Abstract:Magnesium alloy has similar mechanical properties with natural bone, but its high susceptibility to corrosion has limited its application in orthopedics. In this study, a calcium phosphate coating is formed on magnesium alloy (AZ31) to control its degradation rate and enhance its bioactivity and bone inductivity. Samples of AZ31 plate were placed in the supersaturated calcification solution prepared with Ca(NO3)2, NaH2PO4 and NaHCO3, then the calcium phosphate coating formed. Through adjusting the immersion time, the thickness of uniform coatings can be changed from 10 to 20 μm. The composition, phase structure and morphology of the coatings were investigated. Bonding strength of the coatings and substrate was 2–4 MPa in this study. The coatings significantly decrease degradation rate of the original Mg alloy, indicating that the Mg alloy with calcium phosphate coating is a promising degradable bone material.
Preparing Ca-P Coating on Biodegradable Magnesium Alloy by Hydrothermal Method: in Vitro Degradation Behavior

Abstract:Magnesium alloys are potentially attractive biodegradable materials. However, their rapid corrosion rate limits their biomedical application. To slow down the rate of biodegradation, a protective calcium-phosphate coating was formed on a magnesium alloy substrate by a hydrothermal method. Scanning electron microscope results showed that the coating consisted of two layers with different crystalline characteristics. The loose outer layer showed a prism-like crystal structure, while the compact inner layer is a dense ultra-fine regular di-pyramid-like structure with an average grain dimension of ∼200 nm. The compositions of the inner layer and outer layer were calcium-deficient hydroxyapatite (Ca-def HA) and dicalcium phosphate (DCPa), respectively. The coating adhered well to the substrate with a thickness of about 15 μm. Immersion in Hank’s solution indicated that the coating could significantly improve the degradation properties of magnesium alloy. The pH of the solution containing the coated samples increased much more slowly than the untreated control. After 8 d immersion, the uncoated sample had corroded seriously while the coated sample was much less corroded. The Ca/P atom ratio in both the layers of the coating increased and the coating was still protecting the substrate. The two layers of the coating corroded differently because of differences in solubility. The outer layer was more severely attacked and many holes were formed on the surface, the inner layer suffered less attack. In addition, a growth of precipitate on the inner layer was observed, indicating that surface bioactivity was improved by the coating. Thus, magnesium alloys coated with a Ca-P coating prepared by a hydrothermal method are promising candidate biodegradable biomaterials, and further investigation of in vivo degradation behavior is suggested.

Micro-arc oxidation-assisted sol-gel preparation of calcium metaphosphate coatings on magnesium alloys for bone repair

Abstract:Calcium phosphate coating is an attractive surface modification strategy for magnesium alloys, since it can increase their corrosion resistance and endow them with osteogenic function simultaneously. Herein, a calcium metaphosphate (CMP) coating was fabricated on magnesium alloy by using sol-gel approach assisted with micro-arc oxidation pre-treatment. Scanning electron microscopy showed that the micro-pores and cracks in micro-arc oxidation inner layer generated during the pre-treatment process were sealed by the grainy sol-gel outer layer. Energy dispersive spectrometry and X-ray diffraction results demonstrated the identity of the coating as CMP. The cross-cut test showed that the adhesion of CMP coating was strong. Applying bare magnesium alloy substrate as a control, the CMP coating surface was rougher and more hydrophilic. The potentiodynamic polarization test demonstrated that the corrosion resistance was significantly improved by using CMP coating. Hydrogen evolution in immersion test further confirmed that the degradation rate was decelerated within 14 days. Moreover, CMP coating facilitated the adhesion speed, spreading area, and focal adhesion formation of bone marrow stem cells. The number of cells in the active proliferating state and proliferated cells present on the CMP coating also increased. Additionally, CMP coating upregulated alkaline phosphatase activity and osteogenic gene expression in cells. In summary, the micro-arc oxidation assisted sol-gel CMP coatings increased the corrosion resistance and promoted the interfacial cell behavior for magnesium alloy implants, which might inform the further development of surface modifications on magnesium alloys for bone related applications.
In Vivobiocompatibility and Degradation Behavior of Mg Alloy Coated by Calcium Phosphate in a Rabbit Model

