Abstract:A simple and effective high-performance liquid chromatographic (HPLC) method has been developed for simultaneous quantification of three phenolic acids (3,4-dihydroxyphenyllactic acid (Chinese name danshensu), protocatechuic aldehyde, and salvianolic acid B) and four diterpenes (dihydrotanshinone I, cryptotanshinone, tanshinone I, and tanshinone IIA) in radix salviae miltiorrhizae. Chromatography was performed on a 250 mm × 4.6 mm i.d., 5-μm particle size, C18 column. The mobile phase was a linear gradient prepared from 0.1% (v/v) aqueous formic acid and acetonitrile at a flow-rate of 1.0 mL min−1. All the target components were well separated with high resolution and without interference. Good linearity (R 2 > 0.999) was observed over the concentration ranges investigated, and intra-day and inter-day precision were high. Temperature-controlled ultrasound-assisted extraction was used to prevent hydrolysis of thermally unstable components during the sample-extraction procedure, and the extraction conditions were carefully optimized. Recovery of the seven components was from 98.45 to 100.63% and relative standard deviations were always <1.5%. The validated method was successfully used for simultaneous quantification of the three phenolic acids and the four diterpenes in radix salviae miltiorrhizae of different geographic origins.
Simultaneous Chemical Fingerprinting and Quantitative Analysis of Crude and Processed Radix Scrophulariae from Different Locations in China by HPLC.

Abstract:A validated liquid chromatography method was first developed to evaluate the quality of crude and processed Radix Scrophulariae extracts through establishing chromatographic fingerprint and simultaneous determination of five bioactive compounds, namely 5-hydroxymethylfurfural (5-HMF), acteoside, angroside C, harpagoside and cinnamic acid. The chromatographic were separated on an Agilent Zorbax Extend C(18) column (250 mm × 4.6 mm, 5 μm) and detected by diode array detector (DAD). Mobile phase was composed of (A) aqueous phosphoric acid (0.03%, v/v) and (B) acetonitrile using a gradient elution. Analytes were performed at 30 °C with a flow rate of 1.0 mL/min and UV detection at 280 nm. All calibration curves showed good linear regression (r(2) ≥0.9996) within the tested ranges, and the recovery of the method was in the range of 98.12-103.38%, with RSD values ranging from 0.6 to 2.8%. In addition, the contents of those five bioactive compounds in crude and processed Radix Scrophulariae prepared by different locations of China were determined to establish the effectiveness of the method. The results demonstrate that the developed method is accurate and reproducible and could be readily utilized as a suitable quality control method for the quantification of Radix Scrophulariae.

Abstract:OBJECTIVE:To develop an HPLC method to determine the contents of danshensu, hydroxysafflor yellow A, rosmarinic acid, lithospermic acid, salvianolic acid B in the water extract of mixed Salviae Miltiorrhizae Radix et Rhizoma and Carthami Flos simultaneously.METHOD:The separation were carried out at 30 degrees C on a ZORBAX Eclipse Plus C18 column (4.6 mm x 100 mm, 1.8 microm) with formic acid-500 mmol x L(-1) ammonium formate-water solution (0.5:10:90) as mobile phase A and acetonitrile-formic acid solution (100: 0.5) as mobile phase B in gradient mode at a flow rate of 0.5 mL x min(-1). Detection wavelengths were 280 nm for danshensu, rosmarinic acid, lithospermic acid, salvianolic acid B, and 380 nm for hydroxysafflor yellow A.RESULT:The 5 components were separated well with a good linearity (R2 > 0.999 3) in the range of the test concentration. The average recoveries of danshensu, hydroxysafflor yellow A, rosmarinic acid, lithospermic acid, and salvianolic acid B were 99.1%, 102%, 102%, 98.5% and 101%, respectively.CONCLUSION:This method is simple, accurate, and repeatable.

