Abstract:Significance: Untreated methemoglobinemia may cause severe hypoxemia and even death when methemoglobin levels in the blood stream exceed 70%. Although CO-oximetry can be used to monitor the response to treatment for methemoglobinemia, it is costly and requires an invasive procedure for collecting blood samples from patients. A pulse CO-oximeter with a contact probe can be used to continuously and non-invasively measure the percentage of methemoglobin, as well as the percutaneous oxygen saturation. In terms of the prevention of infectious diseases, however, it is desirable to monitor methemoglobin and oxygen saturation levels in a non-contact manner. Diffuse reflectance spectral imaging is promising as a non-contact, non-invasive, and cost-effective clinical diagnostic tool for methemoglobinemia. Aim: To demonstrate the feasibility of visible spectral diffuse reflectance for in vivo monitoring of hemoglobin derivatives and evaluating methemoglobin production and reduction as well as hypoxemia during methemoglobinemia in rats. Approach: A new imaging approach based on the multiple regression analysis aided by Monte Carlo simulations for light transport was developed to quantify methemoglobin, oxygenated hemoglobin, and deoxygenated hemoglobin using a hyperspectral imaging system. An in vivo experiment with rats exposed to sodium nitrite (NaNO2) at different doses was performed to confirm the feasibility of the method for evaluating the dynamics of methemoglobin, oxygenated hemoglobin, and deoxygenated hemoglobin during methemoglobinemia. Systemic physiological parameters, including the percutaneous arterial oxygen saturation, heart rate (HR), and pulse distention, were measured by a commercially available pulse oximeter, and the results were compared to those obtained by the proposed method. Results: Both the methemoglobin concentration and methemoglobin saturation rapidly increased with a half-maximum time of <20 min. They reached their maximal values nearly 60 min after the administration of NaNO2. Tissue oxygen saturation dramatically dropped to a minimum of 33.7 % ± 0.4 % , 23.1 % ± 5.6 % , 8.8 % ± 1.7 % , and 9.7 % ± 5.1 % on average for NaNO2 doses of 25, 37.5, 50, and 75 mg/kg, respectively. Changes in methemoglobin concentration and tissue oxygen saturation are indicative of the temporary production of methemoglobin and severe hypoxemia during methemoglobinemia. Profound increases in the HR and pulse distention implied an elevated cardiac output caused by tachycardia and the resultant increase in peripheral blood volume to compensate for the hypoxia and hypoxemia during methemoglobinemia. This was in agreement with the time course of the peripheral hemoglobin volume concentration obtained by the proposed method. Conclusions: The proposed method is capable of the in vivo non-contact simultaneous evaluation of methemoglobin levels and hypoxemia during methemoglobinemia, and that it has potential as a tool for the diagnosis and management of methemoglobinemia.

Abstract:Microdialysis is a powerful in vivo sampling technique for pharmacokinetic-pharmacodynamic (PK-PD) modeling of drugs in pre-clinical and clinical studies. However, the noticeable limitations of previous studies using microdialysis were that animals anesthesia in the whole experiment and the combination of microdialysis and blood sampling for drug and (or) effect detection, which can obviously influence PK and PD behavior of drugs. In this study, a simple blood microdialysis sampling system in freely moving rats was established for simultaneous study of PK and PD of Shengmai injection (SMI) effect on inducing real-time nitric oxide (NO) release on isoproterenol (ISO) induced myocardial ischemia rats. The LC-MS/MS and HPLC with fluorescence detection (HPLC-FLD) methods were developed to determine ginsenside Rg1, Rg2, Re, Rf, Rb1, Rd and Rc, the main effective components of SMI, and NOx-, the main oxidation products of NO, in dialysates respectively. Through simultaneous determination of drug concentrations and NO efficacy levels in dialysate, the developed methods were successfully applied to set up concentration-time and effect-time profiles followed by PK-PD modeling of SMI effect on inducing NO release after intravenous administration of 10.8 mL kg(-1) SMI in myocardial ischemia rats. The PK-PD modeling characterized the dose-effect relationships of SMI and behaved good prediction ability. The established blood microdialysis in freely-moving rats is an appealing technology for rational PK-PD studies when selecting suitable blood endogenous micromolecule as effect marker. Abstract:The Raman spectra of nitrosyhemoglobin (NOHb), methemoglobin (MetHb), oxygenated hemoglobin (oxyHb), and deoxygenated hemoglobin(deoxyHb) were comparatively studied. The results show that the Raman spectra with 632.8 nm excitation can be used to detect the R state (oxyHb) to T state (deoxyHb) transition. The difference of NOHb and MetHb in their electron spin state was clearly demonstrated in their Raman spectra with 632.8 nm excitation, suggestingthat Raman spectra provide anew simple way to measure and distinguish NOHb and MetHb.

