Abstract:Silver nanoparticles (SNPs) translocate to the brain through the blood stream after they are implanted in vivo. The aim of this study was to investigate the distribution of SNPs that crossed through the blood-brain barrier (BBB). An in vitro BBB model established by co-cultures of rat brain microvessel vascular endothelial cells (BMVECs) with astrocytes (ACs) was cultured with cell culture medium containing 100 microg/mL of either SNPs or silver microparticles (SMPs). After 4 hours of culture, the ultrastructure and its silver content of BBB was evaluated with transmission electronic microscopy (TEM) and inductively-coupled plasma mass spectrometry (ICP-MS) respectively. Results demonstrated that SNPs crossed the BBB and accumulated inside BMVECs, while the SMPs did not. The data indicated a special distribution of SNPs in the BBB and suggested that SNPs pass the BBB mainly by transcytosis of capillary endothelial cells. Further study would be necessary to evaluate the actual biological effects of SNPs on the brain.

Abstract:Nanosilver has been widely used in medical biology; however, the distribution and interaction of nanosilver with cells is still unclear. There have been some reports demonstrating that nanoparticles can cross the blood-brain barrier (BBB). The present study investigated the accumulation of silver nanoparticles in the brain, and the effects of silver nanoparticles on BBB. Nanosilver and microsilver (62.8mg/kg) particles were subcutaneously injected into rats. The rats were sacrificed at predetermined time points and the brains were obtained for ultrastructural observation and silver level detection. The results showed that silver nanoparticles could traverse the BBB and move into the brain in the form of particle. The silver nanoparticles can induce neuronal degeneration and necrosis by accumulating in the brain over a long period of time.

Abstract:Silver nanoparticles (AgNPs) are used as a common ingredient in antiseptic sprays and mists; they can easily come into contact with the upper-airway mucosa. The intranasal pathway represents the only direct connection between the external environment and brain structures, which are generally considered to be well protected. Drugs absorption through this route has been widely studied, but toxicological knowledge is scant. The olfactory bundles are surrounded by meningeal sheets in their path from the nasal mucosa to the olfactory bulb. This study investigated the transmeningeal absorption of 19nm AgNPs, using excised porcine meninges mounted on Franz diffusion cells in vitro. Two donor solutions were used: one containing AgNPs (0.5gL(-1)), and another containing only the water-soluble silver species derived from the ultrafiltration of the first one. Each experiment was carried out separately for 2h. Results showed silver flux permeation through the meninges, with similar values in both experiments (0.78±0.71ngcm(-2)h(-1) and 0.73±0.43ngcm(-2)h(-1), for AgNPs and Ag ions respectively, mean and SD). This study demonstrates that the meningeal barrier is permeable to silver and silver ions, when it is exposed to this metal in nanoparticular form; this might lead to neurotoxic and neurodegenerative effects, as recently shown by other studies. Silver nanoparticles are used by workers and consumers, and potential penetration through meningeal membrane needs to be considered and prevented when it is possible an inhalation exposure.

Abstract:PEGylated PAMAM conjugated fluorescein-doped magnetic silica nanoparticles (PEGylated PFMSNs) have been synthesized for evaluating their ability across the blood–brain barrier (BBB) and distribution in rat brain. The obtained nanoparticles were characterized by transmission electron microscopy (TEM), thermal gravimetry analyses (TGA), zeta potential (ζ-potential) titration, and X-ray photoelectron spectroscopy (XPS). The BBB penetration and distribution of PEGylated PFMSNs and FMSNs in rat brain were investigated not only at the cellular level with Confocal laser scanning microscopy (CLSM), but also at the subcellular level with transmission electron microscopy (TEM). The results provide direct evidents that PEGylated PFMSNs could penetrate the BBB and spread into the brain parenchyma.

Abstract:Silver nanoparticles (SNPs) are widely used in the field of biomedicine, but a comprehensive understanding of how SNPs distribute in the body and the induced toxicity remains largely unknown. The present study was designed to investigate the distribution and accumulation of SNPs in rats with subcutaneous injection. Rats were injected with either SNPs or silver microparticles (SMPs) at 62.8 mg/kg, and then sacrificed at predetermined time points. The main organs of the experimental animals were harvested for ultrastructural analysis by transmission electron microscopy (TEM) and for silver content analysis by inductively coupled plasma mass spectrometry (ICP-MS). Results indicated that SNPs translocated to the blood circulation and distributed throughout the main organs, especially in the kidney, liver, spleen, brain and lung in the form of particles. SMPs, however, could not invade the blood stream, or organ tissues. Ultrastructural observations indicate that those SNPs that had accumulated in organs could enter different kinds of cells, such as renal tubular epithelial cells and hepatic cells. Moreover, SNPs also induced blood-brain barrier (BBB) destruction and astrocyte swelling, and caused neuronal degeneration. The results suggest more cautions needed in biomedical applications of SNPs, in particular, the long-term uses.

