Abstract:OBJECTIVE:To evaluate hippocampal volume changes and neuropsychological performances in patients with either amnestic mild cognitive impairment (aMCI) or Alzheimer's disease (AD). METHODS:Thirty-eight AD dementia, 22 aMCI patients, and 20 healthy controls were enrolled. Bilateral hippocampal volume was measured concurrently with mini-mental state examination (MMSE), auditory verbal learning test (AVLT), Boston naming test (BNT), and activities of daily living (ADL) test. Baseline and two additional follow-up examinations were conducted. RESULTS:Baseline hippocampal volumes were significantly smaller in AD group than that in aMCI and control groups. MMSE, AVLT, ADL, and BNT scores for the AD group were significantly different from that of both aMCI and control groups. Baseline hippocampal volumes were positively correlated with MMSE and AVLT scores in AD and aMCI patients. At follow-up, left hippocampal volume loss was positively correlated with decreased MMSE and AVLT scores both in AD and aMCI groups, while right hippocampal volume loss was positively associated with decreased AVLT performance only in AD group. Increased ADL and decreased BNT scores were positively associated with left hippocampal volume reduction only in the AD group. CONCLUSIONS:Current findings provide evidence of a close relationship between hippocampal volume and cognitive performances in patients with AD and aMCI, both at baseline and over follow-up.
Changes of Learning and Memory in Aged Rats after Isoflurane Inhalational Anaesthesia Correlated with Hippocampal Acetylcholine Level.

Abstract:BACKGROUND AND PURPOSE:It has not been clearly described for mechanisms of postoperative cognitive dysfunction (POCD), but not yet for narcotic in connection with POCD. Acetylcholine plays important roles in learning and memory especially in elderly people. It is not very clear that cholinergic changes in the hippocampus are in relation to spatial memory impairment. The effects of isoflurane, a commonly used inhalation anaesthetic, as well as acetylcholine on learning and memory in the brain tissue of aged rats were observed in present study. We investigated the inhalation anaesthesia drug's effect on cholinergic system to examine whether the regional and progressive cholinergic changes may lead to POCD.METHODS:Seventeen-month-old rats were randomly assigned to either an isoflurane anaesthesia group (n=17) or propofol anaesthesia group (n=17). In isoflurane anaesthesia experiment group, isoflurane (1.4 to 1.7% for 2 hours) was delivered via a ventilator to make freely moving microdialysis model. In propofol group, propofol was administered by continuous infusion via a tail vein catheter to make freely moving microdialysis model. They were all mechanically ventilated. Morris Water Maze test was used to assess the learning and memory abilities of all the two groups' rats twice a day for 5 days. Microdialysis was performed on the freely moving rats to determine the levels of acetylcholine in the brain tissues immediately after each Water Maze test.RESULTS:The isoflurane anaesthesia treatment increased the escape latency contrast to propofol anaesthesia group. The isoflurane anaesthesia's rats were then divided into isoflurane-induced severe learning/memory impairment group (n=6, mean escape latency is 1.96 times more than that of in propofol anaesthesia group) and the isoflurane-induced mild learning/memory impairment group (n=11, mean escape latency is equal or less than 1.96 times of that in propofol anaesthesia group). The results demonstrated that those rats that were categorized in the isoflurane-induced severe learning/memory impairment group had decreased levels of acetylcholine in the brain tissue as compared to those rats categorized in the mild learning/memory impairment group and in propofol anaesthesia group.CONCLUSION:The results indicated that isoflurane may impair learning and memory in aged rats.