Abstract:A gradient reversed-phase HPLC method was developed for the simultaneous separation and determination of rosmarinic acid, lithospermic acid B, caffeic acid, and other phenolic compounds. This method showed a satisfactory result. The limits of detection for these phenolic compounds were less than or equal to 0.05 mg/L. The developed method was applied to the determination of phenolic compounds in the dried roots of Salvia miltiorrhiza (Danshen) and the transformed cells of S. miltiorrhiza. In the transformed cells, rosmarinic acid was the predominant phenolic compound. Small amounts of lithospermic acid B and caffeic acid were also detected. In the dried roots, in contrast, lithospermic acid B was the main phenolic compound and only a very small amount of rosmarinic acid was found. In addition, danshensu and some unidentified phenolic compounds were also detected from the dried roots.
Simultaneous Determination of Five Hydrophilic and Lipophilic Active Components in Radix Et Rhizoma Salviae Miltiorrhizae by HPLC

Abstract:Objective: To develop a new method for simultaneous determination of two hydrophillc components and three lipophillc components of Radix et Rhizoma Salviae Miltiorrhizae.Method:The HPLC-double wavelength detection was employed using a column of Dikma Diamonsil C18(4.6 mm × 250 mm,5 μm) with a mobile phase of acetonitrile(A)-0.05% phosphoric acid(B).The gradient elution program was as follows:0-27 min,20%-25% A;27-55 min,25%-80% A;55-65 min,80%-80% A.The detection wavelength was set at 270 nm and 286 nm.The column temperature was at 30 ℃.Result:Rosmarinic acid was linear in the range of 1.92 × 10-2-0.96 μg;salviamolic acid B was linear in the range of 7.29 × 10-2-3.64 μg;clyptotanshinon was linear in the range of 3.05 × 10-3-0.15 μg;tanshinone was linear in the range of 1.71 × 10-3-8.55 × 10-2 μg and tanshinone ⅡA was linear in the range of 5.80 × 10-3-0.29 μg.The average recovery was 99.88%,104.75%,101.08%,101.41%,102.41% respectively.Conclusion:The present method was simple and had satisfactory efficacy;it can simultaneously determine multiple hydrophilic and lipophilic bioactive components in Salvia miltiorrhiza from different areas.
Comprehensive characterization of active components in Salvia miltiorrhiza using polarity-partitioned two-dimensional liquid chromatography coupled with mass spectrometry

Abstract:Salvia miltiorrhiza, a widely used traditional Chinese medicine, contains a complex matrix of hydrophobic diterpenoids and hydrophilic phenolic acids, presenting significant challenges in comprehensive analysis. In this study, an online polarity-partitioned two-dimensional liquid chromatography coupled with mass spectrometry (2D-LC-MS) method was developed for comprehensive analysis of both lipophilic and hydrophilic active components in Salvia miltiorrhiza. The method integrated hydrophilic interaction liquid chromatography (HILIC) and reversed-phase liquid chromatography (RPLC), facilitating the efficient separation of compounds across a wide range of polarities. An online dilution strategy was implemented, minimizing sample loss and enhancing the method's utility for quality control and chemical characterization of complex herbal matrices. Compared with other LC methods, this approach significantly improved analyte coverage, resolution, and analysis efficiency. Under optimal conditions, 150 active components were successfully identified, including 33 compounds newly discovered in Salvia miltiorrhiza. Additionally, the validated online method was applied to the quantitative determination of 16 quality markers of Salvia miltiorrhiza from different sources. The results demonstrated the online method's potential as a superior alternative to existing techniques, offering broader applicability in traditional Chinese medicine research.

