Bioremediation of oily contamination by free and immobilized microorganisms on laboratory-scale

Abstract:Two oil-degrading bacterial strains (H-1 and B-1) were isolated and obtained from oil-contaminated water samples collected from Shengli Oil Field. H-l and B-l, identified by 16S rDNA sequences and morphological, biochemical and physiological characteristics as Rhosococcus sp. and Bacillus cereus sp. respectively, were tested for their growth behavior and optimal growth conditions in the laboratory. The results showed that the optimal growth conditions were about 35°C, pH=8, salinity 2% and 4%, respectively. The environmental conditions affecting oil biodegradation rates by H-l and B-l were optimized in the media containing 300 mg-L" 'standard oil. The results showed that the optimal degradation conditions were similar to the optimal growth conditions, and the oil degradation rates of H-l and B-1 were about 34.6% and 45.3% after 5 days. An improved microbial immobilization method was described involving the use of sodium alginate carrier containing active carbon. The ability to remove standard oil was used to characterize the biological activity of immobilized H-1 and B-1. The results showed that immobilized H-1 and B-1 degraded 66.3% and 75% standard oil after 5 days, improving 31.7% and 29.7% compared to free microorganisms.