Abstract:Objective To study affect of Isoflurane with inhalation anesthesia on hippocampal RhoA expression and spatial memory function in aged rats. Methods Thirty 22-month-old Sprague-Dawley rats were randomly assigned to control group (group C, 10 cases), anesthesia group Ⅰ (10 cases) and anesthesia group II (10 cases). The inhalation anesthesia model of aged rat was established by using endotracheal intubation, 2% Isoflurane was applied to maintain anesthesia of 2 hours after anesthesia induction with 3% Isoflurane by inhalation in anesthesia groups. Any drug was not used in rat of group C, which inhaled 100% oxygen for 2 hours, oxygen flow rate was 2 L/min. Open-field experi-ment and Y-maze experiment were implemented, performance was calculated after anesthesia of 24, 72 hours in groupⅠ and groupII. Learning and memory ability in rats of three groups were compared. Hippocampal RhoA expression in rats of three groups were detected by using the immune histochemical method. Results ①Y-maze experiment: Y-maze learning time of group Ⅰ [(70.50±16.25) times] was significantly more than that of group II [(61.80±23.49) times] and group C [(49.20±8.56) times] respectively (P<0.05). Y-maze learning time of group C and groupIIwas compared, with no statistical difference (P>0.05). Open-field experiment: there were no statistical difference on the middle of residence time, number of upright, number of across grid, number of total activity among three groups (P>0.05).②Immune histochemi-cal method:RhoA positive cell count of hippocampal CA3 area in groupⅠ(112.66±11.56) was more than that in groupII(97.77±7.47) and group C (99.33±12.10) respectively (P< 0.05). RhoA positive cell count of hippocampal CA3 area in groupII and group C was compared, with no statistical difference (P>0.05). Conclusion Spatial learning and memory ability of 22-month Sprague-Dawley rats received Isoflurane anesthesia of 24 hours decline, basic recovery after anes-thesia of 72 hours. The hippocampus of rat after Isoflurane anesthesia Rho protein signal transduction molecular change mediated synaptic morphology change may be the cellular basis of short-term spatial learning and memory damage.
Sevoflurane Induces Endoplasmic Reticulum Stress Mediated Apoptosis in Hippocampal Neurons of Aging Rats

Abstract:Elderly patients are more likely to suffer from postoperative memory impairment for volatile anesthetics could induce aging neurons degeneration and apoptosis while the mechanism was still elusive. Therefore we hypothesized that ER stress mediated hippocampal neurons apoptosis might play an important role in the mechanism of sevoflurane-induced cognitive impairment in aged rats. Thirty 18-month-old male Sprague-Dawley rats were divided into two groups: the sham anesthesia group (exposure to simply humidified 30-50% O2 balanced by N2 in an acrylic anesthetizing chamber for 5 hours) and the sevoflurane anesthesia group (received 2% sevoflurane in the same humidified mixed air in an identical chamber for the same time). Spatial memory of rats was assayed by the Morris water maze test. The ultrastructure of the hippocampus was observed by transmission electron microscopy (TEM). The expressions of C/EBP homologous protein (CHOP) and caspase-12 in the hippocampus were observed by immunohistochemistry and real-time PCR analysis. The apoptosis neurons were also assessed by TUNEL assay. The Morris water maze test showed that sevoflurane anesthesia induced spatial memory impairment in aging rats (P<0.05). The apoptotic neurons were condensed and had clumped chromatin with fragmentation of the nuclear membrane, verifying apoptotic degeneration in the sevoflurane group rats by TEM observation. The expressions of CHOP and caspase-12 increased, and the number of TUNEL positive cells of the hippocampus also increased in the sevoflurane group rats (P<0.05). The present results suggested that the long time exposure of sevoflurane could induce neuronal degeneration and cognitive impairment in aging rats. The ER stress mediated neurons apoptosis may play a role in the sevoflurane-induced memory impairment in aging rats.
The correlation between spatial memory deficits and the increased cholesterol content of the dorsal hippocampus in aged rats