Abstract:This study is conducted to investigate the biocompatibility and biodegradation behavior of calcium phosphate-coated Mg alloy in vivo. Calcium phosphate (Ca–P) was coated on the Mg alloy (AZ31) by a chemical process. Samples of Ca–P coated rods, the naked alloy rods, and degradable polymer as controls were implanted into the thighbone of rabbits to investigate the bone response at the early stage. The reduction in implant volume was determined by micro-computed tomography and three-dimensional reconstruction of the remaining Mg alloy segmented from the bone matrix. It was observed that the biodegradation rate of naked Mg implant is faster than that of the coated ones. The bone–implant interface was characterized in sections by scanning electron microscopy with energy-dispersive spectroscopy. Biodegradation or reaction layer was formed on the surface of Mg alloy implants and direct contact with the surrounding bone. The layer was mainly composed of Ca, P, O, and Mg. After 8 weeks of post-operation, paraffin sections were generated for histomorphologic analysis; 100% implants were fixed and no inflammation was observed. Histological analysis showed that new bone tissue is formed around the Mg implants, and no fibrous capsule was found. Blood examination showed that the biodegradation of the Mg implant caused little change to blood composition. Ca–P coating on Mg alloy substrate might be an effective method to reduce the biodegradation rate of Mg alloy in vivo and improve the surface bioactivity of Mg alloy implants.

Abstract:Recently, magnesium (Mg) alloys have been attracted as biodegradable implants. However, high degradation rate in human body limits its application. Modifying material surfaces with a coating layer is an effective means to control degradation, and biocompatibility. In this study, thin diamond-like carbon (DLC) films were synthesized on different magnesium alloy substrates by radio frequency plasma assisted chemical vapor deposition (RFPACVD). The morphology, composition and structure of the coating layers were investigated with scanning electron microscopy (SEM) with an energy dispersive spectroscopy (EDS), micro-raman spectra (RAMAN), and Atomic force microscopy (AFM). Static contact angle measurement and nanoindentation test were also used to analyze surface properties. The degradation behavior of naked and coated Mg alloy were evaluated in stimulated body fluid (SBF), and DLC layers on different Mg alloy substrates significantly decreased the degradation rate of substrate, which is useful for the biological applications especially in bone tissue and cardiovascular stent.

Abstract:Magnesium (Mg) and its alloys have exhibited great potential for orthopedic applications; however, their poor corrosion resistance and potential cytotoxicity have hindered their further clinical applications. In this study, we prepared a calcium phosphate (Ca-P) coating with a micro-nanofibrous porous structure on the Mg alloy surface by a chemical conversion method. The morphology, composition, and corrosion performance of the coatings were investigated by scanning electron microscope (SEM), energy-dispersive spectrometer (EDS), X-ray diffraction (XRD), immersion tests, and electrochemical measurements. The effects of the preparation temperature of the Ca-P coatings were analyzed, and the results confirmed that the coating obtained at 60 °C had the densest structure and the best corrosion resistance. In addition, a systematic investigation into cell viability, ALP activity, and cell morphology confirmed that the Ca-P coating had excellent biocompatibility, which could effectively promote the proliferation, differentiation, and adhesion of osteoblasts. Hence, the Ca-P coating demonstrates great potential in the field of biodegradable Mg-based orthopedic implant materials.
Surface Modification by Natural Biopolymer Coatings on Magnesium Alloys for Biomedical Applications

