were cultured in vitro and treated by 0.5 g/L, 0.8 g/L, 1.0 g/L matrine. Then the cell growth curve was protracted, the cell form was observed by light microscope, the invasion and metastasis related factors were measured with cell adhesion test, cell migration test and cell invasion test. The data was collected and analyzed with statistical methods to evaluate roundly the inducing differentiation function of matrine.RESULTTreated by 0.8 g/L, 1.0 g/L matrine, the proliferation of SMMC-7721 was inhibited markedly, and the malignant cell form tended to normal differentiation. It was also found that matrine could inhibit cell invasion and metastasis related factors, including adhesive ability to FN, migrative ability through the polycarbonate filter, and invasive ability through matrigel.CONCLUSIONMatrine is a good differentiation inducer, it can not only inhibit the cell proliferation and induce the cell form differentiation, but also significantly inhibit the invasion and metastasis ability of SMMC-7721 through several steps, involved cell adhesion, migration and invasion.

Abstract:Objectives:To investigate the differentiation of human hepatocellular carcinoma cell line Bel-7404 induced by Matrine in vitro,and discuss its mechanism.Methods:After dealing human hepatocellular carcinoma cell line Bel-7404 with different concentrations of Matrine,we drew cell growth curve by MTT colorimetry,and cell morphology was examined by light microscope.Then we evaluated the tumor cell biological characteristics changes through improved soft agarose clone forming test,cell adhesion assay,cell movement assay and cell invasion assay.Also we detemined cell cycle distributing by flow cytometry.Results:After 72 hours being treated with 0.5,0.75,1.0g/L,in the Matrine test groups,the cell proliferation was inhibited significantly,and cell shape transformed to normal.The amounts of clone were remarkably reduced.The cell adhesion ability increased in different level.At the same time cell ability of movement and invasion differently weakened.Analysis of cell cycle shown that along with rising of Matrine concentration,the cell amounts in G1 phase increased accordingly and cell amounts in G2 and S phase remarkbly decreased.Conclusion:During the appropriate concentration range,Matrine not only inhibited Bel-7404 growth and proliferation,but also induced tumor cell to differentiate.Its probably mechanism may related to cell cycle regulation.

Abstract:OBJECTIVE:To investigate the effect of matrine on the invasiveness and expression of heparanase-mRNA in human malignant melanoma cell line A375. METHODS:The A375 cells were treated by matrine in different concentration. The total RNAs were extracted from the cells 48 hours after treatment and then semi-quantitative RT-PCR were performed to evaluate the heparanase-mRNA expression levels. Effect of matrine on adhesion of treated A375 cells was tested by cell-Matrigel adhesion assay. The invasiveness of treated A375 cells was measured by Matrigel invasion assay. RESULTS:The hepanase-mRNA expression, adhesion and invasiveness of A375 cells treated with matrine of different final concentrations significantly decreased compared with that of the controls (p < 0.01). Besides, the inhibitory effects were signifcantly different when the cells treated with matrine of different concentrations (P < 0.01). CONCLUSION:By down-regulating the expression of heparanase-mRNA, matrine has a significant inhibitory effect on the adhesion and invasiveness of human malignant melanoma cell line in a dose-dependent manner.

Abstract:Background Matrine is a traditional Chinese medicine with significant inhibitory activity against malignant tumors. Its effects on the invasiveness and metastasis of malignant tumors have rarely been reported.Aim To investigate whether matrine can inhibit the metastasis-related activities of the human malignant melanoma cell line A375 in vitro.Methods 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay and Annexin-V-fluorescein isothiocyanate/propidium iodide (Annexin-V-FITC/PI) affinity assay were used to examine the effects of matrine on the proliferation and apoptosis induction of A375 cells. The morphologic changes of A375 cells were observed by light and electron microscopy. Semiquantitative reverse transcription-polymerase chain reaction (RT-PCR) and Western blotting were performed to evaluate the expression of heparanase mRNA and protein. The effect of matrine on the adhesion ability and invasiveness of treated A375 cells was tested by cell-Matrigel adhesion assay and Matrigel invasion assay, respectively.Results Matrine showed significant inhibition of the proliferation of A375 cells in a dose- and time-dependent manner. It also induced apoptosis in a dose-dependent manner. Compared with the control group, the levels of heparanase mRNA and protein expression of A375 cells treated with different concentrations of matrine were decreased significantly, as were their adhesion ability and invasiveness.Conclusions These findings indicate that matrine inhibits the invasiveness and metastasis of A375 cells in vitro. The mechanisms may be linked to the inhibition of cellular proliferation, induction of apoptosis, and downregulation of heparanase mRNA and protein expression.

