Abstract:Foxp3(+) regulatory T cells (Treg) playing a crucial role in the maintenance of immune tolerance and prevention of autoimmune diseases consist of thymus-derived naturally occurring CD4(+)Foxp3(+) Treg cells (nTreg) and those that can be induced ex vivo with TGF-β (iTreg). Although both Treg subsets share similar phenotypes and functional characteristics, they also have potential biologic differences on their biology. The role of iTreg in regulating B cells remains unclear so far. The suppression assays of Treg subsets on activation, proliferation, and Abs production of B cells were measured using a Treg and B cell coculture system in vitro. Transwell and Ab blockade experiments were performed to assess the roles of cell contact and soluble cytokines. Treg were adoptively transferred to lupus mice to assess in vivo effects on B cells. Like nTreg, iTreg subset also directly suppressed activation and proliferation of B cells. nTreg subset suppressed B cell responses through cytotoxic manner related to expression of granzyme A, granzyme B, and perforin, whereas the role of iTreg subset on B cells did not involve in cytotoxic action but depending on TGF-β signaling. Furthermore, iTreg subset can significantly suppress Ab produced by lupus B cells in vitro. Comparison experiments using autoantibodies microarrays demonstrated that adoptive transfer of iTreg had a superior effect than nTreg subset on suppressing lupus B cell responses in vivo. Our data implicate a role and advantage of iTreg subset in treating B cell-mediated autoimmune diseases, boosting the translational potential of these findings.

Abstract:Inflammatory bowel disease (IBD), including Crohn disease and ulcerative colitis, with multifactorial etiologies has led to a global health-associated burden in many countries. Substantial efforts are devoted to understand the pathogenesis, behavioral and environmental triggers, which may be specifically valuable for the treatment of IBD. The specific pathogenesis underlying IBD is as yet incompletely understood. The use of anti-cytokine therapy and small molecule agents targeting the immune system is thought to restore the body's intestinal barrier function and relieve inflammation with manageable adverse effects. In this review, we report recent advances in anti-cytokine therapy and treatment with small molecule agents for the management of IBD.

Abstract:Regulatory T cells (Tregs) are a subset of T cells with immunosuppressive functions. They secrete IL-10, TGF-âand other suppressive cytokines and direct contact among cells to suppress the autoimmunity response of effector T cells, promote immune tolerance, which has a very important role in maintaining the body’s homeostasis. Foxp3 is a specific transcription factor of Treg cells and plays a key role in the regulation of Treg cell development and function. Treg and Th17 cells are derived from the differentiation of the same type of initial CD4+T cells. The direction of differentiation depends on the type of cytokines in the environment. The functions of them are opposite to each other and transform each other under certain conditions. It plays an important role in inducing immune tolerance and preventing the occurrence of autoimmune diseases (AID). Once immune imbalance, it will lead to the occurrence of immune-related diseases. The decrease of its number or function will affect the immune state of the body, cause disorders of autoimmune tolerance and lead to the occurrence of AID. This article reviews the research of Treg and their cytokines in AID in order to understand the role of Treg in the pathogenesis of AID, and open a new way for the treatment of AID.

Abstract:B cells not only can regulate the immune responses by producing antibody, presenting antigens to target immune cells, but also have the function of immune suppression. A small quantity of regulatory B ( Breg) cells in vivo is capable of performing regulatory functions by producing of inhibitory cytokines,such as interleukin -10, or by direct interaction with other immune cells. In vivo, Breg can maintain immune tolerance and inhibit excessive inflammatory responses efficiently. Clinical studies indicate that Breg are associated with some autoimmune diseases, cancers, infections, and transplantation tolerance. The review focuses on molecules associated with the differentiation and function of the Breg. And also, we discuss the mechanisms and the relations of Breg to clinical diseases.

Abstract:Recent studies of immunology have aroused new interest in regulatory T cell for immune responses,and scientists have recognized that suppressor T cells exist in vivo and regulatory T(Treg) cells play a crucial role in mediating immune tolerance and preventing autoimmune diseases.This article reviews the types,characteristics,mechanisms,and roles of Treg in immune tolerance,aiming to offer some important theoretical evidence for the treatment of allograft rejection and autoimmune diseases.

