Preparation of rat models of hepatocellular carcinoma via orthotopic implantation using a cytoskeleton

Abstract:Background: Tumor cell injection has been used to prepare a rat model of hepatocellular carcinoma via orthotopic implantation under ultrasound guidance, but the survival rate is not high and easily forms abdominal cavity transplantation. Objective: To achieve high tumor formation rate and low incidence of abdominal cavity implants by using cell Matrigel to pack tumor cells into rat models of orthotopic transplantation liver cancer under the ultrasonic guidance. Design, Tome And Setting: A verification experiment was performed at the Animal Laboratory, Pharmacology Department, College of Pharmacy, Fujian Medical University from December 2007 to March 2008. Materials: A total of 20 Wistar rats were equally and randomly assigned into a tumor cell suspension injection group and a tumor cell Matrigel injection group. Methods: Walker-256 tumor tissue homogenate of rats following subcutaneous transplantation was used to prepare cell suspension, and infused into the liver parenchyma of 10 rats in the tumor cell suspension group under ultrasonic guidance, 2x106 cells in each rat. After mixing with Matrigel, Walker-256 cells were inoculated into the liver of 10 rats in the tumor cell Matrigel group. Main Outcome Measures: Ultrasonography was used to detect the tumor growth in rats following liver transplantation. On day 21, the rats were sacrificed to perform gross anatomy observation, and the tumors were taken for pathological examination. Results: Liver tumors formed in 9 rats in the tumor cell Matrigel group, and only one rat underwent abdominal cavity implantation. In 10 rats of the tumor cell suspension group, only 6 rats formed liver tumor, and 6 rats occurred abdominal cavity implantation. The pathological examination showed that the orthotopic transplantation tissue displayed typical characteristics of carcinoma. Conclusion: The rat model of hepatocellular carcinoma via orthotopic implantation using Matrigel as cytoskeleton exhibited high tumor formation rate and low incidence of abdominal cavity implants, and was an optimal method for establishing liver cancer models.
