Cdna Microarray Analysis of Metabolism-Related Genes in Inoculated Potato Leaves Expressing Moderate Quantitative Resistance to Phytophthora Infestans

Abstract:Late blight is the most devastating disease of potato (Solanum tuberosum L.). To improve our understanding of the molecular mechanism and metabolic network of potato defenses against Phytophthora infestans, we performed a comprehensive transcriptional analysis using a cDNA microarray that contained 1,009 expressed sequence tags (ESTs) from a subtractive library of potato leaves with moderate quantitative resistance against late blight. Potato leaves were inoculated with P. infestans and sampled at ten time-points from 0 - 72 h after inoculation. A total of 98 metabolism-related genes were identified and the predicted transcriptional activity changes in these genes were observed, implying the metabolic reprogramming of potato from "housekeeping" to defense functions. Some defense-related metabolic pathways such as the biosynthesis of aromatic amino acids, phenylpropanoids, oxylipin, and ethylene, were activated, which resulted in the de novo synthesis of salicylic acid (SA), jasmonic acid (JA), and ethylene (ET), suggesting that these signalling pathways may regulate potato defense responses to P infestans. In addition, certain metabolic pathways associated with energy metabolism (e.g., the tricarboxylic acid cycle and glyoxylate cycle) were also activated, indicating an increased energy demand for plants to fight-off pathogen attack. Our results provide a helpful insight into the complexity of interconnected metabolic pathways during the potato P infestans interaction.