Abstract:Natural biopolymers possess good biocompatibility and exhibit excellent bioactivity due to the components of the extracellular cell matrix, which are abundant in the biomass and are easily accessible from a range of sources, such as forest products, grasses, tunicates, crustacean, and stalks. In this chapter, magnesium alloys modified with a natural biopolymer coating to improve their biocompatibility and degradation properties are introduced. Some natural biopolymers, such as chitosan, phytic acid, collagen, and stearic acid have been studied and coated on the surface of magnesium alloy for biomedical applications. Enhanced corrosion resistance, improved biocompatibility, and a reduced hemolysis ratio of WE43 was achieved after modification with phytic acid solution with different pH values of the conversion coating, especially when the pH value of the modified solution was 5. Various chitosan dip-coated Mg-1Ca alloys also displayed greater corrosion resistance in simulated body fluid. Calcium phosphate/chitosan composite films coated on the surface of micro-arc oxidized (MAO)-AZ91D alloy through electrophoretic deposition or on the surface of AZ31 alloy by aerosol deposition were adopted to enhance the bond strength between the magnesium substrate and hydroxyapatite. Stearic acid coating on hydrothermal-treated Mg samples enhanced the corrosion resistance of Mg. A drug release system on MAO-WE42 was established by a cross-linked gelatin/PLGA particle loaded with paclitaxel. Finally, a comparatively smooth surface of magnesium coated with albumin attached via linker groups was employed for better corrosion property, taking advantage of the presence of an Mg(OH)2-terminated MgO layer via silane coupling chemistry. Although the data and literature are limited in this area, previous works have validated the effectiveness and function of natural polymers on the corrosion resistance and biocompatibility of magnesium alloys, and much more research is expected in the future.
A ceria/calcium-phosphate functional composite coating on magnesium alloy for enhanced adhesion strength, corrosion resistance, and biocompatibility

Abstract:Magnesium (Mg) and its alloys have been regarded as potential orthopedic implant materials due to their suitable properties but face challenges due to rapid biocorrosion and insufficient bio-functionality, necessitating surface treatment to mitigate dysfunction. In this work, a two-step electrodeposition was employed to construct a ceria/calcium-phosphate (Ce/Ca-P) composite coating on Mg alloys. This construction approach endows multi-functions of the coating. The ceria interlayer ameliorates adhesion strength. The pin-like structure on the outer layer of the Ce/Ca-P coating enhances the antibacterial capabilities of the coating in the early stages of the implantation. The calcium-phosphate outer layer and the ceria interlayer induce the deposition of calcium and phosphate ions promoting bone healing in the mid-term. The introduction of the ceria interlayer significantly reduces the corrosion rate of the Mg alloy by 86.7% improving the corrosion resistance of the alloy, which is of great significance to maintain the service reliability of the device during the long-term service period. The results indicate that the Ce/Ca-P coating exhibits good hemocompatibility and cytocompatibility. This type of developed coating with anti-corrosive and bio-functionalized capability helpfully advances the application of Mg-based bone implants.

Abstract:As potential degradable biomaterials, magnesium (Mg) alloys have development prospects in the field of orthopedic load-bearing, whereas the clinical application has encountered a bottleneck due to a series of problems caused by its rapid corrosion. In this study, strontium-substituted calcium phosphate (CaP) coatings with different structures were prepared on the surface of the Mg matrix by a simple one-step electrodeposition method at different temperatures, which enhanced the poor corrosion resistance of the Mg matrix. The coated sample prepared at 65 °C reduced the corrosion current density by 3 orders of magnitude and increased the impedance by nearly 2 orders of magnitude compared with bare Mg alloy, thanks to its dense fibrous structure similar to that of natural bones. Although the coating composition varies with different preparation temperatures, CaP, as an inorganic component similar to natural bone, has good cytocompatibility. Doping the right amount of strontium, which is a trace element in human bones, is beneficial to stimulate osteoblast differentiation, inhibit the activity of osteoclasts, and induce the formation of bone tissues. This provides a new option for modifying the Mg alloy with CaP coatings as a base.

