Abstract:The present Study was undertaken to determinate the cytotoxicity exhibited via multi-walled carbon nanotubes(MWCNTs) in human A549 lung epithelial cells as a model system. Cell mortality was measured by methyl thiazolyl tetrazolium (MTT) assays. The morphological changes of A549 cells were examined by Hoechst 33342 staining. The level of reactive oxygen species(ROS) was detected by means of flow cytometry analysis with a redox-sensitive fluorescent probe 2',7'-dichlorfluorescein-diacetate (DCFH-DA). The mitochondria membrane potential was observed by a confocal laser-scanning microscope with JC-1 fluorescence. The heme oxygenase-1 (HO-1) protein expression was analyzed by immunofluorescence and Western blotting. The result reveal that treatment of A549 cells with MWCNTs lead to loss of cell viability, ROS production, glutathione depletion and decline in mitochondrial membrane potential. MWCNTs were potent toward inducing cellular heme oxygenase-1 (HO-1) expression I a sensitive biomarker for oxidative stress. Our observations suggest that MWCNTs exert toxicity by increasing oxidative stress, decreasing the mitochondrial membrane potential in cultured A549 cells.

Abstract:Carbon nanotubes currently attract intense attention for various biomedical applications due to their large surface areas, high electrical conductivity, and excellent strength. However, these attractive properties of nanomaterials are also the main factors for their potential toxicity. The present study was undertaken to determine the toxicity exhibited by multi-walled carbon nanotubes (MWCNT) in A549 lung epithelial cells. Treatment with increasing doses of MWCNT decreased the cell viability. The glutathione concentration decreased and intracellular reactive oxygen species (ROS) increased in a dose- and time-dependent manner, suggesting that the cytotoxicity on A549 cells was due to oxidative stress. Expression of heme oxygenase (HO)-1, a redox regulator and heat shock protein, increased with dose after MWCNT treatment. Pretreated with zinc protoporphyrin IX (ZnPP IX), a competitive HO inhibitor, underwent cell viability decrease and ROS accumulation increase to a greater extent than cells with MWCNT treatment alone, but these effects were reversed by co-treatment with bilirubin, a product of HO catalysis. Taken together, these findings suggest that MWCNT induce oxidative stress and HO-1 expression in A549 cells, and HO-1 induction may confer a cellular adaptive response against MWCNT-induced cytotoxicity.
Multi-Walled Carbon Nanotubes Induces Nuclear Factor-Kappa B Activation in A549 Cells

Abstract:The present study was undertaken to examine the effects of multi-walled carbon nanotubes (MWCNTs) on nuclear factor(NF)-kappa B activation in human A549 lung epithelial cells. Cultured A549 cells were stimulated with various concentrations of MWCNTs in the presence or absence of antioxidants. The intracellular generation of reactive oxygen species(ROS) was detected by means of flow cytometry analysis with a redox-sensitive fluorescent probe 2',7'-dichlorfluorescein-diacetate. NF-kappa B activity was measured by electrophoretic mobility shift assay(EMSA). The protein levels of p65 and I kappa B alpha were assessed by Western blot analysis. Translocation of p65 into the nucleus was examined by immunofluorescence confocal microscopy. The results showed increased BUS and NF-kappa B activity in response to treatment of A549 cells with MWCNTs. MWCNTs stimulated nuclear translocation of the p65 subunit of NF-kappa B, I kappa B alpha phosphorylation in A549 cells. Treatment of A549 cells with antioxidants prior to adding MWCNTs resulted in a significant reduction in ROS generation, NF-kappa B activity, p65 translocation and I kappa B alpha phosphorylation in A549 cells. These results indicate that MWCNTs are able to induce NF-kappa B activation in A549 cells mediated by oxidative stress.

