The Effect of Puerarin on the Oxidative Stress in Cultured Human Peri-toneal Mesothelial Cells

Abstract:To investigate the effect of Puerarin on the oxidative stress in cultured human peritoneal mesothelial cell.human peritoneal mesothelial cells oltained from patients who underwent surgical operation were cultured in vitro.At first,cells from the third passage were incubated with RPM I1640 culture medium containing 0.1% FBS for 24 hours.Then,they were divided into five groups(RPMI1640 culture medium as control,PDS containing 2.5% glucose as PDS group,and PDS containing 2.5% glucose and 100μg/ml,200μg/ml or 400μg/ml Puerarin as group A,group B and group C).Each group was supplemented with equal volume of RPMI1640 culture medium containing 20% FBS.After 48h incubating,MDA,SOD,and GSH in supernatant of the cell culture medium were detected and compared.Significantly lower level of MDA in supernatant was found in control group(P0.01),as well as in groups A,B and C(all P0.05),as compared with that in PDS group.And,higher level of SOD and GSH were in control group(P0.01) and groups A,B and C(all P0.05),than those in PDS group.Conclusion: Puerarin can reduce the oxidative stress induced by commercial PDS in cultured human peritoneal mesothelial cell.