Abstract:The precise cellular function of peroxynitrite (ONOO-) in biosystems remains elusive, primarily owing to being short of ultrasensitive techniques for monitoring its intracellular distribution. In this work, a novel rhodamine B cyclic 1,2-dimethylhydrazine fluorescent chemodosimeter RDMH-PN for highly specific and ultrasensitive monitoring of basal ONOO- in biosystems was rationally designed. The fluorescence titration experiments demonstrated that RDMH-PN was capable of quantitatively detecting 0-100 nM ONOO- (limit of detection = 0.68 nM). In addition, RDMH-PN has outstanding performances of ultrafast measurement, naked-eye detection, and preeminent selectivity toward ONOO- to accurately detect intracellular basal ONOO-. Finally, it has been confirmed that RDMH-PN could not only map the intracellular basal ONOO- level by inhibition tests but also trace the fluctuations of endogenous and exogenous ONOO- levels with diverse stimulations in live cells and zebrafish.
A Novel Raman Spectrum Detection System Based on an Integrated Optical Biosensor

Abstract:In recent years, there has been a growing demand for hand held and miniaturized spectroscopic Raman systems that can be employed in the field to distinguish and quantify different analytes. In this paper, a novel and integrable system to detect a Raman spectrum is presented. We present the system principle, sensor design, experimental set-up and primary measurement results. In a conventional Raman setup, the four important components are: a light source, sensor, spectrometer and detector. We utilize a tunable laser as light source in the new Raman detecting system to replace the spectrometer by scanning the pump wavelength. A Raman sensor based on silicon nitride platform which has small size and high signal-background ratio is demonstrated in this paper to enable the excitation and the collection of the Raman signal using a plasmonic slot waveguide structure. Besides the tunable laser and the Raman sensor, there are two basic devices in our system, a narrow band-pass filter and a power detector. In this work, the Raman signal of the measured molecule 4-nitrothiophenol (NTP) is obtained by scanning the pump wavelength from 735 nm to 786 nm. The light source and detector in our experiment are implemented by discrete components. Silicon photonics promises the integration of a complete on-chip Raman spectroscope where the tunable laser, detector, sensor and filter can be integrated in a millimeter sized chip. We analyze the primary results measured by the discrete devices and discuss the feasibility of the on-chip integration in the end.

Abstract:The Raman spectrum of a single erythrocyte captured by a microfluidic chip was recorded to determine the conformation of hemoglobin under conditions similar to the hemodynamics of a blood vessel. Amplitude changes in the Raman spectrum at 1355, 1375, 1552, 1620, 1585 and 1637 cm−1 reflect changes in pO2 due to O2 binding to hemoglobin heme.
Microspectrophotometry of nitric oxide-dependent changes in hemoglobin in single red blood cells incubated with stimulated macrophages

Abstract:A highly sensitive microspectrophotometer was developed to measure spectral changes of oxyhemoglobin (oxy Hb) in single red blood cells (RBC) incubated with stimulated macrophages as a model of nitric oxide (NO) dependent cytotoxicity. Our microspectrophotometer, using a modified acousto-optic tunable filter (AOTF) and a 2-dimensional CCD array, allows fast spectrophotometric data acquisition. Human RBC treated with various concentrations of NO showed spectral changes due to the conversion of oxy Hb to methemoglobin (met Hb), in which the change in absorption differences at alpha (557 590 nm) and beta (542-525 nm) bands showed a linear relationship with the concentration of NO up to 100 microM. In contrast to highly diffusible NO, nitrite ions (NO2-) seem to enter RBC very slowly, resulting in negligible formation of met Hb in the presence of 5 mM glucose even during a prolonged incubation period. RBC were incubated with murine macrophages with and without lipopolysaccharide (LPS) in the presence of glucose for 24 and 40 h and subjected to the microspectrophotometric assay. The RBC incubated with LPS-stimulated macrophages showed significant changes in the spectrum due to NO-dependent conversion of oxy Hb to met Hb, which corresponded to the spectral changes of RBC treated with a several times higher concentration of NO than that in the culture medium. The trapping efficiency was calculated from the amounts of the NO released from macrophages and of the met Hb formed in the RBC, which gave a high efficiency (43%). The results suggest that RBC trap NO directly by cell cell interaction with macrophages. This spectrophotometric system is available for use with just a few drops of samples to study NO-specific cytotoxicity as a model of RBC without the use of any chemical reagent, in parallel with microscopic observations on changes of the cellular morphology under physiological conditions, such as membrane damage leading to hemolysis, adherence, and phagocytosis.
Noninvasive Raman Imaging for Monitoring Mitochondrial Redox State in Septic Rats

