Abstract:Nanosized near infrared (NIR) responsive nanohybrids are synthesized by surface-initiated atom transfer radical polymerization (SI-ATRP) of N-isopropylacrylamide (NIPAAm) on gold nanorods (Au NRs). Transmission electron microscopy images demonstrate that the nanohybrids have a clear core/shell structure. Temperature-variable H-1 NMR spectroscopy, quasi-elastic light scattering (QELS), UV-vis spectroscopy, and laser irradiation prove that the nanohybrids are both temperature and NIR responsive. Norvancomycin (NVan) loading and release experiments show that the drug release rate can be modulated by NIR irradiation. Such an intelligent drug-delivery system might find applications in non-invasive controlled drug release in future.

Abstract:Core‐shell structured nanocarriers have come into the scientific spotlight in recent years due to their intriguing properties and wide applications in materials chemistry, biology, and biomedicine. Tailored core‐shell structures to achieve desired performances has emerged as a research frontier in the development of smart drug delivery system. However, a systematic review on the design and loading/release mechanisms of stimulus‐responsive core‐shell structured nanocarriers is seldom available. This review starts with the categories of core‐shell structured nanocarriers with different means of drug payload, and then highlights the controlled release mechanism realized through stimulus‐response processes triggered under different environments. Finally, some multifaceted perspectives on the design of core‐shell structured materials as drug carriers are addressed. We hope this review could provide new enlightenments and prospects in the drug delivery field for further developing advanced and smart nanocarriers. This article is protected by copyright. Abstract:A multifunctional platform with pH-responsive drug release and near-infrared (NIR) light-triggered photodynamic therapy (PDT) was designed and prepared using the novel core-shell-shell structure. The multifunctional platform consists of an upconversion nanoparticle (UCNP) emission core, a photosensitizer methylene blue (MB) embedded dense silica sandwich shell, and a polyethyleneimine conjugated folic acid (PEI-FA) gated mesoporous silica (MS) outmost shell with doxorubicin hydrochloride (DOX) loaded inside. The simulated drug release experiments revealed that DOX will release from the nanoparticles because of the distortion in the PEI-FA layer under acidic conditions. Moreover, under 980 nm NIR irradiation, a 660 nm red light emission was excited, activating MB to generate a singlet oxygen (1O2), which acts as the PDT drug. The multifunctional platform integrated pH-responsive drug release and UCNP-based PDT drug together show promising potential in nanomedicine for future chemotherapy and NIR-triggered PDT.

Abstract:Novel core-shell-shell structured nanoparticles 75 nm in diameter with all-in-one "smart" functional capabilities for simultaneous photoresponsive drug release, photodynamic therapy, and cell imaging are designed and prepared. These nanoparticles consist of an upconversion (UC) emission core, a photosensitizer-embodied silica sandwich shell, and a β-cyclodextrin (β-CD) gated mesoporous silica outmost shell with drugs (Rhodamine B as a model) loaded inside. We show in this proof-of-concept demonstration that, under 980 nm near-infrared irradiation, UC 540 nm green light emissions were emitted for cell imaging, and 660 nm red light emissions were excited for activating photosensitizers to generate singlet oxygen, which could be exploited directly to kill cancer cells and simultaneously dissociate β-CD gatekeeper to release drugs. The preliminary results reported here will shed new light on the future design and applications of multifunctional platforms for cancer therapy and diagnostic.

Abstract:The hexagonal mesoporous silica MCM-41 nanospheres with Au nanorods (AuNRs) as core have been synthesized via a modified Stöber method by a process of hydration and condensation of tetraethoxysilane in a water–ethanol mixture. The AuNR@MCM-41 nanocomposites combine the photothermal characteristic with the mesopore of MCM-41 in one body. We utilized these core–shell materials for ibuprofen encapsulation and release in the simulated body fluid (pH 7.4) for the first time. The results certificated AuNR@MCM-41 nanocomposites as novel dual-functional materials could realize the light-driven release of drug due to the photothermal effect of the AuNRs. Such novel nanomaterials offer a new way for cancer treatment which combine hyperthermia with the chemotherapeutic drugs by synergistic effect.
NIR‐Activated Supersensitive Drug Release Using Nanoparticles with a Flow Core