Abstract:Matrine, an alkaloid isolated from <i>Sophora flavescens</i>, promotes tumor cell apoptosis and strengthens the anticancer capacity of chemotherapeutic drugs. The present study aimed to investigate the inhibitory effect and underlying mechanism of matrine in combination with cisplatin on liver cancer progression. Tumor progression was studied in nude mice. The human liver cancer cell line HepG2 was injected into BALB/c nude mice subcutaneously to establish a tumor model. Mice were subsequently treated with matrine, cisplatin, matrine + cisplatin or normal saline. Nude mice and tumor growth were monitored. Tumors were excised and the expression of survivin, caspase-3, caspase-7 and caspase-9 was detected by immunohistochemistry. Western blotting was used to determine the expression of survivin, caspase-3, caspase-7, caspase-9 and X-linked inhibitor of apoptosis protein (XIAP) in tumor tissues. The results demonstrated that matrine exerted anticancer effects in liver cancer-transplanted tumors, as evidenced by decrease in tumor weight and volume. Furthermore, the tumor inhibition rate in mice treated with matrine + cisplatin was 83.3%, whereas it was of 37.5 and 75% in mice treated with matrine or cisplatin alone, respectively. In addition, the expression of survivin and XIAP was significantly downregulated, whereas the expression of caspase-3, caspase-7 and caspase-9 was significantly upregulated in tumor tissues from nude mice treated with matrine + cisplatin, compared with those treated with cisplatin, matrine or normal saline. These findings suggested that the combination of matrine and cisplatin may promote tumor cell apoptosis in liver cancer by activating the caspase apoptosis pathway and suppressing the survivin-associated inhibition of caspase-9.

Effect of oxymatrine treatment on the invasion of human cervical cancer cells

Abstract:Objective To investigate the anticancer effect of oxymatrine on cervical cancer cell line (HeLa). Methods MTT assay was used to detect the anti-proliferative effect of oxymatrine.Transwell chamber was used to detect the anti-metastatic effect of oxymatrine.Real-time PCR was used to detect the mRNA levels of MMP-2 and MMP-9.Western blot was used to detect the protein levels of MMP-2,MMP-9,AKT,p-AKT and GADPH. Results We found that application of oxymatrine significantly inhibited the growth of HeLa cells at the concentration above 0.8 mg/mL.We also found that oxymatrine (0.1,0.2 and 0.4 mg/mL)inhibited the invasion of HeLa cells under cytotoxic dose,which was (77.07±20.43)%,(53.95±18.17)% and (20.35±11.20)% of cells that migrated through the matrigel when compared with those of non-oxymatrine treatment group (P<0 .0 5 ). Further research found that oxymatrine (0.1,0.2 and 0.4 mg/mL)could reduce the expression of MMP-2 at the mRNA level,i.e.(82.76±8.71)%,(39.51±12.79)% and (21.53±5.38)% of the expression level when compared with that of non-oxymatrine treatment group (P<0 .0 5 ).The protein expression level of MMP-2 in 0 .4 mg/mL group was (64.69 ±16.52)% of non-oxymatrine treatment group (P<0.05).The phosphorylation level of AKT in 0.4 mg/mL group was (41.27±7.13)% of non-oxymatrine treatment group (P<0.05).Conclusion Oxymatrine can inhibit the invasion of HeLa cells by reducing the expression of MMP-2 via inhibiting the activity of AKT signal pathway.All together,our findings bring new insights into the mechanism of the anticancer effects induced by oxymatrine treatment.
Effect of Matrine on Inhibiting Proliferation and Inducing Apoptosis of Human Intestinum Crassum Carcinoma HT29 Cells

