Abstract:Cell adhesion to extracellular matrices (ECM) is critical to physiological and pathological processes as well as biomedical and biotechnological applications. It has been known that a cell can adhere on an adhesive microisland only over a critical size. But no publication has concerned critical adhesion areas of cells on microislands with nanoarray decoration. Herein, we fabricated a series of micro-nanopatterns with different microisland sizes and arginine—glycine—aspartate (RGD) nanospacings on a nonfouling poly(ethylene glycol) background. Besides reproducing that nanospacing of RGD, a ligand of its receptor integrin (a membrane protein), significantly influences specific cell adhesion on bioactive nanoarrays, we confirmed that the concept of critical adhesion area originally suggested in studies of cells on micropatterns was justified also on the micro-nanopatterns, yet the latter exhibited more characteristic behaviors of cell adhesion. We found increased critical adhesion areas of human mesenchymal stem cells (hMSCs) on nanoarrayed microislands with increased RGD nanospacings. However, the numbers of nanodots with respect to the critical adhesion areas were not a constant. A unified interpretation was then put forward after combining nonspecific background adhesion and specific cell adhesion. We further carried out the asymptotic analysis of a series of micro-nanopatterned surfaces to obtain the effective RGD nanospacing on unpatterned free surfaces with densely grafted RGD, which could be estimated nonzero but has never been revealed previously without the assistance of the micro-nanopatterning techniques and the corresponding analysis.

Abstract:The adhesive area is important to modulate cell behaviors on a substrate. This paper aims to semi-quantitatively examine the existence of the characteristic areas of cell adhesion on the level of individual cells. We prepared a series of micropatterned surfaces with adhesive microislands of various sizes on an adhesion-resistant background, and cultured cells of MC3T3-E1 (osteoblast), BMSC (bone mesenchymal stem cell) or NIH3T3 (fibroblast) on those modeled surfaces. We have defined seven characteristic areas of an adhesive microisland and confirmed that they are meaningful to describe cell adhesion behaviors. Those parameters are (1) the critical adhesion area from apoptosis to survival denoted as A∗ or Ac1, (2) the critical area from adhesion of a single cell to adhesion of multiple cells (Ac2), (3) the basic area for one more cell to adhere (AΔ), (4) and (5) the characteristic areas of a microisland most probably occupied by one cell (Apeak(1)) and two cells (Apeak(2)), (6) and (7) the characteristic areas of a microisland occupied by one cell (AN(1)) or two cells (AN(2)) on average. Besides the introduction of those basic parameters, the present paper demonstrates how to determine them experimentally. We further discussed the relationship between those characteristic areas and the spreading area on a non-patterned adhesive surface.

Abstract:Cell adhesions are modulated by the interactions between cells and their surroundings, among which substrate stiffness plays an important role in mediating cellular behaviors and functions. Little is known, however, about the inherent mechanism of how nascent adhesion nucleation, as the precursor of focal adhesions, is regulated by substrate stiffness. This paper presents a microscopic model to imitate integrin clustering kinetics, where integrin diffusion, activation on elastic substrates, receptor–ligand binding and association dynamics are fully considered. Particularly, the contribution of substrate compliance to the activation energy is analyzed, leading to a description of mechanical energy barrier for stretching a substrate-bound integrin molecule from bent to extended conformations. A series of Monte Carlo simulations for integrin clustering dynamics are performed with varied substrate Young's moduli, which demonstrates that more integrins are clustered on stiffer substrates once they begin to assemble over a rigidity threshold, indicating the responsiveness of adhesion nucleation to substrate elasticity, which is in reasonable agreement with results reported previously. Also, these simulations show that the sensitivity of integrin clustering to substrate stiffness is mediated by chemical affinity between receptor–ligand pairs and that between integrins cross-linked by adapter proteins, as well as integrin density on cell membranes. The investigation offers a fascinating insight into the inherent mechanism of mechanosensing concerning integrin-mediated cell–matrix initial adhesions.

