Abstract:Endogenous fluorophores, such as NAD(P)H/FAD and collagen/elastin, have been regarded as in vivo quantitative fluorescence biomarkers for precancerous changes of epithelial tissue. However, the fluorescence signal measured by conventional spectroscopy is a mixture of autofluorescence from the epithelium and deep structures. The dominant fluorescence of collagen/elastin from connective tissue in deep layers creates serious challenge for extracting the epithelial fluorescence of NAD(P)H/FAD that is weak, but important for the characterization of tissue pathology. In this work, we instrumented a confocal fluorescence spectroscopy system and a two-photon excited fluorescence spectroscopy system to measure the depth-resolved single- and two-photon fluorescence spectra from the rabbit esophageal tissues. The excitation wavelengths were 349 nm and 735 nm, respectively. Both systems provided good optical sectioning. The information obtained from depth-resolved fluorescence was generally consistent with the histology of the examined tissue sample. The NAD(P)H signals from epithelial layers were clearly separated from the collagen signal from deep layers. In addition, strong second harmonic generations given by collagen fibers were observed. This work demonstrates that depth-resolved fluorescence spectroscopy may produce more accurate information on the diagnosis of tissue pathology.

Abstract:Two‐photon microscopy (2PM) is one of the most widely used tools for in vivo deep tissue imaging. However, the spatial resolution and penetration depth are still limited due to the strong scattering background. Here we demonstrate a two‐photon focal modulation microscopy. By utilizing the modulation and demodulation techniques, background rejection capability is enhanced, thus spatial resolution and imaging penetration depth are improved. Compared with 2PM, the transverse resolution is increased by 70%, while the axial resolution is increased to 2‐fold. Furthermore, when applied in conventional 2PM mode, it can achieve inertial‐free scanning in either transverse or axial direction with in principle unlimited scanning speed. Finally, we applied 2PFMM in thick scattering samples to further examine the imaging performance. The results show that the signal‐to‐background ratio of 2PFMM can be improved up to five times of 2PM at the depth of 500 μm. Depth-resolved Fluorescence Spectroscopy Reveals Layered Structure of Tissue.

Abstract:A confocal fluorescence spectroscopy system is instrumented to study depth-resolved autofluorescence in biological tissue. The system provides the capability of optical sectioning with the maximal detectiondepth up to 120 m in the examined tissue samples. It was found that the topmost keratinizing epithelial layer produces strong fluorescence similar to collagen. The fluorescence signal from epithelial tissue between the keratinizing layer and stroma can be well resolved. The study results show that depth-resolved fluorescence spectroscopy has the potential to provide more accurate information for the diagnosis of tissue pathology.

Abstract:The depth-resolved autofluorescence ofrabbit oral tissue, normal and dysplastic human ectocervical tissue within l20μm depth were investigated utilizing a confocal fluorescence spectroscopy with the excitations at 355nm and 457nm. From the topmost keratinizing layer of oral and ectocervical tissue, strong keratin fluorescence with the spectral characteristics similar to collagen was observed. The fluorescence signal from epithelial tissue between the keratinizing layer and stroma can be well resolved. Furthermore, NADH and FADfluorescence measured from the underlying non-keratinizing epithelial layer were strongly correlated to the tissue pathology. This study demonstrates that the depth-resolved fluorescence spectroscopy can reveal fine structural information on epithelial tissue and potentially provide more accurate diagnostic information for determining tissue pathology.

Abstract:The linear photophysical, excited state absorption (ESA), superfluorescence, and two-photon absorption (2PA) properties of 4,4'-(1E,1'E)-2,2'-(7,7'(1E,1'E)2,2'(4,4'-sulfonylbis(4,1-phenylene))-bis(ethane-2,1-diyl)bis(9,9-didecy-9H-fluorene7,2-diyl))bis(ethane-2,1-diyl) bis(N,N-diphenylaniline) (1) were investigated in organic and aqueous media with respect to its potential application in biological imaging. The analysis of linear photophysical properties revealed a rather complex nature of the main one-photon absorption band, strong solvatochromic effects in the steady-state fluorescence spectra, single-exponential fluorescence decay, and high fluorescence quantum yields in organic solvents (approximate to 1.0). The ESA spectra of 1 suggested potential for light amplification in nonpolar media while efficient superfluorescence in cyclohexane was demonstrated. The degenerate 2PA spectra of 1 were obtained over a broad spectral range (640-900 nm), using a standard two-photon induced fluorescence method under 1 kHz femtosecond excitation. Two well defined 2PA bands with maximum 2PA cross sections up to 1700 GM in the higher energy, short wavelength band and approximate to 1200 GM in the lower energy, long wavelength band of 1 were shown. The potential use of 1 in bioimaging was demonstrated via one-and two-photon in vitro fluorescence imaging of HCT 116 cells.

