Biocompatibility of collagen sponge versus temperature-sensitive collagen type Ⅰ

Abstract:BACKGROUND: Cardiac structure is extremely complex and muscular tissues show a high mechanical compliance, therefore, myocardium tissue engineering demonstrates a high requirement for the materials. OBJECTIVE: To explore the biocompatibility of collagen sponge or temperature-sensitive collagen type I with bone marrow mesenchymal stem cells (BMMSCs), and provide an ideal material for myocardium tissue engineering. DESIGN, TIME AND SETTING: A contrast observation was carried out in the Laboratory of Cardiology, Xijing Hospital, the Fourth Military Medical University of Chinese PLA from March to October in 2007. METHODS: The in vitro cultured rat BMMSCs were compounded to collagen sponge and temperature-sensitive collagen type I, respectively, followed by the culture for 7 days. Samples were obtained at 1, 3, 5, 7 days of culture. MAIN OUTCOME MEASURES: The morphology and adhesion of BMMSCs were observed under scanning electron microscope and hematoxylin-eosin dyeing; the proliferation of BMMSCs was detected by MTT method. RESULTS: ①Under scanning electron microscope, the BMMSCs on the temperature-sensitive collagen type I adhered, which were better than those on collagen sponge, these cells grew and connected with each other, with a large amount of secretory granules observed on the surface.②MTT result showed that, the BMMSCs activity and the speed of cell division and proliferation were all increased on the temperature-sensitive collagen type I material compared with those on collagen sponge material at 1, 3, 5, 7 days of culture (P < 0.01). CONCLUSION: The biocompatibility of temperature-sensitive collagen type I with BMMSCs is superior to that of collagen sponge.