Abstract:Raman imaging for a sepsis study is reported here. We propose a confocal resonance Raman microscopic imager (CRRMI) to measure in vivo the redox state of mitochondria over a surface area of a septic rat. The CRRMI has excellent performance with spectral and spatial resolutions of 0.1nm and 2 mu m, respectively. It is found that the Raman signal related to the mitochondrial dysfunction in sepsis is abnormally large only locally at many points with some random spatial distribution. Our CRRMI can detect the mitochondrial redox state through the skin of a naturally living rat even without the removal of hairs and overcomes some issues that a pointwise measurement method of Raman signals may encounter when monitoring mitochondrial dysfunction of a sepsis rat, such as the fluorescence of hairs, hitting the points without mitochondrial redox metabolic disorder, etc. The present Raman imager can be used for giving an early warning for sepsis. It provides a new method for noninvasive monitoring of mitochondrial redox state in sepsis.

Abstract:Paper indicators based on colorimetric assays are widely used for nitrite detection, but their application to liquids with strong colours is restricted. We report a novel paper indicator that allows for sensing nitrite by colorimetric assay, Raman spectroscopy, and electron paramagnetic resonance spectroscopy with non-overlapping signal wavelength ranges through non-contact means. The paper indicator was prepared by impregnating poly(4-aminostyrene), 2-naphthol and single-walled carbon nanotubes in a regular filter paper. All three ingredients were essential to realize the triple-modality sensing. This method is simple and inexpensive, and promises to have wider applicability than the existing paper indicators.

Abstract:Blood testing is a crucial application in the field of clinical studies for disease diagnosis and screening, biomarker discovery, organ function assessment, and the personalization of medication. Therefore, it is of the utmost importance to collect precise data in a short time. In this study, we utilized Raman spectroscopy to analyze blood samples for the extraction of comprehensive biological information, including the primary components and compositions present in the blood. Short-wavelength (532 nm green light) Raman scattering spectroscopy was applied for the analysis of the blood samples, plasma, and serum for detection of the biological characteristics in each sample type. Our results indicated that the whole blood had a high hemoglobin content, which suggests that hemoglobin is a major component of blood. The characteristic Raman peaks of hemoglobin were observed at 690, 989, 1015, 1182, 1233, 1315, and 1562–1649 cm−1. Analysis of the plasma and serum samples indicated the presence of β-carotene, which exhibited characteristic peaks at 1013, 1172, and 1526 cm−1. This novel 3D silicon micro-channel device technology holds immense potential in the field of medical blood testing. It can serve as the basis for the detection of various diseases and biomarkers, providing real-time data to help medical professionals and patients better understand their health conditions. Changes in biological data collected in this manner could potentially be used for clinical diagnosis.

Abstract:A miniature wearable Raman spectroscopy system used to achieve noninvasive detection of human blood glucose level was developed. A thallium-doped grin lens was employed as the collection lens and a specially designed wearable fiber optic probe was employed to help the stable and convenient collection of Raman spectrum. The glucose solution, 11 rats and 10 healthy human were studied as subjects. In addition, a method of quantitative analysis of Raman spectrum was proposed which using the peak area as the main reference factor while the peak intensity as the auxiliary reference factor to calculate the target concentration. A non-linearized multivariate dominant factor-based partial least squares model was built for different samples to predict glucose level. The results show that the accuracy are 98.1%, 89.3% and 84.4% for glucose solution, rats and human subjects. The system has the advantages of more compact structure, lower cost, better testing stability and convenient for human body to wear, and is feasible and repeatable to achieve the noninvasive detection of human blood glucose accurately.

