Abstract:The conditions for extracting polysaccharides from tea (Camellia sinensis L.) fruit peel (TFPPs) were studied. Three parameters (temperature, time, and liquid/solid ratio) affecting the extraction of TFPP were optimized using response surface methodology (RSM). Under the optimized conditions, the yield of TFPP was predicted to be 4.98%. The physicochemical properties, in vitro antioxidant activities, and inhibitory effects on α-glucosidase of fractionated TFPPs (TFPP-0, TFPP-20, TFPP-40, and TFPP-60) were investigated. We found that the TFPPs were all acid protein-bound heteropolysaccharides, although with different chemical compositions. They had not only remarkable scavenging activity on 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) and reducing activity, but also excellent inhibitory potential against α-glucosidase in vitro. Our results suggest that tea fruit peel could be treated as a potential bioresource for the development of polysaccharide antioxidants.
Cytotoxicity and polyol pathway inhibitory activities of chemical constituents isolated from the pericarp of Toona sinensis

Abstract:Toona sinensis is a medicinal and edible plant that belongs to the genus Toona of family Meliaceae. Phytochemical investigations carried out on this plant, seven apotirucallane-type triterpenoids (1–7), two cycloartane-type triterpenoids (8–9), four sterols (10–13), two sesquiterpenes (14–15), four phenols (16–19), and one lignin (20) were isolated from the pericarp of T. sinensis by silica gel column and preparative middle pressure liquid chromatography. Their structures were identified by interpretation of NMR and comparison with those reported in the literature. Compounds 11–12, 15–16, and 18 were isolated from the family Meliaceae, compounds 13–14 were obtained from the genus Toona, and compound 19 was obtained from T. sinensis for the first time. Additionally, the cytotoxicity and polyol pathway (PP) inhibitory activities of active constituents were evaluated in rat glomerular mesangial cells cultured under high glucose conditions, suggesting their potential application for a PP inhibitor.Graphical

Abstract:In this study, bioassay-guided fractionation of extracts from the leaves of Morus alba L led to the isolation of 15 bioactive constituents with alpha-glucosidase and tyrosinase inhibitory activities, among which prenylated stilbenes were proved to be a new group of alpha-glucosidase inhibitors apart from iminosugars derived from Morus alba. Their structures were identified on the basis of extensive spectroscopic analysis and chemical evidence, as well as comparing with data from the literature. Among them, compounds (2R)/(2S)-Euchrenone a(7) (6a/6b), Chalcomoracin (7), Moracin C(8), Moracin D (9) and Moracin N (10) exhibited a significant degree of alpha-glucosidase inhibitory activity with IC50 of 6.28, 2.59, 4.04, 2.54 and 2.76 mu M, respectively, while (2R)/(2S)-Euchrenone a(7) (6a/6b), Moracin N (10), Quercetin (13), Norartocarpetin (14), the interconvertible epimeric mixture of (2R)/(2S)-7-methoxyl-8-ethyl-2',4'-dihydroxylflavane-2"-O-beta-D-glucopyranoside (1a/1b) and the interconvertible enantiomers of (2R)/(2S)-7-methoxyl-8-hydroxyethyl-2',4'-dihydroxylflavane (5a/5b) displayed a potent tyrosinase inhibitory effect with IC50 of 0.260, 0.924, 0.523, 0.0824, 0.616 and 0.528 mu M, respectively. Especially, (2R)-7-methoxyl-8-ethyl-2',4'dihydroxylflavane-2"-O-beta-D-glucopyranoside (1a), (2S)-7-methoxyl-8-ethyl-2',4'-dihydroxylflavane-2"-O-beta-D-glucopyranoside (1b), (2S)-8-hydroxyethy1-7,4'-dimethoxylflavane-2'-O-beta-D-glucopyranoside (2), (2R)-7-methoxyl-8-hydroxyethyl-2',4'-dihydroxylflavane (5a) and (2S)-7-methoxyl-8-hydroxyethyl-2',4'-dihydroxylflavane (5b) were identified as new compounds. Abstract:Pueraria lobata (Willd.) Ohwi (Gegen) is a nutritious medicine food homology plant that has been recorded in many ancient remedies for diabetes. However, its hypoglycemic active components and related mechanisms have not been fully elucidated. In this study, potential alpha-glucosidase inhibitors in Pueraria lobata were investigated by liquid-liquid refined extraction, high-resolution profiling and their hypoglycemic activity were evaluated. Ten compounds showing alpha-glucosidase inhibitory bioactivity were separated and identified, including two new compounds, 8-geranyl-7, 3', 4' -trihydroxy-isoflavone (4) and 8-geranyl-7, 2', 4'-trihydroxy-isoflavone (6), and eight known compounds. All screened compounds showed potential hypoglycemic activity, in which compounds 4, 6, 7, 8, 9 and 10 showed strong inhibitory activity (IC50 < 8 mu M). Furthermore, molecular docking simulation revealed binding energy and interacting amino acid residues of the screened compounds with alpha-glucosidase, strongly supporting their potential as alpha-glucosidase inhibitors.

