Abstract:The need for AIDS vaccine has been emphasized by the increase of HIV-1 prevalence in sexual transmission which bridges the spreading of HIV-1 from high-risk population to other populations. After more than two-decade intensive effort on the AIDS vaccine development, it remains elusive whether and how an effective vaccine will be achieved. Recent data released from a phase III trial in Thailand showed a partial protection might be accomplishable by the "prime-boost" combination of two vaccines: ALVAC® HIV vaccine (the prime), and AIDSVAX® B/E vaccine (the boost). This unprecedented large clinical trial observed that the prime-boost combination lowered the rate of HIV infection by 31.2% compared to placebo based on the modified intent-to-treat population (n=51 vs. n=74, respectively; P=0.04). However, debating on the efficacy interpretation of this trial arose among field scientists. Furthermore, how to improve the efficacy will become the most important question to be tackled. Here we reviewed the recent publications and summarized the major progress achieved.

Abstract:Since 1987, when the first AIDS vaccine candidates went into clinical trials to date, there have been hundreds of AIDS vaccines in pre-clinical and clinical trials, but none has been proven to be effective. Especially in and 2008, with high expectation, the V520 vaccine(STEP and PAVE trials) was declared void, and the international AIDS vaccine suffered a major setback. Scholars in this field gathered to discuss problems in the field of AIDS research, including vaccine development and basic research. In order to better summarize the experience of HIV vaccine research and look forward to the future research directions, this review will give a brief introduction to clinical trials of AIDS vaccine and vaccine development strategy, while the reasons for the failure of hot vaccines and new ideas will be focused on.

Abstract:Although the RV144 trial reported an AIDS vaccine regimen conferred certain protection against HIV acquisition,the vaccine is far from being used in clinical practice for the low protection rate. Since both humoral and cellular immune responses play important roles in inhibiting and controlling HIV-1 infection, we believe the efficacy of AIDS vaccine can be further improved if the two arms of adaptive immune systems can work synergistically. Supported by the National Science and Technology Key Project on Major Infectious Diseases, our laboratory designed a vaccine strategy against major circulating HIV-1 subtypes in China. This review briefly introduces the rationale of our design and major achievements.

Abstract:Despite rigorous scientific efforts over the forty years since the onset of the global HIV pandemic, a safe and effective HIV-1 vaccine remains elusive. The challenges of HIV vaccine development have proven immense, in large part due to the tremendous sequence diversity of HIV and its ability to escape from antiviral adaptive immune responses. In recent years, several phase 3 efficacy trials have been conducted, testing a similar hypothesis, e.g., that non-neutralizing antibodies and classical cellular immune responses could prevent HIV-1 acquisition. These studies were not successful. As a result, the field has now pivoted to bold novel approaches, including sequential immunization strategies to drive the generation of broadly neutralizing antibodies and human CMV-vectored vaccines to elicit MHC-E-restricted CD8+ T cell responses. Many of these vaccine candidates are now in phase 1 trials, with early promising results.

Abstract:There are about 35 HIV vaccine candidates around the world entered into early phase of clinical trial at the beginning of 2005. The selected antigens usually could induce both cellular and humoral immune response. Currently the HIV vaccine candidates include traditional vaccine (killed or attenuated vaccine), peptide, protein subunit, DNA vaccine and live vectorial vaccine. The immunization strategies were developed to increase the immunogenicity through antigen priming/boosting, and mucosa immunization. There are both modification, immune adjuvant co-inoculation, numerous opportunities and challenges in the field of Chinese HIV vaccine research, therefore, we should work closely and effectively to facilitate HIV vaccine development.

This review delves into the history of HIV vaccines and the current efforts in HIV prevention, emphasizing pre-clinical vaccine development using the non-human primate model (NHP) of HIV infection. NHP models offer valuable insights into potential preventive strategies for combating HIV, and they play a vital role in informing and guiding the development of novel vaccine candidates before they can proceed to human clinical trials.

Abstract:Objective To discuss the research situation and development trend of AIDS vaccine technology.Methods The research papers about the theme of AIDS vaccine technology from the Web of science database were retrieved,the data was cleaned and managed by TDA software.The development trend from the country/region distribution,country/region cooperation network,cited literature and other aspects were made statistical analysis by the method of co-citation analysis and visual tools such as Ucinet and SPSS.Results The research found that AIDS vaccine technology in the field of papers has been mainly concentrated in the United States,Britain and other countries since 2011,of which,the number of Chinese papers ranked fourth.The international cooperation in the AIDS vaccine field of China was less.There were 9 frontier fields of AIDS vaccine technologies through the research such as structure of a soluble cleaved HIV-1 envelope trimer,the research on animal model of HIV/AIDS.Conclusion In recent years,the research of AIDS vaccine technology has developed rapidly in the world,but the research strength of our country is still relatively weak compared with developed countries such as the United states.