Abstract:Cell-matrix adhesion depends on the collective behaviours of clusters of receptor-ligand bonds called focal contacts between cell and extracellular matrix. While the behaviour of a single molecular bond is governed by statistical mechanics at the molecular scale, continuum mechanics should be valid at a larger scale. This paper presents an overview of a series of recent theoretical studies aimed at probing the basic mechanical principles of focal contacts in cell-matrix adhesion via stochastic-elastic models in which stochastic descriptions of molecular bonds and elastic descriptions of interfacial traction-separation are unified in a single modelling framework. The intention here is to illustrate these principles using simple analytical and numerical models. The aim of the discussions is to provide possible clues to the following questions: why does the size of focal adhesions (FAs) fall into a narrow range around the micrometre scale? How can cells sense and respond to substrates of varied stiffness via FAs? How do the magnitude and orientation of mechanical forces affect the binding dynamics of FAs? The effects of cluster size, cell-matrix elastic modulus, loading direction and cytoskeletal pretension on the lifetime of FA clusters have been investigated by theoretical arguments as well as Monte Carlo numerical simulations, with results showing that intermediate adhesion size, stiff substrate, cytoskeleton stiffening, low-angle pulling and moderate cytoskeletal pretension are factors that contribute to stable FAs. From a mechanistic point of view, these results provide possible explanations for a wide range of experimental observations and suggest multiple mechanisms by which cells can actively control adhesion and de-adhesion via cytoskeletal contractile machinery in response to mechanical properties of their surroundings.

Abstract:Focal adhesions are clusters of specific receptor-ligand bonds that link an animal cell to an extracellular matrix. A capability to control focal adhesions, for which a quantitative description of the collective behavior of multiple molecular bonds is a critical step, is essential for tissue and cellular engineering. While the behavior of single molecular bonds is governed by statistical mechanics at small scale, continuum mechanics should be valid at large scale. How can this transition be modeled and can this tell us something about the mechanics of cell adhesion? Here we develop a stochastic-elasticity model of a periodic array of adhesion clusters between two dissimilar elastic media subjected to an inclined tensile stress, in which stochastic descriptions of molecular bonds and elastic descriptions of interfacial traction are unified in a single modeling framework. A fundamental scaling law of interfacial traction distribution is established to govern the transition between uniform and cracklike singular distributions of the interfacial traction within molecular bonds. Guided by this scaling law, we perform Monte Carlo simulations to investigate the effects of cluster size, cell/matrix modulus and loading direction on lifetime and strength of the adhesion clusters. The results show that intermediate adhesion size, stiff substrate, cytoskeleton stiffening, and low-angle pulling are factors that contribute to the stability of focal adhesions. The predictions of our model provide feasible explanations for a wide range of experimental observations and suggest possible mechanisms by which cells can modulate adhesion and deadhesion via cytoskeletal contractile machinery.

Cell Motility and the Cytoskeleton

Abstract:The morphology and cytoskeletal structure of fibroblasts, endothelial cells, and neutrophils are documented for cells cultured on surfaces with stiffness ranging from 2 to 55,000 Pa that have been laminated with fibronectin or collagen as adhesive ligand. When grown in sparse culture with no cell-cell contacts, fibroblasts and endothelial cells show an abrupt change in spread area that occurs at a stiffness range around 3,000 Pa. No actin stress fibers are seen in fibroblasts on soft surfaces, and the appearance of stress fibers is abrupt and complete at a stiffness range coincident with that at which they spread. Upregulation of alpha5 integrin also occurs in the same stiffness range, but exogenous expression of alpha5 integrin is not sufficient to cause cell spreading on soft surfaces. Neutrophils, in contrast, show no dependence of either resting shape or ability to spread after activation when cultured on surfaces as soft as 2 Pa compared to glass. The shape and cytoskeletal differences evident in single cells on soft compared to hard substrates are eliminated when fibroblasts or endothelial cells make cell-cell contact. These results support the hypothesis that mechanical factors impact different cell types in fundamentally different ways, and can trigger specific changes similar to those stimulated by soluble ligands.