Abstract:Backgrounds Tannic acid (TA), as a plant-derived phenolic substance, is involved in regulating the activity of starch digestive enzymes, but its underlying mechanism remains unclear. Methods and Results In the present study, inhibition rate and inhibition kinetics assays were performed and confirmed that TA had a strong inhibitory effect on both alpha-amylase and alpha-glucosidase with IC50 values of 0.1585 mg/mL and 0.00542 mg/mL, respectively, through a mixed inhibition mode. The secondary structures of both enzymes were confirmed to be modified by TA through circular dichroism (CD) spectra. Fluorescence quenching analysis revealed that the interaction between TA and two enzymes was a static process of pontaneous complex formation. Finally, molecular docking revealed that non-covalent bonds were the main interaction forces between TA and both enzymes. Conclusions Thus, TA was a promising candidate for the inhibition of starch-digesting enzymes, and the present research provided insight into postprandial glucose regulation through polyphenols.
Terpenoids from the Seeds of Toona sinensis and Their Ability to Attenuate High Glucose-Induced Oxidative Stress and Inflammation in Rat Glomerular Mesangial Cells

Abstract:Toona sinensis (A. Juss.) Roem is an edible medicinal plant that belongs to the genus Toona within the Meliaceae family. It has been confirmed to display a wide variety of biological activities. During our continuous search for active constituents from the seeds of T. sinensis, two new acyclic diterpenoids (1–2), together with five known limonoid-type triterpenoids (3–7), five known apotirucallane-type triterpenoids (8–12), and three known cycloartane-type triterpenoids (13–15), were isolated and characterized. Their structures were identified based on extensive spectroscopic experiments, including nuclear magnetic resonance (NMR), high-resolution electrospray ionization mass spectra (HR-ESI-MS), and electronic circular dichroism (ECD), as well as the comparison with those reported in the literature. We compared these findings to those reported in the literature. Compounds 5, 8, and 13–14 were isolated from the genus Toona, and compounds 11 and 15 were obtained from T. sinensis for the first time. The antidiabetic nephropathy effects of isolated compounds against high glucose-induced oxidative stress and inflammation in rat glomerular mesangial cells (GMCs) were assessed in vitro. The results showed that new compounds 1 and 2 could significantly increase the levels of Nrf-2/HO-1 and reduce the levels of NF-κB, TNF-α, and IL-6 at concentrations of 30 μM. These results suggest that compounds 1 and 2 might prevent the occurrence and development of diabetic nephropathy (DN) and facilitate the research and development of new antioxidant and anti-inflammatory drugs suitable for the prevention and treatment of DN.

[Bioactivity Guided Isolation of Alpha-Glucosidase Inhibitor from Whole Herbs of Crossostephium Chinense].