Abstract:Nanotechnology is a highly promising field, with nanoparticles produced and utilized in a wide range of commercial products. Silver nanoparticles (AgNPs) has been widely used in clothing, electronics, bio-sensing, the food industry, paints, sunscreens, cosmetics and medical devices, all of which increase human exposure and thus the potential risk related to their short- and long-term toxicity. Many studies indicate that AgNPs are toxic to human health. Interestingly, the majority of these studies focus on the interaction of the nano-silver particle with single cells, indicating that AgNPs have the potential to induce the genes associated with cell cycle progression, DNA damage and mitochondrial associated apoptosis. AgNPs administered through any method were subsequently detected in blood and were found to cause deposition in several organs. There are very few studies in rats and mice involving the in vivo bio-distribution and toxicity, organ accumulation and degradation, and the possible adverse effects and toxicity in vivo are only slowly being recognized. In the present review, we summarize the current data associated with the increased medical usage of nano-silver and its related nano-materials, compare the mechanism of antibiosis and discuss the proper application of nano-silver particles.
Clearance of Two Organic Nanoparticles from the Brain Via the Paravascular Pathway.

Abstract:The elaboration of nanotechnology offers valuable therapeutic options to overcome the blood-brain barrier and enable the treatment of brain diseases. However, to date, limit work has been done to reveal the fate of nanoparticles within the brain, which largely hinders their safe and effective applications. Here we demonstrated that the commonly-used organic nanoparticles reconstituted high density lipoprotein and poly(ethylene glycol)-b-poly(lactic acid) nanoparticles were cleared relatively fast from the brain (half-life <5 h). Notably, through various transgenic mice and pharmacological inhibition approaches, we revealed that the paravascular glymphatic pathway plays a key role (about 80%) in the brain clearance of the nanoparticles, and disclosed that microglia-mediated transportation is essential for facilitating nanoparticles elimination through the paravascular route. In addition, we witnessed a significant decline in the brain clearance of both of the nanoparticles in Alzheimer's model mice where the glymphatic system is impaired. These findings provide insightful data on the fate of nanoparticles in the brain, which would shed new light into the rational design and safe application of nanoparticles for brain drug delivery.

Abstract:Purpose Nanomedicine-based approaches have shown great potential in the treatment of central nervous system diseases. However, the fate of nanoparticles (NPs) within the brain parenchyma has not received much attention. The complexity of the microstructure of the brain and the invisibility of NPs make it difficult to study NP transport within the grey matter. Moreover, regulation of NP delivery is not fully understood. Methods 2D interstitial system (ISS) models reflecting actual extracellular space (ECS) were constructed. A particle tracing model was used to simulate the diffusion of the NPs. The effect of NP size on NP diffusion was studied using numerical simulations. The diffusion of charged NPs was explored by comparing experimental and numerical simulation data, and the effect of cell membrane potential on the diffusion of charged NPs was further studied. Results The model was verified using previously published experimental data. Small NPs could diffuse efficiently into the ISS. The diffusion of charged NPs was hindered in the ISS. Changes in cell membrane potential had little effect on NP diffusion. Conclusion This study constructed 2D brain ISS models that reflected the actual ECS and simulated the diffusion of NPs within it. The study found that uncharged small NPs could effectively diffuse within the ISS and that the cell membrane potential had a limited effect on the diffusion of charged NPs. The model and findings of this study can aid the design of nanomedicines and nanocarriers for the diagnosis and treatment of brain diseases. Abstract:The blood–brain barrier (BBB) restricts the delivery of many potentially important therapeutic agents for the treatment of brain disorders. An efficient strategy for brain targeted delivery is the utilization of the targeting ligand conjugated nanoparticles to trigger the receptor-mediated transcytosis. In this study, transferrin (Tf) was employed as a brain targeting ligand to functionalize the fluorescein-loaded magnetic nanoparticles (FMNs). The Tf conjugated FMNs (Tf-FMNs) were characterized by transmission electron microscopy, thermal gravimetric analysis, Fourier transform infrared spectroscopy, and X-ray photoelectron spectroscopy. Using fluorescein as an optical probe, the potential of Tf-FMNs as brain targeting drug carriers was explored in vivo. It was demonstrated that Tf-FMNs were able to cross the intact BBB, diffuse into brain neurons, and distribute in the cytoplasm, dendrites, axons, and synapses of neurons. In contrast, magnetic nanoparticles without Tf conjugation cannot cross the BBB efficiently under the same conditions. Therefore, Tf-FMNs hold great potential in serving as an efficient multifunctional platform for the brain-targeted theranostics.
Transport of ultrasmall gold nanoparticles (2 nm) across the blood-brain barrier in a six-cell brain spheroid model

