Amino Acid Mutation Effect on Indole-Hydroxylating Ability of Cytochrome P450 BM-3

Abstract:The triple mutant of cytochrome P450 BM-3(A74G/F87V/L188Q) was demonstrated to have the ability of hydroxylating indole into indigo and imdirubin. In order to further improve its catalytic capability, two mutant libraries, created by saturation site-directed mutagenesis at the amino acid positions 168 and 435, were screened by using the spectroscopic assay based on indigo at 670 nm. For hydroxylating indole, three mutants (D168H, D168L and E435T) show higher catalytic and affinity efficiency than that of parent enzyme. Comparing with the parent enzyme, the kcat value of the mutant D168H is three-fold higher, and the mutant D168H also is higher in indigo forming regioselectivity than that of the parent enzyme (93% versus 72%). Furthermore, the coupling efficiency of the mutant D168H is almost two-fold more than that of the parent enzyme, while the coupling efficiency of the mutants D168L and E435T only increases slightly. All the mutants exhibit higher hydroxylation activity at about pH=8.2, and the distinct activity enhancements of the mutants D168H and D168L only cause a slight loss of their enzyme thermostability. These results demonstrate that the amino acid positions 168 and 435 in P450 BM-3 play a critical role in its structure-function relationships.