Abstract:OBJECTIVE To investigate the chemical constituents of 70% aqueous ethanol extract from the whole plant of Crossostephium chinense for inhibitory activity against alpha-glucosidase. METHOD A bioactivity-guided isolation and purification process was used to identify the alpha-glucosidase activity inhibiting components of the whole plant of C. chinense. The dried whole plants were extracted with 70% aqueous ethanol. The extract was suspended in water and then further fractionated successively with cyclohexane, ethyl acetate, and normal butanol,and tested for their inhibitory activity against alpha-glucosidase in vitro. The chemical structures of isolated compounds were determined by spectroscopic methods including NMR and MS, and comparison with data of authentic samples. All of compounds were assayed for their inhibitory activity against alpha-glucosidase in vitro. RESULT The ethyl acetate and water layer fractions showed the strong inhibitory activity, and were subjected to column chromatography over the various stationary phases. Tricetin 3',4',5'-trimethylether (1), scopoletin (2), tanacetin, hispidulin (3), apometzgerin (4), chrysoeriol (5), quercetagetin 3,6, 7-trimethylether (6), selagin (7) , scopolin (8), and quercetagetin-3,6-dimethylether (9) were isolated and characterized by different spectroscopic techniques. Among them, compounds 2, 3, 5-7 and 9 for inhibitory activity against alpha-glucosidase with IC50 (micromol x L(-1)) values of (34.36 +/- 2.06), (146.28 +/- 12.44), (246.26 +/- 8.73), (74.06 +/- 3.83), (42.19 +/- 5.25) and (136.20 +/- 25.73), respectively, were assayed as the active components. A positive drug, acarbose, showed the inhibitory activity against alpha-glucosidase with IC50 value of (489.25 +/- 38.55) mciromol x L(-1) in the same assay conditions with the above test com-1) pounds. The IC50 values of compounds 1, 4, 8 and tanacetin were all more than 1 000 micromol x L(-1). CONCLUSION The compounds 5 and 9 were isolated from the genus Crossostephium for the first time. The compounds 2, 3, 5-7, and 9 showed the strong inhibitory activity against alpha-glucosidase in vitro in the simple competitive manner. The results indicated that these compounds may be involved in the treatment of diabetes in the whole plant of C. chinense for human.

Abstract:Ampelopsis grossedentata is a valuable medicinal and edible plant, which is often used as a traditional tea by the Tujia people in China. A. grossedentata has numerous biological activities and is now widely used in the pharmaceutical and food industries. In this study, two new flavonoids (1–2) and seventeen known compounds (3–19) were isolated and identified from the dried stems and leaves of A. grossedentata. These isolated compounds were characterized by various spectroscopic data including mass spectrometry and nuclear magnetic resonance spectroscopy. All isolates were assessed for their α-glucosidase inhibitory, antioxidant, and hepatoprotective activities, and their structure–activity relationships were further discussed. The results indicated that compound 1 exhibited effective inhibitory activity against α-glucosidase, with an IC50 value of 0.21 μM. In addition, compounds 1–2 demonstrated not only potent antioxidant activities but also superior hepatoprotective properties. The findings of this study could serve as a reference for the development of A. grossedentata-derived products or drugs aimed at realizing their antidiabetic, antioxidant, and hepatoprotective functions.

Identification and Isolation of α-Glucosidase Inhibitors from Siraitia grosvenorii Roots Using Bio-Affinity Ultrafiltration and Comprehensive Chromatography

Abstract:The discovery of bioactive compounds from medicinal plants has played a crucial role in drug discovery. In this study, a simple and efficient method utilizing affinity-based ultrafiltration (UF) coupled with high-performance liquid chromatography (HPLC) was developed for the rapid screening and targeted separation of α-glucosidase inhibitors from Siraitia grosvenorii roots. First, an active fraction of S. grosvenorii roots (SGR2) was prepared, and 17 potential α-glucosidase inhibitors were identified based on UF-HPLC analysis. Second, guided by UF-HPLC, a combination of MCI gel CHP-20P column chromatography, high-speed counter-current countercurrent chromatography, and preparative HPLC were conducted to isolate the compounds producing active peaks. Sixteen compounds were successfully isolated from SGR2, including two lignans and fourteen cucurbitane-type triterpenoids. The structures of the novel compounds (4, 6, 7, 8, 9, and 11) were elucidated using spectroscopic methods, including one- and two-dimensional nuclear magnetic resonance spectroscopy and high-resolution electrospray ionization mass spectrometry. Finally, the α-glucosidase inhibitory activities of the isolated compounds were verified via enzyme inhibition assays and molecular docking analysis, all of which were found to exhibit certain inhibitory activity. Compound 14 exhibited the strongest inhibitory activity, with an IC50 value of 430.13 ± 13.33 μM, which was superior to that of acarbose (1332.50 ± 58.53 μM). The relationships between the structures of the compounds and their inhibitory activities were also investigated. Molecular docking showed that the highly active inhibitors interacted with α-glucosidase through hydrogen bonds and hydrophobic interactions. Our results demonstrate the beneficial effects of S. grosvenorii roots and their constituents on α-glucosidase inhibition.