Abstract:Stimulated Raman Scattering (SRS) coupled with alkyne tags has been an emerging imaging technique to visualize small-molecule species with high sensitivity and specificity. Here we describe the development of a ratiometric Raman probe for visualizing hydrogen sulfide (H2S) species in living cells as the first alkyne-based sensor for SRS microscopy. This probe uses an azide unit as a selective reactive site, and it targets mitochondria with high specificity. The SRS ratiometric images show a strong response to H2S level changes in living cells.

Abstract:A hybrid "optical tweezers-micro Raman spectroscopy" technique is introduced in this letter. Applying this system, the micro-Raman spectrums of rat erythrocyte and saccharomyces cerevisiae in vivo were obtained. The results show that this technique can obviously improve signal-to-noise ratio compared with conventional approaches.Besides, the damage from lasers on the cells were analyzed. It was revealed that the damage of cell from intense IR laser were much lower than that from the weak Ar~+ laser. Finally, the system was also applied in carbon nano-tube (CNT) analysis and Raman spectra from CNT were obtained.
Detection and monitoring of neurotransmitters--a spectroscopic analysis

Abstract:Objectives: We demonstrate that confocal Raman mapping spectroscopy provides rapid, detailed, and accurate neurotransmitter analysis, enabling millisecond time resolution monitoring of biochemical dynamics. As a prototypical demonstration of the power of the method, we present real-time in vitro serotonin, adenosine, and dopamine detection, and dopamine diffusion in an inhomogeneous organic gel, which was used as a substitute for neurologic tissue. Materials and methods: Dopamine, adenosine, and serotonin were used to prepare neurotransmitter solutions in distilled water. The solutions were applied to the surfaces of glass slides, where they interdiffused. Raman mapping was achieved by detecting nonoverlapping spectral signatures characteristic of the neurotransmitters with an alpha 300 WITec confocal Raman system, using 532 nm neodymium-doped yttrium aluminum garnet laser excitation. Every local Raman spectrum was recorded in milliseconds and complete Raman mapping in a few seconds. Results: Without damage, dyeing, or preferential sample preparation, confocal Raman mapping provided positive detection of each neurotransmitter, allowing association of the high-resolution spectra with specific microscale image regions. Such information is particularly important for complex, heterogeneous samples, where changes in composition can influence neurotransmission processes. We also report an estimated dopamine diffusion coefficient two orders of magnitude smaller than that calculated by the flow-injection method. Conclusions: Accurate nondestructive characterization for real-time detection of neurotransmitters in inhomogeneous environments without the requirement of sample labeling is a key issue in neuroscience. Our work demonstrates the capabilities of Raman spectroscopy in biological applications, possibly providing a new tool for elucidating the mechanism and kinetics of deep brain stimulation.
Applications of Ratiometric Nanoprobe in Endogenous Signal Micromolecules Imaging

Abstract:Endogenous signal micromolecules play important roles in the process of life activities. The changes in concentration and the anomalies of their distribution are related to a variety of physiological and pathological states. The design of specific probes for the real-time sensing and imaging of biomolecules in vivo is significant for disease diagnosis, etiological study and understanding of cell signal transduction mechanism. However, precise quantification by the conventional single-emission-based sensors is fraught with trouble. Ratiometric nanoprobes have unique strength in accurate and quantitative analyses, which have been widely used in the field of biomedicine. In this review, we provide an overview of the design principles of nanoprobes that have been exploited for the ratiometric detection of various pathophysiological substances, including H+, ROS/RNS and gasotransmitter in biological samples.
Online Measurement of Sodium Nitrite Based on Near-Infrared Spectroscopy

Abstract:In this study, a method was developed for the rapid online measurement of sodium nitrite solutions using near-infrared spectroscopy. A series of standard solutions of sodium nitrite at different concentrations were prepared, and the samples were measured in cuvettes and flow cells. Following the preprocessing of raw spectra and band selection, partial least squares were used to establish a prediction model, and the coefficient of determination (R2) of the validation set and the root mean square error of prediction (RMSEP) of the model were 0.9989 and 0.0338. The results demonstrate that the established model can meet the demands of online measurement and perform the rapid, nondestructive detection of sodium nitrite solutions, which provides some basis for the automated formulation of feedstock in spent fuel reprocessing.