Abstract:Multiwall carbon nanotubes (MWCNTs) have been widely applied in many fields due to the excellent physical and chemical properties. As the production and applications of nanotubes expand, public concern about their potential risks to human health has also raised. Cytotoxicity of MWCNTs was evaluated in this study using a cultured human epithelial cell line A549. Uptake of MWCNTs by cultured A549 cells was observed by TEM imaging. Dose-dependent decrease of cell viability showed the cytotoxicity of MWCNTs. Significant reactive oxygen species (ROS) generation and GSH depletion which reduced the cellular antioxidant level could be the major factor of cytotoxicity induced by MWCNTs. MWCNTs seemed to trigger the activation of cell autophagy with the intracellular ATG16L1 level increase as a defense mechanism.
Oxidative stress-mediated pro-inflammatory responses in lung epithelial cells exposed to multiwalled carbon nanotubes

Abstract:Reactive oxygen species (ROS) and activation of redox-sensitive transcription factors such as nuclear factor (NF)-KB have been implicated in pro-inflammatory responses induced by nanoparticles. However, there is still only little knowledge about the pro-inflammatory responses induced by multi-walled carbon nanotubes (MWCNTs). The objective of the present study was to compare the ability of MWCNTs and carbon black (CB) nanoparticles to induce secretion of pro-inflammatory cytokines and to explore possible mechanisms in A549 lung epithelial cells. Exposure of A549 cells to MWCNTs, but not to CB, resulted in a significant increase in interleukin-8 (IL-8) release and gene expression. MWCNTs stimulated NF-κB DNA binding activity, nuclear translocation of the p65 subunit of NF-κB, IκBα, phosphorylation, and ROS generation in A549 cells. MWCNTs-induced IL-8 gene expression and NF-κB activation were abrogated in the presence of ROS scavengers. These results suggest that MWCNTs are able to induce pro-inflammatory responses and NF-κB activation in A549 cells mediated by ROS generation. © 2010 IEEE.

Abstract:Objective To study the in vitro cytotoxicity and oxidative damage effects of Multi-wall Carbon Nanotube on A549 cells. Methods The A549 cells were treated with different concentrations of MWCNTs (2.5,10,25,100 μg/ml),slovent control and saline control for 24 h. MTT assays were applied for testing A549 cell activity in vitro,total protein (TP),lactate dehydrogenase(LDH),nitrogen monoxidum(NO),glutathione(GSH),superoxide dismutase (SOD),and malondialdehyde (MDA) in supernate and intra-cellular were determined to evaluate the in vitro cytotoxicity and oxidative damage effects of NWCNTs on A459 cells. The morphology changes of A549 cells were also observed. Results In comparison with control,the viabilities of A549 cells were decreased significantly after exposure to MWCNTs. The level of TP、LDH、NO and MDA in exposed groups were higher,while GSH and SOD level were lower than control with a significant dose-effect relationship. which accord to the morphology changs of A549 cells exposed to MWCNTs. Conclusion MWCNTs induces cytotoxity and oxidative stress on A549 cells.With the increases of the concentration of MWCNTs.

Abstract:Carbon nanotubes (CNTs) have potential applications in biosensors, tissue engineering, and biomedical devices because of their unique physico-chemical, electronic and mechanical properties. However, there is limited literature data available concerning the biological properties and toxicity of CNTs. This study aimed to assess the toxicity exhibited by multi-walled CNTs (MWCNTs) and to elucidate possible molecular mechanisms underlying the biological effects of MWCNTs in A549 cells. Exposing A549 cells to MWCNTs led to cell death, changes in cell size and complexity, reactive oxygen species (ROS) production, interleukin-8 (IL-8) gene expression and nuclear factor (NF)-κB activation. Treatment of A549 cells with antioxidants prior to adding MWCNTs decreased ROS production and abrogated expression of IL-8 mRNA. Pretreatment of A549 cells with NF-κB inhibitors suppressed MWCNTs-induced IL-8 mRNA expression. These results indicate that MWCNTs are able to induce expression of IL-8 in A549 cells, at least in part, mediated by oxidative stress and NF-κB activation.