Abstract:Magnesium alloy is a potential biodegradable material and is widely used in medicine. However, its rapid degradation causes many negative effects and limits its application, so it is essential to improve corrosion resistance. In this study, a chitosan/polylactic acid composite coating was designed on the AZ31 alloy for medical applications. The composite coating sealed the pores of the chitosan layer to provide strong protection for the AZ31 alloy, with a reduced corrosion current density by 4 orders of magnitude. The excellent biocompatibility of the composite coating was proved by an increased cell viability test and morphology of cells adhered to the chitosan/polylactic acid coated sample. The composite coating provides a feasible method to enhance the corrosion resistance and biocompatibility of magnesium alloys.

Abstract:Calcium stearate based super-hydrophobic coating was deposited on plasma electrolytic oxidation (PEO) pre-treated magnesium substrate. The pre-treated magnesium and super-hydrophobic coating covered sample were characterized by scanning electron microscopy, X-ray diffraction and electrochemical corrosion measurements. The cytocompatibility and degradation resistance of magnesium, pre-treated magnesium and super-hydrophobic coating were analysed in terms of cell adhesion and osteoblast differentiation. The results indicate that the calcium stearate top coating shows super-hydrophobicity and that the surface is composed of micro/nanostructure. The super-hydrophobic coating covered sample shows higher barrier properties compared with the PEO pre-treated magnesium and bare magnesium. Human osteoblast proliferation, but not differentiation is enhanced by the PEO coating. Contrary, the super-hydrophobic coating reduces proliferation, but enhances differentiation of osteoblast, observable by the formation of hydroxyapatite. The combination of corrosion protection and cell reaction indicates that this system could be interesting for biomedical applications.
In vitro and in vivo degradation, biocompatibility and bone repair performance of strontium doped montmorillonite coating on Mg-Ca alloy

Abstract:Abstract Poor bone growth remains a challenge for degradable bone implants. Montmorillonite and strontium were selected as the carrier and bone growth promoting elements to prepare strontium doped montmorillonite coating on magnesium-calcium alloy. The surface morphology and composition were characterized by SEM, EDS, XPS, FT-IR and XRD. The hydrogen evolution experiment and electrochemical test results showed that the magnesium calcium alloy coated with Sr-MMT coating possessed optimal corrosion resistance performance. Furthermore, in vitro studies on cell activity, ALP activity, and cell morphology confirmed that Sr-MMT coating had satisfactory biocompatibility, which can significantly avail the proliferation, differentiation, and adhesion of osteoblasts. Moreover, the results of the 90 days implantation experiment in rats indicated that, the preparation of Sr-MMT coating effectively advanced the biocompatibility and bone repair performance of magnesium calcium alloy. In addition, The Osteogenic ability of Sr-MMT coating may be due to the combined effect of the precipitation of Si4+ and Sr2+ in Sr-MMT coating and the dissolution of Mg2+ and Ca2+ during the degradation of Mg-Ca alloy. By using coating technology, this study provides a late-model strategy for biodegradable Mg alloys with good corrosion resistance, biocompatibility. This new material will bring more possibilities in bone repair.

Abstract:Magnesium alloy is becoming one of the main candidates for biodegradable orthopedic implants due to its excellent biocompatibility and mechanical properties matching with natural bone. However, its rapid degradation rate hinders its clinical application. In this study, the organic (polydopamine) and inorganic (calcium phosphates) composite coating was prepared by chemical conversion and one step impregnation method to improve corrosion resistance and osteogenic differentiation for orthopedic implant materials . The formation mechanism of the coating was discussed by phase analysis and surface morphology observation. The results of anti-corrosion performance showed that the Ca-Mg-P/PDA coating greatly reduced the hydrogen evolution amount compared with WE43 alloy substrate and Ca-Mg-P coating. The biocompatibility and osteogenic differentiation ability of the coated alloys was studied, the alloy with Ca-Mg-P/PDA composite coating exhibited the best osteogenic differentiation ability in both short and long term of osteogenic differentiation.