Abstract:Objective To explore the anti-proliferation and apoptosis-induced effects of matrine on HT29 cell and their possible mechanism.Methods HT29 Cell proliferative activity was measured by MTT assay;Cell cycle and apoptosis were analyzed by Flow Cytometry;Alteration of cellular skeleton was observed by transmission electron microscopy(TEM);Global gene expression profiles were scanned by gene chip.Results After exposure to matrine at concentrations from 0.062 5 to 0.5 mg/mL for 48 h,cellular proliferation was inhibited with increasing the concentration,but this inhibitory effect attenuated at 1 mg/mL and the apoptosis was up-regulated significantly.G2/M and G1/S phases of cell cycle were,to some extent,arrested.Under TEM,the morphological changes could be found.Gene chip showed that matrine could alter a large number of genes,which related to the cell proliferation,cell cycle,and apoptosis.Conclusion The anti-proliferation and apoptosis-induced effects of matrine on HT29 cells are concentration-dependent through changing of many genes involved in proliferation,cell cycle,and apoptosis.
Matrine Promotes Hepatic Oval Cells Differentiation into Hepatocytes and Alleviates Liver Injury by Suppression of Notch Signalling Pathway.

Abstract:AIMS:Recent studies have shown that the hyperactive Notch pathway is involved in cirrhosis and hepatocellular carcinoma (HCC) development by regulating differentiation of hepatic oval cells (HOCs) into cancer cells. The aim of this study was to investigate whether matrine can alleviate liver injury and promote HOC differentiation into hepatocytes by suppression of Notch pathway.MAIN METHODS:We evaluated the expression of Notch-1, Jagged-1, and Hes-1 in HCC tissue by immunohistochemistry. Stem cell characteristics of HOCs were evaluated by CCK-8, cell cycle, and apoptosis. The expression of Notch pathway, HOC markers and albumin (ALB) was detected by immunohistochemistry, QRT-PCR and western blotting. The effects of matrine in protecting liver in vivo were investigated in a rat Solt-Farber precancerous model.KEY FINDINGS:We found an abnormal activated Notch pathway in HCC tissue, and the hyperactive Notch pathway was strongly associated with poor liver function in patients with cirrhosis with HCC. Using siNotch-1 to inhibit Notch pathway confirmed that Notch pathway could maintain stem cell characteristics of HOCs. Matrine inhibited stem cell characteristics of HOCs, the expression of Notch pathway and HOC markers but upregulated ALB. Matrine in combined with siNotch-1 RNA decreased the more potently inhibited HOC markers and Notch pathway. In rat Solt-Farber precancerous model, prophylactic application of matrine alleviated liver injury, downregulated Notch pathway and HOC markers, and upregulated ALB in a dose-dependent manner.SIGNIFICANCE:Matrine could promote the differentiation of HOCs into hepatocytes by inhibiting the Notch signalling pathway and alleviate liver injury.
Matrine Improves the Hepatic Microenvironment and Reverses Epithelial-Mesenchymal Transition in Hepatocellular Carcinoma by Inhibiting the Wnt-1/β-catenin Signaling Pathway.