Abstract:AIMS:This is a pharmacokinetic study of Salviae miltiorrhizae and ligustrazine hydrochloride injection. The study aimed to evaluate the mechanism of action, safety and rational clinical use of Salviae miltiorrhizae and ligustrazine hydrochloride injection.BACKGROUND:Salviae miltiorrhizae and ligustrazine hydrochloride injection is a compound preparation consisted of Salvia miltiorrhiza extract and ligustrazine hydrochloride for the treatment of cardiovascular and cerebrovascular diseases in China.OBJECTIVE:The study aimed to develop a rapid and sensitive high-performance liquid chromatography-diode array detector-tandem mass spectrometry (HPLC-DAD-MS/MS) method for simultaneous determination of six major active ingredients of Salviae miltiorrhizae and ligustrazine hydrochloride injection, namely danshensu, protocatechuic aldehyde, rosmarinic acid, lithospermic acid, salvianolic acid A, and ligustrazine hydrochloride, in rat plasma.METHODS:Plasma samples were precipitated with methanol, which was spiked with ascorbic acid and the supernatant was separated on a Waters Cortecs C18 column, by using a gradient mobile phase system of acetonitrile-water containing 0.05% formic acid (v/v). For internal standards, puerarin was selected for the five salvianolic acids, while isofraxidin was used for ligustrazine hydrochloride. Besides, electrospray ionization in negative mode and multiplereaction monitoring were used to identify and quantify the five salvianolic acids, whereas ligustrazine hydrochloride was quantified at 310 nm using the diode array detector.RESULTS:Noticeably, all calibration curves showed good linearity (R2>0.99) over the concentration range, with a lower limit of quantification between 0.00411 and 0.0369 μg/mL for salvianolic acids, and 1.74 μg/mL for ligustrazine hydrochloride. Next, the precision of the developed method was evaluated by intra- and inter-day assays, and the percentage of relative standard deviation was within 10%. Although the extraction efficiency of some salvianolic acids was not very satisfactory, the sensitivity of the analytical method met the analysis requirements of rat plasma samples. Moreover, the validated method was successfully applied to a pharmacokinetic study of Salviae miltiorrhizae and ligustrazine hydrochloride injection in the rat model.CONCLUSION:Linear pharmacokinetic characteristics were observed for the six active ingredients after intravenous infusion administration in rats within the dose range examined here. In summary, our study proposed a HPLC-DADMS/ MS method with the simultaneous determination of multiple ingredients, and demonstrated its applicability in pharmacokinetic studies.

Abstract:Salvia miltiorrhiza Bunge (Chinese common name Danshen) is one of the most commonly used traditional medicinal materials for treatment of cardiovascular and cerebrovascular diseases in China which was abundant with bioactive ingredients such as diterpenoid quinones and hydrophilic phenolic acids. With the continuous increasing demand, cultivation has become the major form of resource supply. However, S. miltiorrhiza tends to form amounts of fibrous roots under drought and sandy environment with less research and utilization. Herein, a sensitive and reliable combinatorial ultra performance liquid chromatography analysis by integrating the characteristic ultraviolet spectra and optimum absorption wavelengths based on external standards was developed. And seventeen bioactive ingredients in the fibrous roots of S. miltiorrhiza were simultaneous accurate qualitative and quantitative with ideal separation and stability. Analysis show that the chemical constituents of the fibrous roots were similar to those in the roots and rhizomes of S. miltiorrhiza. Moreover, 12 out 17 tested constituents's contents in the fibrous roots were even higher than the latter. In conclusion, the work developed an efficient and precise combinatorial quantitative analysis method and provided insight into the chemical constituents and development of the fibrous roots of S. miltiorrhiza.

Abstract:In this study, a novel infrared-assisted extraction (IRAE) method, in which infrared radiation was employed to extract the active compounds from plant, was developed and coupled with HPLC for simultaneous determination of four phenolic acids and four diterpenoids in Radix Salviae miltiorrhizae (Chinese name Danshen). The extraction conditions of IRAE were optimized and the optimal conditions were as follows: extraction time of 15 min; extraction solvent of 70% v/v methanol in water solution, and solid/liquid ratio of 0.1:15 (g/mL). Chromatography was performed on a 200 mm x 4.6 mm id, 5-mu m particle size, C18 column. Good linearity (r > 0.9994) was observed over the concentration ranges investigated, and intra-day and inter-day precision were high. Recoveries of the eight compounds were from 96.90 to 104.30% and RSD was below 2.5%. By using this novel IRAE method, with a shorter extraction time, the determined amounts of the eight active components in Danshen were comparable with or even higher than those extracted with conventional heat-reflux extraction and ultrasound-assisted extraction methods and microwave-assisted extraction. The developed IRAE method is simple, low-cost and efficient, offering a great promise for quick determination of active compounds in natural plants.
[Simultaneous Determination of Six Components in Water Extract and Alcohol Precipitation Liquid in Astragali Radix by HPLC-UV-ELSD].

