Abstract:Two new 18-norschiartane-type schinortriterpenoids, namely wuwezidilactones Q (1) and R (2) were isolated from the stems of Schisandra lancifolia. Their structures were determined on the basis of extensive spectroscopic analysis. Abstract:Twelve new nortriterpenoids, namely wuweizidilactones J-P (1-7), schindilactones I-K (8-10), preschisanartanin N (11), and schisdilactone J (12), as well as 23 known analogues, were isolated from the leaves and stems of Schisandra chinensis. The absolute configurations of 1 and 11 were established by calculated electronic circular dichroism (ECD) spectra and that of 7 was confirmed by single-crystal X-ray diffraction. The absolute configurations of 2-6, 8, and 9 were determined by an empirical comparison of their experimental ECD spectra with that of 1. It is the first time that the ECD spectra of 18(13 -> 14)-abeoschiartane-, 18-norschiartane-, and schisanartane-type nortriterpenoids possessing an alpha,beta-unsaturated-gamma-lactone moiety are presented, this method providing an alternative means of obtaining absolute configurational assignments for these types of nortriterpenoids. All isolates were evaluated for their anti-acetylcholinesterase and anti-butyrylcholinesterase activities, and compounds 8, 13, 23, and 31 show anti-acetylcholinesterase activity at concentrations of 50 mu M, with 12.7, 10.7, 16.6, and 32.1% inhibition, respectively. Abstract:Schicagenins A-C (1-3), three unprecedented nortriterpenoids characterized with a tetracyclic oxa-cage motif and C(9) side chain, were discovered from the leaves and stems of Schisandra chinensis. Their structures were determined on the basis of extensive spectroscopic analysis, and the absolute stereochemistries were established by single-crystal X-ray diffraction and CD experiments. A plausible biosynthetic pathway of 1-3 was also discussed.

Abstract:Five new dibenzocyclooctadiene lignans, schisphenlignans A-E (1-5), together with eight known ones, were isolated from the stems of Schisandra sphenanthera. The structures of 1-5 were elucidated based on the analysis of their NMR, MS and circular dichroism (CD) spectra. Some isolates were tested for their acute activities on insulin sensitivity in 3T3-L1 differentiated adipocytes, but none of them showed significant bioactivity with 10 mu M administration of the tested compounds.

Abstract:Deoxyschizandrin is an active lignin ingredient originating from Schisandra chinensis (Turcz.) Baill or Schisandrae Sphenantherae Fructus. In the present study, a novel and efficient strategy was developed for the in vivo screening and identification of deoxyschizandrin metabolites using ultra high performance liquid chromatography combined with triple TOF mass spectrometry (UPLC-TOF/MS/MS). This strategy was characterized by the following: a novel and unique multiple mass defect filter (MMDF) combined with an on-line data acquisition method that is dependent on dynamic background subtraction (DBS) was developed to trace all of the probable metabolites of deoxyschizandrin. The MMDF and DBS methods could trigger an IDA scan for the low-level metabolites that are masked by background noise and endogenous components. A combination of data processing methods including extracted ion chromatography (XIC), mass defect filtering (MDF), product ion filtering (PIF) and neutral loss filtering (NLF) were employed to identify the metabolites of deoxyschizandrin. Next, the structures of the metabolites were elucidated based on an accurate mass measurement, the fragmentation patterns of the parent drug and relevant drug bio-transformation knowledge. Finally, an important parameter ClogP was used to estimate the retention time of isomers. Based on the proposed strategy, 51 metabolites (including 49 phase I and 2 phase II metabolites) were identified in rats after the oral administration of deoxyschizandrin. Among these metabolites, 41 metabolites were characterized in the rat urine, and 28 metabolites were identified in the rat bile. The results indicated that oxidization was the main metabolic pathway and that the methoxy group and the biphenyl cyclooctene were the metabolic sites. Conjugation with sulfate and cysteine groups produced two phase-II metabolites. This study firstly reported the description of deoxyschizandrin metabolism in vivo. This study provided a practical strategy for rapidly screening and identifying metabolites, and this methodology can be widely applied for the structural characterization of the metabolites of other compounds.

Abstract:Echinacoside (ECH) is one of the major active phenylethanoid glycosides (PEGs) in famous traditional Chinese medicine, Herba Cistanches. Although it has various bioactivities, such as antioxidation, neuroprotection, and hepatoprotection, knowledge about its metabolic fate is scant. In the present study, eight phase II metabolites, 3,4 -O-dimethyl-ECH-3 -O-beta-d-glucuronide (M1); 4,4 -O-dimethyl-ECH-3 -O-beta-d-glucuronide (M2); 3,4 -O-dimethyl-ECH-4-O-sulfate ester (M3); 4,4 -O-dimethyl-ECH-3-O-sulfate ester (M4); 3,3 -O-dimethyl-ECH (M5); 3,4 -O-dimethyl-ECH (M6); 4,3 -O-dimethyl-ECH (M7); and 4,4 -O-dimethyl-ECH (M8), were isolated from rat bile sample after intravenous administration of ECH and identified by mass spectra and NMR spectroscopy, including (1)H NMR, (13)C NMR, nuclear Overhauser effect difference spectroscopy, and two-dimensional NMR (heteronuclear single quantum correlation, heteronuclear multiple-bond correlation spectroscopy, gradient-selected correlation spectroscopy, and nuclear Overhauser effect spectroscopy). Among them, M5 to M8 were O-di-methylated conjugates; M1 and M2 and M3 and M4 were O-dimethyl glucuronides and O-dimethyl sulfates, respectively. In the three types of metabolites of rat, the major metabolites were the methyl ethers and the glucuronides, whereas the sulfates were minor. The regioselectivity of conjugation for ECH and metabolic pathway of ECH were proposed, which gave insight into the mechanism of ECH for its bioactivities in vivo.

