Effects and Significance of Amiodarone on the Proliferation of Human Hepatocellular Carcinoma Cell Line HepG2 in Vitro

Abstract:Objective: To explore the effects of amiodarone on proliferation and cell cycle distribution of human hepatocellular cancer (HCC) cell HepG2 in vitro, and to discuss the possible molecular mechanism. Methods: The absorbance value and cell inhibition rate of proliferation of HepG2 cells were detected by MTT test, and parameters of cell cycle distribution was measured by flow cytometry using propidium iodide (PI) staining after being treated by amiodarone or 5-fluorouracil (5-Fu); and, accordingly, the characteristics of amiodarone on proliferation and cell cycle distribution of HepG2 were analyzed. Results: HepG2 cells shrank and refractive index decreased gradually along with the increase of concentration of amiodarone from 0.5 μg/ml to 4 μg/ml, while there were gradual growth in quantity observed of dead cells. The proliferation of HepG2 cells was inhibited dramatically, which showed obvious concentration-effect relation and time-effect relation. There were marked differences in parameters of cell cycle distribution and inhibition rates between amiodarone group and control group, which were of statistical significance (P<0.01); however, no statistical significance was observed between amiodarone group and 5-Fu group(P>0.05). The characteristics of cell cycle distribution changed markedly when HepG2 cells were interfered with amiodarone at 0.5-4.5 mg/L in vitro: the percentage of cells in stage G0/G1 and G 2/M increased, while that of cells in stage S reduced. The trend was similar to that of 5-Fu group. Conclusion: Amiodarone can inhibit the proliferation of HepG2 in vitro, which showed concentration-effect relation and time-effect relation. Amiodarone might be a new alternative of neoadjuvant chemotherapy for patients with HCC.
