Abstract:A collagen gradient was constructed to interrogate cell adhesion on a poly(L-lactide) (PLLA) membrane surface. Utilizing a microinfusion pump, gradients of amino groups were generated on the PLLA surface by an aminolysis method. Immobilization of collagen onto the gradient surfaces was performed by glutaraldehyde (GA) coupling to form the collagen gradients. The -NH(2) and immobilized collagen density profiles on the PLLA membranes were quantitatively determined by ninhydrin and hydroproline (Hyp) analysis, respectively. By using fluorescein isothiocyanate (FITC) labeled collagen (FITC-Col), the profile of the as-prepared collagen gradient was directly monitored by a fluorescence microscope. The scanning force microscopy and water contact angle studies revealed that the morphology and wettability of the modified membranes changed progressively as a function of position along the gradient surface. Rabbit auricular chondrocytes were cultured on the collagen gradient membranes to test their cellular response. The attachment and spreading behaviors of the chondrocytes were dependent on the surface collagen density. These results indicate a surface on which the variation of collagen gradient strongly modifies the biological response of chondrocytes.
Preparation of Chitosan Scaffolds Controlling-released bFGF and its Effect on Cell Culture

Abstract:Chitosan scaffolds was prepared by freeze-drying.The bovine serum album and basic fibroblast growth factor(bFGF) were selected as model medicine,respectively the poly(lactic-co-glycolic acid)(PLGA) microspheres were prepared by double emulsion solvent evaporation and encapsulated into chitosan scaffolds.Release behavior of medicine from various chitosan scaffolds was investigated.The effect of controlled released bFGF on L929 was studied by MTT.The morphology of scaffolds containing PLGA microspheres and L929 were observed by using scanning electron microscope.The result showed that medicine released quickly from chitosan scaffolds,while significant sustained release was obtained after the PLGA microsphere was prepared and then encapsulated in scaffolds.Furthermore,the controlled released bFGF could increase the proliferation of L929.

Abstract:Continuous gradients present at tissue interfaces such as osteochondral systems, reflect complex tissue functions and involve changes in extracellular matrix compositions, cell types and mechanical properties. New and versatile biomaterial strategies are needed to create suitable biomimetic engineered grafts for interfacial tissue engineering. Silk protein-based composites, coupled with selective peptides with mineralization domains, were utilized to mimic the soft-to-hard transition in osteochondral interfaces. The gradient composites supported tunable mineralization and mechanical properties corresponding to the spatial concentration gradient of the mineralization domains (R5 peptide). The composite system exhibited continuous transitions in terms of composition, structure and mechanical properties, as well as cytocompatibility and biodegradability. The gradient silicified silk/R5 composites promoted and regulated osteogenic differentiation of human mesenchymal stem cells in an osteoinductive environment in vitro. The cells differentiated along the composites in a manner consistent with the R5-gradient profile. This novel biomimetic gradient biomaterial design offers a useful approach to meet a broad range of needs in regenerative medicine.

Abstract:<p>Advances in engineered hydrogels reveal how cells sense and respond to 3D biophysical cues. However, most studies rely on interfacing a population of cells in a tissue‐scale bulk hydrogel, an approach that overlooks the heterogeneity of local matrix deposition around individual cells. A droplet microfluidic technique to deposit a defined amount of 3D hydrogel matrices around single cells independently of material composition, elasticity, and stress relaxation times is developed. Mesenchymal stem cells (MSCs) undergo isotropic volume expansion more rapidly in thinner gels that present an Arg‐Gly‐Asp integrin ligand. Mathematical modeling and experiments show that MSCs experience higher membrane tension as they expand in thinner gels. Furthermore, thinner gels facilitate osteogenic differentiation of MSCs. By modulating ion channels, it is shown that isotropic volume expansion of single cells predicts intracellular tension and stem cell fate. The results suggest the utility of precise microscale gel deposition to control single cell functions.</p>