Abstract:In the present study, polysaccharides named ALPS and ASPS were isolated from Ampelopsis grossedentata leaves and stems, respectively. Physicochemical properties, in vitro antioxidant activities and the inhibitory effects on alpha-glucosidase of ALPS and ASPS were investigated. It was found that both ALPS and ASPS were acid protein-bound heteropolysaccharides, although with considerably different chemical composition and molecular weight distribution. Meanwhile, in comparison with ALPS, ASPS exhibited stronger antioxidant activity and inhibitory potential against alpha-glucosidase according to the in vitro evaluation. Moreover, our results suggested that protein and uronic acid might, at least partly, contribute positively to the biological behavior of ALPS and ASPS.
Identification, potency evaluation, and mechanism clarification of α-glucosidase inhibitors from tender leaves of Lithocarpus polystachyus Rehd

Abstract:Lithocarpus polystachyus Rehd. known as Sweet Tea in China has attracted lots of interest for its good hypoglycemic effect and the potential as a hypoglycemic agent. Based on affinity separation-UPLC-Q-TOF-MS/MS, 54 potential α-glucosidase inhibitiors were identified and 44 were structurally determined. Out of them, 41 were identified for the first time from this plant including flavonoids, fatty acids, triterpenes, alkaloids, and coumarins. Enzyme assays revealed that flavonoids exhibited higher inhibitory activity against α-glucosidase than others with astilbin (IC50 = 6.14 μg·mL-1), morin (IC50 = 8.46 μg·mL-1), and naringenin (IC50 = 10.03 μg·mL-1) showing 2- to 4-fold higher potency than the positive control acarbose. They were proved as reversible inhibitors with mixed inhibition mechanism. Ki (Ki') values and molecular dockings strongly supported the potency order of astilbin, morin and naringenin that showed in the enzyme assays.

Abstract:Eleven undescribed diarylheptanoids, amomutsaokols A‒K (1–11), together with 13 known ones (13–24), were isolated from the active fraction of the fruits of Amomum tsao-ko. The structures of the undescribed compounds were determined by extensive 1D and 2D NMR, HRESIMS and ECD calculations. Compounds 3–5, 7, 8, 12, 14 and 19 showed obviously α-glucosidase inhibitory activity with IC50 values ranging from 12.9 to 48.8 μM. An enzyme kinetic analysis indicated that compounds 8 and 9 were α-glucosidase noncompetitive inhibitors with Ki values of 18.5 and 213.0 μM, respectively. This study supported diarylheptanoids as the active constituents of A. tsao-ko with α-glucosidase inhibitory effects.

Abstract:Eight α-glucosidase inhibitors including four new compounds were isolated from the 70% aqueous ethanolic extract of leaves of Aquilaria sinensis (Lour.) Gilg by activity-directed fractionation and purification processes. The ethanolic extract was first separated into petroleum ether, ethyl acetate, n-butanol and water soluble fractions and screened for inhibitory activity against α-glucosidase. Further activity-directed investigation lead to the isolation of four new compounds with moderate inhibitory activity, viz, aquilarisinin (1), aquilarisin (2), hypolaetin 5-O-β-d-glucuronopyranoside (3) and aquilarixanthone (4) from the n-butanol fraction, and four known compounds showing potent activity including mangiferin (5), iriflophenone 2-O-α-l-rhamnopyranoside (6), iriflophenone 3-C-β-d-glucoside (7) and iriflophenone 3,5-C-β-d-diglucopyranoside (8) from the most potent ethyl acetate fraction. The structures of these compounds were determined by extensive spectroscopic analyses, including IR, UV, ESIMS, HRESIMS, 1D and 2D NMR.