Abstract:Objective To investigate the relationship between the spatial memory and cholesterol content of the dorsal hippocampus in aged rats.Methods Spatial memory of 21 and 4 months old rats were evaluated by Morris water maze task.And the cholesterol content and the expression of 3-hydroxy-3-methylglutaryl coenzyme A reductase(HMG-CoAR) and cholesterol 24-hydroxylase(CYP46) in dorsal hippocampus were assayed respectively.Results The escape latencies were longer,the total cholesterol content of the dorsal hippocampus was higher,CYP46 mRNA expression in the dorsal hippocampus was decreased in aged rats than those in the young(all P<0.05).But there was no significant difference in HMG-CoAR mRNA expression between the two groups.Conclusions The spatial memory deficits of the aged might be correlated with the increased cholesterol content in the dorsal hippocampus.
Effect of Isoflurane Anesthesia on Expression of Amyloid Β Protein in Hippocampus in Aged Rats

Abstract:Objective To investigate the effect of isoflurane anesthesia on the expression of amyloid β protein (Aβ) in hippocampus in aged rats.Methods Sixty-one healthy male SD rats,aged 18-19 months,weighing 400-500 g,were randomly divided into 3 groups:isoflurane group (group Ⅰ,n =31),control group (groupⅡ,n =20),and blank group (group Ⅲ,n =10).Group Ⅰ inhaled 1.4％ isoflurane for 2 h,and cognitive function was assessed at 5 d before and 2 d after anesthesia by Morris water maze.Group Ⅱ received Morris water maze text only.Group Ⅲ received no treatment.Group Ⅰ was divided into 2 subgroups according to whether the cognitive impairment occurred:cognitive impairment group (P subgroup) and no cognitive impairment group (NP subgroup).The animals were sacrificed immediately after the test.The hippocampus was isolated for determination of the levels of Aβ40 and Aβ42,β-secretase (BACE),insulin degrading enzyme (IDE)and neprilysin (NEP).Results There was no significant difference in the results of Morris water maze test before anesthesia among the 3 groups.There was no significant difference in the swimming velocity between group Ⅰ and group Ⅱ.The time of staying at the original platform quadrant was significantly shorter in P subgroup than in NP subgroup and Ⅱ group,and there was no significant difference in the time of staying at the original platform quadrant between NP subgroup and Ⅱ group.There was no significant difference in the levels of Aβ40 and Aβ42,BACE,NEP and IDE among the 5 groups,and between P subgroup and NP subgroup.Conclusion Isoflurane anesthesia has no effect on the expression of Aβ in the hippocampus,indicating that isoflurane-induced cognitive impairment is not related to Aβ pathway in aged rats.

Abstract:Objective: To investigate the effects of isoflurane on both spatial learning and memory and nNOS expression in the hippocampal CA1 subfield of the aged rats. Methods: Twenty -month -old Sprague -Dawley (SD) male rats (n =36/group) were included in this study. Based on the duration of anesthesia as well as the time-point of behavioral testing post-anesthesia, the aged rats were randomly divided into 6 groups (n =6/group) as follows: control group (no MWM testing, no anesthesia, just hippocampal sections processed); training group (no anesthesia, just MWM testing); isoflurane 2 h/2 d, 2 h/2 week, 4 h/2 d, 4 h/2 week groups in which the aged rats were exposed to 1.2% isoflurane for two or four hours and were tested with MWM at two days or two weeks after isoflurane administered, respectively. Following MWM behavioral testing, animals were killed and hippocampal sections were processed for nNOS expression in the CA1 subfield via immunohistochemistry. Results: Isoflurane exposure induced spatial memory deficits in aged rats since the escape latency increased significantly (P <0.05). Especially, four hours exposure to isoflurane resulted in more severe spatial memory impairment relative to two hours exposure. Moreover, this impairment occurred up to two weeks following the anesthesia (P <0.05). The training groups exhibited a remarkable increase in nNOS expression in the hippocampal CA1 subfield relative to control, non-treated rats (P< 0.05). Exposure to isoflurane inhibited nNOS expression, and this inhibition increased with increasing exposure duration, and that could last for a relative long time (two weeks). Conclusion: The inhalational anesthetic-isoflurane may impair the spatial learning and memory induced by inhibiting nNOS expression in hippocampal CA1 region of the aged rats.
Role of Autophagy in Cognitive Decline Caused by Sevoflurane Anesthesia in Aged Mice: the Relationship with Neurogenesis