Abstract:Tissue engineered scaffolds have demonstrated values in fabricating corneal equivalence. Attempts in this area tend to develop functional scaffold with physiochemical properties and structure resembling the native corneal tissue. Herein, a novel recombinant human collagen (RHC) based scaffold with ordered microstructures is presented to induce the regeneration of corneal stroma. As the RHC is modified with methacrylate anhydride (MA) under optimized concentration and is tailored with hierarchically ordered microstructures of aligned microgrooves and inverse opal nano-scale pores, the derived collagen hydrogels (RHCMA) can possess physiochemical properties resembling native cornea. Diverse topological structures (culture dish, RHCMA hydrogel without patterns, MI-RHCMA hydrogel patch with different width of microgrooves) are determined to study their influence on the growth and differentiation of corneal limbal stromal stem cells (LSSCs), and results demonstrate that the novel MI-RHCMA hydrogel patch could induce LSSCs to grow orderly and differentiate into keratocyte in vitro. Rat intrastromal keratoplasty are performed to examine the tissue repair efficacy of the derived collagen hydrogels, and the results indicate that the novel MI-RHCMA hydrogel patch could induce the regeneration of damaged corneal stroma in vivo. These results indicate that our presented scaffolds can be an effective transplantation graft and will find important values for the damaged cornea in the clinic.

Abstract:The ability to control cell patterning on artificial substrates with various physicochemical properties is of essence for important implications in cytology and biomedical fields. Despite extensive progress, the ability to control the cell-surface interaction is complicated by the complexity in the physiochemical features of bioactive surfaces. In particular, the manifestation of special wettability rendered by the combination of surface roughness and surface chemistry further enriches the cell-surface interaction. Herein we investigated the cell adhesion behaviors of Circulating Tumor Cells (CTCs) on topographically patterned but chemically homogeneous surfaces. Harnessing the distinctive cell adhesion on surfaces with different topography, we further explored the feasibility of controlled cell patterning using periodic lattices of alternative topographies. We envision that our method provides a designer's toolbox to manage the extracellular environment.

Abstract:The construction of gradient scaffolds to mimic the in vivo microenvironment for promoting tissue regeneration is attractive, especially by incorporating biochemical cues with physical guidance to manipulate cell behavior. Here, we report a general method to generate dual concentration-gradient of two types of bioactive components on scaffolds composed of uniaxially aligned electrospun nanofibers through layer-by-layer self-assembly. Different layers of collagen (COL) and chondroitin sulfate (CS) are sequentially coated onto the specific positions of uniaxially aligned electrospun polycaprolactone (PCL) nanofibers upon electrostatic attraction. In this case, dual stepwise gradients of COL and CS are generated along the fiber alignment. In demonstrating the potential application, the dual gradients can effectively promote the migration and proliferation of bone marrow stem cells (BMSCs) and spatially control their osteogenic differentiation. This simple strategy for generating dual gradients can be easily extended to different types of gradient scaffolds for bone regeneration and related applications involving cell migration, stem cell differentiation, and tissue interface.

Abstract:Collective endothelial migration is a hallmark of wound healing, which is regulated by spatial concentration gradients of extracellular biochemical factors. Arginine-glycine-aspartate (RGD) peptides play a vital role in regulating cell migration through specific binding to integrins. In this study, a micro-fluidic technology combined with photopolymerization technique is developed to create gelatin-based substrates with various concentration gradients of RGD peptides. The capability of generating linear and nonlinear RGD concentration gradients was quantitatively verified through numerical simulation and immunohistochemical quantitative experiments. The results of the concentration gradients show a strong concurrence between the immunohistochemical quantification experiments and numerical simulations. Further, endothelial migration experiments were conducted under various concentration gradients of RGD peptides. We have observed that endothelial cells on the surface of gels with a linear concentration gradient exhibit larger cell area, longer cell perimeter, and more stress fiber density. Furthermore, cells demonstrate directional alignment and migration towards regions with higher RGD concentration. High concentration gradients significantly enhance endothelial cell migration, consistent with observations on surfaces of gels with nonlinear concentration gradients. In brief, we proposed a simple and effective microfluidic photopolymerization technique capable of generating diverse concentration gradients of RGD and probing their effects on cell migration. The results suggest that regulating the RGD peptide concentration gradients can alter the migration of endothelial cells, showing the potential in promoting wound healing.

