Antineoplastic effect of tetrandrine on primary cancer cells isolated from ascites

Abstract:Objective: To study the effect of tetrandrine on primary cancer cells isolated from the ascites of patients with metastatic cancers. Methods: Primary cancer cells were isolated from the ascites of patients (n = 21). Compared with culture cell lines BGC-823, LOVO and SMMC-7721, the response of these cancer cells to tetrandrine and chemotherapeutic agents commonly used in clinical were determined by WST-8 assay. Quantitative RT-PCR was used to examine the role of apoptotic associated genes Survivin and Bcl-2 in the antineoplastic effect of tetrandrine. Results: The primary cancer cells isolated from effusions showed a sensitivity to tetrandrine with IC 50 values of 32.96 μm, similar than that in the established cell lines(P>0.05). The antineoplastic effect of tetrandrine on primary cancer cells isolated from gastric, colon cancers and other cancers was found no statistic significance (P > 0.05). Cancer cells of 2 cases isolated from the effusions were resistant to tetrandrine, which also had the increased tolerance to docetaxel, cisplatin and 5-fluorouracil. A significant down-regulation role of tetrandrine on Survivin and Bcl-2 mRNA was obeserved in some primary cancer cells and cell lines (P < 0.05). Conclusion: Tetrandrine may have potential antineoplastic effect on primary cancer cells, especially those chemotherapetic-resistant cells.