Abstract:Objective To evaluate the role of autophagy in cognitive decline caused by sevoflurane anesthesia and the relationship with neurogenesis in aged mice.Methods Forty-five healthy SPF male mice,aged 20-22 months,weighing 25-35 g,were divided into 3 groups (n=15 each) using a random number table method:control group (group C),sevoflurane anesthesia group (group S) and autophagy agonist rapamycin group (group R).Rapamycin 0.2 mg/kg was intraperitoneally injected every day for 7 days in group R,while the equal volume of solvent dimethyl sulfoxide was given instead in S and C groups.In group S and group R,3％ sevoflurane was inhaled for 2 h once a day for 3 consecutive days starting from 5th day of administration,while the mixture of air and oxygen was inhaled instead in group C.Five mice in each group were randomly selected after the last anaesthesia and sacrificed,and the hippocampus was removed for determination of the expression of microtubule-associated protein 1 light chain 3 Ⅱ (LC3 Ⅱ) and Beclin-1 by Western blot.The other mice were sacrificed after Morris water maze test was performed,and hippocampi were isolated for determination of doublecortin (DCX) positive cells in the dentate gyrus by immunohistochemistry.Results Compared with group C,the escape latency was significantly prolonged,the percentage of time spent in the target quadrant was decreased,the expression of LC3 Ⅱ and Beclin-1 was down-regulated,LC3 Ⅱ/LC3 Ⅰ ratio was decreased,and DCX positive cell counts were reduced in S and R groups (P＜0.05).Compared with group S,the escape latency was significantly shortened,the percentage of time spent in the target quadrant was increased,the expression of LC3 Ⅱ and Beclin-1 was up-regulated,LC3Ⅱ/LC3 Ⅰ ratio was increased,and DCX positive cell counts were increased in group R (P＜0.05).Conclusion Autophagy is involved in the process of cognitive decline caused by sevoflurane anesthesia,which is related to inhibiting neurogenesis in the hippocampus of aged mice.

Abstract:Objectives: The molecular mechanism of postoperative cognitive dysfunction is largely unknown. Isoflurane has been shown to promote Alzheimer's disease neuropathogenesis. We set out to determine whether the effect of isoflurane on spatial memory is associated with amyloid-beta (A-beta) levels and tau phosphorylation in aged rats. Methods: Eighteen-month-old male Sprague-Dawley rats were randomly assigned as anesthesia group (n=31, received 1.4% isoflurane for 2 hours and had behavioral testing), training group (n=20, received no anesthesia but had behavioral testing), and control group (n=10, received no anesthesia and had no behavioral testing). Spatial memory was measured before and 2 days after the anesthesia by the Morris water maze. We divided the anesthesia group into an isoflurane-induced severe memory impairment group (SIG, n=6) and a no severe memory impairment group (NSIG, n=25), according to whether the escape latency was more than 1.96 stand deviation of that from the training group. Levels of A-beta and tau in the hippocampus were determined by enzyme-linked immunosorbent assay and quantitative western blot at the end of behavioral testing. Results: We found that isoflurane increased the escape latency in the SIG as compared to that in the training group and NSIG without affecting swimming speed. However, there were no differences in the levels of A-beta and tau among SIG, NSIG, training, and control groups. Conclusions: Isoflurane may induce spatial memory impairment through non-A-beta or tau neuropathogenesis mechanisms in aged rats.
Impaired Spatial Learning Memory after Isoflurane Anesthesia or Appendectomy in Aged Mice is Associated with Microglia Activation.