Abstract:Oxidative stress is a major factor contributing to endothelial cell damage. Single-wall carbon nanotubes (SWCNTs) have oxidative properties; however, the oxidative effects of SWCNTs on endothelial cells are not fully understood. In the present study, we investigated the effects of oxidative stress induced by SWCNTs on rat aortic endothelial cells (RAECs). Various markers of cellular damage were assessed, such as biochemical and ES immunity indexes, and DNA and protein damage. Our findings suggest that RAEC endured oxidative damage following SWCNT exposure. Specifically, after SWCNTs exposure, non-enzymatic antioxidant glutathione was activated prior to superoxide dismutase activation in order to defend against oxidative stress. Additionally, it was found that as SWCNT concentration increased, so did the stress protein, heme oxygenase-1 (HO-1), expression levels. These changes may induce RAEC damage, and result in many serious diseases.

Abstract:The wide application of multi-walled carbon nanotubes (MWCNT) has raised serious concerns about their safety on human health and the environment. However, the potential harmful effects of MWCNT remain unclear and contradictory. To clarify the potentially toxic effects of MWCNT and to elucidate the associated underlying mechanisms, the effects of MWCNT on human lung adenocarcinoma A549 cells were examined at both the cellular and the protein level. Cytotoxicity and genotoxicity were examined, followed by a proteomic analysis (2-DE coupled with LC-MS/MS) of the cellular response to MWCNT. Our results demonstrate that MWCNT induces cytotoxicity in A549 cells only at relatively high concentrations and longer exposure time. Within a relatively low dosage range (30 µg/ml) and short time period (24 h), MWCNT treatment does not induce significant cytotoxicity, cell cycle changes, apoptosis, or DNA damage. However, at these low doses and times, MWCNT treatment causes significant changes in protein expression. A total of 106 proteins show altered expression at various time points and dosages, and of these, 52 proteins were further identified by MS. Identified proteins are involved in several cellular processes including proliferation, stress, and cellular skeleton organization. In particular, MWCNT treatment causes increases in actin expression. This increase has the potential to contribute to increased migration capacity and may be mediated by reactive oxygen species (ROS).
The Role of NADPH Oxidase in Multi-Walled Carbon Nanotubes-Induced Oxidative Stress and Cytotoxicity in Human Macrophages

Abstract:Recent studies suggest reactive oxygen species (ROS) induced in mammalian cells exposed to multi-walled carbon nanotubes (MWCNTs) could mediate the cytotoxicity. This study was conducted to determine the mechanisms responsible for MWCNTs-induced ROS production in human primary macrophages. Our results showed that superoxide levels were significantly increased in a time-dependent manner in blood monocyte-derived macrophages treated with 100 microg/ml MWCNTs for 12 h. Concomitantly, MWCNTs induced membrane translocation of the NADPH oxidase subunits p47phox and p67phox, a signature event for NADPH oxidase activation. Pre-incubation with apocynin, a selective inhibitor of NADPH oxidase, prevented both membrane translocation of p47phox and superoxide production. Treatment with MWCNTs also resulted in an increased cytotoxicity in human primary macrophages that was significantly attenuated by both apocynin and antioxidants. These findings demonstrate that MWCNTs activate NADPH oxidase in human macrophages, which may contribute to ROS generation in MWCNTs treated-macrophages.
In Vitro Evaluation of Cytotoxicity and Oxidative Stress Induced by Multiwalled Carbon Nanotubes in Murine RAW 264.7 Macrophages and Human A549 Lung Cells