Abstract:Based on data from the Chinese Center for Disease Control and Prevention (CDC), there were a total of 861,042 people with human immunodeficiency virus (HIV)/ immune deficiency syndrome (AIDS) as of December 31, 2018 in China, a total of 148,589 new HIV infections, and 38,134 AIDS-related deaths in the year 2018. As of 2017, only 74% of people living with HIV knew their status, 80% of people living with HIV were receiving treatment, and 91% were virally suppressed in China. Although mankind has made great progress in the fight against AIDS in recent years, the vision of ending the AIDS epidemic still faces many challenges in China. Due to the huge population and the imbalance in the prevalence of HIV/AIDS in China, expanding HIV screening and early detection remains the key to Chinas response to HIV. Limitations of antiviral therapy (ART), rejection or discontinuation of an immediate ART strategy by people infected with HIV, and the difficult search for a cure for AIDS all limit the coverage and quality of treatment. The high price of drugs and lack of vaccines present enormous challenges; social discrimination still exists, and participation by non-governmental organizations in prevention, treatment, and care is limited. As part of the future response to HIV, HIV eradication programs should continue to be explored, and attention should be paid to long-term care for people living with HIV.

Abstract:Background: The effort to develop an effective preventive vaccine against HIV-1 infection is challenged by the wide genetic diversity of HIV-1 among different isolates. Objectives: To explore a new vaccination strategy by using heterologous HIV immunogens derived from different clades for sequential priming and boosting. Methods: HIV Env and Gag immunogens derived from Thailand B (B'), C/B' recombinant and A/E recombinant were selected as these three clades account for 29%, 45% and 15% of HIV-1 prevalence in China, respectively. Three humanized fusion genes of env and gag derived from those three clades were synthesized and inserted into DNA and recombinant Tiantan vaccinia vectors as model vaccines. C57BL/6 and Balb/c mice were used as animal model. Peptides spanning the entire Env and Gag were used as stimuli and Elispot assay was used to assess the T cell immunity. Results: Sequential priming and boosting was observed with heterologous HIV immunogens predominantly stimulated T cell immunity against conserved epitopes, whereas a single vaccine derived from one clade or the mixture of multiple vaccines from different clades primarily raised T cells against less conservative or non-conservative epitopes. Conclusions: This is the first demonstration of a practical strategy to raise immune responses against conserved epitopes. This strategy has important implications for vaccine development against HIV and other pathogens that have high genetic diversity, such as influenza. (c) 2006 Lippincott Williams & Wilkins.

Abstract:ABSTRACT: In the past 37 years, human immunodeficiency virus/acquired immunodeficiency syndrome (HIV/AIDS) has undergone various major transmission routes in China, with the world most complex co-circulating HIV-1 subtypes, even the prevalence is still low. In response to the first epidemic outbreak of HIV in injecting drug users and the second one by illegal commercial blood collection, China issued the Anti-Drug Law and launched the Blood Donation Act and nationwide nucleic acid testing, which has avoided 98,232 to 211,200 estimated infections and almost ended the blood product-related infection. China has been providing free antiretroviral therapy (ART) since 2003, which covered >80% of the identified patients and achieved a viral suppression rate of 91%. To bend the curve of increasing the disease burden of HIV and finally end the epidemic, China should consider constraining HIV spread through sexual transmission, narrowing the gaps in identifying HIV cases, and the long-term effectiveness and safety of ART in the future.

Abstract:Despite progress in antiretroviral therapy, pre-exposure prophylaxis, microbicides, and other preventive strategies, a vaccine to prevent HIV-1 infection remains desperately needed. Development of an effective vaccine is challenged by several immunologic features of HIV-1 evidenced by the failure of five of the six HIV-1 candidate vaccine efficacy trials to date. This chapter reviews these efficacy trials with a focus on the Phase 3 RV144 trial in Thailand, the only HIV-1 vaccine efficacy trial to show a moderate protective effect of 31% with respect to placebo administration. Although modest, this protection has allowed for the study of potential immunologic correlates of protection to improve development of future HIV-1 pox-protein and other vaccine strategies. Trials in Thailand and South Africa have built upon the RV144 framework to provide additional immunologic insights which enable current and future efficacy testing of related vaccine candidates.