Abstract:Glucose performs many essential functions associated with metabolic processes in the living system, and is closely related to many diseases such as diabetes and hypoglycemia. Most of the existing glucose concentration detection methods require complex instruments, which undoubtedly limit its widespread use. Here, we have designed a glucose colorimetric detection system composed of glucose, glucose oxidase (GOD), manganese dioxide nanosheets (MnO2 NSs) and 3,3',5,5' -tetramethylbenzidine (TMB) to achieve colorimetric detection with the naked eye. Compared with the single-color change of the colorimetric method in previous studies, multiple-color changes have been realized. MnO2 NSs, as a kind of nanomaterial imitating oxidase, can directly oxidize TMB to oxTMB. Because oxTMB showed a dark yellow color when strongly oxidized and light blue when weakly oxidized, this feature can achieve multiple-color changes rather than a single-color change, which is helpful for colorimetric observation with the naked eye. Finally, we successfully used MnO2 NSs for colorimetric detection of glucose and realized multiple-color changes, making it easier to achieve colorimetric observation with the naked eye. The linear detection range is 0-4000 mu M and limit of detection is 5.0 mu M. This is not only useful for glucose, but also has an important significance for other experiments considering colorimetric experiments with the naked eye.

Abstract:Scattering can seriously affect the highly sensitive detection and quantitative analysis of chemical substances in scattering media and becomes a significant challenge for <i>in vivo</i> application of Raman spectroscopy. In this study, we demonstrated a proof of concept for using the self-reconstructing Bessel beam for Raman spectroscopic sensing of the chemicals in the handmade scattering media and biological tissue slices. The homebuilt Bessel beam Raman spectroscopy (BRS) was capable of accurately detecting the Raman spectra of the chemicals buried in the scattering media, and had a superiority in quantitative analysis. The feasibility of the developed technique was verified by detecting the Raman spectra of pure samples in air. Compared with the spectra acquired by the Gaussian beam Raman spectroscope, the performance of the BRS system in terms of Raman spectrum detection and Raman peak recognition was confirmed. Subsequently, by employing the technique for the detection of acetaminophen buried in the scattering media, the application of the new technology in detecting and quantitating the chemicals in the scattering media were underlined, offering greater detection depth and better linear quantification capability than the conventional Gaussian beam Raman spectroscopy. Finally, we explored the potential of the BRS system for chemical sensing of acetaminophen in biological tissue slices, indicating a significant development towards the evaluation of drug <i>in vivo</i>.

Abstract:Nonmetal plasmonic semiconductors have shown promising surface plasmon properties with advantages including abundance, low cost, and flexibility in synthesis and tuning. However, it is urgently desired to explore semiconductor materials for improving the plasmonic effect. Herein, a plasmonic oxygen-deficient sodium tungsten bronze Na y WO3–x nanosheet array has been facilely achieved on the glass substrate by in situ Na intercalation, followed by hydrogen reduction. The Na y WO3–x nanosheet array manifested a visible localized surface plasmon resonance band centered at 558 nm, which, to the best of our knowledge, was the bluest absorption peak for sodium tungsten bronze. Based on this unique surface plasmon property, the Na y WO3–x nanosheet array exhibited a good surface-enhanced Raman scattering (SERS) effect, and a detection limit of 10–7 M for methylene blue was achieved. In addition, as a SERS substrate, the Na y WO3–x nanosheet arrays not only guaranteed high SERS signal stability but also had a much easier and facile operation. Therefore, the plasmonic oxygen-deficient Na y WO3–x nanosheet array with low cost and easy availability shows great potential for applications in light harvesting and optoelectronic devices in addition to sensing devices.

Abstract:A novel label-free method for the in-situ monitoring of protein tyrosine nitration (PTN) was explored based on surface-enhanced Raman spectroscopy (SERS). Benefiting from the relative weak binding ability of sulfate to silver surface, the Raman signals of nitrated peptides were boosted well with sulfate-aggregated silver nanoparticles (Ag NPs). The distinction of the SERS spectra between non-nitrated peptides and nitrated peptides was obtained by directly comparing SERS bands at 330–400cm−1, allowing the rapid identification of PTN. Furthermore, without any pretreatments, the established method was successfully applied in the rapid in-situ dynamic monitoring of the mimic hemin-catalyzed PTN process in synthetic peptide, bovine serum albumin (BSA), and original human blood serum samples. The results indicated that the proposed approach could be a promising in-situ label-free tool for observing PTN process, which may be quite helpful to deeply understand the mechanism of post-translation modification.