Abstract:OBJECTIVE:To investigate in vitro cytotoxicity and oxidative stress response induced by multiwalled carbon nanotubes (MWCNTs).METHODS:Cultured macrophages (murine RAW264.7 cells) and alveolar epithelium cells type II (human A549 lung cells) were exposed to the blank control, DNA salt control, and the MWCNTs suspensions at 2.5, 10, 25, and 100 μg/mL for 24 h. Each treatment was evaluated by cell viability, cytotoxicity and oxidative stress.RESULTS:Overall, both cell lines had similar patterns in response to the cytotoxicity and oxidative stress of MWCNTs. DNA salt treatment showed no change compared to the blank control. In both cell lines, significant changes at the doses of 25 and 100 μg/mL treatments were found in cell viabilities, cytotoxicity, and oxidative stress indexes. The reactive oxygen species (ROS) generation was also found to be significantly higher at the dose of 10 μg/mL treatment, whereas no change was seen in most of the indexes. The ROS generation in both cell lines went up in minutes, reached the climax within an hour and faded down after several hours.CONCLUSION:Exposure to MWCNTs resulted in a dose-dependent cytotoxicity in cultured RAW264.7 cells and A549 cells, that was closely correlated to the increased oxidative stress.
Celastrol alleviates oxidative stress induced by multi-walled carbon nanotubes through the Keap1/Nrf2/HO-1 signaling pathway

Abstract:Multi-walled carbon nanotubes (MWCNTs) mainly induce oxidative stress through the overproduction of reactive oxygen species (ROS), which can lead to cytotoxicity. Celastrol, a plant-derived compound, can exert antioxidant effects by reducing ROS production. Our results indicated that exposure to MWCNTs decreased cell viability and increased ROS production. Nrf2 knockdown (kd) led to increased ROS production and enhanced MWCNT-induced cytotoxicity. Keap1-kd led to decreased ROS production and attenuated cytotoxicity. Treatment with celastrol significantly decreased ROS production and promoted Keap1 protein degradation through the lysosomal pathway, thereby enhancing the translocation of Nrf2 from the cytoplasm to the nucleus and increasing HO-1 expression. The in vivo results showed that celastrol could alleviate the inflammatory damage of lung tissues, increase the levels of the antioxidants, GSH and SOD, as well as promote the expression of the antioxidant protein, HO-1 in MWCNT-treated mice. Celastrol can alleviate MWCNT-induced oxidative stress through the Keap1/Nrf2/HO-1 signaling pathway.

Abstract:Growing evidence has indicated the potential adverse effects on cardiovascular system of some nanomaterials, including fullerenes. In this study, we have evaluated the biological effects of multiwall carbon nano-onions (MWCNOs) (average size of 31.2 nm, ζ potential of 1.6 mV) on human umbilical vein endothelial cells (HUVECs). It was found that MWCNOs exhibited a dose-dependent inhibitory effect on cell growth; EC50 was 44.12 μg/mL. Thus, three concentrations were chosen (0.2, 1, and 5 μg/mL) for further experiments. Flow cytometry analysis revealed that 1 and 5 μg/mL MWCNOs could induce apoptosis in HUVECs, the apoptotic rates were 12% and 24% at 24 h after exposure. On the other hand, MWCNOs did not affect the cell cycle distribution during 24 h period. Using γH2AX foci formation as an indicator for DNA damage, it was shown that 5 μg/mL MWCNOs can induce γH2AX foci formation in HUVECs at 6, 12, and 24 h after treatment, whereas 0.2 μg/mL MWCNOs induced γH2AX foci formation only at 6 h after treatment. In addition, all three concentrations of MWCNOs induced the generation of reactive oxygen species (ROS), and inhibition of ROS generation can partially decrease the γH2AX foci formation induced by MWCNOs. Taken together, these data first suggested that MWCNOs can induce DNA damage and apoptosis in HUVECs, and that ROS might be involved in this process.