Abstract:Vaccines play important roles in preventing infectious diseases caused by different pathogens. However, some pathogens such as HIV-1 challenge current vaccine strategy. Poor immunogenicity and the high mutation rate of HIV-1 make great difficulties in inducing potent immune responses strong enough to prevent infection via vaccination. Epitope-vaccine, which could intensively enhance predefined epitope-specific immune responses, was suggested as a new strategy against HIV-1 and HIV-1 mutation. Epitope-vaccines afford powerful approaches to elicit potent, broad and complete immune protection against not only primary homologous viral isolates but also heterologous viral mutants. Although most studies are still preliminary now, epitope-vaccine as a novel strategy against the AIDS epidemic has great developmental potential. To trigger T-cell-dependent IgG antibody responses and improve affinities of the epitope-specific antibodies, approaches such as recombinant multi-epitope-vaccination and prime-boosting vaccination were suggested. Cellular immune responses, especially CTL responses, could also be elicited and enhanced in addition to humoral immune responses. Developed epitope-vaccines activating both arms of the immune system would benefit prevention and immunotherapy not only against HIV but also other chronic infections.

Abstract:This systematic review aims to provide with information for the future clinical trials of human immunodeficiency virus(HIV) preventive vaccine in China on the phase Ⅱ/Ⅲ clinical trials of HIV preventive vaccines worldwide.Related literatures were collected by the joint use of medical subject headings (Mesh) and free terms from Medline.The types of vaccination,research subjects,immunogenicity and the efficacy of the clinical trials were sorted out and analyzed.According to the inclusive and exclusive criteria,total twenty five literatures were selected.There were more and more various types and the strategies of the vaccination with stronger immunogenicity,yet RV144 was the only clinical trial with significant efficacy.Based on the previous worldwide researches' findings,while many types of HIV preventive vaccines have been developed,there is still the need to explore potential candidates for HIV has various mutation,and there are confusions about protective immune response to HIV and pre-immune response to viral vectors effecting the vaccine efficacy.According to the previous experiences,it is necessary to target the local epidemic HIV genotypes,to apply the regimens of multiple vaccines and to investigate the HIV incidence rate,follow-up rate and willing-to-participate rate of high-risk populations in advance if large clinical trials of HIV preventive vaccines will be conducted in China.

Abstract:COVID-19 pandemic has severely impacted the public health and social economy worldwide. A safe, effective, and affordable vaccine against SARS-CoV-2 infections/diseases is urgently needed. We have been developing a recombinant vaccine based on a prefusion-stabilized spike trimer of SARS-CoV-2 and formulated with aluminium hydroxide and CpG 7909. The spike protein was expressed in Chinese hamster ovary (CHO) cells, purified, and prepared as a stable formulation with the dual adjuvant. Immunogenicity studies showed that candidate vaccines elicited robust neutralizing antibody responses and substantial CD4+ T cell responses in both mice and non-human primates. And vaccine-induced neutralizing antibodies persisted at high level for at least 6 months. Challenge studies demonstrated that candidate vaccine reduced the viral loads and inflammation in the lungs of SARS-CoV-2 infected golden Syrian hamsters significantly. In addition, the vaccine-induced antibodies showed cross-neutralization activity against B.1.1.7 and B.1.351 variants. These data suggest candidate vaccine is efficacious in preventing SARS-CoV-2 infections and associated pneumonia, thereby justifying ongoing phase I/II clinical studies in China (NCT04982068 and NCT04990544). Abstract:The acquired immunodeficiency syndrome (AIDS) has now become one of the most serious infectious disease in the world, the safe and effective AIDS vaccines would be the best tools to control the disease. Fowlpox virus has been studied recently as a potential vector for the delivery of heterologous vaccine antigen. In this study, a recombinant fowlpox virus coexpressing gp120 of Chinese prevalent HIV-1 strain and IL-2 was constructed and the cellular immune responses against HIV-1 gp120 in BALB/c mice were evaluated. Chinese vaccine strain 282E4 of fowlpox virus was used as the vector to construct the recombinant fowlpox virus (rFPV) coexpressing HIV-1 gp120 and IL-2 via homologous recombination, and the recombinant virus was identified by PCR and Western blotting. The specific DNA fragment was amplified by PCR from the genomes of rFPV. Western blotting analysis showed that HIV-1 gp120 and IL-2 was expressed not only in chicken embryo fibroblast (CEF) cells infected by rFPV, but also in mammalian cells infected by rFPV. After the recombinant fowlpox virus was inoculated into BALB/c mice, specific CTL activities in the immunized mice were detected in the spleen. The results demonstrated that rFPV had good immunogenicity and could induce BALB/c mice to produce specific cellular immunity. IL-2 played a role of immunoadjuvant and enhanced the cellular immune response. The study provides the basis for the preparation of a Chinese vaccine candidate against HIV-1.