Abstract:A new triterpene glycoside, 3-O-[(alpha-L-rhamnopyranosyl)(1 -> 2)]-[beta-D-glucuronopyranosyl-6-O-methyl ester]-olean-12-ene-28-olic acid (1) and a new indole alkaloid, 5-methoxy-2-oxoindolin-3-acetic acid methyl ester (5) were isolated from the leaves of Acanthopanax senticosus Harms along with six known compounds. The structures of the new compounds were determined by means of 2D-NMR experiments and chemical methods. All the isolated compounds were evaluated for their glycosidase inhibition activities and compound 6 showed significant alpha-glucosidase inhibition activity.
Anti-α-Glucosidase Activities and Bioactive Components of Tibetan Hull-Less Barley Bran Extracts

Abstract:The hexane,ethyl acetate and methanol extracts of Tibetan hull-less barley (THB) bran displayed dose-dependent inhibitory activities on α-glucosidase in vitro.The hexane and ethyl acetate extracts had higher inhibitory activities than did acarbose,which is used in clinical treatment of type 2 diabetes.Gas chromatography-mass spectrometry (GC-MS) analysis showed that linoleic acid,oleic acid,palmitic acid and α-linoleic acid were the main fatty acids in the hexane extracts,accounting for 96.5％ of the total fatty acids.All of these four fatty acids have been reported to be potent inhibitors of α-glucosidase.Moreover,65 peaks were separated from the unsaponifiable fractions,with 17 of them identified as sterols,triterpenes and long chain fatty alcohols,respectively.The total free phtosterol content was 7 357.72 mg/kg,which presented β-sitosterol,campesterol,desmosterol,stigmasterol,and fucosterol as the primary phytosterols.In addition,β-sitosterol and campesterol accounted for 83％ of the total phtosterols.These results show that THB can be a natural source of potent α-glucosidase inhibitors.

Abstract:α-Glucosidase plays an important role in catalyzing the hydrolytic cleavage of disaccharides into monosaccharides. In this study, a phytochemical investigation of the potential α-glucosidase inhibitory fraction from the aerial parts of Euonymus fortunei led to the isolation and identification of two new tetracyclic triterpenoids, fortunenones A and B (1-2), together with 11 known triterpenoids (3-13). Fortunenones A and B are rare C32 triterpenoids possessing a 24,24-dimethyl group. The partial isolated compounds were evaluated their effects on α-glucosidase, of which echinochlorin D (5), lupenone (7), wilforlide B (12), and wilforlide A (13) exhibited remarkable inhibitory effects with the half inhibitory concentration ranged from 207.2 × 10-6 M to 388.3 × 10-6 M compared with the positive control, acarbose. An enzyme kinetics analysis by Lineweaver-Burk plots revealed that the inhibition types of the four active compounds were all mixed inhibition. Molecular docking further revealed that hydrophobic interactions and hydrogen bonds play an important role in the inhibition of α-glucosidase activity. Our results demonstrate the potential of E. fortunei extract and its constituents to inhibit α-glucosidase.

Comprehensive Screening and Characterization of α-glucosidase Inhibitory Components in the Edible Medicinal Plant Pholidota cantonensis Rolfe Using UPLC-Q-TOF-MS/MS Analysis and Molecular Docking

Abstract:Pholidota cantonensis Rolfe is an edible medicinal plant in the genus Pholidota of the family Orchidaceae. This plant is used to prepare medicated food in China and has been reported to possess anti-α-glucosidase activity. To date, little is known about the active substances responsible for the observed anti-α-glucosidase activity. In the present study, we aimed to screen and characterize the α-glucosidase inhibitory fraction of P. cantonensis using ultraperformance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS/MS) analysis and molecular docking. As a result, the 50% ethanol fraction obtained from D101 macroporous adsorption resin column chromatography (D50 fraction) had the highest total phenol content (353.83 ± 6.06 mg GAE/g) and the most prominent α-glucosidase inhibitory activity (IC50 = 30.01 ± 7.30 µg/mL). Forty-five compounds were identified from the D50 fraction by using UPLC-Q-TOF-MS/MS analysis. Molecular docking results showed that six main constituents, namely, crepidatin, 2,7-dihydroxy-4-methoxyl-9,10-dihydrophenylene, 4,4',5,6-tetrahydroxystilbene, 4,7-dihydroxy-2-methoxyl-9,10-dihydrophenylene, (-)-lariciresinol, and thunalbene, in the D50 fraction occupied the catalytic sites of α-glucosidase through strong hydrophobic interactions, hydrogen bonding, and other patterns. The binding energies were between - 29.95 and - 11.41 kJ/mol, indicating good binding between the tested compounds and α-glucosidase. The active ingredients responsible for the α-glucosidase inhibitory activity may include phenanthrenes, stilbenes, dibenzyls, and lignans. The D50 fraction has potential value for developing innovative drugs for the prevention and treatment of diabetes mellitus (DM) and is worthy of in-depth research.
Structural Identification of Compounds from Toona Sinensis Leaves with Antioxidant and Anticancer Activities

