Biological characteristics of spermatogonial stem cells under defined culture environment with chinese traditional medicine

Abstract:Objective: To observe the biological characteristics of mouse spermatogonial stem cells (SSCs) under defined culture environment with Chinese traditional medicine, and provide some evidences for multipotential differentiation of SSCs. Methods: The male mice with 5-7 days age were executed by cervical vertebra disjointing, and their testes were isolated and collected for culture and identify of SSCs. After three days culture, SSCs were divided into two groups including the control group with complete medium of DMEM/F12(include: 10%FBS, 4mmol/L L-glutamine, 0.12%NaHCO3, 0.5 mmol/L sodium pyruvate, 15 mmol/L HEPES, 50u/ml penicillininverted commas 50u/ml streptomycin) and experiment group with defined medium (DMEM/F12 + Chinese traditional medicine). The morphological changes, proliferation and DNA content of SSCs were detected by inverted microscope and ELISA and FCM. Results: The proliferation of SSCs was faster in experiment group than that in control group, SSCs grew rapidly under defined medium after 3-7 days culture and presented a colony-like growth manner. Three-dimensional structure of SSCs was enhanced, and intercellular bridge was not obvious; After 15 days culture, the proliferation peak of SSCs was observed and the intercellular bridge of SSCs was still unclear. There was a significant difference in OD values and DNA content between two groups (P<0.01). Conclusion: SSCs showed some biological characteristics under defined culture environment with Chinese traditional medicine such as rapid growth and proliferation, similar with proliferation of normal cells. These results will provide clues for study of SSCs' multipotential differentiation and some proofs for treatment of oligozoospermia or asthenozoospermia with Chinese traditional medicine.