Abstract:Intravenous Administration of Multiwalled Carbon Nanotubes Affects the Formation of Atherosclerosis in Sprague-Dawley Rats: Yu-Ying Xu, et al. Zhejiang University School of Medicine, China-Background: Carbon nanotubes (CNTs) have many potential applications, including as delivery systems for a variety of diagnostic or therapeutic agents. However, it has been suggested that exposure to carbon nano-materials may be a risk for the development of vascular diseases due to its impact on the vascular endothelium. Materials and Methods: Male Sprague-Dawley rats (180-200 g) were used to generate an atherosclerosis (AS) model, and the effect of intravenous administration of multi-walled carbon nanotubes (MWCNTs) on AS was studied. To further understand the underlying mechanisms, the effects of exposure of human umbilical vascular endothelial cells (HUVECs) to MWCNTs were examined. Results: Exposure to 200 mu g/kg MWCNTs aggravated AS in this model. In addition, exposure to 50, 100 and 200 mu g/kg MWCNTs increased the calcification of the aorta in the model. Short-term exposure also revealed that 200 mu g/kg MWCNTs injured the endothelium in the aorta. MWCNTs disrupted the endothelial tight junction and induced endothelial cell death. Conclusion: The results demonstrated that MWCNTs could induce structural and functional changes in the endothelium, probably through vascular endotheliocyte injury, which eventually affected the development of AS in SD rats. Oxidative stress and cell cycle arrest induced by short-term exposure to dustfall PM2.5 in A549 cells

Abstract:It was reported that in vitro short-term exposure to PM2.5 caused different lung diseases through inflammatory response, immune toxicity, oxidative stress, and genetic mutations. However, the complex molecular biological mechanism for its toxicity had not been fully elucidated. Therefore, the present study investigated the cytotoxicity, oxidative damage, mitochondria damage, apoptosis, and cell cycle arrest of NX and QH PM2.5 in A549 cells. Further, cell cycle arrest-related gene levels in PM2.5-induced A549 cells were also detected. Our results suggested that PM2.5 reduced the cell viability in A549 cells. Simultaneously, excessive ROS decreased MMP levels and damaged mitochondrial membrane integrity and induced mitochondrial oxidative damage through the oxygen-dependent killer route, resulting in mitochondrial damage and cell apoptosis. Besides, the results also showed that PM2.5 induced A549 cell cycle alteration in G2/M phase after co-culture for 24 h. G2/M phase arrest was induced by upregulation of p53 and p21 and downregulation of CDK1 mRNA expression. In addition, lncRNA Sox2ot might play an important role as the specific oncogenes and it participated in G2/M phase arrest by regulating the expression of EZH2.

Abstract:The use of nano-sized materials offers exciting new options in technical and medical applications. Single-walled carbon nanotubes are emerging as technologically important in different industries. However, adverse effects on cells have been reported and this may limit their use. We previously found that 200μg/mL of single-walled carbon nanotubes induce apoptosis in rat aorta endothelial cells. The current study aimed to determine the signaling pathway involved in this process. We found that reactive oxygen species generation was involved in activation of the mitochondria-dependent apoptotic pathway. The finding of apoptosis was supported by a number of morphological and biochemical hallmarks, including chromatin condensation, internucleosomal DNA fragmentation, and caspase-3 activation. In conclusion, our results demonstrate that single-walled carbon nanotubes induce apoptosis in rat aorta endothelial cells and that reactive oxygen species are involved in the mitochondrial pathway.

Abstract:Nuclear factor erythroid 2-related factor 2 (Nrf2) is a redox-sensitive transcription factor regulating the expression of a battery of cytoprotective genes. Using a cell-based ARE-reporter assay we discovered that the flavonoid luteolin is a potent Nrf2 inhibitor. Luteolin inhibited ARE-driven gene expression redox-independently. In non-small-cell lung cancer A549 cells, which possess constitutively active Nrf2, luteolin elicited a dramatic reduction in Nrf2 at both the mRNA and the protein levels, leading to decreased Nrf2 binding to AREs, down-regulation of ARE-driven genes, and depletion of reduced glutathione. After transcription was blocked with actinomycin D, 1μM luteolin decreased the Nrf2 mRNA level by 34% in 30min, indicating its role in accelerating Nrf2 mRNA turnover. At physiological concentrations, luteolin significantly sensitized A549 cells to the anticancer drugs oxaliplatin, bleomycin, and doxorubicin. However, knockdown of Nrf2 using siRNA essentially abolished the induced sensitivity by the flavonoid, implying the importance of inhibiting Nrf2 for its activity. Our study demonstrates that an Nrf2 inhibitor can enhance the responsiveness of cancer cells to chemotherapeutic drugs and indicates the potential application of luteolin as a natural sensitizer in chemotherapy.

