Repairing injured optic nerve using olfactory ensheathing cells and autologus sciatic nerve transplantation

Abstract:Background: Previous research has proved that methylprednisolone exhibits protective effects on retinal ganglion cells in the early stage of injury. Objective: To investigate the changes of morphology and count in the damaged optical fiber and myelin sheath after transplanted methylprednisolone with olfactory ensheathing cells and autologus sciatic nerves. Design, time and setting: The randomized control experiment of animals were performed at the Department of Anatomy, Peking University Health Science Center between September 2005 and March 2006. Materials: Forty adult, male, Sprague Dawley rats, four from them were served as the normal control group. Methods: The other thirty-six rats were randomly divided into the optic nerve injury, methylprednisolone, olfactory ensheathing cells (OECs) transplantation, OECs transplantation plus methylprednisolone, sciatic nerve transplantation and sciatic nerve transplantation plus methylprednisolone groups, with 6 animals in each group. Main outcome measures: At days of 14 and 21, frozen section of the optic nerves was prepared and randomized selected 10 slides from each group to observe the morphous change of optic nerves and the number change of myelin sheath with biotinylated dextran amine (BDA) staining and toluidine blue staining, respectively. The number of optical fiber myelin sheath was count under the electron microscope. Results: The DAB staining mainly appeared in the OECs transplantation plus methylprednisolone, sciatic nerve transplantation plus methylprednisolone groups, in which the nerve fibers arranged in order compared with the optic nerve injury group. At days of 14 and 21, the number of optical fiber myelin sheath in the methylprednisolone group was significant greater than that of the optic nerve injury group (P < 0.05, 0.01). The sciatic nerve transplantation plus methylprednisolone group was obvious higher than that of the sciatic nerve transplantation group (P < 0.05). Meanwhile, the OECs transplantation plus methylprednisolone was notable greater than the OECs transplantation group (P < 0.05). Conclusion: The morphology result indicates that combined OECs and autologus sciatic nerves transplantation with methylprednisolone can stimulate, further more, enhance the self-protection and self-repair mechanism of the injury central nerve, which plays an delaying effects of degeneration on the myelin sheath.