Preparation and Degradation Behavior of Composite Bio-Coating on ZK60 Magnesium Alloy Using Combined Micro-Arc Oxidation and Electrophoresis Deposition

Abstract:In this work, ZK60 magnesium alloy was employed as the substrate substratum for producing micro-arc oxidation (MAO) coating which contains Ca and P. Electrophoretic deposition (ED) process was conducted on the micro-arc oxidized sample to prepare a hydroxyapatite (HA) layer on the original coating. X-ray diffraction (XRD), scanning electron microscopy (SEM), energy dispersive spectrometer (EDS) and Fourier transform infrared spectroscopy (FT-IR) were used to analyze the phase constituents and microstructures of both MAO and MAO-ED coatings. Corrosion resistance and degradation behavior of the coatings in SBF were investigated by electrochemical tests and simulated body fluid (SBF) immersion tests. The results indicate that a dense HA layer about 5 μm in thickness had been successfully prepared on the MAO coating. After going through the ED process, the porosity of the MAO-ED coating had decreased from 5.63 to 0.81%. The Ca/P ratio of the MAO-ED coating had become 1.60, which indicates a potential biocompatibility of the material. Therefore, the MAO-ED coating can induce the deposition of bioactive products from SBF. During an immersion test of SBF for 10 days, the weight gain of MAO-ED sample continued to increase, showing that the deposition rate of the induced products is always higher than that of corrosion products before experiencing the 10 days of immersion. The deposition of induced products protects the substrate from being corroded so effectively that it ensures the good mechanical properties and biocompatibility during the initial stage of implantation. According to the changes in sample morphologies and the electrochemical measurements in SBF, a relevant degradation model has been suggested and the underlying mechanism for degradation behavior is discussed.

Abstract:A strategy of suppressing the fast degradation behaviour of Mg-based biomaterials by the introduction of one of Mg degradation products Mg(OH)2 was proposed according to the following degradation mechanism, Mg + 2H2O ⇋ Mg(OH)2 + H2↑. Specifically, Mg(OH)2 submicron particles were mixed into poly (L-lactic acid) (PLLA) to synthesize a composite coating onto hydrofluoric acid-pretreated Mg-Nd-Zn-Zr alloy. The in vitro degradation investigations showed that the addition of Mg(OH)2 particles not only slowed down the corrosion of Mg matrix, but also retarded the formation of gas pockets underneath the polymer coating. Correspondingly, cytocompatibility results exhibited significant improvement of proliferation of endothelial cells, and further insights was gained into the mechanisms how the introduction of Mg(OH)2 particles into PLLA coating affected the magnesium alloy degradation and cytocompatibility. The present study provided a promising surface modification strategy to tailor the degradation behaviour of Mg-based biomaterials.

Abstract:Bio-ceramic coatings were synthesized on ZK60 magnesium alloys by micro-arc oxidation (MAO). The degradation behavior of the ZK60 alloys with and without MAO coating in the simulated body fluid (SBF) was studied. The samples were characterized by means of scanning electron microscopy (SEM), laser scanning confocal microscopy (CLSM), and X-ray diffraction (XRD). Electrochemical impedance spectroscopy (EIS) was used to study the degradation behavior. The results showed that the porous MAO coating mainly consisted of MgO, Mg2SiO4, Mg-3(PO4)(2), and CaCO3. The pH values of both coated and uncoated samples increased over time. However, the pH values of the SBF for coated samples always maintained a lower level compared with those for the uncoated samples. Thereby, the coated samples showed a much lower degradation rate. After immersion in SBF for 5 days, corrosion product containing Ca and P was found on both samples, while the deposition was more active on the coated samples. The degradation models for the uncoated and coated samples in the SBF are also proposed and discussed.
Polydopamine mediated assembly of hydroxyapatite nanoparticles and bone morphogenetic protein-2 on magnesium alloys for enhanced corrosion resistance and bone regeneration