Abstract:Near infrared (NIR) light-activated supersensitive drug release via photothermal conversion is of particular interest due to its advantages in spatial and temporal control. However, such supersensitive drug release is rarely reported for polymeric nanoparticles. In this study, polymeric nanoparticles observed with flowable core can achieve NIR-activated supersensitive drug release under the assistance of photothermal agent. It is demonstrated that only 5 s NIR irradiation (808 nm, 0.3 W cm(-2)) leads to 17.8% of doxorubicin (DOX) release, while its release is almost completely stopped when the NIR laser is switched off. In contrast, the control, poly(D, L-lactide) nanoparticles with rigid cores, do not exhibit such supersensitive effect. It is demonstrated that intraparticle temperature is notably increased during photothermal conversion by detecting fluorescein lifetime using a time-correlated single photon counting (TCSPC) technique, which is the main driving force for such supersensitive drug release from hydrophobic flow core. In contrast, rigid chain of nanoparticular core hinders drug diffusion. Furthermore, such NIR light-activated supersensitive drug release is demonstrated, which significantly enhances its anticancer efficacy, resulting in overcoming of the resistance of cancer cells against DOX treatment in vitro and in vivo. This simple and highly universal strategy provides a new approach to fabricate NIR light-activated supersensitive drug delivery systems.

Abstract:Nanocarriers are essential for targeted and enhanced drug delivery. However, the use of toxic solvent and the complex of chemical functionalization have seriously limited their clinical translation. Here, a one-step strategy is developed to direct the self-assembly of biocompatible core-shell nanocapsules with tunable surface functionality by microfluidics. Upon rapid mixing and solvent exchange of ethanol with water in a microfluidic device, drug, oil, polymer, and polymer-PEG-function co-precipitate and self-assemble into core-shell nanocapsules with desired surface functionality as driven by energy minimization. The prepared PCL-PEG-FA core-shell nanocapsules demonstrate a high encapsulation efficiency and loading capacity and exhibit excellent tumor-targeting drug delivery performances both in vitro and in vivo via the binding of functional group on the nanocapsule surface with corresponding receptor on the tumor cell membrane. The immune activation in the tumor microenvironment is investigated in detail by flow cytometry and multiplex immunohistochemistry staining to reveal the underlying mechanism for enhanced anti-tumor performances. The developed strategy is green, facile, versatile, scalable and offers a promising platform for the design of advanced nanocarriers with hierarchical structure and desired function for enhanced drug delivery. Core-shell nanocapsules with tunable surface functionality are prepared by a one-step self-assembly method in a microfluidic device without any toxic solvent and additional chemical functionalization. Abstract:Multifunctional core-shell nanoparticles are widely used for biomedical and catalytic applications. In this work, bilayers of chitosan (Cs) and phosphorylated polyvinyl alcohol (PPVA) were sequentially deposited on 3-Aminopropyltri-ethoxysilane-modified SiO2 nanoparticles via layer-by-layer electrostatic self-assembly. The good spherical shape and size distribution were observed by DLS and transmission electron microscope analysis. 7-Hydroxycoumarin (7-HC) and rhodamine B (RhB) as model drugs were loaded in the core and shell of the nanoparticles separately. Confocal laser scanning microscopy shows the core-shell structure of HC-SiO2(PPVA/Cs)(n)-RhB nanoparticles and the embedded location of 7-HC and RhB. The pH-sensitive release investigation of RhB indicates that the release profiles of RhB from HC-SiO2(PPVA/Cs) 3PPVA-RhB core-shell nanoparticles are totally different at pH values of 2.0, 7.4, and 9.2. These results predict that the multifunctional nanoparticle SiO2(PPVA/Cs)(n) has a great potential for drug delivery.

Abstract:Stimuli triggered drug delivery systems enable controlled release of drugs at the optimal space and time, thus achieving optimal therapeutic effects. As one of the most important stimuli used in bioapplications, near-infrared (NIR) light possesses unique advantages such as deep tissue penetration with minimum auto-fluorescence & tissue scattering and high biosafety. Mesoporous silica nanoparticles (MSNs) are one of the most studied nanocarriers; apart from having a high surface area and large pore volume for loading of drugs, they can be easily functionalized with inorganic nanomaterials and stimuli responsive polymers or organic switch molecules, creating possibilities for designing complex stimuli triggered drug delivery systems. Considering the high tissue penetration depth of NIR light and the unique mesoporous structure of MSNs, NIR responsive inorganic nanoparticle functionalized MSNs can be further combined with stimuli responsive materials to form smart "nano-devices" for controlled drug delivery toward tumors, and to date much progress has been made. In this article, recent advances in the design of NIR triggered mesoporous silica drug delivery systems are systematically summarized and some outstanding studies are highlighted. We will also discuss the shortcomings, challenges and opportunities in the field.