Abstract:Combinatorial material synthesis is a powerful approach for creating composite material libraries for the high-throughput screening of cell-material interactions. Although current combinatorial screening platforms have been tremendously successful in identifying target (termed "hit") materials from composite material libraries, new material synthesis approaches are needed to further optimize the concentrations and blending ratios of the component materials. Here we employed a microfluidic platform to rapidly synthesize composite materials containing cross-gradients of gelatin and chitosan for investigating cell-biomaterial interactions. The microfluidic synthesis of the cross-gradient was optimized experimentally and theoretically to produce quantitatively controllable variations in the concentrations and blending ratios of the two components. The anisotropic chemical compositions of the gelatin/chitosan cross-gradients were characterized by Fourier transform infrared spectrometry and X-ray photoelectron spectrometry. The three-dimensional (3D) porous gelatin/chitosan cross-gradient materials were shown to regulate the cellular morphology and proliferation of smooth muscle cells (SMCs) in a gradient-dependent manner. Cell‐laden Gradient Microgel Suspensions for Spatial Control of Differentiation During Biofabrication

Abstract:During tissue development, progenitor cells form functional tissue with high cellular diversity and intricate micro and macro architecture. Current approaches have attempted to replicate this process with materials cues or through spontaneous cell self‐organization. However, cell‐directed and materials‐directed organization are required simultaneously to achieve biomimetic structure and function. Here, we show how integrating live adipose derived stem cells with gradient microgel suspensions steers divergent differentiation outcomes. Microgel matrices composed of small particles were found to promote adipogenic differentiation, while larger particles fostered increased cell spreading and osteogenic differentiation. Tuning the matrix formulation, or using a pharmacological disruptor of actomyosin contractility, demonstrated that early cell adhesion and spreading dictate differentiation outcome. Combining small and large microgels into gradients leads to associated changes in cell behavior, with the properties of the microgels directing proliferation and differentiation spatially over time. After 21 days of culture, osteogenic conditions fostered significant mineralization within the individual microgels, thereby providing cell‐directed changes in composition and mechanics within the gradient porous scaffold. Freeform printing of high‐density cell suspensions was performed across these gradients to demonstrate the potential for hierarchical tissue biofabrication. Microgel properties were shown to guide cell behavior, where porosity influences cell expansion from the print and microgel size guides spatial differentiation, thereby providing scope to fabricate tissue gradients at multiple scales through integrated and printed cell populations. Effects of surface functionalization of PLGA membranes for guided bone regeneration on proliferation and behavior of osteoblasts.

Abstract:Covalent immobilization of bioactive compounds onto modified poly lactic-co-glycolic acid (PLGA) surfaces is being rapidly developed in tissue engineering, but the compounds and the grafting procedure require optimization. Here, PLGA membranes were grafted with various ratios of collagen/chitosan (COL/CHI) composites after modification by polydopamine and then analyzed using attenuated total reflectance Fourier transform infrared spectroscopy, field emission scanning electron microscopy, and a contact angle meter. Mechanical properties of the membranes were examined by tensile testing. Proliferation of osteoblastic cell line MC3T3-E1 cultured on the membranes was examined by MTT (3-(4, 5-dimethylthiozole-2-yl)-2, 5-diphenyltetrazolium bromide) and flow cytometric analysis. Effects on cell behavior, including cytotaxis, adhesion, and migration, were further investigated by continuous time-lapse imaging for 8 h. The COL/CHI composites were successfully immobilized onto PLGA surfaces. PLGA mainly contributed the mechanical properties, while COL/CHI played a major role in bioactivity. COL facilitated cell adhesion and spread, but the addition of CHI decreased both. A critical ratio of COL/CHI (2:1) above which the addition of CHI only slightly impacted cell proliferation was found. The results should be useful for combining versatile materials from different origins to construct guided bone regeneration membranes and to further optimize the ratio of COL/CHI composites. Ultrahigh pressure field: A friendly pathway for regulating the cellular adhesion and migration capacity of collagen