Abstract:OBJECTIVE:Hepatocellular carcinoma (HCC) is a malignant tumor with high morbidity and mortality. Despite rapid progress in targeted therapy and immunotherapy for HCC over the past 10 years, the overall efficacy remains unsatisfactory. This is mainly due to the presence of an intrahepatic microenvironment of cirrhosis in HCC patients, leading to cancer recurrence and drug resistance.METHODS:In this study, we investigated the correlations between the Wnt-1/β-catenin signaling pathway and the prognosis as well as liver function of HCC patients. Additionally, we conducted in vitro experiments using different concentrations of matrine on HuH-7 cells. Furthermore, we verified the associations between the Wnt-1/β-catenin signaling pathway, inflammation, and epithelial-mesenchymal transition (EMT) in a rat model of pre-hepatocellular carcinoma. Finally, matrine was employed to treat pre-hepatocellular carcinoma in rats and patients with advanced hepatocellular carcinoma.RESULTS:The results demonstrated the activation of the Wnt-1/β-catenin signaling pathway, the occurrence of EMT, and exacerbated inflammation in human HCC tissues. In HuH-7 cell experiments, matrine effectively downregulated the Wnt-1/β-catenin pathway, reversed EMT, and suppressed migration and invasion of HCC cells. In the rat model of pre-hepatocellular carcinoma, matrine dose-dependently inhibited the activation of the Wnt-1/β-catenin signaling pathway, reversed the occurrence of EMT, and alleviated liver inflammation. Matrine analogues exhibited promising hepatoprotective effects in patients with advanced HCC.CONCLUSIONS:Matrine can reverse EMT, alleviate intrahepatic inflammation, and counteract immune depletion by inhibiting the Wnt-1/β-catenin signaling pathway in HCC.
Matrine Prevents the Early Development of Hepatocellular Carcinoma Like Lesions in Rat Liver

Abstract:Matrine (C15H24N2O) is an alkaloid extracted from the Chinese herb Sophora flavescens that has anti-fibrotic and anti-cancer properties. The aim of the present study was to determine the chemopreventive effect of matrine on the development of primary hepatocellular carcinoma (HCC) and its possible association with the suppression of the Notch signaling pathway. The rats were randomly divided into four groups: Control, model, low-dose matrine and high-dose matrine groups. The model was established by combining a partial hepatectomy with diethylnitrosamine (DEN) + 2-acetylaminofluorene (2-AAF). Low- and high-dose matrine groups received intragastric administration of matrine (0.25 and 2.5 g/l of matrine, respectively). DEN + 2-AAF injections and hepatectomy were not performed in the control group. All rats were sacrificed 2, 4 and 7 weeks after hepatectomy. HCC-like histopathological lesions were detected using hematoxylin and eosin staining. The expression levels of α-1-fetoprotein (AFP), albumin (ALB), Notch1 and Hes1 were analyzed using immunohistochemistry. Hepatic lobule structure loss, liver tissue necrosis and inflammatory cell infiltration, and edema degeneration were observed in the model group. By contrast, hepatocyte cord structure was restored and hepatocyte edema degeneration was significantly reduced after 7 weeks of treatment with matrine. In addition, compared with the model group, matrine reduced the expression of AFP, increased the expression of ALB and reduced the expression of Notch1 and Hes1 (only for high-dose matrine; all P<0.05). The findings suggested that matrine could prevent the early development of HCC-like lesions in a rat model, possibly by modulating Notch pathway activation.

Abstract:Matrine is one of the main bioactive alkaloids of Sophora flavescens Aiton, which has been widely used to treat various diseases in China. These diseases include viral hepatitis, liver fibrosis, cardiac arrhythmia, skin diseases, and tumors. However, matrine is also the main toxic compound of this herb, and the available biomarkers are not reliable in detecting or quantifying matrine risk. Metabolomics is a powerful tool used to identify early toxicity biomarkers that are specific indicators of damage to biosystems. This study aimed to find the potential biomarkers of the matrine-induced toxic effects in rats and HepG2 cells. The toxicological effects of rats induced by matrine could be derived from the elevated taurine and trimethylamine N-oxide levels and the depletion in hippurate and tricarboxylic acid cycle intermediates, such as 2-oxoglutarate, citrate, and succinate in the urine. Cell metabolomics revealed that the levels of alanine, choline, glutathione, lactate, phosphocholine, and cholesterol showed dose-dependent decreases, whereas the levels of taurine, fatty acid, and unsaturated fatty acid showed dose-dependent increases. Overall, a significant perturbation of metabolites in response to high dose of matrine was observed both in vivo and in vitro, and the selected metabolites particularly represent an attractive marker for matrine-induced toxicity.