Abstract:In the past 20 years,the TH1 and TH2 cell classification has provided the basic knowledge for CD4+T cell differentiation and interplay between innate and adaptive immunity.Until recently,novel immune studies have proved that,under the condition of different cytokines and environmental factors,innate T cell can be differentiated into several CD4+T cell effectors——TH1,TH2,TH17 and Treg cell,which are controlled by their respective transcription factors and may thus play important roles in the body.The uncover on the differentiation of TH17 cell may be related to the research on auto-immunity research.And TH17 lineage can be controlled by several factors in the body,for example,it can be induced from innate T cell by the control of both TGF-β and IL-6.In this paper,different regulative mechanism for TH17 differentiation,including T cell effectors TH1,TH2,Treg cell and DC cell,will be reviewed.This promises to deepen the understanding on the new CD4+ T cell subset and its immune regulation,and may provide important basic knowledge for the treatment on auto-immunity and inflammation.

Abstract:Dysregulation of mucosal immune system has been proposed to be critical in the pathogenesis of inflammatory bowel diseases (IBDs). Regulatory T cells (Tregs) play an important role in regulating immune responses. Tregs are involved in maintaining intestinal homeostasis and exerting suppressive function in colitis. Our previous studies showed that a novel forkhead box protein P3 (Foxp3) negative Tregs (Treg-of-B cells), induced by culturing naïve CD4+ T cells with B cells, could protect against colitis and downregulate T helper (Th) 1 and Th17 cell cytokines in T cell-mediated colitis. In the present study, we aimed to induce Treg-of-B cells in the CD8+ T-cell population and investigate their characteristics and immunomodulatory functions. Our results showed that CD8+ Treg-of-B cells expressed Treg-associated markers, including lymphocyte-activation gene-3 (LAG3), inducible co-stimulator (ICOS), programmed death-1 (PD-1), cytotoxic T-lymphocyte-associated protein-4 (CTLA-4), tumor necrosis factor receptor superfamily member-4 (TNFRSF4, OX40), and tumor necrosis factor receptor superfamily member-18 (TNFRSF18, GITR), but did not express Foxp3. CD8+ Treg-of-B cells produced higher concentration of inhibitory cytokine interleukin (IL)-10, and expressed higher levels of cytotoxic factor granzyme B and perforin after stimulation, compared to those of CD8+CD25- T cells. Moreover, CD8+ Treg-of-B cells suppressed T cell proliferation in vitro and alleviated colonic inflammation in chronic dextran sulfate sodium (DSS)-induced colitis. In conclusion, our study identified a novel subpopulation of CD8+ Tregs with suppressive effects through cell contact. These CD8+ Treg-of-B cells might have therapeutic potential for IBDs.

Abstract:Inflammatory bowel disease (IBD) is caused by an uncontrolled immune response in the intestinal lumen, leading to inflammation in genetically predisposed individuals. Immunotherapy may be a promising approach to the treatment of IBD. Here, we show that transforming growth factor-β1 (TGF-β1) gene-modified immature dendritic cells (imDCs) could enhance the inhibitory function of imDCs and delay the progress of IBD induced by dextran sodium sulfate in mice. The results of fluorescence-activated cell sorter (FACS) demonstrated that this protective effect is mediated partially by inducing CD4+Foxp3+ regulatory T cells (Tregs) in mesentery lymph nodes to control inflammation. In vitro experiments also supported this hypothesis. In conclusion, we provide evidence that TGF-β1-modified bone marrow-derived imDCs may have a therapeutic effect to IBD.

Abstract:Research into the suppressive activity of CD4(+)FoxP3(+) T regulatory cells (Treg) has defined a sublineage of CD4(+) cells that contribute to self-tolerance and resistance to autoimmune disease. Much less attention has been given to the potential contribution of regulatory sublineages of CD8(+) cells. Analysis of a small fraction of CD8(+) cells that target autoreactive CD4(+) cells through recognition of the MHC class Ib molecule Qa-1 in mouse and HLA-E in human has revitalized interest in CD8(+) Treg. Here we summarize recent progress and future directions of research into the role of this CD8(+) sublineage in resistance to autoimmune disease.
Control of intestinal homeostasis by regulatory T cells and dendritic cells

Abstract:Many different pathways contribute to the maintenance of tolerance to harmless antigens in the intestine. When these important pathways are compromised, chronic intestinal inflammation can develop. In particular, naturally occurring CD4+CD25+ regulatory T cells have been shown to play an important role in the prevention and cure of colitis in animal models of intestinal inflammation. These regulatory T cell responses may be influenced by the local environment in the intestine. For example, functionally specialised populations of dendritic cells exist in the intestine which may favour regulatory type responses. Understanding how these pathways intersect may lead to the development of more specific therapies for the treatment of inflammatory bowel disease.

