Comparison of permeation rates in vitro between indometacin ultradeformable nanoliposomes and general nanoliposomes

Abstract:Background: Oral administration of indomethacin causes great side effects, while local application of indomethacin unguentum is limited by slow permeation and diffusion and poor absorption. Ultradeformable nanoliposomes are new percutaneous medication vector and can promote drug permeation. Objection: To explore the effect of indomethacin ultradeformable nanoliposomes on permeation rate. Design, Time and Setting: An in vitro study was performed at Key Laboratory of Nanobiotechnology, Ministry of Public Health, Central South University between July 2008 and January 2009. Materials: Indomethacin ultradeformable nanoliposomes were prepared using Film-ultrasonic wave dissolving techniques. Methods: Indomethacine permeation tests in vitro through the skin of rats were performed in Franz diffusion cells. Drug permeation rate was compared between indomethacin ultradeformable nanoliposomes and the general liposome. Main Outcome Measures: Particle size, recovery rate, encapsulation efficiency, and drug cumulative permeation rate. Results: Nano-liposomes as a carrier of small size, high encapsulation efficiency, and variable type, could successfully carry drugs through the skin. Compared with general nanoliposomes, ultradeformable nanoliposomes could increase permeation rate of indomethacine through the skin of rats. Conclusion: Ultradeformable nanoliposomes can significantly enhance the permeation rate of indomethacine through the skin of rats.