Abstract:A HPLC-UV-ELSD method was established for simultaneous determination of six components in two intermediates of Shenqi Fuzheng injection (SFI) and the feasibility of establishing quantitative analysis of multi-components by single marker (QAMS) methods on different detectors was further explored. Calycosin-7-O-β-D-glucoside and astragloside Ⅳ were selected as internal reference substances for respectively flavonoids and saponins, and relative correlation factors (RCF) of formononetin-7-O-β-D-glucoside, 9, 10-dimethoxypterocarpan-3-O-β-D-glucopyranoside, 2'-dihydroxy-3', 4'-dimethoxyisoflavan-7-O-β-D-glucopyranoside and astragloside Ⅱ were calculated. Eventually, quantitative results of the 14 samples were compared between QAMS and external standard method. The sample concentrations calculated by QAMS were similar with concentrations calculated by external standard method, and the absolute values of relative deviations were generally less than 5% according to the UV detection of flavonoids. On the basis of ELSD detection for saponins, however, the absolute values of relative deviation of the two methods ranged from 0.48% to 23.17%. The QAMS method built on ultraviolet (UV) detectors was stable and can be used as a substitute method to reduce the consumption of standard compounds; meanwhile, the accuracy of QAMS method built on evaporative light scattering detector (ELSD) was inferior to that of external standard method, and the working principle of ELSD and feasible concentration range remain to be further studied.
A Liquid Chromatographic-Tandem Mass Spectrometric Method for the Quantitation of Eight Components Involved in Lithospermic Acid B Biosynthesis Pathway in Salvia Miltiorrhiza Hairy Root Cultures

Abstract:Elucidation of regulatory mechanisms of plant secondary metabolism requires quantitation of metabolic fluxes. A method of high-performance liquid chromatography-tandem mass spectrometry (LC-MS/MS) has been developed for the trace analysis of eight components (rosmarinic acid, lithospermic acid B, L-phenylalanine, t-cinnamic acid, 4-coumaric acid, L-tyrosine, 4-hydroxyphenylpyruvic acid and homogentisic acid) involved in lithospermic acid B biosynthesis pathway in Salvia miltiorrhiza hairy root cultures. The separation was performed by a ZORBAX SB-C-18 column (3.5 mu m, 2.1 x 150 mm, I.D. Agilent Corporation, MA, USA) and a C-18 guard column (5 mu m, 4.0 x 2.0 mm, Agilent Corporation, MA, USA) with an isocratic mobile phase consisting of acetonitrile-water (55: 45, v/v) at a flow rate of 0.3 ml/min. The components were detected by an Agilent G6410A triple quadrupole LC/MS system equipped with a MassHunter interface. The components were detected using electrospray ionization (ESI) in negative-ion mode and quantified by multiple-reaction monitoring (MRM) mode using the transition mass of m/z 359 -> 161, 717 -> 519, 164 -> 147, 147 -> 103, 163 -> 119, 180 -> 119, 179 -> 107 and 167 -> 123 for rosmarinic acid, lithospermic acid B, L-phenylalanine, t-cinnamic acid, 4-coumaric acid, L-tyrosine, 4-hydroxyphenylpyruvic acid and homogentisic acid, respectively. The recovery of the method was in the range of 95.4 to 105.5%, and all the components showed good linearity (r>0.995) in a relatively wide concentration range. This method has been successfully applied to the determination of the eight components in different elicitor (Methyl jasmonate and silver ions) treated S. miltiorrhiza hairy root cultures, for describing elicitor-based regulatory effect to lithospermic acid B metabolic fluxes.

Abstract:To evaluate the quality of Salvia miltiorrhiza Bunge, high-performance liquid chromatography-diode array detector (HPLC/UV-PAD), near infrared (NIR) spectroscopy, and chemometrics were used to discriminate nine components of samples from four different geographical locations. HPLC was performed with a C18 (5 μ m, 4.6 mm × 250 mm) column and 0.1% formic acid aqueous solution-acetonitrile with a gradient elution system. Orthogonal partial least squares discriminant analysis was used to identify the amounts of salvianolic acid B. NIR was used to distinguish rapidly S. miltiorrhiza Bunge samples from different geographical locations. In this assay, discriminant analysis was performed, and the accuracy was found to be 100%. The combination of these two methods can be used to quickly and accurately identify S. miltiorrhiza Bunge from different geographical locations.

Identification of Phenolic Constituents in Radix Salvia Miltiorrhizae by Liquid Chromatography/electrospray Ionization Mass Spectrometry.