Abstract:A near-infrared light-responsive drug delivery platform based on Au-Ag nanorods (Au-Ag NRs) coated with DNA cross-linked polymeric shells was constructed. DNA complementarity has been applied to develop a polyacrylamide-based sol-gel transition system to encapsulate anticancer drugs into the gel scaffold. The Au-Ag NR-based nanogels can also be readily functionalized with targeting moieties, such as aptamers, for specific recognition of tumor cells. When exposed to NIR irradiation, the photothermal effect of the Au-Ag NRs leads to a rapid rise in the temperature of the surrounding gel, resulting in the fast release of the encapsulated payload with high controllability. In vitro study confirmed that aptamer-functionalized nanogels can be used as drug carriers for targeted drug delivery with remote control capability by NIR light with high spatial/temporal resolution.

Abstract:Here we report a novel spiropyran-based organic-inorganic composite nanogel, which was prepared by upconversion nanoparticles, spiropyran, acrylic acid and N,N'-(bisacryloyl)cystamine compounds under emulsion polymerization. Compared with other polymer nanoparticles, the cross-linked multi-stimulus-responsive nanogels can adjust the release rate through more parameters and can meet the needs of complex biological environment to control the release of drugs. Doxorubicin hydrochlorides were used as a simulated drug to test the drug loading performance and controllable drug release performance of the composites nanogels (NGs). Under near-infrared light irradiation, acidic environment or reducing agent, the loaded drugs could be controlled release in 24 hours. Under mild triple combined stimulation (near-infrared light & pH 6 & 4 m M reducing agent), the loaded drug could be more efficiently released. The organic-inorganic composite NGs with highly-efficient and controllable release performance for loaded drugs provide multiple choices for novel stimulus-responsive nanocarriers.

Abstract:<p>Drug-delivery technology is an effective way to promote drug absorption and efficacy. Mesoporous hollow silica material and small-molecule drug ibuprofen were used as a carrier model and as model drug, respectively. By quantum chemical calculation (density functional theory and frontier orbital theory), it was found that the content of geminal silanols on the material surface played a decisive role in the release of the different drugs. The rough hollow materials are easily adsorbed and have a large loading capacity, and so we fabricated a mesoporous hollow silica material (R-nCHMSNs) with a rough surface and rich geminal silanols by using hydroxyl-rich nanocellulose as a template. The content and types of hydroxyl groups on the material surface were studied by <sup>29</sup>Si NMR. The loading and delivery of ibuprofen and lysozyme were studied in detail. Materials with rich geminal silanols exhibited excellent delivery properties for different drugs, which shows great potential and research value for drug delivery.</p>

Abstract:Ti3C2 MXenes, a novel two-dimensional material, have attracted lots of attention in biomedical filed for its large surface area and excellent near-infrared (NIR) responsiveness. In this paper, a pH/near-infrared (NIR) multi-responsive drug delivery platform consisted of hollow hydroxyapatite (HAP), chitosan (CS)/hyaluronic acid (HA) multilayers, gold nanorods (AuNRs) and MXene had been fabricated via a layer-by-layer (LbL) approach. Chitosan/hyaluronic acid multilayers were deposited on the surface of hollow HAP to retard the burst release of DOX in the initial delivery stage. MXenes and AuNRs equipped on the surface of hybrid matrix greatly enhanced the photothermal conversion efficiency of the microcapsules. Due to the collapse of electrostatic force among chitosan/hyaluronic acid multilayers and the dissolution of HAP under acidic condition, as well as the synergistically enhanced photothermal effect between MXene and AuNRs, HAP/CS/HA/MXene/AuNRs microcapsules exhibited outstanding pH-/NIR-responsive drug delivery properties. The present paper provides an attractive method to prepare chitosan/hyaluronic acid based pH/NIR multi-responsive hybrid microcapsules with excellent photothermal conversion efficiency and biocompatibility, which has great potential in the field of remotely controlled drug delivery.
Hierarchical hydroxyapatite/polyelectrolyte microcapsules capped with AuNRs for remotely triggered drug delivery

