A Rat Model Of Congestive Heart Failure Following Myocardial Infarction.

Abstract:Background: Left anterior descending branch ligation of rat can cause consistent myocardial infarction at infarction site, and the left ventricle function and pathophysiologic alterations are similar to clinical myocardial infarction. The study is based on the animal model of congestive heart failure after myocardial infarction. Objective: To establish and evaluate a rat model of congestive heart failure following myocardial infarction. Design, time and setting: Paired sample observation was performed at the University of Southern California from January to August 2007. Materials: Ten 8-month-old healthy male SD rats, weighing (500±25) g, were randomly divided into experimental group (n=10) and sham operation group (n=8). Methods: The left anterior descending coronary artery of SD rats in the experimental group was ligated to induce myocardial infarction. In sham operation group, the animals only underwent open-chest surgery, but not ligation. Main outcome measures: Electrocardiogram was recorded. Four weeks later, echocardiography was used to estimate the effect of ligation; A catheter was advanced from the right carotid artery into the left ventricle to measure heymodynamics parameters such as left ventricle diastolic pressure, the rate of left ventricle pressure increase at 40 mm Hg (dP/ dt40) and the rate of left ventricle pressure decline (-dP/ dt). Blue ink was injected from the root of aorta to assess the area of infarction; frozen sections were performed to detect the thickness of left ventricular anterior wall. Results: Myocardial infarction manifestations were induced successfully in rats including changes of electrocardiogram, ejection fraction reduction by echocardiography measurement, left ventricle diastolic pressure elevation, dP/dt40 and -dP/dt reduction. Ink injection indicated large area of myocardial infarction and attenuation of left ventricular anterior wall. Conclusion: The results of the present study indicate that ideal animal model of congestive heart failure following myocardial infarction is induced by ligation of left anterior descending coronary artery.