Abstract:The blood-brain barrier (BBB) is an efficient barrier for molecules and drugs. Multicellular 3D spheroids display reproducible BBB features and functions. The spheroids used here were composed of six brain cell types: Astrocytes, pericytes, endothelial cells, microglia cells, oligodendrocytes, and neurons. They form an in vitro BBB that regulates the transport of compounds into the spheroid. The penetration of fluorescent ultrasmall gold nanoparticles (core diameter 2 nm; hydrodynamic diameter 3-4 nm) across the BBB was studied as a function of time by confocal laser scanning microscopy, with the dissolved fluorescent dye (FAM-alkyne) as a control. The nanoparticles readily entered the interior of the spheroid, whereas the dissolved dye alone did not penetrate the BBB. We present a model that is based on a time-dependent opening of the BBB for nanoparticles, followed by a rapid diffusion into the center of the spheroid. After the spheroids underwent hypoxia (0.1% O2; 24 h), the BBB was more permeable, permitting the uptake of more nanoparticles and also of dissolved dye molecules. Together with our previous observations that such nanoparticles can easily enter cells and even the cell nucleus, these data provide evidence that ultrasmall nanoparticle can cross the blood brain barrier.

Abstract:Nowadays, more and more nanotechnology products and nanomaterials are being applied in our lives. Silver nanoparticles (SNPs) are used in infection prevention and treatment due to their antimicrobial activity. However, as a kind of nanomaterial, the toxicology of SNPs has not been completely studied. The mechanism of cytotoxicity of SNPs in vitro to mouse's fibroblast cells (L929) was investigated in this study. As a contrast, silver microparticles (SMPs) were also studied. Propidium iodide (PI) single staining and Annexin-V/PI staining were carried out to unveil the influence of SNPs and SMPs on the cells. A transmission electron microscope (TEM) was used to observe SNPs' distribution in the cells. The results of cell cycle analysis indicated that the cells treated with SNPs were arrested in the G2M phase. Meanwhile, SNPs lead to apoptosis of more cells compared to SMPs at the same dose as a result of apoptosis analysis. Analysis of the cells' ultrastructure showed that SNPs could be phagocytized into the cells while SMPs could not. The mechanism of cytotoxicity of SNPs in vitro to L929 cells may be that SNPs are phagocytized into the cells and they interact with mitochondria or other organelles, even nuclei, which results in cells' apoptosis or necrosis.
Bioavailability and Distribution of Nanosilver after Oral Administration in Rats

Abstract:Due to the unique antimicrobial properties,nanosilver (AgNP) are widely applied in many fields.However,its bioavailability,tissue distribution and excretion in rats are less known.Female Sprague-Dawley rats were treated by a single intragastic administration of 10 mg·kg-1 polyvinylpyrrolidone-AgNPs,and rat tissues,feces and urine were collected periodically to analyze total Ag concentrations by ICP-MS.The results showed that after uptake by gastrointestine,AgNP was transported via the blood circulation and distributed to tissues such as liver,kidney,spleen,lung,and brain.At 1 h after AgNP exposure,total silver accumulation in those tissues peaked (i.e.0.29 ± 0.13 mg·kg-1,0.23 ± 0.04 mg·kg-1,0.17 ± 0.05 mg·kg-1,0.11 ± 0.01 mg·kg-1 and 0.06 ± 0.02 mg·kg-1 for liv er,kidney,spleen,lung and brain,respectively),and declined to background levels over time.Our calculation showed that the bioavailability of AgNP to Sprague-Dawley rats via intragastric administration was 8.5％.Especially,73％ of the dosed AgNP was excreted via feces.

Abstract:In order to study the potential bad biological effect of nanosilver, the equal quantity of nanosilver particles and microsilver particles were implanted into the rat’s back subcutaneous tissue. After 90 days, the method of plasma mass spectrometry (ICP-MS) was used to determine the silver distribution in several tissues of rats. ICP-MS analysis showed that the cumulation silver of nanosilver in the rat’s organ was more greater than microsilver. ICP-MS tests validated that nanosilver particles and microsilver particles appeared different distribute characteristic.

Abstract:Along with the increasing applications of nanomaterials in medical fields, to know the systemic distribution of nanomaterials in the body through a precise method is required for the biosafety assessment of nanomaterials. In this study, we firstly have established a reliable inductively coupled plasma mass spectrometry (ICP-MS) method for concentration measurement of silver (Ag) and gold (Au) in biological tissues. Then, based on this method, the Ag or Au distribution in rat blood and almost all of the organs were analyzed after an i.v. or s.c. administration of Au@Ag NRs. Both the time-dependent contents of Ag and Au in blood and two pharmacokinetic models confirmed the rapid clearance of Ag from blood. At 24 h after i.v. injection, there was the highest level of Ag in liver, followed by portal nodes, spleen, lung, bone marrow and pancreas. In addition, we also found there were gender-related distributions of Ag and Au in some organs, especially after s.c. injection. Therefore, these more comprehensive and important results would give fundamental information for the biological risk assessment of nanomaterials.