Abstract:Cell interactions with extracellular matrix, such as cell adhesion and spreading, are crucial for many biological functions and processes. Recent experimental progresses have demonstrated that substrate rigidities exert a remarkable influence on cell–substrate interfacial adhesion and spreading behaviors. The underlying biophysical mechanism, however, remains elusive. Based on the classical Bell-Dembo’s theory, this paper develops a mechano-chemical coupling model to physically describe cell adhesion and spreading mediated by substrate stiffness. By investigating the competitive nature between cell–substrate specific attraction and non-specific repulsion, the kinetic relation of receptor–ligand interplay is established, in which the influences of receptor–ligand separation, substrate elasticity and non-specific repulsion on cell adhesions are especially addressed. According to mechanical equilibrium conditions between cell membranes and underlying elastic substrates, an analytical expression is then deduced to relate the cell–substrate interfacial adhesion strength to the substrate rigidity. Moreover, by means of the conventional wetting theory, the dependence of steady-state cell spreading on substrate stiffness is also quantitatively studied. Comparisons with the existing experimental data show that the proposed model can be used to explore cell–substrate interactions regulated by substrate rigidities.
Characterization of Cell Response on Patterned Stiffness Substrate by AFAM

Abstract:The stiffness of extracellular matrix strongly influences cells behavior, but characterization of substrate stiffness patterns is a challenge. In our study, atomic force acoustic microscopy (AFAM) was used to study the cell responses on substrate stiffness patterns. The nano-stripe stiffness pattern was imprinted on SU-8 film. Our results show that AFAM acoustic images can detect both the cell surfaces and the stiffness pattern on the SU-8 substrate. We find that the cells significant deformation on the patterned stiffness substrate. Moreover, when using cytochalasin-D to interfere actin filaments, there is a significantly decrease in cell area for cells seeded on the patterned stiffness substrate. This study provides new information for the design and characterization of extracellular matrix interfaces, and contributes to the understanding of cell-substate interactions.
Optimal shapes and stresses of adherent cells on patterned substrates

Abstract:We investigate a continuum mechanical model for an adherent cell on two dimensional adhesive micropatterned substrates. The cell is modeled as an isotropic and homogeneous elastic material subject to uniform internal contractile stresses. The build-up of tension from cortical actin bundles at the cell periphery is incorporated by introducing an energy cost for bending the cell boundary, resulting in a resistance to changes in the local curvature. Integrin-based adhesions are modeled as harmonic springs that pin the cell to adhesive patches of a predefined geometry. Using Monte Carlo simulations and analytical techniques we investigate the competing effects of bulk contractility and cortical bending rigidity in regulating cell shapes on non-adherent regions. We show that the crossover from convex to concave cell edges is controlled by the interplay between contractile stresses and boundary bending rigidity. In particular, the cell boundary becomes concave beyond a critical value of the contractile stress that is proportional to the cortical bending rigidity. Furthermore, the intracellular stresses are found to be largely concentrated at the concave edge of the cell. The model can be used to generate a cell-shape phase diagram for each specific adhesion geometry.

Abstract:The ability to control cell patterning on artificial substrates with various physicochemical properties is of essence for important implications in cytology and biomedical fields. Despite extensive progress, the ability to control the cell-surface interaction is complicated by the complexity in the physiochemical features of bioactive surfaces. In particular, the manifestation of special wettability rendered by the combination of surface roughness and surface chemistry further enriches the cell-surface interaction. Herein we investigated the cell adhesion behaviors of Circulating Tumor Cells (CTCs) on topographically patterned but chemically homogeneous surfaces. Harnessing the distinctive cell adhesion on surfaces with different topography, we further explored the feasibility of controlled cell patterning using periodic lattices of alternative topographies. We envision that our method provides a designer's toolbox to manage the extracellular environment.

Abstract:The mechanically heterogeneous extracellular matrix (ECM) or tissues widely exist in biological systems and are capable of significantly regulating directional cell migration. However, prior to whole cell movement, how the cell senses these cues from mechanical heterogeneities of the ECM or substrate remains unclear at the molecular bond level. To address this issue, we theoretically investigate interface adhesion between a non-uniform stiffness substrate and a rigid plate via a series of receptor–ligand bonds subjected to a tensile loading by integrating substrate surface deformation described by continuum mechanics approach into the stochastic events of bond dissociation and association govern by Markov processes. Interestingly, it is found that, during stretching adhesion interface, due to the large collective contact forces near the stiff edge of the adhesion area, the crack first develops at this stiff edge and then grows to another relatively soft adhesion edge until the completed detachment achieved, which is distinct from the cracks growing from both two edges to center of adhesion area in the case of uniformly elastic solid–solid or solid–fluid interface. Moreover, the lifetime of the bond cluster, interface adhesion strength, and the effect of inter-bond distance are examined, respectively. The corresponding mechanism of dependence of the lifetime and adhesion strength on the non-uniform stiffness of the substrate and inter-bond distance is also analyzed. These findings provide a detailed mechanistic understanding of the adhesion interface responding to the mechanical heterogeneities of the substrate at the molecular bond level.

