Abstract:An improved process was reported to solve the problems such as low yield and high costs in the synthesis of 3-n-propyl-2, 4-dihydroxyacetophenone, which is a key intermediate of Leukotriene receptor antagonist. The target compound was synthesized starting from m-dihydroxybenzene, via acylation, allylation, Claisen rearrangement and hydrogenation reduction. The optimal reaction conditions were as follows. The acylation reaction which used acetic acid as acylation agent and ZnCl 2 as catalyst was carried out under reflux for 5 h, and 77.9% yield was obtained. The allylation reaction using allyl chloride as allylation reagent and NaBr, KI as catalyst was carried out at 40°C for 2 h and continued at 70°C for 14 h, to give 98.6% yield. In Claisen rearrangement carried out at 205°C for 4.5 h, diphenyl ether was used as reaction solvent and NaCl was used as catalyst, and the yield was improved to 67.4%. In the final step, the hydrogenated reduction with Pd/C (w(Pd) is 5%) as catalyst was carried out at 45°C for 6 h, and 88.6% yield was obtained. The process is much of industrial value because of high yield, cheap and available materials, and convenient operations. The structure of the main compounds was confirmed by 1H-NMR, GC-MS and IR.

Abstract:Pyridazine was prepared by hydrogenolysis of 3,6-dichloropyridazine obtained via chlorination of 3,6-pyridazine diol with phosphorus oxychloride.3,6-Pyridazine diol was prepared by cyclization of hydrazine dihydrochloride and maleic anhydride.The feasible reaction conditions were as follows: cyclization: the mole ratio of maleic anhydride to hydrazine dihydrochloride was 1∶1,100°C,3 h;chlorination: the mole ratio of 3,6-pyridazine diol to phosphorus oxychloride was 1∶12.5,100～105°C,5 h;catalytic hydrogenolysis: the mole ratio of 3,6-dichloropyridazine to triethylamine was 1∶2.6,50°C,H_2 0.5 MPa,6 h.The overall yield was 62.2%(mol).The product was identified by melting point test,()~1H NMR and elemental analysis.

Abstract:An improved synthetic approach to tenatoprazole 1 is described. It started from 2,3,5-trimethyl-4-nitropyridine-N-oxide 2 with acetic anhydride via rearrangement and hydrolysis to give 3, Chlorination with SOCl2 yielded 2-chloromethyl-3,5-dimethyl-4-nitropyridine hydrochloride 4, then 4 condensed with 2-mercapto-5-methoxy imidazole [4,5-b]pyridine 5 to give 5-methoxy-2-[(4-nitro-3,5-dimethyl-2-pyridinyl)methylthio]imidazole[4,5-b]pyridine 6. At last the title compound 1 was produced by two methods: 6 was oxidized with MCPBA and then methoxylated with CH3ONa to give 1 and 6 was first methoxylated with CH3ONa and then oxidized with MCPBA to give 1. The overall yield is around 26% for both five-step syntheses.

Abstract:A new and efficient method for the synthesis of 4-methoxyl-2,3,5-trimethylpyiidine with an overall yield of 76% from cheap and easily available 2,3,5-trimethylpyridine was developed. The structure of related compounds was identified by IR, H-1 NMR and C-13 NMR. The method could be carried out economically on a huge scale.

Abstract:A radiochemical, propyl 4-[2-(4,6-dimethoxy-2-pyrimidinyloxy)benzylamino]benzoate with all six benzoic ring carbons labeled by 14 C was made from 4-aminobenzoic acid with all six benzoic ring carbons labeled by C-14 through esterification, condensation, reduction and substitution, and purified by preparative HPLC. Its structure was identified by LC-MS and H-1 NMR. HPLC test showed its chemical purity exceeded 98%. HPLC-LSC and TLC-IIA demonstrated that its radiochemical purity exceeded 98% and specific activity was 1.089 +/- 0.011 mCi/mmol. The total chemical yield and radiochemical yield were all 53%.