Abstract:Postoperative cognitive dysfunction (POCD) has been one of the most common problems in elderly patients following surgery. But the specific mechanism of POCD is still not clear. To further understand the reason of these postoperative behavioral deficits, we evaluated the spatial learning memory of both adult (3 months) and aged (18 months) male mice, 3 or 28 days after isoflurane (Iso) exposure for two hours or appendectomy (App). Hippocampal microglia activation and IL-1β, TNF-α, and IFN-γ expression were also evaluated at day 3, day 14 and day 28 after Iso exposure or appendectomy. Results showed that spatial learning memory of aged, but not adult, mice was impaired after Iso exposure or appendectomy, accompanied with more hippocampal microglia activation and IL-1β, TNF-α, and IFN-γ overexpression. These findings suggest that the cognitive deficits of elderly patients who have undergone surgeries are quite possibly caused by hippocampal microglia overactivation and the subsequent inflammation.

Abstract:Objective To investigate the effects of isoflurane anesthesia on β-amyloid protein (Aβ) generation and phosphorylation of c-Jun N-terminal kinase (JNK) and cyclic AMP responsive element binding protein (CREB) in the hippocampus of aged rats.Methods Ninety-six male Sprague-Dawley rats,aged 18 months,weighing 450-500 g,were randomly divided into control group (group C,n =24),1％ isoflurane anesthesia group (group S1,n =36) and 2 ％ isoflurane anesthesia group (group S2,n =36).Groups S1 and S2 inhaled 1％ and 2％ isoflurane for 4 h,respectively.Twelve rats were chosen in each group and Morris water maze was performed 1 day after anesthesia.Twelve rats in each group were chosen on day 1 and 7 after anesthesia (T1,T7) and open field test was performed.The rats were sacrificed after open field test and hippocampi were isolated for determination of the expression of Aβ,phosphorylated JNK (p-JNK) and phosphorylated CREB (p-CREB) in hippocampal tissues by Western blot.Results Compared with group C,the escape latency was significantly prolonged and target quadrant dwell time ratio was decreased significantly in groups S1 and S2,the total movement distance,leave border distance and inner toroid distance were prolonged,Aβ expression was up-regulated,and p-CREB expression was down-regulated at T1 and leave border distance and inner toroid distance were prolonged,Aβ expression was up-regulated at T7 in group S1,and the total movement distance,faster movement distance,leave border distance and inner toroid distance were prolonged,the expression of Aβ and p-JNK was up-regulated,and p-CREB expression was down-regulated at T1,and the total movement distance,faster movement distance,leave border distance and inner toroid distance were prolonged,the expression of Aβ was up-regulated at T7 in group S2 (P ＜ 0.05 or 0.01).Conclusion The mechanism by which isoflurane anesthesia induces cognitive dysfunction is related to promotion of Aβ generation and JNK activation and inhibition of CREB activation in the hippocampus of aged rats.
Effects of Isoflurane on Learning and Memory in Aged Rats

Abstract:Objective To investigate the role of isoflurane inhalation on learning and memory in aged SpragueDawley(SD) rats.Methods A total of 36 SD rats were randomly assigned to control group and anesthesia group with 18 rats in each group.Rats in the anesthesia group were given 2% isoflurane for the purpose of anesthesia which was induced by 40% oxygen and last for 3 hours.Rats in the control group inhaled the mixture of air and 40% oxygen for 3 hours.Learning and memory abilities of these rats were assessed daily in Y-maze 24 hours after the anesthesia intervention.Results There were no statistically significant differences between the two groups in numbers of wrong reaction(P > 0.05).The voltage when rats raised their feet was not significantly different between the two groups except that on the 7th day,it was higher in the anesthesia group than in the control group(P < 0.05).The voltage when rat began to run was also not significantly different between the two groups except that on the 3rd day,it was higher in the anesthesia group than in the control group(P < 0.05).Conclusion Two percent isoflurane anesthesia could be of no effect on postanesthetic learning and memory in aged SD rats.
Neural Correlates of Associative Memory in the Elderly: A Resting-State Functional MRI Study.

Abstract:The neural correlates of associative memory in healthy older adults were investigated by examining the correlation of associative memory performance with spontaneous brain oscillations. Eighty healthy older adults underwent a resting-state functional MRI and took a paired-associative learning test (PALT). Correlations between the amplitude of low-frequency fluctuations (ALFF) as well as fractional ALFF (fALFF) in the whole brain and PALT scores were calculated. We found that spontaneous activity as indexed by both ALFF and fALFF in the parahippocampal gyrus (PHG) was significantly positively correlated with associative memory performance, suggesting that the PHG plays a critical role in associative memory in older people.

