Abstract:Electrospun nanofibers could be used to mimic the nanofibrous structure of the extracellular matrix (ECM) in native tissue. In tissue engineering, the ECM could be used as tissue engineering scaffold to solve tissue engineering problems. In this paper, poly(L-lactid-co-ɛ-caprolactone) [P(LLA-CL)] nanofibers and collagen-chitosan complex nanofibers were fabricated by electrospinning. Results of the experiments showed that the mechanical properties of the collagen-chitosan complex nanofibers varied with the collagen content in the complex. It was also found that the biodegradability of P(LLA-CL) nanofibers was faster than its membrane and that smooth muscle cells (SMC) grow faster on collagen nanofibers than on P(LLA-CL) nanofibers.

Abstract:Converting common biomass materials to high-performance biomedical products could not only reduce the environmental pressure associated with the large-scale use of synthetic materials, but also increase the economic value. Chitosan as a very promising candidate has drawn considerable attention owing to its abundant sources and remarkable bioactivities. However, pure chitosan materials usually exhibit insufficient mechanical properties and excessive swelling ratio, which seriously affected their in vivo stability and integrity when applied as tissue engineering scaffolds. Thus, simultaneously improving the mechanical strength and biological compatibility of pure chitosan (CS) scaffolds becomes very important. Here, inspired by the fiber-reinforced construction of natural extracellular matrix and the porous structure of cancellous bone, we built silk microfibers/chitosan composite scaffolds via ice-templating technique. This biomimetic strategy achieved 500% of mechanical improvement to pure chitosan, and meanwhile still maintaining high porosity (> 87%). In addition, the increased roughness of chitosan pore walls by embedded silk microfibers significantly promoted cell adhesion and proliferation. More importantly, after subcutaneous implantation in mice for four weeks, the composite scaffold showed greater structural integrity, as well as better collagenation, angiogenesis, and osteogenesis abilities, suggesting its great potential in biomedicine.

Abstract:The collagen–chitosan complex nanofibers have been prepared for the first time by electrospinning. The mixed HFP/TFA (the volume ratio of 90/10) was found to be the appropriate solvent for electrospinning. The concentration of the spinning solution and the ratio of chitosan/collagen were varied and adjusted to get smooth nanofibers. It was found that the diameter of the spun fibers became thick with the concentration of the solution increasing and became fine with the ratio of the chitosan/collagen increasing. We have characterised the molecular interactions in collagen–chitosan complex by Fourier transform infrared spectroscopy. The spun fibers are designed to mimic the native extracellular matrix for tissue engineering and to develop functional biomaterials.

Abstract:Collagen and chitosan blends were fabricated into ultrafine fibers to mimic the native extracellular matrix (ECM). So far less mechanical property investigation of electrospun fibers has been reported because of the small dimensions of micro and nanostructures that pose a tremendous challenge for the experimental study of their mechanical properties. In this paper, the electrospun collagen–chitosan complex single fibers and fibrous membrane were collected and their mechanical properties were investigated with a nano tensile testing system and a universal materials tester, respectively. The mechanical properties were found to be dependent on fiber diameter and the ratio of collagen to chitosan in fibers. Fibers with a smaller diameter had higher strength but lower ductility due to the higher draw ratio that was applied during the electrospinning process. For the electrospun single fibers, the fibers demonstrated excellent tensile ductility at chitosan content of 10% and 20% and the highest tensile strength and Young's modulus at chitosan content from 40% to 60%. For the electrospun fibrous membrane, the ultimate tensile strength of the fibrous membrane decreased with the increase of chitosan content in fibers and the trend in the ultimate tensile elongation is similar to that of the single fiber.

Abstract:Electrospinning is a very effective method to fabricate nanofibers. This technique has been achieved more attention due to its cheap cost, simple device and obtaining nanofibers similar to extracellular matrix.Silk fibroin(SF) is a natural protein with good biodegradability and low immunogenicity, Therefore, it can be widely applied in tissue engineering. In this paper, we summarize the research of electrospun silk fibrion nanofibers in regenerative medicine. Furthermore, the future research work of SF nanofibers was suggested.