Abstract:We report the imaging of single-molecule fluorescence induced by two-photon excitation in ambient conditions. Using an inverted fluorescence microscope, we obtained the two-photon images of different single fluorophores (Rhodamine B, Sulforhodamine 101, Coumarin 535 on poly-methyl methacrylate films) and biological membrane fragments by Faster scanning the sample with respect to a diffraction limited focus of a mode-locked Ti: sapphire laser beam. The signal to background ratio was as high as 50:1 and the full width at half maximum (250nm) of a single-molecule peak was significantly shorter than that for one photon excitation. With its high sensitivity and simplicity, the two-photon experiment offers a valuable approach for spectroscopic studies on individual immobilized molecules.

Abstract:The fluorescence power from biological tissue excited by a femtosecond laser pulse compared with excitation power does not appear to obey a simple quadratic relationship given by the steady non-linear theory. A more reliable analysis is developed based on transient two-photon absorption because the response time of two-photon absorption is longer than the width of a femtosecond pulse. Good agreement is obtained between the theoretical analysis and the experimental results of fluorescence power versus excitation power. This letter offers potential value to non-linear optics in biological tissues.

Abstract:Two-photon excitation fluorescence imaging provides thin optical sections from deep within thick, scattering specimens by way of restricting fluorophore excitation (and thus emission) to the focal plane of the microscope. Spatial confinement of two-photon excitation gives rise to several advantages over single-photon confocal microscopy. First, penetration depth of the excitation beam is increased. Second, because out-of-focus fluorescence is never generated, no pinhole is necessary in the detection path of the microscope, resulting in increased fluorescence collection efficiency. Third, two-photon excitation markedly reduces overall photobleaching and photodamage, resulting in extended viability of biological specimens during long-term imaging. Finally, localized excitation can be used for photolysis of caged compounds in femtoliter volumes and for diffusion measurements by two-photon fluorescence photobleaching recovery. This review aims to provide an overview of the use of two-photon excitation microscopy. Selected applications of this technique will illustrate its excellent suitability to assess cellular and subcellular events in intact, strongly scattering tissue. In particular, its capability to resolve differences in calcium dynamics between individual cardiomyocytes deep within intact, buffer-perfused hearts is demonstrated. Potential applications of two-photon laser scanning microscopy as applied to integrative cardiac physiology are pointed out.

Abstract:The noninvasive two-photon excitation autofluorescence imaging of cellular and subcellular structure and dynamics in live tissue could provide critical in vivo information for biomedical studies. However, the two-photon microscopy of short-wavelength endogenous fluorophores, such as tryptophan and hemoglobin, is extremely limited due to the lack of suitable imaging techniques. In this study, we developed a short-wavelength excitation time- and spectrum-resolved two-photon microscopy system. A 520-nm femtosecond fiber laser was used as the excitation source, and a time-correlated single-photon counting module connected with a spectrograph was used to provide time- and spectrum-resolved detection capability. The system was specially designed for measuring ultraviolet and violet-blue fluorescence signals and thus was very suitable for imaging short-wavelength endogenous fluorophores. Using the system, we systematically compared the fluorescence spectra and fluorescence lifetimes of short-wavelength endogenous fluorophores, including the fluorescent molecules tyrosine, tryptophan, serotonin (5-HT), niacin (vitamin B3), pyridoxine (vitamin B6), and NADH and the protein group (keratin, elastin, and hemoglobin). Then, high-resolution three-dimensional (3D) label-free imaging of different biological tissues, including rat esophageal tissue, rat oral cheek tissue, and mouse ear skin, was performed in vivo or ex vivo. Finally, we conducted time-lapse imaging of leukocyte migration in the lipopolysaccharide injection immunization model and a mechanical trauma immunization model. The results indicate that the system can specifically characterize short-wavelength endogenous fluorophores and provide noninvasive label-free 3D visualization of fine structures and dynamics in biological systems. The microscopy system developed here can empower more flexible imaging of endogenous fluorophores and provide a novel method for the 3D monitoring of biological events in their native environment.

Abstract:Observing microvasculature in its native environment provides invaluable information to understand the initiation and development of microcirculatory related diseases. However, the lack of a high-resolution three-dimensional (3D) imaging technique hinders in vivo investigation of the microvasculature. Recently, we found that the red blood cells can emit autofluorescence signals with short-wavelength two-photon excitation. In this study, we exploited this property and developed a time-resolved two-photon excitation microscopy system using a homemade 520 nm femtosecond fiber laser as the excitation source. Using this system, we could achieve intravital high-resolution 3D imaging of a microvascular network noninvasively. In a mouse tumor model, tumorous blood vessels could be observed and distinguished clearly from the normal vessels.