Abstract:Previous studies have shown multiple mechanisms and pathophysiological changes after anesthesia, and genome-wide studies have been implemented in the studies of brain aging and neurodegenerative diseases. However, the genome-wide gene expression patterns and modulation networks after general anesthesia remains to be elucidated. Therefore, whole transcriptome microarray analysis was used to explore the coding gene expression patterns in the hippocampus of aged rats after sevoflurane anesthesia. Six hundred and thirty one upregulated and 183 downregulated genes were screened out, then 44 enriched terms of biological process, 16 of molecular function and 18 of the cellular components were identified by Gene Ontology (GO) and KEGG analysis. Among them, oxidative stress, metabolism, aging, and neurodegeneration were the most enriched biological processes and changed functions. Thus, involved genes of these processes were selected for qPCR verification and a good consistency was confirmed. The potential signaling pathways were further constructed including mitochondrion and oxidative stress-related Hifs-Prkcd-Akt-Nfe2l2-Sod1 signaling, multiple metabolism signaling (Scd2, Scap-Hmgcs2, Aldh18a1-Glul and Igf1r), as well as aging and neurodegeneration related signaling (Spidr-Ercc4-Cdkn1a-Pmaip1 and Map1lc3b). These results provide potential therapeutic gene targets for brain function modulation and memory formation process after inhaled anesthesia in the elderly, which could be valuable for preventing postoperative brain disorders and diseases, such as perioperative neurocognitive disorders (PND), from the genetic level in the future.
Resveratrol Alleviates Learning And Memory Impairment In Aged Rats After General Anesthesia With Sevoflurane And Nitrous Oxide Via Sirt1-P53 Signaling Pathway

Abstract:The aim of this study was to evaluate the effects of resveratrol on learning and memory impairment in aged rats after general anesthesia with sevoflurane and nitrous oxide (N2O), and explore the potential mechanism. 18-month-old Sprague-Dawley rats were randomly divided into anesthesia group (receiving 1.3% sevoflurane and 50% N2O for 4 hours) and control group. 48 hours after anesthesia, spatial learning and memory were tested by the Morris Water Maze experiment. The neurotoxicity and expression of silent information regulator 1 (SIRT1) in the hippocampus were measured. Moreover, the effect of resveratrol on spatial learning and memory after anesthesia in aged rats was also investigated. The results showed that general anesthesia triggered neuronal apoptosis and, subsequently, induced impairment of learning and memory ability. Neuronal apoptosis induced by anesthetics triggered endogenous SIRT1 expression. Moreover, the SIRT1 expression and p53 deacetylation was increased in rats pre-treated with resveratrol, which subsequently inhibited neuronal apoptosis and improved behavioral performance. In conclusion, our results demonstrated that general anesthesia with sevoflurane plus 50% N2O impaired learning and memory ability in aged rats, induced neuronal apoptosis and increased SIRT1 expression. Pretreatment with resveratrol improved the ability of learning and memory, inhibited neuronal apoptosis by increasing the expression of SIRT1 in aged rats after general anesthesia.

Abstract:General anesthesia can impact a patient's memory and cognition by influencing hippocampal function. The CA1 and dentate gyrus (DG), serving as the primary efferent and gateway of the hippocampal trisynaptic circuit facilitating cognitive learning and memory functions, exhibit significant differences in cellular composition, molecular makeup, and responses to various stimuli. However, the effects of isoflurane-induced general anesthesia on CA1 and DG neuronal activity in mice are not well understood. In this study, utilizing electrophysiological recordings, we examined neuronal population dynamics and single-unit activity (SUA) of CA1 and DG in freely behaving mice during natural sleep and general anesthesia. Our findings reveal that isoflurane anesthesia shifts local field potential (LFP) to delta frequency and reduces the firing rate of SUA in both CA1 and DG, compared to wakefulness. Additionally, the firing rates of DG neurons are significantly lower than CA1 neurons during isoflurane anesthesia, and the recovery of theta power is slower in DG than in CA1 during the transition from anesthesia to wakefulness, indicating a stronger and more prolonged impact of isoflurane anesthesia on DG. This work presents a suitable approach for studying brain activities during general anesthesia and provides evidence for distinct effects of isoflurane anesthesia on hippocampal subregions.