Abstract:Extracellular matrix (ECM) derived from decellularized tissues and organs has been used as a biological scaffold in a variety of pre-clinical and clinical applications. However, the lack of mechanical properties and shape controllability is a drawback. In contrast, synthetic polymers can be easily engineered with good mechanical performance, but they have limited biological functionality. In this study, we explored a new method to electrospun cartilage-derived extracellular matrix (cECM) and polycaprolactone (PCL) composite nanofibrous membranes. The cartilage is a compact tissue for which electrospinning is difficult. To overcome this problem, the cartilage was sliced into pieces, milled into powders, and digested into a looser structure. The cECM/PCL (mass ratio 50:50) hybrid nanofibers appeared to be smooth, thinner, and uniform with enhanced mechanical properties and wettability compared to the electrospun PCL. Meanwhile, the presence of cECM in the cECM/PCL nanofibrous membranes significantly promoted chondrocyte proliferation in vitro and facilitated cartilage regeneration in vivo. All these results indicate that the cECM/PCL nanofibrous membranes fabricated with good mechanical properties and biocompatibility may represent a promising scaffold for cartilage regeneration. Moreover, this work may offer a convenient and cost-effective method to synthesize ECM-based hybrid nanofibrous scaffolds for other tissue applications.
Novel shish-kebab structured nanofibrous decorating chitosan unidirectional scaffolds to mimic extracellular matrix for tissue engineering

Abstract:Electrospun nanofibrous scaffolds are renowned for their ability to mimic the microstructure of the extracellular matrix (ECM). However, they often fail to replicate the geometry of target tissues, and the biocompatibility of these scaffolds those made from synthetic polymers is always limited due to the lack of cell binding sites. To address these issues, we proposed an innovative approach that combined unidirectional freeze-drying and electrospinning. During this process, electrospun polycaprolactone (PCL) nanofibers were chopped into nanofibrils, which range in size up to several hundred micrometers, and were incorporated into the chitosan scaffolds via unidirectional freeze-drying. In these scaffolds, the chitosan phase was responsible for maintaining the structural integrity at the macroscale, while the embedded nanofibers enhanced the surface topography at the microscale. The resulting scaffolds exhibited a high porosity of 90% and an impressive water uptake capacity of 2500%. Furthermore, 3T3 fibroblast cells showed strong interactions with the scaffolds, characterized by high rates of cell proliferation and viability. The cells also displayed significant orientation along the direction of the pores, suggesting that the scaffolds effectively guided cellular growth.

Abstract:A nanofabrication method for the production of flexible core-shell structured silk elastin nanofibers is presented, based on an all-aqueous coaxial electrospinning process. In this process, silk fibroin (SF) and silk-elastin-like protein polymer (SELP), both in aqueous solution, with high and low viscosity, respectively, were used as the inner (core) and outer (shell) layers of the nanofibers. The electrospinnable SF core solution served as a spinning aid for the nonelectrospinnable SELP shell solution. Uniform nanofibers with average diameter from 301 ± 108 nm to 408 ± 150 nm were obtained through adjusting the processing parameters. The core-shell structures of the nanofibers were confirmed by fluorescence and electron microscopy. In order to modulate the mechanical properties and provide stability in water, the as-spun SF-SELP nanofiber mats were treated with methanol vapor to induce β-sheet physical crosslinks. FTIR confirmed the conversion of the secondary structure from a random coil to β-sheets after the methanol treatment. Tensile tests of SF-SELP core-shell structured nanofibers showed good flexibility with elongation at break of 5.20% ± 0.57%, compared with SF nanofibers with an elongation at break of 1.38% ± 0.22%. The SF-SELP core-shell structured nanofibers should provide useful options to explore in the field of biomaterials due to the improved flexibility of the fibrous mats and the presence of a dynamic SELP layer on the outer surface.