Abstract:The immune system is pivotal in mediating the interactions between host and microbiota that shape the intestinal environment. Intestinal homeostasis arises from a highly dynamic balance between host protective immunity and regulatory mechanisms. This regulation is achieved by a number of cell populations acting through a set of shared regulatory pathways. In this review, we summarize the main lymphocyte subsets controlling immune responsiveness in the gut and their mechanisms of control, which involve maintenance of intestinal barrier function and suppression of chronic inflammation. CD4 + Foxp3 + T cells play a nonredundant role in the maintenance of intestinal homeostasis through IL-10- and TGF-β-dependent mechanisms. Their activity is complemented by other T and B lymphocytes. Because breakdown in immune regulatory networks in the intestine leads to chronic inflammatory diseases of the gut, such as inflammatory bowel disease and celiac disease, regulatory lymphocytes are an attractive target for therapies of intestinal inflammation.

Abstract:In the intestinal tract, only a single layer of epithelial cells separates innate and adaptive immune effector cells from a vast amount of antigens. Here, the immune system faces a considerable challenge in tolerating commensal flora and dietary antigens while preventing the dissemination of potential pathogens. Failure to tightly control immune reactions may result in detrimental inflammation. In this respect, 'conventional' regulatory CD4(+) T cells, including naturally occurring and adaptive CD4(+) CD25(+) Foxp3(+) T cells, Th3 and Tr1 cells, have recently been the focus of considerable attention. However, regulatory mechanisms in the intestinal mucosa are highly complex, including adaptations of nonhaematopoietic cells and innate immune cells as well as the presence of unconventional T cells with regulatory properties such as resident TCRgammadelta or TCRalphabeta CD8(+) intraepithelial lymphocytes. This review aims to summarize the currently available knowledge on conventional and unconventional regulatory T cell subsets (Tregs), with special emphasis on clinical data and the potential role or malfunctioning of Tregs in four major human gastrointestinal diseases, i.e. inflammatory bowel diseases, coeliac disease, food allergy and colorectal cancer. We conclude that the clinical data confirms some but not all of the findings derived from experimental animal models.

Abstract:CD4⁺CD25⁺Foxp3⁺ T lymphocytes, known as regulatory T cells or T(regs), have been proposed to be a lineage of professional immune suppressive cells that exclusively counteract the effects of the immunoprotective "helper" and "cytotoxic" lineages of T lymphocytes. Here we discuss new concepts on the mechanisms and functions of T(regs). There are several key points we emphasize: 1. Tregs exert suppressive effects both directly on effector T cells and indirectly through antigen-presenting cells; 2. Regulation can occur through a novel mechanism of cytokine consumption to regulate as opposed to the usual mechanism of cytokine/chemokine production; 3. In cases where CD4⁺ effector T cells are directly inhibited by T(regs), it is chiefly through a mechanism of lymphokine withdrawal apoptosis leading to polyclonal deletion; and 4. Contrary to the current view, we discuss new evidence that T(regs), similar to other T-cells lineages, can promote protective immune responses in certain infectious contexts (Chen et al., 2011; Pandiyan et al., 2011). Although these points are at variance to varying degrees with the standard model of T(reg) behavior, we will recount developing findings that support these new concepts.

Abstract:Regulatory T cells (Treg) play a key role in the negative regulation of the immune system, which can inhibit inappropriate self-reactive T cells, as well as limit the range, extent and lasting time of immune responses, so as to inhibit the proliferation of CD4(+) and CD8(+) effector T cells. In view of these properties, there is little doubt about the enormous potential value of Tregs in molecular mechanisms and clinical treatment of human autoimmune disorders, graft-versus-host disease (GVHD) and allergies. In this review, the molecular mechanisms of CD4(+)CD25(+) Treg and CD8(+) Treg were mainly discussed, and challenge with which study on Treg cells is faced and prospects were briefly described.