Abstract:Customized control of the biological response between the material matrix and cells is a crucial aspect in the development of the next generation of collagen materials. This study aims to investigate the effects of ultrahigh pressure treatment on the interaction between collagen and cells by subjecting bovine tendon collagen to different intensities of ultrahigh pressure field. The results indicate that ultrahigh pressure treatment alters the spatial folding of collagen, causing distortion of its triple helical conformation and exposing more free amino groups and hydrophobic regions. As a result, collagen's cell adhesion capability and ability to promote cell migration are significantly enhanced. Optimal cell adhesion and migration capabilities are observed in collagen samples treated at 500 MPa for 15 min. However, further increasing the intensity of the ultrahigh pressure treatment leads to severe damage to the triple-helical structure of collagen, along with re-aggregation of free amino groups and hydrophobic moieties, thereby reducing collagen's cell adhesion capability and ability to promote cell migration. Therefore, ultrahigh pressure treatment offers a promising method to effectively regulate collagen-cell adhesion and promote cell migration without the need for external components. This provides a potential means for the customized enhancement of collagen-based material interfaces.

Micropatterning of proteins and mammalian cells on biomaterials

Abstract:Controlling the spatial organization of cells is vital in engineering tissues that require precisely defined cellular architectures. For example, functional nerves or blood vessels form only when groups of cells are organized and aligned in very specific geometries. Yet, scaffold designs incorporating spatially defined physical cues such as microscale surface topographies or spatial patterns of extracellular matrix to guide the spatial organization and behavior of cells cultured in vitro remain largely unexplored. Here we demonstrate a new approach for controlling the spatial organization, spreading, and orientation of cells on two micropatterned biomaterials: chitosan and gelatin. Biomaterials with grooves of defined width and depth were fabricated using a two-step soft lithography process. Selective attachment and spreading of cells within the grooves was ensured by covalently modifying the plateau regions with commercially available protein resistant triblock copolymers. Precise spatial control over cell spreading and orientation has been observed when human microvascular endothelial cells are cultured on these patterned biomaterials, suggesting the potential of this technique in creating tissue culture scaffolds with defined chemical and topographical features.

Abstract:Abstract The construction of polymer-based mimicry on cell surface to manipulate cell behaviors and functions offers promising prospects in the field of biotechnology and cell therapy. However, precise control of polymer grafting sites is essential to successful implementation of biomimicry and functional modulation, which has been overlooked by most current research. Herein, we report a biological site-selected, in situ controlled radical polymerization platform for living cell surface engineering. The method utilizes metabolic labeling techniques to confine the growth sites of polymers and designs a Fenton-RAFT polymerization technique with cytocompatibility. Polymers grown at different sites (glycans, proteins, lipids) have different membrane retention time and exhibit differential effects on the recognition behaviors of cellular glycans. Of particular importance is the achievement of in situ copolymerization of glycomonomers on the outermost natural glycan sites of cell membrane, building a biomimetic glycocalyx with distinct recognition properties.

Abstract:Collagen fibrillogenesis is of special significance for the maintenance of collagen scaffold's mechanical stability and biological performance. Comprehensive information about the mechanism of collagen fibrillogenesis in vitro, as well as the effect of fibrillogenesis on deposited layers of ordered collagen molecules for cellular behavior regulation is thus crucial. In the current study, the pH, phosphate ion as well as reconstitution time impacting on the in vitro fibrillogenesis was systematically investigated, including the zeta potential and turbidity measurement. Furthermore, the fibrillogenesis impacting on the π-a isotherms of collagen assembly at the air/water interface was then fully evaluated. By applying LB technique, collagen fibril-assembling arrays structure can be successfully transferred to form surface deposition onto the mica and glass substrate. The morphology and collagen content were subsequently assessed by atomic force microscopy (AFM) and hydrolyzing examination respectively. Effect of collagen LB deposition on the adhesion and proliferation of SD rat bone marrow mesenchymal stem cells were estimated by Rhodamine Phalloidin/DIPI staining and CCK8 proliferation assays. The results show that highly oriented and collagen-abundant thin film can further facilitate cell adhesion and proliferation, indicating an innovative direction for tissue engineering.