Abstract:The collagen-chitosan complex nanofibers have been prepared here by electrospinning. Intermolecular interactions in electrospun collagen-chitosan complex fibers have been studied by Fourier transform infrared spectroscopy (FT-IR), differential scanning calorimetry (DSC) and the mechanical measurements of single ultratine fiber. It was found that the -OH group, the -NH2 group and the amide 1, 11 and III characteristic absorption bands in FT-IR spectra of electrospun collagen and chitosan blends were shifted and modified with the difference of chitosan content in electrospun fibers. DSC measurements showed the existence of intermolecular interactions enthalpy between collagen and chitosan of electrospun fibers. The mechanical measurements of single nanofiber showed that intermolecular interactions varied with various chitosan content in electrospun fibers. The results have shown that intermoleculars interactions exist in electrospun collagen-chitosan complex fibers.These interactions make collagen and chitosan be miscible at the molecular level. Electrospinning of collagen and chitosan blends may give the possibility of producing new materials for potential biomedical applications. (C) 2007 Elsevier Ltd. All rights reserved.

Abstract:The collagen-chitosan nanofibers have been prepared by electrospinning from a mixed HFP/TFA(the volume ratio of 90/10) solution. The intermolecular interactions in collagen-chitosan nanofibers were characterized by Fourier transform infrared spectroscopy (FTIR). XRD patterns show that the collagen-chitosan nanofibers have become amorphous phases after electrospinning. Mechanical characterization indicates that the ratio of collagen to chitosan will affect the mechanical properties of electrospun collagen-chitosan fibers. Both the ultimate tensile elongation and the ultimate tensile strength of fibrous. membrane decrease with the increase of chitosan content. All these would be useful to a biomaterial design by electrospinning method.

Abstract:AIM: Electrospunning is a process that the charged polymer solution or melt jet efflux in the electrostatic field to prepare the polymer nanofibers. This study aimed to prepare electrospun chitosan/collagen composite nanofibers and evaluate their cell biocompatibility. METHODS: The experiment was carried out in the Laboratory of Biomaterials and Tissue Engineering, College of Chemistry, Chemical Engineering and Biotechnology at Donghua University between November 2006 and October 2007. ① Taking hexafluoroisopropanol/trifluoroacetic acid as solvent, the composite nanofibers were prepared by electrospunning according to the mass ratio of chitosan/collagen as 100:0, 80:20, 50:50, 20:80 and 0:100.②The composite nanofibers were inoculated with porcine iliac artery endothelial cells in vitro. The cell adhesion and proliferation as well as cell morphologies were evaluated by MTT method and HE stain, respectively. RESULTS: The porcine iliac artery endothelial cells adhered tightly on surface of electrospun chitosan/collagen composite nanofibers, exhibited plump and fusiform shape, as well as good growth. MTT results showed that nanofibers could obviously enhance the adhesion and proliferation of endothelial cells on the surface of materials, and the cells had the best spreading and attachment on the composite nanofibers with a ratio of 20:80, then 50:50. CONCLUSION: The electrospun chitosan/collagen composite nanofibers hold great potential for tissue engineering application due to their good cell biocompatibility.
Electrospun nanofiber-reinforced three-dimensional chitosan matrices: Architectural, mechanical and biological properties

Abstract:The poor intrinsic mechanical properties of chitosan hydrogels have greatly hindered their practical applications. Inspired by nature, we proposed a strategy to enhance the mechanical properties of chitosan hydrogels by construction of a nanofibrous and cellular architecture in the hydrogel without toxic chemical crosslinking. To this end, electrospun nanofibers including cellulose acetate, polyacrylonitrile, and SiO2 nanofibers were introduced into chitosan hydrogels by homogenous dispersion and lyophilization. With the addition of 30% cellulose acetate nanofibers, the cellular structure could be maintained even in water without crosslinking, and integration of 60% of the nanofibers could guarantee the free-standing structure of the chitosan hydrogel with a low solid content of 1%. Moreover, the SiO2 nanofiber-reinforced chitosan (SiO2 NF/CS) three-dimensional (3D) matrices exhibit complete shape recovery from 80% compressive strain and excellent injectability. The cellular architecture and nanofibrous structure in the SiO2 NF/CS matrices are beneficial for human mesenchymal stem cell adhesion and stretching. Furthermore, the SiO2 NF/CS matrices can also act as powerful vehicles for drug delivery. As an example, bone morphogenetic protein 2 could be immobilized on SiO2 NF/CS matrices to induce osteogenic differentiation. Together, the electrospun nanofiber-reinforced 3D chitosan matrices exhibited improved mechanical properties and enhanced biofunctionality, showing great potential in tissue engineering.