Abstract:3, 3-Dimethoxypropanenitrile was synthesized from 2-chloro-3-phenylsulfonylpropanenitrile by elimination and addition reactions. 3-phenylsulfonylpropanenitrile was obtained by eliminating HCl from 2-chloro-3-phenylsulfonylpropanenitrile at 0-20℃ with a yield of 88%. 3-Phenylsulfonylpropanenitrile was reacted with sodium methoxide in methanol, giving 3, 3-dimethoxypropanenitrile with a yield of 98%. Elimination reaction was innovated by using THF as a solvent instead of ethyl ether and addition reaction was innovated by using an excess of sodium methoxide in methanol, so as to have the reaction condition optimized.

Abstract:SYP-3343, (E)-2-(2-((3-(4-chlorophenyl)-1-methyl-1H-pyrazole-5-yloxy)methyl)) phenyl)-3-methoxyacrylate, is a novel fungicide with broad-spectrum and high activity against fungi. In this paper, radioactive pyraoxystrobin, labeled in the pyrazole ring system was achieved from 14C labeled 4-chlorobenzoic acid through substitution, cyclization, and condensation, in an overall chemical and radiochemical yield of 44%. The chemical and radiochemical purity of the title compound was 98.5% and 98.2%, respectively. Metabolic Profiles of 20(S)-Protopanaxadiol in Rats after Oral Administration Using Ultra-Performance Liquid Chromatography/quadrupole Time-of-flight Tandem Mass Spectrometry.

Abstract:RATIONALE:20(S)-Protopanaxadiol (PPD), a dammarane-type triterpenoid sapogenin, acts as the pharmacophore of ginsenosides which are considered as the principal bioactive components in Chinese ginseng. To fully understand the mechanism of action of PPD, it is important to study its metabolic profiles in vivo.METHODS:Plasma, urine, fece and bile were collected after administration of PPD formulated in 0.5% aqueous Tween-80 to rats (150 mg/kg). Samples were analyzed by using a sensitive and reliable method based on ultra-performance liquid chromatography/quadrupole time-of-flight tandem mass spectrometry (UPLC/Q-TOF-MS/MS) in both positive and negative ion mode. The chemical structures of metabolites were elucidated by comparing the retention time, accurate molecular mass, and fragmentation patterns of analytes with those of PPD.RESULTS:In total 29 metabolites, including 10 new metabolites (M20-M29), were tentatively identified and characterized. Among them, two metabolites (M3 and M4) were unambiguously identified by matching their retention times and fragmentation patterns with their standards. Principal metabolites, namely, 20, 24-oxide metabolites (M3 and M4), 26/27-carboxylic acid derivatives (M22 and M23) and a glucuronidated product (M28), were found in the rat plasma.CONCLUSIONS:The results showed that phase I metabolites are monooxygenation, dioxygenation and oxidative dehydrogenation metabolites, and phase II metabolic pathways were demonstrated to be cysteine conjugation and glucuronidation. The newly identified metabolites are useful to understand the mechanism of elimination of PPD and, in turn, its effectiveness and toxicity.
Tentative identification of 20(S)-protopanaxadiol metabolites in human plasma and urine using ultra-performance liquid chromatography coupled with triple quadrupole time-of-flight mass spectrometry