Abstract:The phenolic components from Radix Salvia miltiorrhizae Bunge, a well-known herbal medicine (Dan-Shen in Chinese), have been investigated by high-performance liquid chromatography/electrospray ionization tandem mass spectrometry (HPLC/ESI-MS/MS). HPLC analyses were performed on a reversed-phase C-18 column using gradient elution. In the ESI mass spectra a predominant [M-H](-) ion was observed in negative mode and provided molecular mass information. ESI-MS/MS spectra of the [M-H](-) ions were used for structural analysis, based on the spectra of standards. It was found that caffeic acid and its monomeric analogs containing a carboxyl group readily lost CO2, while dimers, trimers and tetramers of caffeic acid expelled successively danshensu or caffeic acid or their esters. Twenty-eight phenolic compounds in S. miltiorrhizae were characterized, of which eight compounds were positively identified by comparison with standards. The remaining twenty phenolics for which standards were not available were tentatively identified based on their UV spectra and MS/MS fragmentation characteristics. Abstract:This paper describes an improved quality assessment method for Radix Salviae Miltiorrhizae (Root of Salvia miltiorrhiza BGE.) which was established using chromatographic fingerprinting and quantification of multiple marker compounds in the crude drug. High-performance thin-layer chromatography (HPTLC) fingerprinting of water-soluble phenolics and nonpolar tanshinones was performed separately and the authentication of Radix Salviae Miltiorrhizae was achieved by comparing the fingerprints of the samples with those of the reference crude drug and by comparing the Rf values of the bands in TLC fingerprints with those of reference compounds. HPLC fingerprints were obtained by simultaneous separation of phenolics and diterpenoids in Radix Salviae Miltiorrhizae. The HPLC fingerprints of seven batches of samples from different regions of China showed similar chromatographic patterns, and seven peaks were selected as characteristic peaks. The relative retention time of these characteristic peaks in the HPLC fingerprints was established as an important parameter for the identification of this herbal medicine. The pharmacologically active marker compounds salvianolic acid B, rosmarinic acid, and tanshinone IIA in herbal medicine were quantitatively determined using reverse-phase HPLC techniques. The HPLC quantitation methods of the three marker compounds were validated and the measurement uncertainty, which is important for setting the proposed content limit of the marker compounds in herbal medicine, were further evaluated.

Abstract:Phenolic acids are the major water-soluble components in Salvia miltiorrhiza (>5%). According to previous studies, many of them contribute to the cardiovascular effects and antioxidant effects of S. miltiorrhiza. Polymeric phenolic acids can be considered as the tanshinol derived metabolites, e.g., dimmers, trimers, and tetramers. A strategy combined with tanshinol-based expected compounds prediction, total ion chromatogram filtering, fragment ion searching, and parent list-based multistage mass spectrometry acquisition by linear trap quadropole-orbitrap Velos mass spectrometry was proposed to rapid profile polymeric phenolic acids in S. miltiorrhiza. More than 480 potential polymeric phenolic acids could be screened out by this strategy. Based on the fragment information obtained by parent list-activated data dependent multi-stage mass spectrometry acquisition, 190 polymeric phenolic acids were characterized by comparing their mass information with literature data, and 18 of them were firstly detected from S. miltiorrhiza. Seven potential compounds were tentatively characterized as new polymeric phenolic acids from S. miltiorrhiza. This strategy facilitates identification of polymeric phenolic acids in complex matrix with both selectivity and sensitivity, which could be expanded for rapid discovery and identification of compounds from complex matrix.
Multi-component HPLC Fingerprinting of Radix Salviae Miltiorrhizae and Its LC-MS-MS Identification.

Abstract:The multi-component fingerprinting method of Radix Salviae Miltiorrhizae (Root of Salvia miltiorrhiza BGE.), an important and popular medicinal herb in traditional Chinese medicine, was studied using reverse-phase HPLC and LC-MS-MS. Extract containing both the water-soluble phenolic compounds and nonpolar diterpenoid compounds known to be the herb's main bioactive components was prepared by a two-step extractive procedure. An HPLC fingerprinting method which can simultaneously separate these two types of compounds was established with gradient elution mode and photodiode array detection at 280 nm. Eighteen peaks in this HPLC fingerprint were structurally identified by employing LC-MS-MS techniques. The electrospray ionization (ESI) MS-MS spectra of most salvianolic acids displayed a characteristic behavior of loss of danshensu and caffeic acid moieties, while those of tanshinones showed a particular behavior of loss of H2O which is quite different from the fragmentation pattern in electron ionization mass spectrometry (EI-MS). The HPLC fingerprints of 7 batches of crude drugs showed similar separation pattern and provided much chemical information of the pharmacologically-active compounds in the crude drugs, which is useful for the authentication and quality evaluation of this medicinal herb.