Abstract:The collagen–hyaluronic acid oligosaccharides (collagen–HA) complex was obtained by covalent attachment using EDC/NHS. The cross-linked product collagen–HA had been confirmed by Fourier transform infrared spectra (FTIR) and the sulfuric acid–carbazole colorimetry, and factors that affect the cross-linking process were investigated. Electrospinning of collagen–HA complex in hexafluoroisopropanol was investigated for the biomimetic nanostructure and biocompatible vascular tissue-engineered inner-layer scaffold. The morphology of the electrospun collagen–HA nanofibers with a series of concentrations was observed by scanning electron microscopy. The thermal behavior and chemical properties of the fabricated scaffolds were characterized using dynamic scanning calorimetry and FTIR after being stabilized by glutaraldehyde vapor. The cell attachment, morphology and proliferation of porcine iliac artery endothelial cells (PIECs) on the electrospun scaffolds were also evaluated. The results indicate that the biomimetic matrix collagen–HA electrospun nanofibrous scaffold could promote PIECs proliferation and may be a better candidate with endothelial potential for the vascular tissue-engineered inner-layer scaffold.

Abstract:The collagen-chitosan complex with a three-dimensional nanofiber structure was fabricated to mimic native ECM for tissue repair and biomedical applications. Though the three-dimensional hierarchical fibrous structures of collagen-chitosan composites could provide more adequate stimulus to facilitate cell adhesion, migrate and proliferation, and thus have the potential as tissue engineering scaffolding, there are still limitations in their applications due to the insufficient mechanical properties of natural materials. Because poly (vinyl alcohol) (PVA) and thermoplastic polyurethane (TPU) as biocompatible synthetic polymers can offer excellent mechanical properties, they were introduced into the collagen-chitosan composites to fabricate the mixed collagen/chitosan/PVA fibers and a sandwich structure (collagen/chitosan-TPU-collagen/chitosan) of nanofiber in order to enhance the mechanical properties of the nanofibrous collagen-chitosan scaffold. The results showed that the tensile behavior of materials was enhanced to different degrees with the difference of collagen content in the fibers. Besides the Young's modulus had no obvious changes, both the break strength and the break elongation of materials were heightened after reinforced by PVA. For the collagen-chitosan nanofiber reinforced by TPU, both the break strength and the Young's modulus of materials were heightened in different degrees with the variety of collagen content in the fibers despite the decrease of the break elongation of materials to some extent. In vitro cell test demonstrated that the materials could provide adequate environment for cell adhesion and proliferation. All these indicated that the reinforced collagen-chitosan nanofiber could be as potential scaffold for tissue engineering according to the different mechanical requirements in clinic.
Effect of Electrospun Collagen/Chitosan Composite Nanofibrous Membrane on Osteogenesis for Bone Regeneration

Abstract:The aims of this study are to prepare collagen/chitosan composite nanofibrous membranes,and to examine their biocompatibility and effect of osteogenic differentiation for bone marrow mesenchymal stem cells (BMSCs).Take acetic acid as solvent,poly(ethylene oxide) (PEO) as plasticizer,the collagen nanofibrous membranes (the mass rate of collagen/PEO,4∶1) and various collagen/chitosan nanofibrous membranes (the mass rate of collagen/chitosan/PEO,5∶1 ∶ 4,5∶ 2∶ 3,5∶ 4∶1) were fabricated through electrospinning technique.The surface morphology of membranes were observed by scanning electron microscopy (SEM).BMSCs were cultured on the collagen nanofibrous membranes and the optimized collagen/chitosan nanofibrous membranes,the cell proliferation and osteogenic makers including alkaline phosphate(ALP),collagen content,osteocalcin (OCN) and mineralized matrix deposits were detected respectively.SEM showed collagen nanofibers and collagen/chitosan nanofibers of mass ratio of 5 ∶ 1 ∶ 4 had a mean fiber diameter.Both collagen nanofibrous membranes and collagen/chitosan nanofibrous membranes supported adhesion,proliferation and osteogenic differentiation of the BMSCs.The collagen/chitosan nanofibrous membranes induced higher proliferation of BMSCs than collagen nanofibrous membranes.The collagen contents were higher in cells cultured on thecollagen/chitosan nanofibrous membranes for 14 days as compared to collagen nanofibrous membranes (p ＜ 0.05).Similarly,cells cultured on collagen/chitosan nanofibrous membranes had the higher expression of ALP,OCN and mineralized matrix deposits than that on collagen nanofibrous membranes.Our results demonstrated that the collagen/chitosan composite nanofibrous membranes promoted adhesion,proliferation and osteogenic differentiation of BMSCs,implying their promising applications for bone regeneration.