Abstract:Abstract Background 20( S )-Protopanaxadiol (PPD), the aglycone part of 20( S )-protopanaxadiol ginsenosides, possesses antidepressant activity among many other pharmacological activities. It is currently undergoing clinical trial in China as an antidepressant. Methods In this study, an ultra-performance liquid chromatography coupled with triple quadrupole time-of-flight mass tandem mass spectrometry method was established to identify the metabolites of PPD in human plasma and urine following oral administration in phase IIa clinical trial. Results A total of 40 metabolites in human plasma and urine were identified using this method. Four metabolites identified were isolated from rat feces, and two of them were analyzed by NMR to elucidate the exact structures. The structures of isolated compounds were confirmed as (20 S ,24 S )-epoxydammarane-12,23,25-triol-3-one and (20 S ,24 S )-epoxydammarane-3,12,23,25-tetrol. Both compounds were found as metabolites in human for the first time. Upon comparing our findings with the findings of the in vitro study of PPD metabolism in human liver microsomes and human hepatocytes, metabolites with m/z 475.3783 and phase II metabolites were not found in our study whereas metabolites with m/z 505.3530, 523.3641, and 525.3788 were exclusively detected in our experiments. Conclusion The metabolites identified using ultra-performance liquid chromatography coupled with triple quadrupole time-of-flight mass spectrometry in our study were mostly hydroxylated metabolites. This indicated that PPD was metabolized in human body mainly through phase I hepatic metabolism. The main metabolites are in 20,24-oxide form with multiple hydroxylation sites. Finally, the metabolic pathways of PPD in vivo (human) were proposed based on structural analysis.

Preparation of 4-(2′-Methoxyethyl)phenol

Abstract:The titled compound (1), which is an important intermediate of metoprolol, was synthesized in overall 29% yield from cheap p-nitrotoluene and paraformaldehyde via condensation, alkylation, reduction, diazotisation and hydrolysis. The key step (p-nitrotoluene condensation with paraformaldehyde) was investigated in detail, including effects of catalysts, solvent, and reactants mole ratio on yield. Thus, 2 mol p-nitrotoluenes reacted with 1 mol paraformaldehyde in DMSO, catalyzed by potassium phenol to give p-nitrophenyl ethanol (4) with the conversion of p-nitrotoluene of 31% and selectivity of 72%. The remaining p-nitrotoluene could be recycled.
Simultaneous Determination of 20(S)-Protopanaxadiol and Its Three Metabolites in Rat Plasma by LC-MS/MS: Application to Their Pharmacokinetic Studies.

Abstract:The aim of this study was to develop an LC-MS/MS method for simultaneous determination of 20(S) protopanaxadiol (PPD) and its three metabolites, PPD-glucuronide (M1), (20S,24S)-epoxy-dammarane-3,12,25-triol (M2) and (20S,24R)-epoxydammarane-3,12,25-triol (M3), in rat plasma. Precipitation with acetonitrile was employed for sample preparation and chromatographic separations were achieved on a C-18 column. The sample was detected using triple quadrupole tandem mass spectrometer with selected reaction monitoring mode. The monitored precursor-to-product ion transitions were m/z 459.4375.3 for PPD, m/z 635.4113.0 for M1, m/z 477.4441.4 for M2 and M3 and m/z 475.4391.3 for IS. The developed assay was validated according to the guidelines of the US Food and Drug Administration. The calibration curves showed good linearity over the tested concentration ranges (r>0.9993), with the LLOQ being 1ng/mL for all analytes. The intra- and inter-day precisions (RSD) were<9.51% while the accuracy (RE) ranged from -8.91 to 12.84%. The extraction recovery was >80% and no obvious matrix effect was detected. The analytes were stable in rat plasma with the RE ranging from -12.34 to 9.77%. The validated assay has been successfully applied to the pharmacokinetic study of PPD as well as its metabolites in rat plasma. According to the pharmacokinetic parameters, the in vivo exposures of M1, M2 and M3 were 11.91, 47.95 and 22.62% of that of PPD, respectively.

Abstract:(2R,3R,4S)-2-Hydroxymethylpyrrolidine-3,4-diol (1,4-dideoxy-1,4-imino-D-ribitol) was synthesized in five steps from N-protected (2S)-3,4-dehydroproline methyl esters. The stereoselective reaction of osmium tetraoxide with dehydroproline derivatives gave high yields of (2S,3R,4S)-3,4-dihydroxyprolines (2,3-trans-3,4-cis-3,4-dihydroxy-L-prolines) accompanied by small amounts (< 15%) of the diastereomeric (2S,3S,4R)-3,4-dihydroxyprolines (2,3-cis-3,4-cis-3,4-dihydroxy-L-prolines). The mixture of the diastereomeric glycols was converted into the isopropylidene acetals, and the isomers separated efficiently on a preparative scale. The resulting protected (2S,3R,4S)-3,4-dihydroxyproline methyl ester was reduced (LiBH4) to the 2-hydroxymethylpyrrolidine and deprotected, resulting in the production of (2R,3R,4S)-2-hydroxymethylpyrrolidine-3,4-diol in high yield and in high purity. The 1H and 13C NMR signals of the product have been unambiguously assigned using two-dimensional NMR techniques, and the identity of the title pyrrolidine confirmed by comparisons of its spectra with those reported for the authentic material.