Abstract:The inflammatory bowel diseases (IBDs), represented mainly by Crohn disease and ulcerative coil-tis but also including noninfectious inflammations of the bowel, are characterized by persistent intestinal inflam-mation which is due to disordered immune response. Although the pathogenesis of IBD remains unknown, it is generally accepted that IBD is the results of the combinational effect of environmental, genetic, and immunolog-icnl factors in which an uncontrolled immune response within the intestinal lumen leads to inflammation in ge-netically predisposed individuals. Existing conventional treatments are not ideal and have severe side effects. Advances in understanding the immunological pathogenesis of IBD have allowed the development of novel thera-pies.
[Research advance on the interaction between gut microbes and cytokines in inflammatory bowel disease]

Abstract:Dysregulation of interaction between the gut microbiota and the host immune system leads to the development of chronic inflammation, such as inflammatory bowel disease (IBD), including Crohn's disease (CD) and ulcerative colitis (UC). Numerous studies have shown that the host-microbiota of IBD can directly or indirectly affect the homeostasis of the immune system. Furthermore, the balance between pro-inflammatory and anti-inflammatory cytokines involves in maintaining a healthy microbiota community and enhances epithelial barrier functions. Understanding the interactions between gut microbiota and cytokines (IL-10, IL-17, IL-18, TNF-α) provides crucial insights into the pathogenesis of inflammatory bowel disease.

Abstract:Foxp3+ T regulatory cell (Treg) subsets play a crucial role in the maintenance of immune homeostasis against self-antigens. The lack or dysfunction of these cells contributes to the pathogenesis and development of many autoimmune diseases. Therefore, manipulation of these cells may provide a novel therapeutic approach to treat autoimmune diseases. In this review, we provide current opinions concerning the classification, developmental, and functional characterization of Treg subsets. Particular emphasis will be focused on the therapeutic role of TGF-β-induced CD4M+ Foxp3+ cells (iTregs) in established autoimmune disease. Moreover, the similarity and diversity of iTregs and naturally occurring, thymus-derived CD4+ CD25+ Foxp3+ regulatory T cells (nTregs) will be discussed, including the finding that the pro-inflammatory cytokine IL-6 can convert nTregs to IL-17-producing cells, whereas iTregs induced by TGF-β are resistant to the effects of this cytokine. Understanding these aspects may help to determine how Tregs can be used in the treatment of autoimmune diseases.

Abstract:Regulatory B (Breg) cells represent a population of suppressor B cells that participate in immunomodulatory processes and inhibition of excessive inflammation. The regulatory function of Breg cells have been demonstrated in mice and human with inflammatory diseases, cancer, after transplantation, and particularly in autoinflammatory disorders. In order to suppress inflammation, Breg cells produce anti-inflammatory mediators, induce death ligand-mediated apoptosis, and regulate many kinds of immune cells such as suppressing the proliferation and differentiation of effector T cell and increasing the number of regulatory T cells. Central nervous system Inflammatory demyelinating diseases (CNS IDDs) are a heterogeneous group of disorders, which occur against the background of an acute or chronic inflammatory process. With the advent of monoclonal antibodies directed against B cells, breakthroughs have been made in the treatment of CNS IDDs. Therefore, the number and function of B cells in IDDs have attracted attention. Meanwhile, increasing number of studies have confirmed that Breg cells play a role in alleviating autoimmune diseases, and treatment with Breg cells has also been proposed as a new therapeutic direction. In this review, we focus on the understanding of the development and function of Breg cells and on the diversification of Breg cells in CNS IDDs.

Abstract:Inflammatory bowel disease (IBD) is a chronic, inflammatory, and autoimmune disorder. The pathogenesis of IBD is not yet clear. Studies have shown that the imbalance between T helper 17 (Th17) and regulatory T (Treg) cells, which differentiate from CD4+ T cells, contributes to IBD. Th17 cells promote tissue inflammation, and Treg cells suppress autoimmunity in IBD. Therefore, Th17/Treg cell balance is crucial. Some regulatory factors affecting the production and maintenance of these cells are also important for the proper regulation of the Th17/Treg balance; these factors include T cell receptor (TCR) signaling, costimulatory signals, cytokine signaling, bile acid metabolites, and the intestinal microbiota. This article focuses on our understanding of the function and role of the balance between Th17/Treg cells in IBD and these regulatory factors and their clinical significance in IBD.