Abstract:Bottom-up or modular tissue engineering is one of the emerging approaches to prepare biomimetic constructs in vitro, involving fabrication of small tissue units as building blocks before assembling them into functional tissue constructs. Herein, we reported a microscale tissue engineering approach to generate tubular tissue units through cellular contractile force induced self-folding of cell-laden collagen films in a controllable manner. Self-folding of cell-laden collagen films was driven by film contraction resulted from intrinsic contractile property of adherent mammalian cells seeded in collagen films. We explored in detail independent effects of collagen gel concentration, cell density, and intrinsic cellular contractility on self-folding and tubular structure formation of cell-laden collagen films. Through both experiments and theoretical modeling, we further demonstrated the effectiveness of integrating ridge array structures onto the backside of collagen films in introducing structural anisotropy and thus controlling self folding directions of collagen films. Our approach of using ridge array structures to introduce mechanical anisotropy and thus promote tubular tissue unit formation can be extended to other biomaterial systems and thus provide a simple yet effective way to prepare tubular tissue units for modular tissue engineering applications.

Abstract:Directional migration of cells in vitro can mimic corresponding biological events in vivo, which provides a way to determine the cascade responses in the tissue regeneration process and develop novel criteria for the design of tissue-inductive biomaterials. In this work, a density gradient of methoxy poly(ethylene glycol) brushes (from 0.37 to 0.95 chains/nm2) on plain and striped pattern surfaces was fabricated, using a dynamically controlled reaction process, and characterized by X-ray photoelectron spectroscopy and quartz crystal microbalance with dissipation. Adhesion and migration behavior of vascular smooth muscle cells were studied on the gradient and gradient-patterned surfaces. The vascular smooth muscle cells exhibited preferential orientation and enhanced directional migration on the gradient surface toward the lower end of the methoxy poly(ethylene glycol) density. By introducing methoxy poly(ethylene glycol) chemical striped patterns in parallel with the gradient direction on the surface, the extent of cell orientation and directional migration were significantly improved. Due to the synergetic effects of surface methoxy poly(ethylene glycol) striped patterns and gradient cues, almost all cells were oriented, and 67% of the cells were observed to move unidirectionally.
Surface Engineering of Poly(d,l‐lactic Acid) by Entrapment of Chitosan‐based Derivatives for the Promotion of Chondrogenesis

Abstract:Chitosan and chitosan-amino acid derivatives were explored to engineer poly(D,L-lactic acid) (PDL-LA) as an extracellular matrix-like surface to promote cell adhesion and growth. Four kinds of chitosan-amino acid derivatives were prepared to mimic the carbohydrate moieties of cell matrix glycoprotein. The chitosan-amino acid derivatives were characterized by using Fourier transform infrared and ultraviolet spectra. The amino acid content on chitosan-amino acid derivatives was determined by using a ninhydrin-ultraviolet method. A new strategy, entrapment, was therefore used to modify the PDL-LA membrane with chitosan and chitosan-amino acid derivatives. The results of X-ray photoelectron spectroscopy, attenuated total reflectance-Fourier transform infrared, and contact angle confirmed that a stable thin film of chitosan and its derivatives can be entrapped on the surface of the PDL-LA membrane. From the results of chondrocyte cytocompatibility, MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide] assays, and cell morphology, the chitosan-amino acid derivative modified PDL-LA membranes were shown to promote chondrogenesis. The novel surface treatment method combines the good mechanical property of PDL-LA with the good cytocompatibility of chitosan derivatives, which may have potential for tissue engineering.