Abstract:Key Points Two main techniques have been used to analyse the immune system. In vivo experiments examine cell populations in their natural environment, but they cannot provide information about individual cells. In vitro experiments determine subcellular and molecular details, but they cannot replicate adequately the physiological environment. Therefore, new techniques, involving fluorescent labels, are required to analyse single cells in intact tissues in real time. Three main problems limit optical resolution when attempting to visualize fluorescently labelled cells in living tissues. First, the high-numerical-aperture lenses that are required have a narrow depth of field, so that fluorescent label from above and below the focus obscures the image. Second, biological tissues scatter light, which reduces image contrast. Third, the intense excitation light causes photodamage and photobleaching. A partial solution is provided by confocal microscopy, which provides a sharp 'optical section' at a given depth by rejecting out-of-focus fluorescence from above or below the focal plane. But, light scattering limits the depth of imaging and the specimen is susceptible to photodamage. Two-photon microscopy has the advantages of greater imaging depth (owing to reduced light scattering of the long wavelengths that are used for excitation) and minimal photobleaching (as excitation is confined to the focal plane). Two-photon excitation involves the simultaneous absorption by a fluorophore of energy from two photons, each of which contributes one half of the total energy required to induce fluorescence. The probability of two-photon excitation falls off rapidly from the focal point. Pulsed lasers (such as the titanium-sapphire laser) are required to provide the necessary high photon density at the focal spot without vaporizing the specimen. The peak power during a pulse is extremely high, but the average laser power is relatively low. The choice of objective lens is also important to maximize the numerical aperture while maintaining imaging depth. 'Dipping' water-immersion objectives provide the best compromise. Two-photon microscopy allows imaging in as many as six dimensions (three spatial dimensions, time, intensity and wavelength of more than one probe). This creates problems in terms of storage, analysis and representation of the data that will require new solutions, such as the use of virtual-reality technology. As an example, two-photon techniques have been used to examine the movement of T cells in lymph nodes and their response to antigen. These studies have indicated that both the single-encounter model and the serial-encounter model of antigen presentation might be relevant physiologically.

Steady State and Time-Resolved Autofluorescence Studies of Human Colonic Tissues

Abstract:Steady state and time-resolved autofluorescence spectroscopies are employed to study the autofluorescence characteristics of human colonic tissues in vitro. The excitation wavelength varies from 260 to 540 nm, and the corresponding fluorescence emission spectra are acquired from 280 to 800 nm. Significant difference in fluorescence intensity of excitation-emission matrices (EEMs) is observed between normal and tumor colonic tissues. Compared with normal colonic tissue, low nicotinamide adenine dinucleotide (phosphate) (NAD(P)H) and flavin adenine dinucleotide (FAD), and high amino acids and protoporphyrin IX (PpIX) fluorescences characterize high-grade malignant tissue. Moreover, the autofluorescence lifetimes of normal and carcinomatous colonic tissues at 635 nm under 397-nm excitation are about 4.32+-0.12 and 18.45+-0.05 ns, respectively. The high accumulation of endogenous PpIX in colonic cancers is demonstrated in both steady state and time-resolved autofluorescence spectroscopies.
Autofluorescence Lifetime Imaging of Lipofuscin and Oxidized Melanin in Retinal Pigment Epithelium Cells

Abstract:We report on our investigation of the spatial distribution and autofluorescence lifetime characterization of lipofuscin and oxidized melanin in the retinal pigment epithelium cells of the pig eye using a two-photon excitation fluorescence lifetime imaging microscopy (TPE-FLIM) system, which that is based on a time-correlated single photon counting technique. In particular, we analyzed the difference of autofluorescence lifetimes of these pigment granules in light-induced oxidizing environment. The experimental results showed that the fluorescence lifetime imaging can provide an effective differentiation of multi-component fluorophores, and fluorescence decay can be used to distinguish normal from abnormal fluorescence. TPE-FLIM has the potential to provide a high sensitive imaging instrument for the clinical diagnosis and pathological studies in ophthalmology, and is also of significance to the study of aging mechanism of cells in the fundus.