Abstract:[Objective]To study the inhibiting proliferation effects and the apoptosis-promoting effects of Oxymatrine serum on hepatic stellate cell(HSC-T6),trying to find the function of Oxymatrine.which might be one of the mechanisms of Oxymatrine to treat alcohol liver fibrosis.[Methods]The experiments adopted hepatic stellate cell(HSC-T6)builded model by acetaldehyde as the model for study in vitro.By means of MTT,the inhibiting proliferation effect of differentconcentration of medicated serum was examined,so as to find the best functional point.By means of flow cytometry,the apoptosis-promoting effects were examined.[Results]The results showed that the culture medium containing 800 μg/ml medicated serum had significant inhibiting effects and could promote cell apoptosis.HSC-T6 cell's proportion was increased in G0/G1 period and decreased in the S period.[Conclusion]Oxymatrine can inhibit HSC-T6 proliferation and induce apoptosis.

Abstract:Objective To observe matrine's inhibitory effect on the growth of ACHN human renal cell carcinoma in vitro and explore its possible mechanism.Methods The cell line of ACHN human renal cell carcinoma was treated with matrine of different concentrations for 24,48,72 and 96 h,respectively.MTT assay was used to evaluate the cytotoxic effects of matrine on ACHN cells.The ACHN cell apoptosis induced by matrine was detected with transmission electron microscopy and flow cytometry.The expressions of Bcl-2 and Bax proteins were evaluated with immunocytochemical technique.Results Matrine of different concentrations had a cytotoxic effect on ACHN cell line in obvious dose-and time-dependent manners.Matrine could obviously induce apoptosis of ACHN cell line.Bax protein expression was significantly increased while the expression of Bcl-2 protein declined but with no significant difference.Conclusion Matrine has an obvious inhibitory effect on proliferation of ACHN renal cell carcinoma in vitro,which may be related to down-regulating the ratio of Bcl-2/ Bax protein expression and promoting cell apoptosis.
Effect of Oxymatrine on Proliferation of Hepatic Stellate Cells of Rat Liver

Abstract:Objective To study the effect of oxymatrine on proliferation of hepatic stellate cells (HSC) of rat liver. Methods HSCs were isolated and cultured from liver of Wistar rats by in situ perfusion with pronase and collagenase and density gradient centrifugation with Nycodenz. MTT colorimetric assay was used for detection of HSC proliferation.Results Oxymatine had an effect on proliferation of HSC. 0.5 ～16μg/ml oxymatrine inhibited HSC proliferation ( P 0.01). Conclusion Oxymatrine can prevent hepatic fibrosis by inhibiting HSC proliferation. (Shanghai Med J, 1999,22∶598 599)

Abstract:We investigated the effects of Matrine on proliferation by trypan blue exclusion and differentiation by benzidine staining positive cells in K-562 cells, assayed the telomerase activity using PCR-ELISA assay, analyzed cell cycle by fluorescence-activated cell sorter analysis of the DNA content, and also determined the gene expression level of c-myc, N-ras and p53 by northern blot and dot blot analysis. The results showed that with the addition of 0.1 mg/ml Matrine, cell growth was inhibited significantly by 4 days, benizidine-positive cells rose from 1% to 2% in control cells to 15% in treated cells on day 5; treatment of K-562 cells with 0.1 mg/ml Matrine for 5 days resulted in a marked inhibition in telomerase activity, in a manner that correlated with the extent of differentiation; after exposure to Matrine for 72 h, 64.6% cells were arrested in the G1-phase of the cell cycle, the fraction of cells in S-phase had decreased from 56.9% in control cells to 24.4% in differentiated cells, and the levels of N-ras and p53 mRNA were remarkably increased for 24 and 48 h, respectively, c-myc mRNA expression level declined for 24 h and was inhibited significantly for 48 h. Our study confirmed that Matrine plays a significant effect on the inhibition of proliferation cells and inducing differentiation in K-562 cells.