METHOD: The chemical constituents of Schisandra sphenanthera were isolated with chromatographic techniques, and their chemical structures were elucidated by spectral analysis. RESULT and CONCLUSION: Sphenanlignan(1), a new 2,3-dimethyl-1,4-diarylbutane lignan was isolated with 6 known lignans (+)-anwulignan(2), schisantherin A(3), deoxyschizandrin(4), tigloylgomisin O(5), 6-O-benzoylgomisin O(6) and schisandrone(7). 6-O-benzoylgomisin O(6) was obtained from Schisandra sphenanthera for the first time.

Abstract:Two new sesquiterpenoid derivatives, elgonenes M (1) and N (2), and a new shikimic acid metabolite, methyl 5-O-acetyl-5-epi-shikimate (3), were isolated from the mangrove sediment-derived fungus Roussoella sp. SCSIO 41427 together with fourteen known compounds (4–17). The planar structures were elucidated through nuclear magnetic resonance (NMR) and mass spectroscopic (MS) analyses. The relative configurations of 1–3 were ascertained by NOESY experiments, while their absolute configurations were determined by electronic circular dichroism (ECD) calculation. Elgonene M (1) exhibited inhibition of interleukin-1β (IL-1β) mRNA, a pro-inflammatory cytokine, at a concentration of 5 μM, with an inhibitory ratio of 31.14%. On the other hand, elgonene N (2) demonstrated inhibition at a concentration of 20 μM, with inhibitory ratios of 27.57%.

Abstract:Two new monoterpenoid indole alkaloids, named alstoscholines A and B (1 and 2), along with three known ones (3–5) were isolated from the stems of Alstonia scholaris. The new structures including their absolute configurations were elucidated on the basis of HRMS and NMR spectroscopic data and computational calculation methods. Abstract:Schsphenines A ( 1 ) and B ( 2 ), two new lignans were isolated from the fruit of Schisandra sphenanthera , together with six known ones respectively identified as schisanhenol ( 3 ), methylgomisin O ( 4 ), wuweilignan E ( 5 ), neglschisandrin E ( 6 ), schisandrin A ( 7 ) and schisandrin B ( 8 ). The structures of isolated compounds were determined by UV, IR, HR-ESI-MS, NMR, ORD analysis and quantum chemical calculation. The in vitro cardioprotective activities of all compounds were studied on H 2 O 2 -injury H9C2 cells using the MTT method. Compound 1 exhibited a considerable protective effect with cell viability in 79.94 ± 2.36%. This research provided useful information for the utilisation of S. sphenanthera as the TCM.

Abstract:Two new ent-kaurane-derived diterpene derivatives, phyllostacins A (1) and B (2), were isolated from the aerial parts of Isodon phyllostachys, together with two known compounds, irroratin A (3) and serrin B (4). Both 1 and 2 were found to be present as diastereoisomers. In the case of 1, the corresponding diastereoisomeric diacetates 5 and 6 were prepared and separated. The structures of the new compounds were elucidated by extensive 1D- and 2D-NMR spectroscopic methods, in combination with MS experiments. In (D-5)pyridine solution, the two epimers of 1 are present in equal amounts, but in CDCl3 or CD3OD, the (S)-epimer predominates in the mixture of hemiacetals.

Abstract:Melia. toosendan Sieb.et Zucc., a potent herbal medicine, boasts diverse therapeutic properties. As the main components, the isotoosendanin blocks protective autophagy in chemotherapy-induced cultured cancer cells and xenograft tumor tissue to significantly enhancing anticancer activity, and the fraxinellone contributes to pesticidal activity, anti-inflammatory and immunomodulatory effects. Until now, the metabolic profiles remained unknown. In the present study, 13 metabolites of isotoosendanin and 13 metabolites of fraxinellone were characterized in the blood, urine, and feces of rats by UPLC/ESI/qTOF-MS analysis. Five metabolites (F-M3, F-M5, I-M1, I-M5, I-M8) were fully characterized. The isotoosendanin and fraxinellone were mainly involved in hydrogenation, acetylation, and methylenation metabolic reactions. This is the first study illuminates the two components in vivo metabolism, laying the foundation for future pharmacodynamic and mechanistic investigations. It is necessary to deeply understand the process of drug activation, and deactivation, rationally design new drugs, and guide the research and development of new drugs.

Abstract:Eleven undescribed schinortriterpenoids (SNTs) and one known analogue (12) were isolated from the stems and leaves of Schisandra henryi. Their diverse structures included preschisanartane (1), 18-norschiartane (2–5, 12), schiartane (6 and 7), and schisanartane (8–11) skeletons, which were elucidated by comprehensive NMR, MS, electronic circular dichroism analyses, single crystal X-ray diffraction and biogenetic considerations. 1 was the first case of preschisanartane-type SNT with six-membered lactone ring, and 2 was one of the most highly oxygenated 18-norschiartane SNTs. Three types of the highly oxygenated SNTs, 1, 4, 10 and 11, effectively prevent apoptosis induced by corticosterone in PC12 cells. In addition, 11 showed neurite outgrowth-promoting activity.

Abstract:Four new, highly oxygenated nortriterpenoids with unique schisanartane skeletons, micrandilactones D-G (1-4), have been isolated from the leaves and stems of Schisandra micrantha, and their structures have been elucidated on the basis of extensive spectral studies. The postulated biogenetic sequences of sixteen highly oxygenated nortriterpenoids and bisnortriterpenoids with new skeletons from three Schisandra species are discussed and have been compared from a chemotaxonomic standpoint.