Abstract:AIM:To observe the characteristics of memory behavior, free radical contents and ultrastructural morphometry of pyramidal cells of hippocampal CA1 sector of aged rats in order to study the neurobiological mechanism of age related memory deficiency. METHODS:Forty male SD rats were randomly divided into young and aged groups, with 20 rats for each group. Morris water maze test was used to observe learning and memory behavior of all the rats. The homogenates of rat hippocampus was used to determine the amount of lipid peroxide using the barbituric acid reactive substances assay, and the protein content was determined by spectrophotometer.The ultrastructural morphology of hippocampal CA1 pyramidal cells was observed using transmission electron microscope (TEM), and the morphometrical quantification and stereological technique were used to compare the numbers and sizes of cell nuclei, rough endoplasmic reticulum and mitochondria between the two groups. RESULTS:Compared with the young rats, the aged rats showed the longer latency (38 vs 12 s, t=8.04, P< 0.01) in place navigation during the place training experiment and less crossing numbers over the platform (50% vs 82% , t=10.17, P< 0.05) during spatial probe test. The amount of lipid peroxide(146.2 vs 178.4 nmol/g, t=5.61, P< 0.01 ), tissue total protein (127.9 vs 168.8 μ g, t=8.25, P< 0.01) and sedimental protein (21.1 vs 34.3 μ g, t=9.53, P< 0.01 )were all increased in aged rats, compared with those in the young rats. A lot of pyramidal cells in 250 μ m CA1 sector (51.9 vs 40.7, t=6.55, P< 0.01 ) lost with smaller volume of nucleoli (5.34 vs 4.02 μ m3, t=6.55, P< 0.01 ). The rough endoplasmic reticulum density (1.47 vs 0.94 μ m2/ μ m3, t=10.44) and mitochondria of pyramidal cells were less and smaller in the aged rats than those in the young rats. CONCLUSION:Degenerative changes are present in pyramidal cells in hippocampal CA1 sector. The degeneration is one of the causes that lead to spatial memory deficient of the aged rats. The overdose lipid peroxide may result in the degeneration of pyramidal cells.

Abstract:Postoperative cognitive decline is a clinical concern especially for senior patients. It is generally recognized that glutamatergic system plays a crucial role in the physiopathologic process of neurocognitive deterioration. However, alterations of glutamatergic system in prolonged isoflurane-induced learning/memory decline are still unclear. This study investigates the question whether glutamate concentration and corresponding transporters or receptors display any alternations in aged rat suffering from isoflurane-induced learning/memory impairment. 111 male Sprague–Dawley rats (>18 months) were randomly divided into two main groups: hippocampal microdialysis group (n = 38) and western blotting group (n = 73). Each group was subdivided into three subgroups including (1) control subgroup (n = 6 and 10, receiving no behavioral trial, anesthesia or air exposure); (2) air-exposed subgroup (n = 7 and 15, receiving behavioral trial and air exposure but not anesthesia); (3) isoflurane anesthesia subgroup (n = 25 and 48, receiving both behavioral trial and anesthesia). The isoflurane-exposed rats were further divided into a learning/memory-impaired subgroup and a non-learning/memory-impaired subgroup according to their behavioral performance, which was measured using Morris water maze. Hippocampal glutamate concentrations in microdialysates were determined by high-performance liquid chromatography. Expression levels of GLAST, GLT-1, NMDAR1, NMDAR2A/B, AMPAR and tau in hippocampus were assessed via quantitative Western blotting. The incidences of learning/memory impairment of isoflurane-exposed rats in hippocampal microdialysis group and western blotting group were 12.0 (3/25) and 10.4 % (5/48) respectively. The intra-anesthesia hippocampal glutamate levels were significantly lower than those of non-anesthesized rats. The learning/memory-impaired rats showed a long-lasting increased glutamate level from 24 h after isoflurane exposure to the end of the study, but the other 22 isoflurane-exposed rats did not. The learning/memory-impaired subgroup displayed a significantly higher GLAST level than the other three subgroups ( p = 0.026, 0.02 and 0.032 respectively). The expression levels of GLT-1, NMDAR1, NMDAR2A/B and AMPAR of every subgroup were comparable. We found a continuous raised hippocampal glutamate and an up-regulation of GLAST rather than GLT-1, NMDAR1, NMDAR2A/B, AMPAR or tau in hippocampus of aged rats associated with isoflurane-induced learning/memory impairment.

