Influence of osteogenic growth peptide microsphere on osteoblast proliferation

Abstract:Background: It is a new domain to use biodegradable polymer polylactide-co-glycolide (PLGA) as scaffold to prepare delayed release microspheres by carrying polypeptide and therapeutic grade protein. Objective: To explore the bioactivities of osteogenic growth peptide (OGP) microspheres on the cultured osteoblasts. Design, time and setting: Comparative observation was performed at the Laboratory of School of Life Science and Technology, Xi'an Jiaotong University. Materials: New Zealand rabbits were used to isolate osteoblasts; PLGA was provided by Shandong Medical Apparatus Research Institute. Methods: OGP was microcapsulated with the multiple emulsion encapsulative method using PLGA as coating material. Its morphology, particle size distribution, drug loading-embedding rate and in vitro release property were studied. In OGP group, osteoblasts were cultured in DMEM containing 0.1 volume fraction calf serum and 50 μ g/L OGP; in OGP microsphere group, cells were cultured in DMEM containing 0.1 volume fraction calf serum and 50 μ g/L OGP-PLGA microsphere. Main outcome measures: Morphology, particle size distribution, drug loading-embedding rate and in vitro release property of microsphere; influence of OGP rnicrosphere on cell proliferation. Results: The morphology of the OGP-PLGA microspheres were even and good; the particle size was (19.6±4.47) μ m; the drug-loading and drug-embedding rate of microspheres were (83.9±4.21)% and (72.9±5.13)%; the release property of microspheres in vitro was good and the overall release rate was 85.43% in 56 days. On culture days 1 and 2, the cell viability of OGP group was much better than that of OGP-PLGA group (P < 0.05); on days 3 and 4, the cell viability of OGP group and OGP-PLGA group were not statistically different (P > 0.05), but on clays 6 and 8, the cell viability of OGP-PLGA group was much better than OGP group (P < 0.05). Conclusion: Multiple emulsion encapsulative method is practical to prepare the OGP-PLGA microspheres. OGP-PLGA microspheres can preserve the bioactivities of OGP effectively and promotes the proliferation of osteoblasts in a long period because of the controlled release of OGP from microspheres.
