Abstract:Purpose: Kashin-Beck Disease (KBD) is a type of endemic, chronic and disabling osteoarthropathy with high disability. Most previous studies reported that environmental factors played an important role in KBD development, however, the etiology and pathogenesis has not been fully unraveled. In the present study, we firstly conducted a comprehensive collection of genes that were reported to be associated with KBD. Then a comprehensive analysis focusing on biological function and interactions of KBD-related genes was performed to understand KBD's pathogenesis. Methods: By reviewing the literature deposited in PubMed, we identified 922 genes genetically associated with KBD. Biological function and network analyses were conducted with Cytoscape software. Moreover, KBD specific molecular network analysis was conducted by Cytocluster using the Molecular Complex Detection Algorithm (MCODE) (Degree cutoff = 2, Node score cutoff = 0.2, K-core = 2, MAX. Depth = 100). Results: The biological function enrichment analysis suggested that apoptosis, ECM-receptor interaction, and the p53 signaling pathway played important roles in KBD development. The pathway enrichment analysis identified the immune system and the body's oxidative stress response as being related to KBD development. The apoptosis pathway, NF-kappa B signaling pathway, and the glutathione metabolism pathway were significantly enriched in the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway network (Fig. 1). MCODE clusters showed that in top 3 clusters, 54 of KBD-related genes (red ellipses) were included in human interactome network and the extracted KBD-specific network and 110 candidate genes (green diamonds) were discovered (Fig. 2). Conclusions: Our systematic and comprehensive results on the key biomarkers related to KBD indicated again apoptosis and oxidative stress response may play an important roles in the occurrence and development of KBD, which was similar to some previous experiments. Our results will not only improve our understanding of the genetic mechanism of KBD but will also help us to identify potential biomarkers for KBD in the future.View Large Image Figure ViewerDownload Hi-res image Download (PPT)