Abstract:Human immunodeficiency virus type 1 (HIV-1) has been a global epidemic since 1983; yet, the virology and immunology related to HIV-1 remain elusive. Furthermore, as there is still no effective chemoprophylaxis or vaccine to treat patients with HIV-1, most research focuses on strategies to prevent HIV-1 infection, such as with antiviral drugs, novel therapeutics, or improved diagnostic kits. The HIV-1 Gag precursor protein (p55)-comprising the matrix (MA/p17), capsid (CA/p24), and nucleocapsid (NC/p7) protein domains-is the main structural HIV-1 protein, and is uniquely responsible for virion assembly within the virus life cycle. Recently, the immature and mature capsid structures were solved; however, the precursor protein structure is still unknown. Here, we expressed two subtypes of HIV-1 MA-CA stretch of the Gag protein, referred to as p41, in a bacterial expression system. We characterized the purified p41 protein, and showed its superior antigenicity over that of p24, highlighting the potential influence of the p17 domain on p24 structure. We further showed that p41 has good immunogenicity to induce an antibody response in mice. These results will aid future investigations into the HIV-1 capsid precursor structure, and potentially contribute to improving the design of diagnostic kits.

Abstract:Remarkable progress has been achieved for prophylactic and therapeutic interventions against human immunodeficiency virus type I (HIV-1) through antiretroviral therapy. However, vaccine development has remained challenging. Recent discoveries in broadly neutralizing monoclonal antibodies (bNAbs) has led to the development of multiple novel vaccine approaches for inducing bNAbs-like antibody response. Structural and dynamic studies revealed several vulnerable sites and states of the HIV-1 envelop glycoprotein (Env) during infection. Our review aims to highlight these discoveries and rejuvenate our endeavor in HIV-1 vaccine design and development.

Abstract:Since 1985, we have tested several immunological approaches to suppressing HIV replication in HIV-infected patients and to prevent HIV acquisition in uninfected people. Here, after briefly reviewing our studies on immunosuppressive treatments and therapeutic dendritic cell-based therapies, we examine in more detail our work on the tolerogenic vaccines we developed against AIDS in Chinese macaques. The vaccine consisted of inactivated SIVmac239 particles adjuvanted with the Bacillus of Calmette and Guerin (BCG), Lactobacillus plantarum (LP), or Lactobacillus rhamnosus (LR). Without adjuvant, the vaccine administered by the intragastric route induced the usual simian immunodeficiency virus (SIV)-specific humoral immune responses but no post-challenge protection. In contrast, out of 24 macaques that were immunized with the adjuvanted vaccine and challenged intrarectally with SIVmac239 or SIVB670, 23 were sterilely protected for up to 5 years, while all control macaques were infected. On the other hand, all macaques of Indian origin that were immunized with the same adjuvanted vaccine were not protected. We then discovered that vaccinated Chinese macaques developed a previously unrecognized class of non-cytolytic MHC-Ib/E-restricted CD8+ T cells (or CD8+ T-Regs) that suppressed the activation of SIV RNA-infected CD4+ T cells and thereby inhibited the (activation-dependent) reverse transcription of the virus and prevented the establishment of SIV infection. Finally, we found a similar population of HLA-E-restricted CD8+ T-Regs in human elite controllers (a small group of HIV-infected patients whose viral replication is naturally inhibited). Ex vivo, their CD8+ T-Regs suppressed viral replication in the same manner as those of vaccinated Chinese macaques. It is noteworthy that all of these elite controllers had a homo- or heterozygous HLA-Bw4-80I genotype. Taking into account the longevity and the high percentage of vaccine-protected Chinese macaques together with the concomitant identification of a robust ex vivo correlate of protection and the discovery of similar CD8+ T-Regs in human elite controllers, preventive and therapeutic HIV vaccines should be envisaged in humans.
