Protective effect of adipose stromal cells on the apoptosis of cardiomyocytes following hypoxia preconditioning

Abstract:Background: Transplantation of newly found adipose-derived stromal cells into the infarcted myocardium can improve heart function. Adipose-derived stromal stem cells can differentiate into myocardium and endothelial cells. Whether adipose-derived stromal stem cells have directly effects on host cardiomyocytes deserves further studies. Objective: To explore the effects and mechanisms of adipose stromal stem cells on the apoptosis of cardiomyocytes following hypoxia preconditioning. Design, time and setting: The randomized, cell molecule experiment was performed at the Institute of Hypertensive Disease, First Affiliated Hospital, Nanchang University from December 2007 to June 2008. Materials: Collagenase I and erythrocyte lysis buffer (Invitrogen, USA), Dulbecco modified Eagle medium, fetal bovine serum and trypsin (Gibco, USA) were used in this study. Methods: Adipose tissues from ten female Sprague Dawley rats were digested with collagenase type I solution and adipose stromal stem cells were derived by culture. The cells surface phenotype was examined by flow cytometry. Cardiomyocytes from five neonatal rats were cultured and divided into 4 groups, which were respectively incubated in DMEM, adipose stromal stem cell supernatant under normal conditions, adipose stromal stem cell supernatant under hypoxia, and wortmannin. DMEM group served as a control group. All the 4 groups of adipose stromal stem cells were cultured under hypoxia conditions (O2, CO2, N2 with volume fraction of 0.02, 0.05, 0.93) for 3 days. Main outcome measures: Anexin V/P1 double staining was used to investigate the apoptotic rate of cardiomocytes. Western blotting was used to test the expression of protein kinase and phosphorylated protein kinase. Results: Adipose stromal stem cells were derived by culture and expressed CD44, CD90, while did not expressed CD31 or CD45. The apoptotic rate of cardiomyocytes in the control group was significantly higher than in the conventional adipose stromal stem cell and hypoxia adipose stromal stem cell groups (P < 0.01). The rate in hypoxia adipose stromial stem cell group was significantly lower than in conventional adipose stromal stem cell group (P < 0.05). No significant differences were detected in apoptotic rate between the wortmannin group and the control group (P > 0.05). The ratio of phosphorylated protein kinase to protein kinase was significantly higher in the hypoxia adipose stromal stem cells than in the routine adipose stromal stem cells (P < 0.01). No significant difference was found in the ratio of phosphorylated protein kinase to protein kinase between the wortmannin group and the control group (P > 0.05). Conclusion: Adipose stromal stem cells following hypoxia preconditioning can elevate the protective effect on cardiornyocyte apoptosis. This mechanism may be associated with adipose stromal stem cells secreting cytokines which effect the P13-K/ protein kinase signaling pathway.
