Effects of Polypeptide Extract from Scorpion Venom on Invasive Ability of Prostatic Carcinoma Cells in Vitro

Abstract:OBJECTIVE:To observe the effects of pomypeptide extract from scorpion venom (PESV) on the invasive ability of prostatic carcinoma DU-145 cells, and investigate the potential mechanism.METHODS:After the treatment of DU-145 cells with PESV, MTT assay was used to test the effect on the proliferation.The invasive ability was assessed with a transwell cell culture chamber. Immunohistochemistry stain and real-time PCR analysis were used to investigate the expressions of MMP-9 and TIMP-1.RESULTS:The growth of cancer cells was inhibited in a dose dependent manner.The cells moved through the membrane were (51.29±7.56)/field in the control group,while decreased to (20.43±1.51),(12.29±2.75) and (5±1.73)/field after being treated with 5,10 and 20 μg/mL PESV, respectively. The differences between each group among the four groups were all significant,F=38.933,P=0.000. Compared with the control group,the cells moved through the membrane of PESV groups decreased significantly,P=0.000. After being treated with PESV, the expression of MMP-9 was decreased and TIMP-1 was increased.CONCLUSION:PESV can inhibit the invasiveness of DU-145 cells, and the mechanism is related to the down regulation of MMP-9 and the up regulation of TIMP-1 in DU-145 ceI1s.