Abstract:Magnesium alloys have the great potential to be used as orthopedic implants due to their biodegradability and mechanical resemblance to human cortical bone. However, the rapid degradation in physiological environment with the evolution of hydrogen gas release hinders their clinical applications. In this study, we developed a novel functional and biocompatible coating strategy through polydopamine mediated assembly of hydroxyapatite nanoparticles and growth factor, bone morphogenetic protein-2 (BMP-2), onto the surface of AZ31 Mg alloys. Such functional coating has strong bonding with the substrate and can increase surface hydrophilicity of magnesium alloys. In vitro electrochemical corrosion and hydrogen evolution tests demonstrate that the coating can significantly enhance the corrosion resistance and therefore slow down the degradation of AZ31 Mg alloys. In vitro cell culture reveals that immobilization of HA nanoparticles and BMP-2 can obviously promote cell adhesion and proliferation. Furthermore, in vivo implantation tests indicate that with the synergistic effects of HA nanoparticles and BMP-2, the coating does not cause obvious inflammatory response and can significantly reduce the biodegradation rate of the magnesium alloys and induce the new bone formation adjacent to the implants. © 2017 Wiley Periodicals, Inc. Abstract:Magnesium (Mg) based alloys are good candidates for developing degradable metallic implants. Mg can easily degrade in a physiological environment without producing any toxic by-products. However, the degradation rate needs to be controlled to make it compatible with the tissue growth rate. Achieving enhanced degradation resistance and biological performance in the physiological environment without sacrificing its mechanical and physical properties has been a great challenge. In this work, a combination of surface pre-treatment and polymer nanocomposite coating is attempted for reducing degradation rate and improving biomineralization. The alloy surface was pickled in phosphoric acid and then coated with polydioxanone/hydroxyapatite nanocomposite. The samples were characterized using scanning electron microscopy (SEM) with energy dispersive spectroscopy (EDS) and x-ray diffraction (XRD). The corrosion resistance was studied using potentiodynamic polarization (PDP) test in 8.035 g/l NaCl solution. An immersion test in simulated body fluid (SBF) was also carried out to evaluate the biodegradation and biomineralization on the sample surfaces. Phosphoric acid pickling increased the corrosion resistance and adhesion between the coating and substrate. Polymer nanocomposite coating resulted in further control of degradation rate and enhanced biomineralization. Thus, pre-treated Mg-Ca alloys coated with polydioxanone/nHA composites can be considered as promising materials for developing degradable metallic implants.

Functionalized Polymeric Membrane with Enhanced Mechanical and Biological Properties to Control the Degradation of Magnesium Alloy

Abstract:To achieve enhanced biological response and controlled degradation of magnesium alloy, a modified biodegradable polymer coating called polycaprolactone (PCL) is fabricated by a thermal approach in which the heat treatment neither alters the chemical composition of the PCL membrane nor the rate of magnesium ion release, pH value, or weight loss, compared with the untreated sample. The changes in the crystallinity, hydrophilicity, and oxygen content of heat-treated PCL coating not only improve the mechanical adhesion strength between the coating and magnesium substrate but also enhance the biological properties. Moreover, the thermally modified sample can lead to higher spreading and elongation of osteoblasts, due to the enhanced hydrophilicity and CO to CO functional group ratio. In the analyses of microcomputed tomography from one to four weeks postoperation, the total volume of new bone formation on the heat-treated sample is 10%-35% and 70%-90% higher than that of the untreated and uncoated controls, respectively. Surprisingly, the indentation modulus of the newly formed bone adjacent to the heat-treated sample is ≈20% higher than that of both controls. These promising results reveal the clinical potential of the modified PCL coating on magnesium alloy in orthopedic applications.