Abstract:Our study in aged mice unveils sevoflurane‐induced changes in gene expression within excitatory neurons, which is associated with synaptic function and cognitive impairment. Immunofluorescence staining confirms reduced hippocampal glutamatergic synapses, emphasizing their central role in age‐related cognitive dysfunction. Aims Perioperative neurocognitive disorders (PND), including postoperative delirium (POD) and postoperative cognitive dysfunction (POCD), are common following anesthesia and surgery in older patients and significantly increase morbidity and mortality. However, the underlying mechanism of PND is unclear. Our study aims to analyze the differentially expressed genes (DEGs) in excitatory neurons and investigate the role of hippocampal glutamatergic synaptic alterations in sevoflurane‐induced cognitive dysfunction in aged mice. Methods We performed single‐nucleus RNA sequencing (snRNA‐seq) technology to examine the alterations of excitatory neurons in hippocampus induced by sevoflurane in aged mice. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis of DEGs were performed in excitatory neurons. At last, immunofluorescence staining was used to validate sevoflurane‐induced alternation of glutamatergic synapses in the hippocampus of aged mice. Results This study demonstrates that DEGs in excitatory neurons are associated with reduction of glutamatergic synapses and cognitive dysfunction. After immunofluorescence staining validation, we also confirmed that sevoflurane anesthesia decreased the density of glutamatergic synapses in the hippocampus of aged mice. Conclusions Our findings demonstrated a key role of hippocampal glutamatergic synaptic alterations in sevoflurane‐induced cognitive dysfunction in aged mice.

Effect of Penehyclidine Hydrochloride on Cognitive Function in Aged Rats

Abstract:Objective:To investigate the effect of Penehyclidine hydrochloride on spatial learning and memory and the changes of Bax/bcl-2levels and acetylcholinesterase(AChE)in the hippocampus of aged rats.Methods:Forty-eight 9-month old SD rats weighing 300-350g were randomly divided into four groups(n=12):control group(group C,intraperitoneal injection of normal saline), 0.64,1.28,and 2.56mg/kg Penehyclidine hydrochloride treatment groups(P1,P2,and P3 groups respectively).Twenty-four hours after treatment,all rats underwent Morris water-maze test(MWM test).The hippocampus samples were harvested to determine the expression of Bax and bcl-2 by using immunohistochemical technique,and the activities of AchE in the hippocampus were examined by colorimetric method.Results:Both the latent period and swimming distance in group P3 and P2 were significantly longer than those in group C and P1(all P0.05).The numbers of Bax and Bcl-2 positive cells were significantly more and the ratio of Bax to Bcl-2 was higher in group P3 and P2 than in group C and P1 respectively(all P0.05).The activities of AChE in group P3 and P2 were significantly higher than in group C and P1(all P0.05).Conclusion:Repeated large dosage administration of Penehyclidine hydrochloride can impair spatial learning and memory of rats maybe through its effects on the apoptosis of hippocampal neurons and themodulations on cholinergic system.
