Effect of advanced glycation end products and its receptor on oxyradical metabolization in rat cavernosum

Abstract:Objective: To illuminate the role of advanced glycation end products (AGEs) in the development of diabetic mellitus erectile dysfunction (DMED), and investigate the effect of AGEs and RAGE on oxyradical metabolization in rat cavernosum. Methods: Forty rats were randomly taken out from 60 male Sprague-Dawley rats for diabetes mellitus (DM) models. One group of DM rats was given freely access to water(DM group) and another group was given 1g/L aminoguanidine hydrochloride strating from the occurrence of DM (DM+AG group). Two groups of rats that were not DM acted as age-matched group CO and CO+AG. After eight weeks cavernosum tissue of rate were harvested for analysis of distribution and expression of AGEs and RAGE, content of AGE-peoptide (AGE-P), malondialdehyde (MDA) as well as the activity of superoxide dismutase (SOD). Results: Compared with that of CO group, the expression of AGEs, RAGE, the content of AGE-P, MDA in rat cavernosum tissue of DM group were significantly higher P<0.05, but the activity of SOD cavernosum tissue of ACE-P and MDA were downregulated distinctly under the administration of AG, whereas activity of SOD enhanced; There was no difference between CO and CO+AG groups in all datas (P>0.05). Conclusion: In the DM condition, the combination of AGEs and RAGE results in the overproduction of oxyradical in rat cavernosum. All these changes may promote the development of DMED.