Arenobufagin Promoted Oxidative Stress-Associated Mitochondrial Pathway Apoptosis in A549 Non-Small-Cell Lung Cancer Cell Line

Abstract:Arenobufagin (ARE) has demonstrated potent anticancer activity in various types of tumor, but the role and mechanism of ARE for lung cancer remain unclear. Oxidative stress exists under normal conditions and is an inevitable state in the body. A variety of noxious stimuli can break the equilibrium state of oxidative stress and promote apoptosis. Here, we used a CCK-8 assay to examine cell viability. We determined oxidative stress damage by measuring levels of intracellular ROS and levels of GSH, SOD, and MDA. Annexin V-FITC/PI double staining assay, as well as the Hoechst 33258 staining, was used to detect ARE-induced apoptosis in A549 cell. Evaluation of the expression level of the specified molecule was indicated by Western blot and qRT-PCR. Loss of function experiment was carried out using NAC pretreatment. The experimental results show that ARE significantly declines in the viability of A549 cells and increases the apoptosis rate of A549 cells. As reflected in cell morphology, the A549 cells showed features of shrinkage and had incompletely packed membranes; the same phenomenon is manifested in Hoechst 33258 staining. Following ARE treatment, the ROS level in A549 cells was rising in a concentration-dependent manner, and so were MDA and GSH levels, while the SOD level was decreasing. Moreover, we found that ARE can decrease mitochondrial membrane potential (MMP), and a cascade of apoptotic processes can be triggered by decreased MMP. Importantly, we found significant changes in protein expression levels and mRNA levels of apoptosis-related proteins. Furthermore, when we used NAC to restrain oxidative stress, the expression levels of apoptosis-related proteins have also changed accordingly. Our data demonstrate that apoptosis in the non-small-cell lung cancer (NSCLC) cell line A549 is caused by oxidative stress due to ARE. Our research also shows that ARE may have the potential to become a targeted therapeutic for the treatment of NSCLC in the future.

Abstract:After emitted from incomplete combustion of fossil fuels and biomass, ambient black carbon (BC) was then undergone photochemical oxidization processes in the air to form aged BC particles, also called oxidized BC (OBC). This study aimed to investigate the interactions between oxidative stress, autophagy and apoptosis induced by OBC in A549 cells and to explore associated molecular mechanisms. First, OBC could stimulate oxidative stress, autophagy and apoptosis dose-dependently, as evidenced by increased intercellular reactive oxygen species (ROS) levels, up-regulated autophagosome markers (light chain 3, LC3), and elevated apoptosis rate. Inhibitors of oxidative stress (N-acetylcysteine, NAC), autophagy (bafilomycin A1, Baf) and apoptosis (Z-DEVD-FMK) were used to investigate their interactions. NAC pretreatment could significantly reduce autophagy and apoptosis. Additionally, pretreatment with Baf or Z-DEVD-FMK could also significantly suppress the other two biological effects. Furthermore, OBC up regulated the expressions of DNA-dependent protein kinase catalytic subunit (DNA-PKcs), phosphorylated protein kinase B (Akt) and mammalian target of rapamycin (mTOR). The Akt inhibitor (MK-2206) significantly reduced both autophagy and apoptosis. Taken together, dual-direction regulation existed between each two of oxidative stress, autophagy, and apoptosis in A549 cells exposed to OBC. In addition, the autophagy process is modulated by the PI3K/Akt pathway regardless of mTOR activity.