Abstract:Two-photon absorption (TPA) fluorescence of biomarkers has been decisive in advancing the fields of biosensing and deep-tissue <i>in vivo</i> imaging of live specimens. However, due to the extremely small TPA cross section and the quadratic dependence on the input photon flux, extremely high peak-intensity pulsed lasers are imperative, which can result in significant photo- and thermal damage. Previous works on entangled TPA with spontaneous parametric downconversion light sources found a linear dependence on the input photon-pair flux, but are limited by low optical powers, along with a very broad spectrum. We report that by using a high-flux squeezed light source for TPA, a fluorescence enhancement of <span class="equationTd inline-formula"><math> ∼ 47</math></span> is achieved in fluorescein biomarkers as compared to classical TPA. Moreover, a polynomial behavior of the TPA rate is observed in the the laser dye 4-dicyanomethylene-2-methyl-6-(p(dimethylamino)styryl)-4H-pyran in dimethyl sulphoxide.

Abstract:The steady-state and time-resolved autofluorescence spectroscopic differences between normal and carcinomatous colonic tissues and the optimal excitation wavelengths were studied. The fluorescence excitation wavelengths varying form 260 to 540 nm were used to induce tissue autofluorescence, and the corresponding emission spectra were measured in the range from 280 to 800 nm. These spectra then can be assembled into an excitation-emission matrix (EEM). Significant changes in fluorescence intensity of excitation-emission matrices were observed between normal and tumor colonic tissues. Low NAD(P)H and FAD, and high protoporphyrin IX fluorescence characterize high-grade malignant tissue when compared with normal colonic tissue, and the most marked difference being at the excitation wavelengths of 340, 380, 460 and 540 nm. Furthermore, the average lifetimes for the normal and carcinomatous colonic tissues were about 4.12 and 18.8 ns, respectively. The promising applications of laser-induced autofluorescence for colonic tissue diagnosis are indicated.
A single-shot approach for measuring two-photon action cross-section of fluorescent markers.

Abstract:We report on a novel method designed for measuring two photon action cross sections spectra in a single shot without tuning the excitation wavelength. Our technique is based on (i) using a nonlinear photonic crystal fiber to broaden the spectrum of the femtosecond excitation pulses and (ii) exploiting angular dispersion to focus different wavelengths to different lateral positions. As a result, two-photon fluorescence signal at different excitation wavelengths can be obtained simultaneously. As a proof of principle, the relative two-photon action cross sections of rhodamine green and DiI-C-18 are measured over 740 - 860 nm range using fluorescein as a reference. Our results are in good agreement with that obtained using conventional tunable mode-locked laser. (c) 2006 Optical Society of America

Abstract:Fibre-optic based time-resolved fluorescence spectroscopy (TRFS) is an advanced spectroscopy technique that generates sample-specific spectral-temporal signature, characterising variations in fluorescence in real-time. As such, it can be used to interrogate tissue autofluorescence. Recent advancements in TRFS technology, including the development of devices that simultaneously measure high-resolution spectral and temporal fluorescence, paired with novel analysis methods extracting information from these multidimensional measurements effectively, provide additional insight into the underlying autofluorescence features of a sample. This study demonstrates, using both simulated data and endogenous fluorophores measured bench-side, that the shape of the spectral fluorescence lifetime , or fluorescence lifetimes estimated over high-resolution spectral channels across a broad range, is influenced by the relative abundance of underlying fluorophores in mixed systems and their respective environment. This study, furthermore, explores the properties of the spectral fluorescence lifetime in paired lung tissue deemed either abnormal or normal by pathologists. We observe that, on average, the shape of the spectral fluorescence lifetime at multiple locations sampled on 14 abnormal lung tissue, compared to multiple locations sampled on the respective paired normal lung tissue, shows more variability; and, while not statistically significant, the average spectral fluorescence lifetime in abnormal tissue is consistently lower over every wavelength than the normal tissue.

Abstract:Light-induced autofluorescence spectra of nasopharyngeal carcinoma and normal tissue in vitro were compared to that of known endogenous fluorophores to explore the possible causes of tissue autofluorescence and to further determine the optimal excitation wavelengths for optical biopsy in vivo. Nasopharyngeal carcinoma and normal tissues were obtained from the suspected patients during pathological biopsy. A FL/FS920 combined TCSPC spectrofluorimeter and a lifetime spectrometer system was used for autofluorescence spectra measurement. Fluorescence excitation wavelengths varying from 260 to 480 nm were used to induce tissue autofluorescence, and the corresponding fluorescence emission spectra were recorded from a range starting 20 nm above the excitation wavelength and extending to 700 run. The autofluorescence excitation-emission pairs of nasopharyngeal carcinoma and normal tissues occur at 300-330, 340-460 and 450-520 nm, and the optimal diagnostic excitation wavelengths for detection of nasopharyngeal carcinoma were 340 and 450 run. The results abtained in this study could be treated as a reference for the development of optical biopsy system for nasopharyngeal carcinoma.