Abstract:The aim of this study was to isolate key active compounds from Toona sinensis leaves (TSL) with antioxidant and anticancer activities. Four compounds, quercetin-3-O-α-L-rhamnopyranoside (QLR), kaempferol-3-O-α-L-rhamnopyranoside (KLR), 1,2,3,4,6-penta-O-galloyl-β-D-glucopyranose (PGG) and ethyl gallate (EG), were purified from the ethyl acetate fraction of TSL by column chromatography. Their structures were identified by mass spectrometry (MS) and nuclear magnetic resonance (NMR) spectrometry analyses. All four compounds exhibited strong antioxidant activities (including ABTS radical scavenging activities and ORAC values). PGG showed the strongest antioxidant activity. KLR showed the strongest anticancer activities against human hepatoma HepG2 and human breast cancer MCF-7 cells. QLR, PGG, and EG showed relatively weak anticancer activities. These results suggest that four compounds are key chemical compositions valuable for preparing functional foods in the food industry.
Natural α-glucosidase inhibitors from Aquilaria sinensis leaf-tea: Targeted bio-affinity screening, identification, and inhibition mechanism

Abstract:Aquilaria sinensis leaves have long been consumed as a popular replacement tea for lowering postprandial blood glucose levels, but their specific functional components remain unclear. In this study, Aquilaria sinensis leaf-tea 70 % ethanol extract (ALTE) exhibited excellent anti-α-glucosidase activity (IC50 = 6.93 ± 1.91 μg/mL) and promoted glucose consumption ability in 3 T3-L1 preadipocyte cells. Subsequently phenolic compositions of ALTE were identified for the first time. After that, five potential α-glucosidase inhibitors (α-GIs) including cynaroside-3,5-diglucose, malvidin 3-glucose, epicatechin, epigallocatechin gallate, and dihydromyricetin in ALTE were screened using a targeted bio-affinity ultrafiltration-HPLC/MS method. Moreover, these five α-GIs all showed good anti-α-glucosidase effects and glucose consumption-promoting ability. Furthermore, the binding properties and inhibition mechanisms of five α-GIs to α-glucosidase were further analyzed via enzyme inhibition kinetics, molecular docking, and molecular dynamics simulation. This study confirms that Aquilaria sinensis leaf-tea is effective in preventing post-hyperglycemia in vitro models, suggesting potential for future research in human trials.
The chemical composition, antioxidative and antimicrobial potentials of Toona sinensis essential oils from different regions of China

Abstract:Toona sinensis is one of the traditional Chinese medicinal materials with rich physiological activities. This study aimed to investigate the chemical composition differences of the essential oils from the tender leaves and shoots of T. sinensis , collected from four main producing areas in China, and to verify their anti-listeria and antioxidant properties. Gas chromatography-mass spectrometry (GC-MS) analysis was conducted on the essential oils obtained by hydro-distillation. The results indicated that compounds such as β-elemene (3.5∼8.1%), α-caryophyllene (3.7∼7.2%), and trans -caryophyllene (4.0∼9.7%) were the main common components of T. sinensis essential oils (TSEO) from four producing areas. The results of DPPH and ABTS showed that the essential oil from Dazhou, Sichuan, China had the best antioxidant effect. At a concentration of 1.25 mg/mL, the antioxidant activity of TSEO-DZ from Sichuan was comparable to 0.08 mg/mL of BHT and was only slightly lower than 0.06 mg/mL of VC. The broth dilution method showed that the essential oil from Tai 'an, Shandong, China, showed better antibacterial effect, with the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MLC) values of 1.25 μL/mL and 2.54 μL/mL, respectively. This study demonstrates that TSEO possesses promising application potential in antioxidant and anti-listeria activities, and it confirms that the growth environment of T. sinensis significantly influences the quality, content, and biological activity of the essential oil.