Abstract:In the present paper, a smart drug delivery platform combining the hollow hierarchical hydroxyapatite (HAP), chitosan/hyaluronic acid polyelectrolyte multilayers and Au nanorods (AuNRs) had been prepared via a layer-by-layer (LbL) technique. AuNRs capped on the surface of HAP/polyelectrolyte hybrid microcapsules acted as the "gate-keeper" to obstruct the leakage of loaded drug and endow the hybrid microcapsules with excellent near-infrared (NIR)-triggered drug release property. In vitro drug release results indicated that the hybrid microcapsules exhibited distinguished pH- and NIR-responsive drug release properties. More interestingly, relatively high DOX release could be observed at pH 4.5 upon NIR laser irradiation owing to the hybrid matrix disintegration in acidic media and the noteworthy photothermal transition effect of AuNRs. The cell viability results demonstrated that the hybrid microcapsules possessed excellent biocompatibility. The present paper provides a facile and green route to fabricate hierarchical multi-responsive drug carriers with high drug loading capacity and outstanding biocompatibility, which are highly attractive for remotely controllable drug delivery.

Abstract:As a naturally-abundant biopolymer, chitosan (CS) exhibit pH-sensitive structural transformation within a narrow pH range. Integrating hydrophobic groups to CS molecules gives modified CS polymers with more adjustable pH responsiveness. In this paper, near-infrared (NIR) photoluminescent Ag2S QDs capped by long-chain carboxylic acid were synthesized and then conjugated with CS via esterification reaction. The anticancer drug doxorubicin (DOX) has an affinity for the hydrophobic oleoyl groups and was entrapped by them to produce Ag2S(DOX)@CS nanospheres. A variety of experiments were performed to characterize the nanospheres. In vitro and in vivo experiments showed that the nanospheres can release DOX at lowered pH in tumor cells and have high antitumor efficacy. In addition, the strong NIR signal derived from the encapsulated Ag2S QDs makes real-time monitoring of the nanosphere distribution in a body possible. This study provides a new CS-based nanocomposite drug carrier for efficient cancer therapy.

Abstract:Drug-delivery vehicles have garnered immense interest in recent years due to unparalleled progress made in material science and nanomedicine. However, the development of stimuli-responsive devices with controllable drug-release systems (DRSs) is still in its nascent stage. In this paper, we designed a two-way controlled drug-release system that can be promoted and prolonged, using the external stimulation of near-infrared light (NIR) and protein coating. A hierarchical nanostructure was fabricated using upconversion nanoparticles (UCNPs)—mesoporous silica as the core-shell structure with protein lysozyme coating. The mesoporous silica shell provides abundant pores for the loading of drug molecules and a specific type of photosensitive molecules. The morphology and the physical properties of the nanostructures were thoroughly characterized. The results exhibited the uniform core-shell nanostructures of ~four UCNPs encapsulated in one mesoporous silica nanoparticle. The core-shell nanoparticles were in the spherical shape with an average size of 200 nm, average surface area of 446.54 m2/g, and pore size of 4.6 nm. Using doxorubicin (DOX), a chemotherapy agent as the drug model, we demonstrated that a novel DRS with capacity of smart modulation to promote or inhibit the drug release under NIR light and protein coating, respectively. Further, we demonstrated the therapeutic effect of the designed DRSs using breast cancer cells. The reported novel controlled DRS with dual functionality could have a promising potential for chemotherapy treatment of solid cancers.

Abstract:Metal-organic frameworks (MOFs) are permanently microporous materials. MOFs have become promising candidates as drug delivery hosts because of high specific surface area. In this work, we control preparation of a new core-shell microsphere NaYF4:Yb,Er@NaYF4:Nd@MIL-53 by layer-by-layer self-assembly method. Here upconverting fluorescent nanoparticles (UCNPs) NaYF4:Yb,Er@NaYF4:Nd has potential for bioimaging due to excellent NIR optical property. MIL-53 as a drug carrier for DOX delivery. Folic acid as targeting agent is encapsulated into the above core-shell structured to form new nanocomposite (UCNPs@ MIL-53/FA). We have found that the new core-shell nanocomposites are nontoxic toward human cervix adenocarcinoma (HeLa) and can simultaneously achieve targeted therapy and drug delivery are used for the treatment of cancer. This result is expected to provide new ideas and perspectives for the treatment of cancer of the tumor.