Abstract:A new route for the synthesis of( 3S,4S)-4-amino-3-hydroxy-5-phenylpentanoic acid derivatives was reported. The target compounds with overall yields of 5. 8% ~ 6. 7% were synthesized by condensation with Weinreb amide,reduction,aldol condensation reaction and bromine imitation reaction,using amino-protected L-phenylalanine as starting material. The structures were confirmed by1 H NMR,13 C NMR and ESI-MS. The exploration of reaction conditions showed that the loading amount of D-proline had no evident effect on the yield,but on stereoselectivity. Larger size protecting groups in substrate can enhanced the stereo-selectivity.
Synthesis of Trinitropyridine and Its N-Oxide

Abstract:2,2-Dinitropropane-1,3-diol was synthesized from nitromethane in low temperature by oxidative nitration method.Silver was successfully recycled with a yield upon 93% and reused in the synthesis of 2,2-dinitropropane-1,3-diol.Potassium 2,2-dinitroethanol,the key intermediatecompound for synthesis of trinitropyridine(TNPy),was prepared through one-pot method with a yield upon 62%,which is 10% higher than conventional method.Controlling the pH value in the range of 1-2,trinitropyridine N-oxide(TNPyO) was synthesized by the cyclization of potassium 2,2-dinitroethanol in 68% nitric acid at 50-60℃.Maintaining the temperature between 80-90 ℃,TNPyO can be further reduced to produce TNPy by refluxing for 1 h;The yields of TNPyO and TNPy are 50% and 15%,respectively.TNPyO can also be prepared from potassium 2,2dinitroethanol under phosphoric acid and sulfuric acid.The products were characterized by IR,nuclear magnetic resonance,elemental analysis and EI-MS.

Abstract:In the present study, a novel sample preparation method based on magnetic core-mesoporous shell microspheres with C8-modified interior pore walls (C8-Fe3O4@mSiO2) was established for the identification of 20(S)-protopanaxadiol (PPD) metabolites in rat plasma by UPLC-Q-TOF-MS/MS analysis. C8-Fe3O4@mSiO2 allowed selective extraction of PPD metabolites from rat plasma by excluding macromolecules in the plasma owing to size exclusion effect. Five extraction conditions including the amount of C8-Fe3O4@mSiO2 microspheres used, extraction time, elution solvents, elution volume, and elution time were investigated and optimized. The present method was compared with two conventional sample preparation methods: protein precipitation and C8 solid phase extraction (C8-SPE). Our method provided higher UPLC intensity of result than protein precipitation method. While the resulting intensity of our method and that of C8-SPE were not significantly different, it consumed less processing time (15min 55s for C8-Fe3O4@mSiO2, and 27min 30s for C8-SPE). Finally, the proposed method was successfully applied in the identification of PPD metabolites in vivo, in which a total of 17 metabolites and the parent drug were identified in rat plasma.
New synthetic route for D-prolinamide

Abstract:Aim To establish a new method for the synthesis of D-prolinamide.Method D-proline was obtained from the asymmetric transformafion of L-proline via the formation of(S)-tartaric acid salt by using butanal as catalyst.Then,D-prolinamide was synthesized by amino protection,esterification,amidation and deprotection.Results and conclusion The yield of final product was 78.4% and optical purity was 99.3%.Comparing with patent synthetic procedure for D-prolinamide,it is a new synthetic procedure with better yield and purity and suitable for industrial manufacture.