Abstract:This article described establishment of improved quality assessment methods for Radix Salviae Miltiorrhizae (Danshen) by using chromatographic fingerprinting and quantitation of multiple marker compounds in this herb. HPTLC fingerprinting of phenolics and diterpeniods was performed separately to authenticate Radix Salviae Miltiorrhizae. HPLC fingerprints were obtained by simultaneous separation of both phenolics and diterpenoids in Radix Salviae Miltiorrhizae, and relative retention time of each characteristic peak was employed as an important parameter for the identification of this herbal medicine. Salvianolic acid B, rosmarinic acid and tanshinone IIA in this herb were quantitatively determined using HPLC analytical method. The quantitation methods were validated and the measurement uncertainty (MU) which is important for setting the proposed content limit of the marker compounds in this herbal, was evaluated. Furthermore, LC-MS-MS method was developed for the structural identification of the peaks in HPLC fingerprints and for the quantitative determination of four major diterpenoids in Radix Salviae Miltiorrhizae.

Abstract:A liquid chromatographic method was applied to determine trifolirhizin, kushenol K, kushenol L, kushenol N, kushenol X, kurarinone, norkurarinone, isokurarinone and kushenol A in the roots of Sophora flavescens, namely Kushen in China. The samples were separated on a YMC-C18 column (250 × 4.6 mm, 5 μm) with a gradient of methanol and 0.3% aqueous acetic acid (v/v) at a flow rate of 0.8 mL min−1 and detected at 295 nm. The complete separation was achieved within 45 min for the nine major flavonoids. All calibration curves expressed good linearity (r 2 > 0.999) within the test range. The recovery of this method was 92.3–106.9%. The assay was successfully applied to the quantification of nine flavonoids in 26 samples of Kushen. The results indicated that this developed LC assay could be readily utilized as a quality control method for the Chinese herb medicine Kushen.

Abstract:A simple high performance liquid chromatographic method has been developed for the determination of nine flavonoids in Herba Epimedii, including quercetin-3-O-glucoside, epimedin A, hexandraside A, epimedin B, epimedin C, icariin, icariside I, icariside II, and icaritin. The chromatographic separation was performed on a Diamonsil C(18) column (5 microm, 250x4.6 mm) with gradient elution by acetonitrile-0.5% acetic buffer (adjusted to pH 5.00 with triethylamine). Methodological validation gave acceptable linearities (r(2) >0.9992) and recoveries (ranging from 98.9 to 102.4%). The limits of detection of these flavonoids ranged from 16.3 to 83.3 ng. The results indicated that the contents of flavonoids in Herba Epimedii varied significantly from habitat to habitat with contents ranging from 0.05 to 39.0 mg/g. Twenty five Herba Epimedii samples prepared from five different botanical materials were investigated by the established method, and the results showed the influence of the botanical material and the habitat on the quality of Herba Epimedii. The proposed method is simple, effective, and suitable for the evaluation of this traditional Chinese medicine.

Abstract:An HPLC-ESI-MS-MS method has been developed for the quantitative determination of four diterpenoids, dihydrotanshinone I, cryptotanshinone, tanshinone I, and tanshinone IIA in Radix Salviae Miltiorrhizae (RSM, the root of Salvia miltiorrhiza B-GE.). The diterpenoids were chromatographically separated on a C18 HPLC column, and the quantification of these diterpenoids was based on the fragments of [M+H](+) under collision-activated conditions and in Selected Reaction Monitioring (SRM) mode. The quantitative method was validated, and the mean recovery rates from fortified samples (n=5) of dihydrotanshinone I, cryptotanshinone, tanshinone I, and tanshinone IIA were 95.0%, 97.2%, 93.1 %, and 95.9% with variation coefficient of 6.0%, 4.3%, 3.7%, and 4.2%, respectively. The established method was successfully applied to the quality assessment of seven batches of RSM samples collected from different regions of China.