Abstract:An effective strategy to study the gastrointestinal fate of particulate Ag following oral exposure is developed by using Au–Ag core‐shell nanoparticles (Au@AgNPs), which opens an avenue toward the examination of oral bioavailability of NPs and may have significant implications in exploring the oral delivery technologies of nanoagents. The silver‐based formulation has become one of the most widely used nanoagents for agrifood industry and clinical translation. However, the in vivo fate of silver nanoparticles (AgNPs) via oral exposure remains poorly understood. Here, a specific strategy is shown for tracking the gastrointestinal transformation, intestinal absorption, and organ distribution of particulate Ag after oral exposure by using the gold (Au)‐Ag core−shell NPs (Au@AgNPs). Taking advantage of both plasmonic and elemental characteristics of Au@AgNPs, the transformation of particulate Ag in the gastrointestinal tract (GIT), and later on, intestinal absorption are visualized at the single‐particle level. Furthermore, quantitative analysis reveals that the clearance rate of ingested particulate Ag from the blood and organs is much higher than that of ionic Ag, and at 6 h post‐oral administration, the former only contributed 0.08%, 1.10%, and 0.64% of the total Ag in the liver, spleen, and kidneys. Finally, the transcellular and paracellular pathways are identified as two possible mechanisms responsible for the transepithelial transport of particulate Ag. This study opens an avenue toward the examination of oral bioavailability of NPs and may have important implications in exploring the oral delivery technologies of nanoagents.

Optimization and Evaluation of Pretreatment Method for Sp-Icp-ms to Reveal the Distribution of Silver Nanoparticles in the Body

Abstract:The prevalent use of engineered nanoparticles (ENPs) has increased our exposure to these particles. The current available analytical techniques fail to simultaneously quantify and analyze the physical properties of ENPs in biological tissues. Therefore, new methods are required to evaluate the exposure conditions to ENPs. Single particle inductively coupled plasma-mass spectrometry (sp-ICP-MS) is an attractive approach that can perform quantitative and qualitative analyses of ENPs. However, the application of this approach for biological samples is limited because of the lack of pretreatment methods for effectively recovering ENPs from biological tissues. In this study, we evaluated various pretreatment methods and identified the optimal pretreatment conditions for sp-ICP-MS analyses of ENPs in biological tissues using silver nanoparticles (nAg) as a model. We screened five reagents as pretreatment solvents (sodium hydroxide, tetramethylammonium hydroxide, nitric acid, hydrochloric acid, and proteinase K). Our results showed that treatment with sodium hydroxide was optimal for detecting nAg in the mouse liver. Moreover, this pretreatment method can be applied to other organs, such as the heart, lung, spleen, and kidney. Finally, we evaluated the applicability of this method by analyzing the quantity and physical properties of silver in the mouse blood and liver, after intravenous administration of nAg or silver ion. Our sp-ICP-MS method revealed that nAg administered into the blood was partially ionized and tended to be distributed in the particle form (approximately 80%) in the liver and in ionic form (approximately 95%) in the blood. In conclusion, we optimized pretreatment strategies for sp-ICP-MS evaluation of ENPs in biological tissues and demonstrated its applicability by evaluating the changes in the physical properties of nAg in the liver and blood. We also showed that partial changes from the particle form to the ionic form of nAg influences their kinetics and distribution when administered to mice.

In this paper, integrated analytical techniques with high sensitivity were used to study the brain translocation and potential impairment induced by intranasally instilled copper nanoparticles (CuNPs). Mice were exposed to CuNPs in three doses (1, 10, 40 mg/kg bw). The body weight of mice decreased significantly in the 10 and 40 mg/kg group (p<0.05) but recovered slightly within exposure duration. Inductively coupled plasma mass spectrometry (ICP-MS) analysis showed that CuNPs could enter the brain. Altered distribution of some important metal elements was observed by synchrotron radiation X-ray fluorescence (SRXRF). H&E staining and immunohistochemical analysis showed that CuNPs produced damages to nerve cells and astrocyte might be the one of the potential targets of CuNPs. The changes of neurotransmitter levels in different brain regions demonstrate that the dysfunction occurred in exposed groups. These data indicated that CuNPs could enter the brain after nasal inhalation and induced damages to the central nervous system (CNS). Integration of effective analytical techniques for systematic investigations is a promising direction to better understand the biological activities of nanomaterials.