Abstract:Abstract Cell adhesion is a basic requirement for anchorage-dependent cells to survive on the matrix. It is the first step in a series of cell activities, such as cell diffusion, migration, proliferation, and differentiation. In vivo , cells are surrounded by extracellular matrix (ECM), whose physical and biochemical properties and micromorphology may affect and regulate the function and behavior of cells, causing cell reactions. Cell adhesion is also the basis of communication between cells and the external environment and plays an important role in tissue development. Therefore, the significance of studying cell adhesion in vitro has become increasingly prominent. For instance, in the field of tissue engineering and regenerative medicine, researchers have used artificial surfaces of different materials to simulate the properties of natural ECM, aiming to regulate the behavior of cell adhesion. Understanding the factors that affect cell behavior and how to control cell behavior, including cell adhesion, orientation, migration, and differentiation on artificial surfaces, is essential for materials and life sciences, such as advanced biomedical engineering and tissue engineering. This article reviews various factors affecting cell adhesion as well as the methods and materials often used in investigating cell adhesion.

Quantitative Analyses of Dynamic Features of Fibroblasts on Different Protein-Coated Compliant Substrates.

Abstract:Cell response to substrate rigidity, closely related to extracellular matrix protein composition, requires actomyosin-generated contractility. By introducing coefficients describing cell spreading and traction dynamics, and a revised high-resolution traction force microscopy, we analyzed the static and dynamic features of fibroblasts on fibronectin- or collagen- coated stiff or soft substrates. Large cell spreading area and branchlike morphology were more favorable on fibronectin than collagen. Cell spreading on fibronectin-coated substrates was more sensitive to rigidity compared with collagen. Low concentration fibronectin-coated substrate induced more dynamic lamellipodia movement than other conditions. Interestingly, the static average cell traction on high concentration fibronectin-coated stiff and soft substrates showed no difference. However, the lamellipodium traction dynamics was sensitive to rigidity on fibronectin. Particularly, lamellipodia on fibronectin-coated soft substrate performed much higher local traction dynamics compared with other groups. Together, dynamics of cell adhesion and traction are regulated by extracellular matrix protein composition, coupled with substrate rigidity.
Mechanics of Cell Mechanosensing on Patterned Substrate

Abstract:It has been recognized that cells are able to actively sense and respond to the mechanical signals through an orchestration of many subcellular processes, such as cytoskeleton remodeling, nucleus reorientation, and polarization. However, the underlying mechanisms that regulate these behaviors are largely elusive; in particular, the quantitative understanding of these mechanical responses is lacking. In this study, combining experimental measurement and theoretical modeling, we studied the effects of rigidity and pattern geometry of substrate on collective cell behaviors. We showed that the mechanical force took pivotal roles in regulating the alignment and polarization of cells and subcellular structures. The cell, cytoskeleton, and nucleus preferred to align and polarize along the direction of maximum principal stress in cell monolayer, and the driving force is the in-plane maximum shear stress. The higher the maximum shear stress, the more the cells and their subcellular structures preferred to align and polarize along the direction of maximum principal stress. In addition, we proved that in response to the change of in-plane shear stresses, the actin cytoskeleton is more sensitive than the nucleus. This work provides important insights into the mechanisms of cellular and subcellular responses to mechanical signals. And it also suggests that the mechanical force does matter in cell behaviors, and quantitative studies through mechanical modeling are indispensable in biomedical and tissue engineering applications.
Role of Stiffness versus Wettability in Regulating Cell Behaviors on Polymeric Surfaces

