Modified method for in vitro culture of rat bone marrow mesenchymal stem cells

Abstract:Background: Mesenchymal stem cells (MSCs) are few in bone marrow, and need to be isolated, purified and proliferated in vitro. Objective: To establish a stable method to proliferate and culture bone marrow mesenchymal stem cells in vitro, and to elevate the pick-up rate and amplification rate of bone marrow mesenchymal stem cells. Design, Enforcement and Setting: The cell observation experiment was performed at the Laboratory of Department of Hematology, Zhujiang Hospital, Southern Medical University from March 2007 to February 2008. Materials: Clean male rats aged 6-8 weeks, weighing 130-150 g were used for this study. Methods: Bone marrow mesenchymal stem cells wore incubated by modified adherence method. Rat humerus, femurs and tibias were remained. Tissues surrounding bones were completely removed to discard miscellaneous cells. 2.5 mL syringe was used to wash medullary cavity to remove the bone marrow mesenchymal stem cells from medullary cavity and to enhance pick-up rate. 3/4-4/5 medium was changed. Primary cells were separately digested in two bottles, and combined into a bottle to ensure the number of cells and to avoid cell over ageing during passage. Main Outcome Measures: The morphologic changes in MSCs were detected under an inverted optical microscope and by hematoxylin-eosin (HE) dyeing. Cell cycle was analyzed by flow cytometry. Growth curve and clone formation of P1 and P5 was measured by MTT and experiment of clone formation Identification was performed by flow cytometry and alkaline phosphatase (AKP) staining. Results: Bone marrow mesenchymal stem cells wore uniform, showing spindle or star-shaped. After passage, cells were rapidly proliferated. Passage was conducted once every 3-4 days. Cells with uniform shape were fibroblasts. Flow cytometry demonstrated that 80% of MSCs was in G0G1 phase. Growth curve demonstrated that cells entered duplication phase at 3 days, and platform phase at 5 days. Cell proliferation became slow and clone ability decreased over time. Flow cytometry showed CD44+, CD99+, CD45-, AKP(+). Conclusion: Bone marrow mesenchymal stem cells with uniform shape and good function are obtained by the modified method. Pick-up rate and proliferation rate are effectively enhanced.