Abstract:Natural extracellular matrixs are net-like being composed of fibrin and fibers in nanoscale(50~500nm).From the biomimetic viewpoint,electrospinning technique would be a proper method to produce fibrous network mimicking the natural extracellular matrixes.Diameters of electrospun fibers range from tens of nanometers to micrometers,porous structures resulted from the loosely deposited fibers are of high porosity and high surface area,which are good choice for tissue engineering by providing proper environment for cells proliferation and differentiation.In this paper,an introduction to the preparation and biomedical application of several functional nanofibers is summarized basing on recent literature reports and the research works in our lab,focusing on developing novel regeneration materials for dental and hard tissue restoration.

Abstract:The protein-polysaccharide nanofibers have attracted intensive attention in promoting wound healing, due to their components and nanoscale fibrous structure that mimics the native extracellular matrix (ECM). For the full-thickness wounds, in addition to promoting healing, hemostatic property and antibacterial activity are also of critical importance. However, currently, protein-polysaccharide-based nanofiber membranes exhibit poor mechanical properties, lack inherent hemostatic and antibacterial capabilities, as well as the ability to promote tissue repair. In this study, we developed composited membranes, which were composed of collagen (Col) and chitosan (Chs), through solvent alteration and post-processing, the membranes showed enhanced stability under physiological conditions, proper hydrophilic performance and improved mechanical property. Appropriated porosity and water vapor transmission rate, which benefit to wound healing, were detected among all the membranes except for Col membrane. Aimed at wound dressing, hemocompatibility, antibacterial activity and cell proliferation of the electrospun membranes were evaluated. The results indicated that the Col/Chs composited membranes exhibited superior blood clotting capacity, and the membranes with Chs exceeding 60% possessed sufficient antibacterial activity. Moreover, compared with Chs nanofibers, significant increase in cell grow was detected in Col/Chs (1:3) membrane. Taken together, the electrospun membrane with multiple properties favorable to wound healing, superior blood coagulation, sufficient antibacterial performance and promoting cell proliferation property make it favorable candidate for full-thickness skin wound healing.

Abstract:AbstractElectrospinning is a simple and versatile technique to fabricate continuous fibers with diameter ranging from micrometers to a few nanometers. To date, the number of polymers that have been electrospun has exceeded 200. In recent years, electrospinning has become one of the most popular scaffold fabrication techniques to prepare nanofiber mesh for tissue engineering applications. Collagen, the most abundant extracellular matrix protein in the human body, has been electrospun to fabricate biomimetic scaffolds that imitate the architecture of native human tissues. As collagen nanofibers are mechanically weak in nature, it is commonly cross-linked or blended with synthetic polymers to improve the mechanical strength without compromising the biological activity. Electrospun collagen nanofiber mesh has high surface area to volume ratio, tunable diameter and porosity, and excellent biological activity to regulate cell function and tissue formation. Due to these advantages, collagen nanofibers have been tested for the regeneration of a myriad of tissues and organs. In this review, we gave an overview of electrospinning, encompassing the history, the instrument settings, the spinning process and the parameters that affect fiber formation, with emphasis given to collagen nanofibers’ fabrication and application, especially the use of collagen nanofibers in skin tissue engineering.