Abstract:Substrate wettability and stiffness, two factors impacting cell behaviors simultaneously, have been attracting much attention to elaborate which one dominates. In this study, hydrophilic poly(2-hydroxyethyl methacrylate) brushes were grafted onto the surfaces of poly(dimethylsiloxane) (PDMS) with elastic moduli of 3.66, 101.65 and 214.97 MPa and decreasing water contact angle from 120.4° to 38.5°. Cell behaviors of three cell lines including mBMSCs, ATDC-5, and C28/I2 were then investigated on the hydrophilic and hydrophobic PDMS with different stiffness, respectively. The proliferation of three cell lines was faster on the hydrophilic PDMS than the hydrophobic PDMS, but the stiffness of the hydrophilic or hydrophobic PDMS did not have a significant impact on cell proliferation. The increase of the stiffness enhanced cell migration, the cell spread and the gene expression proportion of extracellular matrix/intercellular adhesion molecules (integrin + FAK/NCAM + N-cadherin) for all three cell lines, but the increase of the wettability showed small enhancement in cell migration, cell spread and gene expression. Moreover, the cartilage-specific gene expression of SOX9 and COL2 downregulated for all three cell lines with the increasing stiffness. The interpretation of the effect of substrate wettability and stiffness on cell behaviors would function as very useful guideline to direct scaffold fabrication.
Modulating Particle Adhesion with Micro-Patterned Surfaces.

Abstract:We report the first experimental study on the modulation of adhesion force distribution by surface micro-patterns and its impact on particle attachment. The effect of substratum topography on particle adhesion was evaluated using well-defined microscopic surface patterns consisting of orthogonal arrays of cuboid pillars or pits with different sizes and spacing fabricated by the conventional photolithography and reactive ion etching (RIE). Adhesion of carboxyl modified poly(styrene-co-acrylic-acid) particles of 6 μm in diameter under favorable deposition conditions was found to be markedly lower on all the micro-patterned surfaces compared with that on the smooth control surface, and particle adhesion depended on the characteristic dimensions of the surface micro-structures relative to the particle size. Particle adhesion was minimal when the pillar cross-sectional dimension was below a critical value close to the diameter of the particle while the spacing between pillars was less important. Meanwhile, particles adhered displayed unique distribution on the micro-patterned surfaces. The majority of particles preferentially adhered on or close to the edge of the pillars (in the valley). Atomic force microscopy measurements using a colloidal probe revealed that the surface features strongly modulated the spatial and probability distribution of adhesion forces on the micro-patterned surfaces. Micro-sized pillars changed the adhesion force probability distribution from monomodal to bimodal, with significantly reduced maximum adhesion force. This was hypothesized to be responsible for the reduced total particle adhesion.

Abstract:There is compelling evidence that substrate stiffness affects cell adhesion as well as cytoskeleton organization and contractile activity. This work was designed to study the cytoskeletal contractile activity of cells plated on microposts of different stiffness using a numerical model simulating the intracellular tension of individual cells. We allowed cells to adhere onto micropost substrates of various rigidities and used experimental traction force data to infer cell contractility using a numerical model. The model discriminates between the influence of substrate stiffness on cell tension and shows that higher substrate stiffness leads to an increase in intracellular tension. The strength of this model is its ability to calculate the mechanical state of each cell in accordance to its individual cytoskeletal structure. Abstract:Many studies have demonstrated that the mechanical properties of the extracellular matrix can significantly influence the morphology, strength and lifetime of focal adhesions. However, how the morphology of the contact surface affects the pattern formation of the molecular bonds still remains largely unknown. Here, by simplifying the cell and extracellular matrix to two opposing elastic bodies and considering the lateral diffusion as well as the bonding/debonding of molecular bonds, we study the clustering behavior of receptor-ligand bonds between curved surfaces and the phase diagrams of cluster patterns. We reveal the important role of surface morphology and bond kinetics in regulating the patterns of bond clusters. We further investigate the segregation dynamics of the interfacial bonds under various loading speeds, and we show that the average interfacial stress is rate-dependent while the rupture stress is rate-independent. Finally, we demonstrate that programmable patterning of bond clusters can be achieved through the designed surface morphology.

Abstract:We present a minimal continuum model of strongly adhering cells as active contractile isotropic media and use the model to study the effect of the geometry of the adhesion patch in controlling the spatial distribution of traction and cellular stresses. Activity is introduced as a contractile, hence negative, spatially homogeneous contribution to the pressure. The model shows that patterning of adhesion regions can be used to control traction stress distribution and yields several results consistent with experimental observations. Specifically, the cell spread area is found to increase with substrate stiffness and an analytic expression for the dependence is obtained for circular cells. The correlation between the magnitude of traction stresses and cell boundary curvature is also demonstrated and analyzed.
