In vitro culture of microencapsulated adult rabbit Schwann cells

Abstract:BACKGROUND: It is studied that a large number of Schwann cells in vitro amplification plant into nerve conduit up planting guide regeneration of peripheral nerve effect, the current concern microencapsulated immunoisolation technology, in overcoming the issue of immune rejection has obvious advantages and practicality. OBJECTIVE: To observe the changes in cell morphology and proliferation between microencapsulated and non-microencapsulated Schwann cells in vitro culture. DESIGN, TIME and SETTING: Experiments were performed at the Molecular Laboratory in Second Affiliated Hospital of Nanchang University between September 2005 and May 2006. MATERIALS: Healthy adult rabbits were supplied by Animal Center, Medical College, Nanchang University. Microcapsule generator was prepared by the Institute of Neurobiology, Medical College, Nanchang University. METHODS: Bilateral sciatic nerve developed Wallerian denaturation. Schwann cells were cultured in modified repeated differential adherence method, rapidly isolated and purified. Schwann cells at passage 3 grew well and packed by alginic acid-barium chloride. MAIN OUTCOME MEASURES: Morphological changes in Schwann cells was observed under the inverted microscope. MTT was used to detect the proliferation change between microencapsulated Schwann cells and free Schwann cells at different time phases. RESULTS: Microencapsulated Schwann cells lived well in vitro, with complete structure. Following microencapsulation, Schwann cells were suspended. There was significant difference in absorbance between microencapsulated and non-microencapsulated Schwan cells at 0, 1, 3, 5, 7 days (P < 0.05). CONCLUSION: Microencapsulated Schwann cells can survive for a long time in vitro. The proliferation speed of microencapsulated Schwann cells was slower than non-microencapsulated Schwann cells.
